Brucella infections mainly occur through mucosal surfaces. Thus, the development of mucosal administered vaccines could be instrumental for the control of brucellosis. Here, we evaluated the usefulness of recombinant Lactococcus lactis secreting Brucella abortus Cu-Zn superoxide dismutase (SOD) as oral antigen delivery system, when administered alone or in combination with L. lactis expressing IL-12. To this end, mice were vaccinated by oral route with L. lactis NZ9000 transformed with pSEC derivatives encoding for SOD (pSEC:SOD) and IL-12 (pSEC:scIL-12). In animals receiving L. lactis pSEC:SO

The replication region of the 7.8 kilobase (kb) citrate plasmid pSL2 from Lactococcus lactis ssp. lactis biovar. diacetylactis Bu2 was identified. Deletion derivatives of pSL2 were introduced into plasmid-free strain Bu2-60 and tested for their ability to replicate autonomously. The region necessary for replication was identified by comparison of the pSL2 derivatives, cloned and sequenced. No homologies were detected by comparing the putative Rep protein of pSL2 with replicons of other plasmids of Gram-positive bacteria. A part of an IS-element flanking the replication region was found.

Intractable functional constipation (IFC), a severe form of chronic constipation characterized by slow transit and resistance to conventional treatments, posed a significant clinical challenge. Here, we identified Lactococcus formosensis (Lf), a Gram-positive bacterium prevalent in IFC patients, as a novel contributor to intestinal motility impairment. Clinically, IFC patients exhibited increased colonic mucosal colonization of Lf and significant myenteric neuronal loss and pyroptosis, particularly in excitatory choline acetyltransferase (ChAT)+ neurons, but not inhibitory neuronal nitric oxid

AIMS: Anti-tumor vaccines that target the early proteins E6 and E7 of human papillomavirus (HPV) type 16 represent a potential immunotherapy for cervical cancer, although underlying mechanisms remain unclear. Lactococcus lactis (L.L) is generally regarded as safe (GRAS) which has potential as vehicle. Study investigated the immunoregulatory effect of L.L on dendritic cells (DCs) activation. Besides, antigen delivery and anti-tumor effects of DC-based vaccine prepared with recombinant L.L vaccine (L.L-De) carrying prokaryotic-eukaryotic HPV-16 E6E7 fusion antigen were explored.

Lactococcus lactis, widely used in the manufacture of dairy products, encounters various environmental stresses both in natural habitats and during industrial processes. It has evolved intricate machinery of stress sensing and defense to survive harsh stress conditions. Here, we identified a novel TetR/AcrR family transcription regulator, designated AcrR1, to be a repressor for acid and antibiotic tolerance that was derepressed in the presence of vancomycin or under acid stress. The survival rates of acrR1 deletion strain ΔAcrR1 under acid and vancomycin stresses were about 28.7-fold (pH 3.0,

Aquaculturists use polyploid fish to maximize production albeit with some unintended consequences including compromised behaviors and physiological function. Given benefits of probiotic therapies (e.g., improved immune response, growth, and metabolism), we explored probiotic supplementation (mixture of Bifidobacterium, Lactobacillus, and Lactococcus), to overcome drawbacks. We first examined fish gut bacterial community composition using 16S metabarcoding (via principal coordinate analyses and PERMANOVA) and determined probiotics significantly impacted gut bacteria composition (p = 0.001). A colovesical fistula is a pathological communication between the colon and bladder. The symptoms include pneumaturia, fecaluria, and a lower urinary tract infection. The diagnosis is based on clinical symptoms, but the symptoms are not specific. Therefore, confirming the diagnosis is challenging. Urine cultures performed in patients with colovesical fistulas usually show growth of Escherichia coli or mixed growth of bowel organisms. Urinary tract infections caused by Lactococcus lactis are very rare, as it is rarely considered pathogenic in humans. We report the case of a 70-year-old woman wh

Amino acid conversion to aroma compounds by Lactococcus lactis is limited by the low production of alpha-ketoglutarate that is necessary for the first step of conversion. Recently, glutamate dehydrogenase (GDH) activity that catalyzes the reversible glutamate deamination to alpha-ketoglutarate was detected in L. lactis strains isolated from a vegetal source, and the gene responsible for the activity in L. lactis NCDO1867 was identified and characterized. The gene is located on a 70-kb plasmid also encoding cadmium resistance. In this study, gdh gene inactivation and overexpression confirmed th

Fish mucosal tissues lack the organized lymphoid structures present in mammals. Although this is also true for the gills, with the gill-associated lymphoid tissue (GIALT) being mainly composed of B and T cells scattered throughout the epithelium and the lamellae, in some species such as rainbow trout (Oncorhynchus mykiss) or Atlantic salmon (Salmo salar), a more organized structure designated as the interbranchial lymphoid tissue (ILT) is also identified. Yet, although many studies have investigated the immune response of the gill to stimulation or pathogen encounter, which have revealed the a

INTRODUCTION: The immune system's defense against pathogens involves innate and adaptive responses, crucial in maintaining overall health. Immunosuppressed states render individuals more susceptible to potential diseases, indicating the need for effective strategies to bolster immune functions.
OBJECTIVES: Although the immunostimulatory effects of various probiotics have been studied, the specific effects and molecular mechanisms of Lactococcus lactis OTG1204 (OTG1204) remain unknown. In this study, the aim was to investigate the molecular mechanisms of OTG1204 in RAW 264.7 macrophages, the k

Most of the current approaches to influenza vaccine design focus on antibodies against influenza (HA). However, these influenza vaccines typically provide strain-specific protection against mostly homologous subtypes. There is an urgent need to develop a universal vaccine that confers cross-protection against influenza viruses. Of note, the HA stalk domain (HAsd) is a promising target for such an influenza vaccine. In this study, we generated recombinant Lactococcus lactis (L. lactis)/pNZ8150-phosphatidylglycerophosphate synthetase A (pgsA)-HAsd, in which pgsA was used as an anchor protein, an

Upon prolonged exposure to cholate and other toxic compounds, Lactococcus lactis develops a multidrug resistance phenotype that has been attributed to an elevated expression of the heterodimeric ABC-type multidrug transporter LmrCD. To investigate the molecular basis of bile acid resistance in L. lactis and to evaluate the contribution of efflux-based mechanisms in this process, the drug-sensitive L. lactis NZ9000 DeltalmrCD strain was challenged with cholate. A resistant strain was obtained that, compared to the parental strain, showed (i) significantly improved resistance toward several bile

The novel goose parvovirus (NGPV), an emerging variant of goose parvovirus, is the causative agent of short beak and dwarfism syndrome (SBDS) in ducks, posing a substantial threat to the global waterfowl industry. There is an urgent need for safe and convenient vaccines that can effectively induce mucosal immunity. This study aimed to develop and evaluate the immunogenicity and protective efficacy of an oral vaccine based on recombinant Lactococcus lactis (L. lactis) expressing the major capsid protein VP3 of NGPV, with or without fusion to duck interferon-alpha (IFNα). Interferon-alpha (IFN-α) mediates innate antiviral immunity. Although it comprises multiple subtypes, their functional divergence and application potential remain incompletely characterized. In this study, we identified 13 BoIFN-α subtypes (A1-A13) in the bovine genome through bioinformatic analysis. Prokaryotic expression and functional comparison revealed substantial differences in antiviral activity among these subtypes (up to 77.7-fold), with BoIFN-A8 being the most potent. Based on these findings, we designed a novel consensus interferon, BoIFN-Acons, by multiple sequence alignment and su

Sea perch iridovirus (SPIV) causes severe mortality in aquaculture. Lactococcus lactis M48, a strain isolated from the intestine of sea perch, exhibits tolerance to acidic conditions, bile salts, and hyperosmotic stress, along with excellent intestinal colonization capacity and high safety. In this study, a recombinant strain surface-displaying the SPIV major capsid protein (MCP) was constructed based on L. lactis M48, designated RE SPIV MCP M48. Fluorescence microscopy and Western blot analyses confirmed that L. lactis M48 successfully expressed the pgsA-anchored MCP-eGFP fusion protein. Oral

Viruses, like influenza and Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), remain major causes of upper respiratory tract infections worldwide, with symptoms ranging from asymptomatic to lethal outcomes. While antivirals and vaccines have helped ameliorate disease morbidity and mortality, these infections still pose significant challenges. Probiotics, including Lactococcus lactis strain plasma (LC-Plasma), have recently shown antiviral effects by activating plasmacytoid dendritic cells (pDCs), though their detailed mechanism remains unclear. In this study, we stimulated peripher

In the global food industry, fermented dairy products are valued for their unique flavors and nutrients. Lactococcus lactis is crucial in developing these flavors during fermentation. Meeting diverse consumer flavor preferences requires the careful selection of fermentation agents. Traditional assessment methods are slow, costly, and subjective. Although electronic-nose and -tongue technologies provide objective assessments, they are mostly limited to laboratory environments. Therefore, this study developed a model to predict the electronic sensory characteristics of fermented milk. This model

Brucella infections typically occur in mucosal membranes, emphasizing the need for mucosal vaccinations. This study evaluated the effectiveness of orally administering Lactococcus lactis (L. lactis) for producing the Brucella abortus multi-epitope OMPs peptide. A multi-epitope plasmid was generated through a reverse vaccinology method, and mice were administered the genetically modified L. lactis orally as a vaccine. The plasmid underwent digestion, synthesizing a 39 kDa-sized protein known as OMPs by the target group. The sera of mice that were administered the pNZ8124-OMPs-L. lactis vaccine
