Pharmacological targeting of ion channels for cancer therapy: In vivo evidences.

Since the discovery of the participation of various ion channels in the regulation of cell proliferation and programmed cell death two decades ago, the field exploring ion channel function in relation to cancer has undergone rapid development. Although the mechanisms accounting for the impact of ion channel modulators on cancer growth have not been fully clarified in all cases, numerous in vivo experiments targeting diverse ion channels in various cancer models illustrate the great potentiality of this approach and promote ion channels to the class of oncological targets. In the present review

Crystallographic studies of pharmacological sites in pentameric ligand-gated ion channels.

BACKGROUND: Pentameric ligand-gated ion channels (pLGICs) mediate fast chemical transmission of nerve signals in the central and peripheral nervous system. On the functional side, these molecules respond to the binding of a neurotransmitter (glycine, GABA, acetylcholine or 5HT3) in the extracellular domain (ECD) by opening their ionotropic pore in the transmembrane domain (TMD). The response to the neurotransmitter binding can be modulated by several chemical compounds acting at topographically distinct sites, as documented by a large body of literature. Notably, these receptors are the target

CRAC ion channels and airway defense reflexes in experimental allergic inflammation.

Calcium release-activated calcium channels (CRAC) play unambiguous role in secretory functions of mast cells, T cells, and eosinophils. Less knowledge exists about the role of CRAC, widely distributed in airway smooth muscle (ASM) cells, in airway contractility. The presented study seeks to determine the possible participation of CRAC in ASM-based inflammatory airway disorders in guinea pigs. The acute and long-term administration (14 days) of the CRAC antagonist 3-fluoropyridine-4-carboxylic acid was used to examine the ASM contractility and associated reflexes in the guinea pig model of alle

A novel sea anemone peptide that inhibits acid-sensing ion channels.

Sea anemones produce ion channels peptide toxins of pharmacological and biomedical interest. However, peptides acting on ligand-gated ion channels, including acid-sensing ion channel (ASIC) toxins, remain poorly explored. PhcrTx1 is the first compound characterized from the sea anemone Phymanthus crucifer, and it constitutes a novel ASIC inhibitor. This peptide was purified by gel filtration, ion-exchange and reversed-phase chromatography followed by biological evaluation on ion channels of isolated rat dorsal root ganglia (DRG) neurons using patch clamp techniques. PhcrTx1 partially inhibited

The human corneal endothelium: new insights into electrophysiology and ion channels.

The corneal endothelium is a monolayer that mediates the flux of solutes and water across the posterior corneal surface. Thereby, it plays an essential role to maintain the transparency of the cornea. Unlike the epithelium, the human endothelium is an amitotic cell layer with a critical cell density and the risk of corneal decompensation. The number of endothelial cells subsequently decreases with age. Moreover, the endothelial cell loss is accelerated after various impairments such as surgical trauma (e.g. cataract extraction) and following corneal transplantation. This cell loss is associate

Thermosensitive TRP ion channels mediate cytosolic calcium response in human synoviocytes.

The transient receptor potential (TRP) channels are important membrane sensors, responding to thermal, chemical, osmotic, or mechanical stimuli by activation of calcium and sodium fluxes. In this study, three distinct TRP channels were detected and their role established in mediating cytosolic free calcium concentration ([Ca(2+)](cyt)) response in tumor-derived SW982 synoviocytes and primary cultures of human synovial cells from patients with inflammatory arthropathies. As shown by fura-2 ratio measurements while cells were incubated in a temperature-regulated chamber, significant [Ca(2+)](cyt

Phoneutria nigriventer venom: a cocktail of toxins that affect ion channels.

1. We review the pharmacological actions of toxins present in the venom of the aggressive spider Phoneutria nigriventer. 2. This venom is rich in toxins that affect ion channels and neurotransmitter release. Voltage-gated sodium, calcium, and potassium channels have been described as the main targets of these toxins. 3. In addition to these classical actions Phoneutria toxins have also been shown to affect glutamate transporter. 4. It is expected that molecular genetics in addition to biochemical, biophysical and pharmacological approaches will help to further define Phoneutria toxins and thei

Genetics, an alternative way to discover, characterize and understand ion channels.

1. The conventional approach to understanding the structure and properties of ion channels has been to use physiological characterization. 2. Purification and molecular cloning of ion channel genes has enabled more detailed structure-function analyses to be undertaken. 3. An alternative approach to the identification of genes of pathophysiological importance has been the use of genetic linkage approaches and positional cloning or positional candidate analysis of ion channel genes. 4. Using genetic approaches, mutations have been described that cause inherited neurological disorders of neurons

Roles of ion channels in carotid body chemotransmission of acute hypoxia.

In this review, we have highlighted the roles of ion channels in carotid body chemotransmission of acute hypoxia. With the application of new technologies, significant breakthroughs have been made in the last decade. The discovery of oxygen-sensitive K(+) channels in rabbit glomus cells has generated the membrane model of hypoxic chemotransmission: the inhibition of oxygen-sensitive K(+) channels by hypoxia initiates the depolarization of glomus cells and increases the firing frequency of glomus cells. The depolarization of glomus cells activates voltage-gated Ca(2+) channels, elevating intrac

Oxygen-sensing by ion channels and the regulation of cellular functions.

From bacteria to mammals, ambient O2 tension influences such diverse cellular functions as gene expression, secretion, contraction and the patterns of electrical activity. Some of the effects of O2 are attributed to its interaction with various classes of voltage-dependent ion channels. In glomus cells of the carotid body, the differential properties of O2-sensitive K+ and Ca2+ channels help us to understand the basic features of O2 chemoreception. Modifications of ion-channel activity in response to changes in the partial pressure of O2 are also involved in the adjustments of vascular tone to

Block of acetylcholine-activated ion channels by an uncharged local anaesthetic.

It is now thought that amine local anaesthetic compounds (procaine, lignocaine and related molecules) depress electrical activity in nerve and muscle cells by binding to sites within ion channels and blocking current flow. Such mechanisms have been proposed to account for the effects of these local anaesthetics on both the voltage-dependent sodium current and the postsynaptic actylcholine (ACh)-activated ionic current. Recently, strong evidence for block of ion channels by cationic drug molecules has been obtained by recording current from single ACh-activated channels in the presence of perma

Ion fluxes through the sodium channels of garfish olfactory nerve membranes.

1. The efflux of (22)Na from garfish olfactory nerves, after treatment with ouabain and equilibration with (22)Na, occurs in two components of almost equal size. One component represents efflux from the extracellular space and the other, much slower, component represents efflux from the intracellular space through the axonal membrane.2. The rate of efflux of (22)Na through the membrane is increased from 0.038 to 0.055 min(-1) by veratrine, and restored back to 0.036 min(-1) by the additional application of tetrodotoxin or saxitoxin (10(-7)M).3. 50% inhibition of this increased sodium ion flux

Multi-targeting sodium and calcium channels using venom peptides for the treatment of complex ion channels-related diseases.

Venom peptides are amongst the most exquisite group of bioactive molecules able to alter the normal physiology of organisms. These bioactive peptides penetrate tissues and blood vessels to encounter a number of receptors and ion channels to which they bind with high affinity and execute modulatory activities. Arachnid is the most diverse class of venomous animals often rich in peptides modulating voltage-gated sodium (NaV), calcium (CaV), and potassium (KV) channels. Spider venoms, in particular, contain potent and selective peptides targeting these channels, with a few displaying interesting

Enhancement of Calcium Ion Permeation via Resonant Coupling of Ion and Terahertz Waves in Voltage Gated Calcium Channels.

Dysregulated calcium ion (Ca2+) influx is implicated in diverse channelopathies. Terahertz (THz) waves have been explored as a promising approach to modulate the influx, which primarily target resonant interactions with chemical groups in channels. Here, we demonstrate a strategy that directly regulates the motion of confined Ca2+ ions within the selectivity filter via resonant THz waves. We characterize the ions' axial oscillations, identifying a distinct intrinsic frequency of 1.65 THz and two coherent modes. By tuning a THz electric field to this frequency, we induce a remarkable resonance

Multi-ion permeation and dynamic conductance modulation in connexin gap junction channels.

Gap junction channels formed by connexins mediate direct intercellular communication and are essential for electrical signaling and tissue homeostasis. Despite their large, solvent-accessible pores, connexin channels exhibit distinct conductance, selectivity, and rectification properties, but the molecular mechanisms underlying these behaviors remain incompletely understood. Here, we performed ∼67 μs of all-atom computational electrophysiology simulations of connexin-46 (Cx46), connexin-50 (Cx50), and heterotypic Cx46/50 gap junction channels based on high-resolution open-state structures, ena

Sodium ion channels as therapeutic targets in epilepsy: a drug discovery perspective.

Traditional broad-spectrum sodium channel blockers have long served as the mainstay of epilepsy therapy, yet their clinical utility is constrained by dose-limiting adverse effects and the risk of exacerbating certain genetic epilepsies. Targeting the specific Nav subtypes, including Nav1.1, Nav1.2, Nav1.3, and Nav1.6, has emerged as a promising therapeutic strategy to mitigate these liabilities. In this Perspective, we highlight the structure, physiological functions, and their pathological roles in epilepsies, and analyze the clinical status of Pan-Nav inhibitors and the recent advances in dr

Signaling pathways and ion channels in osteoarthritis: a review of recent advances.

The understanding of osteoarthritis (OA) has gradually evolved from the traditional perspective of it being a mere "wear-and-tear" joint disease to a more comprehensive view that characterizes it as a degenerative joint disease marked by chronic low-grade inflammation and immune imbalance. The progression of OA is closely linked to abnormal mechanical loading and the activation of the innate immune system. This narrative review examines the role of mechanosensory-immune signaling networks in OA, with a specific focus on ion channels as receptors for upstream mechanical and chemical stimuli. Th

Antiseizure Medications Impact Mitochondrial Ion Channels via Novel Bioenergetic and Neural Mechanisms.

Antiseizure medications (ASMs) have traditionally been characterized by their modulation of neuronal ion channels and synaptic processes; however, accumulating evidence indicates that numerous ASMs also directly modulate mitochondrial function. Specifically, several ASMs interact with ion channels located in both the inner and outer mitochondrial membranes, including the voltage-dependent anion channel (VDAC), the mitochondrial calcium uniporter (MCU), the mitochondrial Na+/Ca2+ exchanger (NCLX), the mitochondrial permeability transition pore (mPTP), and mitochondrial ATP-sensitive potassium c

The role of ion channels in the regulation of dendritic cell function.

Ion channels, membrane proteins that facilitate the transport of various inorganic ions across hydrophobic cellular lipid membranes, are ubiquitous in a wide variety of cell and tissue types. They are involved in establishing the cell membrane potential and play a role in various physiological activities by regulating ion concentrations within the cell. Dendritic cells (DCs) are specialised antigen-presenting cells found mainly on the surface of the body (skin and mucous membranes), in the mesenchyme of most organs, in the T-cell compartment of the spleen and in lymph nodes. DCs exert an impor
