Spontaneously fermented rennet-coagulated whey from milk of different ruminants: from microbiota characterisation to isolation of lactic acid bacteria with proteolytic potential.

Cheese whey is a highly nutritious by-product of cheesemaking, whose microbiota has been insufficiently studied, particularly in spontaneous fermentation systems. This study aimed to characterize the microbiota of rennet-coagulated whey obtained from raw cow, goat and sheep milks, collected from artisanal farms differing in altitude and sampling season, and to explore spontaneous fermentation as a strategy to recover bacteria with technological potential for whey valorisation. Culturomic and metataxonomic approaches were combined, and spontaneous fermentations at different temperatures were pe

Multi Omics Analysis Reveals That Compound Radix Pulsatillae and Lactic Acid Bacteria Reprogram the Microbiome Metabolome Network in Oat Silage.

Oat (Avena sativa L.) silage fermentation often fails due to insufficient lactic acid bacteria (LAB) and low water-soluble carbohydrate content. We investigated the effects of Compound Radix Pulsatillae (CRP; 40 g/kg FM) alone or combined with a commercial LAB inoculant (containing L. plantarum, L. buchneri, and Enterococcus faecium, CRP_LA) on oat silage after 60 days. Compared to control (CK), both CRP and CRP_LA increased dry matter and water-soluble carbohydrate retention while reducing fiber components and ammonia nitrogen (p < 0.05). CRP_LA exhibited superior fermentation quality (lowest

Malodorous gases production from food wastes decomposition by indigenous microorganisms.

Volatile organic compounds (VOCs) produced during the degradation of food wastes may harm to the health of people and create annoyance in adjacent communities. In this work, the VOCs emitted from the decomposition food wastes including fruit, meat and vegetable, and their microbial communities were measured in three individual 57-L reactors for 61 days. Total of 232.8, 373.5, and 191.1 μg·kg-1·h-1 VOCs with oxygenated VOCs (57.6%), volatile organic sulfur compounds (VOSCs, 58.6%) and VOSCs (54.9%) as the main group were detected during fruit, meat and vegetable fermentation, respectively. 2-Bu

Peroxyl radical- and photo-oxidation of glucose 6-phosphate dehydrogenase generates cross-links and functional changes via oxidation of tyrosine and tryptophan residues.

Protein oxidation is a frequent event as a result of the high abundance of proteins in biological samples and the multiple processes that generate oxidants. The reactions that occur are complex and poorly understood, but can generate major structural and functional changes on proteins. Current data indicate that pathophysiological processes and multiple human diseases are associated with the accumulation of damaged proteins. In this study we investigated the mechanisms and consequences of exposure of the key metabolic enzyme glucose-6-phosphate dehydrogenase (G6PDH) to peroxyl radicals (ROO•)

A Synbiotic of Lacto-N-tetraose and Bifidobacterium animalis subsp. lactis MN-Gup Attenuates High-Fat Diet-Induced Obesity by Modulating Metabolism and Gut Microbiota in Mice.

Background/Objectives: Obesity is closely associated with gut microbiota dysbiosis, intestinal barrier dysfunction, and impaired glucose and lipid metabolism. However, single probiotic or prebiotic interventions often yield only limited metabolic improvements. This study aimed to evaluate the effects of a synbiotic formulation comprising Lacto-N-tetraose (LNT) and Bifidobacterium animalis subsp. lactis MN-Gup (MN-Gup) in a high-fat diet (HFD)-induced obese mouse model. Methods: In this study, an HFD-induced obese mouse model was used to investigate whether the synbiotic formulation of LNT and

Fate of Clostridium estertheticum in biofilms and in response to conventional and novel sanitizers.

Blown pack spoilage (BPS) is a major concern to the meat industry and is primarily caused by Clostridium estertheticum, a psychrophilic spore-forming, anaerobic bacterium. We investigated the survival mechanisms of C. estertheticum and the efficacy of various sanitizers against C. estertheticum spores. When cultured alone or with Pseudomonas poae at 10°C in a biofilm reactor, viable C. estertheticum vegetative cells were recovered only sporadically from stainless steel coupons. However, they were consistently and constantly recovered from planktonic cultures under various conditions for up to

Probiotic potential and partial characterizations of exopolysaccharide from lactic acid bacteria isolated from camel rumen and dairy products.

Lactic acid bacteria (LAB) isolated from camel rumen and traditional dairy products were assessed for their probiotic potentials and exopolysaccharide (EPS) production. EPS were partially characterized, and their safety assessed by hemocompatibility testing in human, sheep, rabbit, and chicken red blood cells. EPS-producing isolates exhibited notable antibacterial activity against Salmonella paratyphi, but showed limited effects against Staphylococcus aureus and Enterococcus faecalis. All isolates demonstrated low resistance under acidic conditions (2.5 and 4.5) with survival rates ranging fro

Entamoeba histolytica - binding of the lipopeptidophosphoglycan - specific monoclonal antibody EH5 to dextrans.

The human intestinal parasite Entamoeba histolytica remains the cause of significant morbidity and mortality. Glycans often play an important role in its interactions with the host. A major surface component of E. histolytica, the lipopeptidophosphoglycan (LPPG), is a glycosylphosphatidylinositol- (GPI)- linked glycoprotein with a large amount of O-glycosylation with dextran-like oligoglucosyl side chains. We had previously isolated an LPPG-specific monoclonal antibody named EH5. This antibody protected SCID (severe combined immunodeficient) mice from amoebic liver abscess. To date, the sequen

New potentially antihypertensive peptides liberated in milk during fermentation with selected lactic acid bacteria and kombucha cultures.

Compounds with the ability to inhibit angiotensin-converting enzyme (ACE) are used medically to treat human hypertension. The presence of such compounds naturally in food is potentially useful for treating the disease state. The goal of this study was to screen lactic acid bacteria, including species commonly used as dairy starter cultures, for the ability to produce new potent ACE-inhibiting peptides during milk fermentation. Strains of Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus helveticus, Lactobacillus paracasei, Lactococcus lact

Head injury profoundly affects gut microbiota homeostasis: Results of a pilot study.

OBJECTIVES: Head injury (HI) induces a hypercatabolic state, dysimmunity, and septic complications that increase morbidity and mortality. Although compromised immune function is usually incriminated in infection occurrence, gut dysbiosis could also be involved in this phenomenon and, to our knowledge, has never been considered. To assess if HI could affect microbiota, we explored the impact of HI on intestinal microbiota in a rodent model of fluid percussion. METHODS: Nineteen rats were randomly assigned to two groups: Healthy rats fed ad libitum (n = 7) and HI rats (n = 12), which received s

ortho-Substituents govern aryl aldehyde reactivity: toward lysine-targeted, tunable inhibitors of glucose-6-phosphate dehydrogenase.

Certain drugs achieve highly potent and long-lasting enzyme inhibition by forming covalent bonds with nucleophilic amino acid functionalities. Aldehyde-containing inhibitors are able to react with lysine ε-amino functionalities to form reversible-covalent imine (Schiff base) adducts. ortho-Substituted aryl aldehydes have been used to further stabilize these imines through intramolecular interactions. However, relationships between aryl aldehyde structure, reactivity, and enzyme inhibition remain poorly defined. To address this gap, we characterized imine formation across a diverse panel of ary
