Association between thyroid autoimmunity and fibromyalgia.

Evidence exists that autoimmune thyroiditis is present in a high percentage of fibromyalgia (FM) and associated with the presence of typical symptoms of FM. However, the role of thyroperoxidase antibody (TPO Ab) in the manifestation of FM is still unclear. The goal of this study was to investigate the prevalence of positive TPO Ab in euthyroid FM patients, and whether TPO Ab positivity is associated with the clinical manifestations in euthyroid FM patients.Thyroid assessment was done by free T4, TSH and TPO Ab. The clinical parameters including Fibromyalgia Impact questionnaire (FIQ), pain vis

Autoimmune thyroid disease--a continuous spectrum.

AIM: to present a case report revealing a variant of natural history of autoimmune thyroid disease. CASE REPORT: I.F., a 39 years old woman, had a previous medical history of Graves' disease treated with antithyroid drugs (ATD). After 1 year of treatment, the remission was confirmed one and two years after ATD withdrawal. Twenty years after the initial hyperthyroidism, spontaneous subclinical hypothyroidism was diagnosed. The patient presented both anti TSH-receptor (TSH-R) antibodies (Ab), antithyroperoxidase (TPO) and antithyroglobulin (antiTgl) antibodies in elevated titres. CONCLUSION: B

Ovarian iodide uptake and triiodothyronine generation in follicular fluid. The enigma of the thyroid ovary interaction.

Since 1928, the iodine concentration in the ovary has been known to be higher than in every other organs except the thyroid. The ovarian iodide uptake varies with sexual activities, is enhanced by estrogens and a hypothyroid state and blocked by goitrogens. The recent discovery of a sodium iodide symporter (NIS) in ovaries has offered a possible mechanism for ovarian iodide uptake and other functional similarities to its thyroid counterpart. Nevertheless, the physiological significance of ovarian iodine uptake and accumulation remains unknown. The presence of thyroid hormones (TH) in follicula

Clinical value of a bispecific antibody binding to thyroglobulin and thyroperoxidase (TGPO-aAb) in various thyroid diseases.

TGPO-aAb is a bispecific antibody which binds to thyroglobulin as well as thyroid peroxidase. It is supposed to be raised in some patients with autoimmune thyroid disease. We investigated 205 patients suffering from Graves' disease (n = 81), Hashimoto's thyroiditis (n = 36), toxic nodular goitre (n = 50), differentiated carcinoma of the thyroid (n = 10), and autoimmune thyropathy of unknown origin (n = 28). An immunoradiometric assay was used to measure serum TGPO-aAb. Eighty-nine of 205 patients had elevated titres of TGPO-aAb. If TGPO-aAb were raised then autoantibodies against thyroglobulin

[Immunohistochemical identification of biochemical defect of dyshormonogenetic goiter].

This is a report of two cases of congenital goiter in which it was possible to indicate, through immunohistochemistry method, the disorder in the hormonal synthesis. One of these cases was of a sixteen-year-old woman, with goiter since childhood who used thyroid hormone replacement for ten years. Pathology showed coloid goiter with follicular adenoma. Immunohistochemistry showed strong mainly apical follicular cell thyroglobulin. Thyroglobulin was not found in the coloid. This finding is compatible with a malfunction of thyroglobulin exocitosis. The reaction for thyroid peroxidase was adequate

[Anti-TPO autoantibodies].

Thyroid peroxidase (TPO) is a member of the thyroid autoantigens. Anti-thyroid autoantibodies, anti-TPO,-thyroglobulin and -TSH receptor autoantibodies, are important in diagnosing autoimmune thyroid diseases and for judging treatment efficacy. To diagnose chronic thyroiditis, anti-TPO autoantibody detection is the most sensitive among the three anti-thyroid autoantibodies. Eighty-five to ninety percent of patients with chronic thyroiditis have anti-TPO autoantibodies. Using recombinant human TPO protein, a kit measuring anti-TPO antibodies specifically have been developed. The developmental p

Thyroid antibodies are not a risk factor for pregnancies with Down syndrome.

The observation that thyroid disease is frequent in mothers of children with Down syndrome (DS) has suggested that maternal thyroid antibodies could be a factor predisposing to trisomy 21 in their offspring. In this study, the incidences of thyroglobulin (Tg) and thyroid peroxidase (TPO) antibodies were analysed with a sensitive solid-phase immunosorbent radioassay in sera from 29 mothers giving birth to children with trisomy 21 and 87 control mothers. The serum samples were collected at delivery. There was no statistical difference regarding the proportion of thyroid antibodies (against Tg an

No association between Alport's syndrome and antithyroid antibodies.

The presence of circulating antithyroid antibodies in a small number of patients with Alport's syndrome has led some to suggest that the two abnormalities are associated. If confirmed, such an association could have important diagnostic as well as pathophysiological implications. Using sensitive methods, we assessed the prevalence of antithyroid antibodies in 40 patients with Alport's syndrome from 28 families. Antithyroglobulin antibodies were absent in all patients. Antimicrosomal and antithyroperoxidase antibodies were absent in all but one patient. Three patients with Alport's syndrome fro

Interaction of halides with the cyanide complex of myeloperoxidase: a model for substrate binding to compound I.

EPR spectra of the low-spin cyanide complex of myeloperoxidase have been measured in the absence and presence of halide substrates; chloride, bromide and iodide. Halide-dependent spectral changes are found at acidic pH. The electronic structure of the low-spin ferric iron in cyanide complex appears to be modulated by halide binding to a protonated amino acid in the distal heme cavity. These findings suggest halide substrates can interact with ferryl oxygen in compound I during enzyme catalysis to form hypohalous acid.

Nocturnal increase in pineal melatonin production in two lemming species, Dicrostonyx hudsonius and D. groenlandicus.

Pineal N-acetyltransferase (NAT) activity and melatonin levels and thyroxine 5'-deiodinase (5'D) activity in brown adipose tissue (BAT) were compared during the day and night in two species of lemmings, Dicrostonyx hudsonius and D. groenlandicus. In both species, NAT and melatonin exhibited significant night-time increases in the pineal gland. Thyroxine 5'D activity in BAT did not show a statistically significant day-night difference in either species of lemming, suggesting the night-time rise in melatonin does not induce a similar increase in BAT 5'D.

The effect of fasting on thyroidal T4-5' monodeiodinating activity in mice.

Complete fasting induces a significant decrease of serum T3 and a fall in TSH in rodents and man. To evaluate the effect of starvation on thyroidal T4 5'monodeiodinating activity, in vitro conversion of T4 to T3 by thyroid and liver homogenate from one to three days fasted mice was compared with homogenates from control mice on animal chow. 5'monodeiodinating activity was significantly lower in thyroid homogenates of fasted mice than in those of chow-fed control [100 +/- 5.0 and 92 +/- 5.0 pmol T3.(mg protein)-1.h-1 at 48 and 72 h fasting, respectively, vs 132 +/- 5.0 pmol T3.(mg protein)-1.h-

Formation of 3,3'-diiodothyronine and 3',5',3-triiodothyronine (reverse T3) in thyroid glands of rats and in enzymatically iodinated thyroglobulin.

It was observed in the present investigation that labeled thyroxine (T4) comprised less than 2% of the total 131I in the thyroids of severely iodine-deficient rats labeled with 131I for 18-24 h, a much lower value than had previously been reported for iodine-deficient rats. This low value was attributable to two factors: 1) the use of a diet low enough in iodine content to produce extreme iodine deficiency, and 2) the use of a paper chromatography system that successfully separates T4 from the minor iodothyronines, 3,3'-diiodothyronine (T2) and 3',5',3-triiodothyronine (reverse T3; T3'). Relationship of Anti-thyroid Peroxidase Antibody with Recurrent Pregnancy Loss in Euthyroid Women.

Recurrent pregnancy loss is a common issue in Obstetrics&Gynaecology practice. Thyroid autoimmunity has been associated with pregnancy loss. Thyroid peroxidase antibodies (TPO-Ab) is the most common antithyroid autoantibodies observed in women with recurrent pregnancy loss history. Although the precise mechanisms of the TPO-Ab's role in recurrent pregnancy loss are not fully understood. The study was aimed to assess the relationship of Anti-TPO antibody with recurrent pregnancy loss. This cross-sectional comparative study was conducted in the Department of Obstetrics and Gynecology at Mymensin

Serotonergic and Chemosensory Brain Areas and Sensory Ganglia Expressing Type 3 Deiodinase Mapped With Dio3Cre drivers.

Thyroid hormone (triiodothyronine, T3) promotes neurodevelopment but under strict control because unconstrained exposure to T3 impairs brain and sensory functions. Thyroid hormone-inactivating type 3 deiodinase, encoded by Dio3, critically limits T3 signaling and controls diverse neural functions. Accordingly, understanding the cellular basis of T3 action requires identification of Dio3-expressing cell types but this is difficult because of low level, transient expression within the complexity of the nervous system. Here, we derived a knock-in Dio3Cre driver that sensitively labels Dio3-expres

Daily and seasonal changes in steroidogenic markers in the hypothalamus and testes of tree sparrow (Passer montanus).

The population responds to environmental variability largely determined by the dynamic interactions between fitness components within- and among-individual variation in the expression of the environmentally sensitive phenotype. The study was conducted on daily and seasonal changes in the expression of steroidogenic gene markers and corresponding seasonal changes in the physiological characters in adult male tree sparrows. Two experiments were performed. In experiment one, birds (n = 5/time points) were sampled during the breeding season at 6-time points, i.e., ZT1, ZT5, ZT9, ZT13, ZT17, and ZT
