We studied a group of patients with myelodysplastic syndromes (MDS) for surface markers and cytotoxic activities of peripheral blood mononuclear cells (PBMNC). The results indicate a significant increase in the total count of CD11b+, Leu7+ and CD16+ with a percent reduction in CD4+. A reduction in PHA-induced cellular cytotoxicity (PHA-ICC) and NK activity were found. A similar phenotype was found both in refractory anemia (RA) and (RA) with excess of blasts (RAEB/RAEB-t). Lectin-binding patterns were examined in epithelial walls of 65 jaw cysts (30 post-operative maxillary cysts: POMCs, 20 radicular and 15 follicular cysts), and characteristic lectin staining for each kind of jaw cysts is presented. A battery of 18 fluorescein isothiocyanate (FITC) labeled lectins was used to histochemically define the features of epithelial cells in the body, neck and the cystic duct of the human gallbladder. A lectin in gluten was detected by agglutination of papain-treated human erythrocytes. A partially purified lectin preparation was obtained by chromatography on immobilized ovalbumin. The number of binding sites for Axinella lectin (specific for D-galactose) on monkey erythrocytes containing schizonts of Plasmodium knowlesi was found to be reduced by about a third, from approximately 11 X 10(6) sites on normal erythrocytes to approximately 8 X 10(6) sites on parasitized erythrocytes. In contrast, only minor differences were found in the amounts of iodinatable external proteins. The results indicate that there is a general reduction of accessible sugars to which lectins may bind on parasitized erythrocytes.
