Infection of cattle with Cryptosporidium parvum: mast cell accumulation in small intestine mucosa.

Mast cells might play an important role as the major effector cells in the immune response against Cryptosporidium parvum. C. parvum is a protozoan parasite that causes cryptosporidiosis in animals and humans worldwide. To investigate the interaction between C. parvum and mast cells during infection, nine 3-day-old male calves were orally challenged with 10(6) oocysts of C. parvum per calf. The distribution of mast cells in the mucosa of the small intestine was analyzed by toluidine blue staining. The concentrations of histamine and the cytokines interferon-γ, interleukin-4, interleukin-2, and

Microscopy and microanalysis study of the indium (In) behavior in the intestinal mucosa, the liver, the kidney and the testicle.

Several studies have demonstrated that In used in medicine has several impacts on organs like spleen and lungs after its systemic administration. In the present study, ultrastructural and microanalytical methods were used to investigate the impact of the presence of this element in the intestinal mucosa, the liver, the kidney and the testicle after its administration in two ways. After intraperitoneal administration, In was selectively concentrated in the lysosomes of hepatocytes, of tubular proximal convoluted cells and of Sertoli and Leydig cells. After intragastric administration, ultrastru

Expression of retinoic acid receptors in intestinal mucosa and the effect of vitamin A on mucosal immunity.

OBJECTIVE: To explore the mechanism of vitamin A (VA) modulation of mucosal immunity, the expression of retinoic acid receptors in intestinal mucosa was measured, and the effect of VA on intestinal dendritic cells (DCs) and mucosal cytokine production was examined. METHODS: The expression of retinoic acid receptor (RAR-alpha, RAR-beta, RAR-gamma, RXR-alpha, RXR-beta, RXR-gamma) mRNA, the distribution and number of DCs, and the protein secretion of interleukin (IL)-12, T-helper type 1/2 cells (interferon [IFN]-gamma/IL-4), and regulatory (IL-10) cytokines in the mucosa of terminal ileum in nor

Paneth cells, defensins, and the commensal microbiota: a hypothesis on intimate interplay at the intestinal mucosa.

Mucosal surfaces are colonized by a diverse and dynamic microbiota. Much investigation has focused on bacterial colonization of the intestine, home to the vast majority of this microbiota. Experimental evidence has highlighted that these colonizing microbes are essential to host development and homeostasis, but less is known about host factors that may regulate the composition of this ecosystem. While evidence shows that IgA has a role in shaping this microbiota, it is likely that effector molecules of the innate immune system are also involved. One hypothesis is that gene-encoded antimicrobia

[Immunohistochemical study of the apoptotic and proliferative mechanisms in the intestinal mucosa during coeliac disease].

Mechanisms leading to morphological changes of the small intestine during coeliac disease are not yet completely recognized, however, two main processes have been suggested recently: remodelling of mucosa by matrix metalloproteinases, and mucosal atrophy by apoptosis. The aim of this study was to analyze the expression of proteins regulating apoptosis and some markers of proliferation in the mucosa of the small intestine of children with active (ACD) and latent form (LCD) of coeliac disease (CD). Intestinal biopsies of 43 children with ACD and LCD were analyzed by standard indirect immunohisto

Basement membrane and connective tissue proteins in intestinal mucosa of patients with coeliac disease.

AIMS: Gluten ingestion in coeliac disease is associated with alterations of the intestinal mucosa, especially the expansion of the lamina propria. Antiendomysium and antireticulin antibodies may result from interactions between gliadin and extracellular matrix components. By behaving as autoantigens, connective tissue proteins could initiate mucosal damage. This study evaluates changes in the distribution of laminin, type IV collagen, and fibronectin in the mucosa of patients with coeliac disease in an attempt to explain the alterations of mucosal morphology. METHODS: Intestinal biopsies were

Multiple organ mucosa-associated lymphoid tissue lymphomas often involve the intestine.

BACKGROUND: Low grade mucosa-associated lymphoid tissue (MALT) lymphomas usually are confined to single extranodal organs. Although some case reports have been published, clinicopathologic characteristics of multiorgan MALT lymphomas remain unclear. METHODS: The authors evaluated 7 MALT lymphoma cases involving multiorgans in the past 7 years. In this period, they experienced 304 cases of MALT lymphomas. They analyzed the clinicopathologic features of these cases, including examination of clonal comparison among the lesions. RESULTS: The patients, 4 females and 3 males, were aged 55-68 years

T cell receptor repertoire in the peripheral blood and intestinal mucosa of coeliac patients.

The alpha beta and gamma delta T cell receptor (TCR) repertoire in the peripheral blood and intestinal mucosa of six coeliac and six age-matched controls was analysed by reverse transcription and polymerase chain reaction (PCR). No TCR alpha and gamma delta restriction was observed in coeliacs and controls. However, V gamma 3 was expressed only in coeliac peripheral and intestinal T cells. V delta 2 was strongly expressed in coeliacs and scarcely transcribed in control cells. The unique expression of these gamma delta TCR in coeliac patients suggests that V gamma 3 and perhaps V delta 2 TCR-be

Leukotriene B4 and C4 generation by small intestinal mucosa in children with coeliac disease.

The capacity of the small intestinal mucosa to generate leukotriene B4 (LTB4) and C4 (LTC4) in children with coeliac disease (CD) was investigated by measuring the production of LTB4 and LTC4 in intestinal biopsy specimens after stimulation with 10 microM calcium ionophore A23187. In addition, we examined the relationship between the production of LTB4 and LTC4 in the small intestinal mucosa and symptoms of diarrhoea. LTB4 and LTC4 production was significantly higher in biopsies from patients with active CD than from controls (p < 0.05 and p < 0.01, respectively). There was no significant diff

Intestinal metaplasia of the gastric mucosa: a study of its incidence in various disease states.

The incidence of intestinal metaplasia (IM) was studied in 159 specimens from gastric mucosa. Rare in histologically normal specimens (4%), the IM showed the highest incidence in patients with gastric carcinoma (75%). Intermediate values were observed in chronic atrophic gastritis (25%) and peptic ulcer (21%). Histochemical data shows that type complete-IM was most common in benign conditions; type incomplete-IM and particularly subtype secreting sulphomucins revealed a higher frequence in the mucosa bordering carcinomas.

Epithelial-mesenchymal interactions can influence the phenotype of carcinoma metastases in the mucosa of the intestine.

In this paper we report two cases in which morphological differentiation of gastric adenocarcinoma has occurred when metastatic tumour was present within the lamina propria of the intestinal mucosa. In one case, there were metastases in the small intestinal mucosa and in the second case, in the mucosa of the appendix. The morphology of the intramucosal tumour simulated that of an adenomatous neoplasm. The recognition of this occurrence may be important in the differential diagnosis of primary and secondary adenocarcinomas of the gastrointestinal tract. The phenomenon is not only of diagnostic

[Clinico-immunomorphological characteristics of the mucosa of the distal segments of the large intestine in children with gastroduodenitis].

The clinical, x-ray, endoscopic and morphological studies performed in 156 children afflicted with gastroduodenitis have shown that concomitant lesions of the colon are of paramount importance in the genesis of this illness. The immunomorphological assay is the most informative one in the assessment of the pattern of structural alterations in the colonic mucosa. Examination of 71 colonic biopsy specimens has disclosed that 38 patients had minimal alterations in the mucosa characteristic of colodyskinesias and 22 had the signs of chronic inflammation, which plays an important part in goal-orien

[T lymphocyte populations of the intestinal mucosa in celiac disease in children. Immunohistochemical study].

In order to study the distribution of lymphocyte subpopulations in a pathologic intestinal mucosa, the authors, instead of using the classic method by counting the number of lymphocytes, present an original method permitting the exploitation of quantified data from labelled surface cells by texture analyser coupled with a computerized system. We investigated 25 children presenting with chronic diarrhea and villous atrophy and 5 control subjects. Fifteen of the 25 children had celiac disease (10 active with total villous atrophy and 5, celiac disease in remission with healing mucosa), 5 cow's m

[Effect of vitamin A deficiency on the macromolecular permeability of small intestine mucosa in the adult rat].

Adult rats with experimental vitamin A deficiency and control animals were intraperitoneally injected with chicken ovalbumin (OA) solution and the entrance of native OA into the blood was assessed 3 hours later by competitive radioimmunoassay. The OA amounts circulating in the blood of control animals averaged (0.39 +/- 0.06) X 10(-4)% of the consumed dose, while in the experimental group it averaged (1.33 +/- 0.42) 10(-4)%. Electron microscopy, using colloid lanthanum hydroxide, has shown vitamin A deficiency to give rise to an abrupt reduction in glycocalix layer, as compared to the control,

[Ultrastructural changes in the epithelial cells of the small intestine mucosa in chronic liver diseases].

Electron microscopy of epithelial cells of the mucous membrane of the duodenum and jejunum was performed in 20 patients with chronic hepatitis and liver cirrhosis to reveal changes in the structure of intracellular formations of epithelial cells of the cryptae and villi, of goblet and enterochromaffin cells. The most essential differences were found in mitochondria, granular cytoplasmic network, ribosomes, microvilli and glycocalix. There were no differences in the pattern of cellular changes in chronic hepatitis and liver cirrhosis. However, the degree of lesions increased in patients with li

The authors traced the dynamics of appearance, increase and disappearance of O-antibody-containing cells in the mucosa of the distal portion of the large intestine in patients suffering from acute Sonne and Flexner dysentery (aspiration biopsy of the mucosa). The antibodies were revealed by the indirect Coons method. The total number of cells per 1 mm2 of 4 mum of the mucosal section the number of lymphoid cells and the content of the antibody-containing cells in % per total cell count of the lymphoid tissue was calculated. It was revealed that the antibody-containing cells appeared on the 4th

The dynamic structure of a flat small intestinal mucosa studied on the explanted rat jejunum.

Small pieces of jejunum with an intact blood supply were explanted to the anterior abdominal wall in rats. Six weeks after explantation the mucosa appeared totally flat in many areas, both histologically and under the dissecting microscope. The structure of the flattened mucosa was shown to be identical to that in coeliac disease with hypertrophied intervillous ridges. A dynamic study with tritium-labelled thymidine demonstrated a considerably increased turnover in the flat mucosa with some disorganization of cell production and migration.
