Lipidomic Profiling and Biological Aging in Patients with Coronary Microvascular Dysfunction.

BACKGROUND: Coronary microvascular dysfunction (CMD) was frequently encountered in patients with angina in the absence of epicardial coronary stenosis. CMD was associated with adverse outcomes while the etiology was unclear. OBJECTIVE: In this study, we aimed to investigate the association between lipidomic abnormalities, biological aging and CMD. METHODS: We consecutively enrolled 43 patients with angina exhibiting no epicardial coronary stenosis on angiography. CMD was defined by a coronary angiography-derived index of microvascular resistance (caIMR) >25. Targeted lipidomic profiling quan

Lysosome-derived methylated arginine is a signalling metabolite controlling the lipidome.

Lysosomes are integral organelles that communicate cellular status to an entire tissue through mechanisms that are poorly defined. Here we developed an unbiased platform, integrating human plasma metabolomes and single-lysosome metabolomics, and show the byproducts of proteolysis are an unexpected class of signalling molecules. We show that dimethylarginine is a lysosomal-derived metabolite and a predictor of patient morbidity. Genetic depletion of a lysosomal exporter, cystinosin, accumulated dimethylarginine in lysosomes. Leveraging a lysosomal storage disease with cystinosin mutations, we s

Lipidomic profile of meningiomas harboring different NF2 mutation status.

INTRODUCTION: Meningiomas are mainly benign brain tumors, but they can evolve to higher grades. The phenomena of these changes are not well-known. Therefore, more basic research is needed. This study attempted to assess the lipidome profile in meningiomas harboring different NF2 mutation statuses (wildtype and mutated). Solid-phase microextraction (SPME) probes were used to sample and extract the metabolites and reduce the invasiveness of lipidomic analysis. OBJECTIVES: This study aimed to select the set of lipids distinguishing meningiomas with different genotypes using two chromatography me

Integrated lipidomic and transcriptomic profiling of the host response in human malaria.

BACKGROUND: Identifying the mechanisms of host-parasite interactions in vivo in malaria is essential for the development of antimalarial strategies tailored to clinically and physiologically relevant contexts. RESULTS: We analyzed 396 paired global serum lipidomes from pediatric patients sampled before and during blood-stage malaria infection in Burkina Faso, spanning three ethnic groups (Gouin, Mossi, and Fulani). Consistent infection-induced remodeling of the host lipidome was identified across populations, including depletion of 47 host-derived lipid species that correlated with parasitemi

Lipidomic Signature of Abdominal Aortic Aneurysm and Peripheral Artery Disease.

Vascular diseases are powerful predictors of cardiovascular mortality, but they are typically under-recognized and undertreated. There is no effective treatment for either abdominal aortic aneurysm (AAA) or peripheral artery disease (PAD). Lipids are key molecules in cardiovascular diseases and good candidates for diagnosis, monitoring, and risk prediction; nonetheless, there is very limited information on the lipidomic profile of patients with AAA and PAD. We hypothesize that lipids can be used as important prognostic biomarkers of these diseases. To achieve this, we conducted a comprehensive

Serum Lipidome as an Early Peripheral Indicator in Familial Alzheimer's Disease.

Protein biomarkers in biofluids are highly sensitive indicators of prodromal cognitive impairment yet remain limited for primary prevention. Lipids, essential to brain structure and function, offer untapped prognostic value. Here, we identify a lipidomic signature in serum from asymptomatic PSEN1-E280A mutation carriers aged 6-40 years, that differentiate carriers from non-carriers with an AUC 80-90%. Similarly, to symptomatic carriers (≥41 years; 93%) and sporadic AD cases (85%), using high-resolution mass spectrometry. Latent profile analysis revealed lipid-based signatures of dementia risk

A Causal Relationship Between the Lipidome and Central Nervous System Tumors.

BACKGROUND: The incidence of central nervous system (CNS) tumors is increasing. However, despite advances in treatment, the etiological factors and mechanisms remain poorly understood. Recent studies have increasingly linked the lipidome to the development of CNS tumors. However, the actual association between liposomes and nervous system tumors remains speculative and lacks definitive conclusions. METHODS: To address this gap, we used a Mendelian randomization approach to systematically evaluate the association between the lipidome and 5 common types of CNS tumors. RESULTS: Our findings rev

Lipidomic Profiling in Cancer: Phospholipid Alterations and their Role in Tumor Progression.

Phospholipids play a crucial role in various aspects of cancer biology, including tumor progression, metastasis, and cell survival. Recent studies have highlighted the signifi-cance of phospholipid metabolism and signaling in multiple cancer types, such as breast, cer-vical, prostate, bladder, colorectal, liver, lung, melanoma, mesothelioma, and oral cancer. Al-terations in phospholipid profiles, particularly in phosphatidylcholine and phosphatidylethan-olamine, have been identified as potential biomarkers for cancer diagnosis and prognosis. Moreover, specific phospholipids and their metabolic

The shifting lipidomic landscape of blood monocytes and neutrophils during pneumonia.

The lipidome of immune cells during infection has remained unexplored, although evidence of the importance of lipids in the context of immunity is mounting. In this study, we performed untargeted lipidomic analysis of blood monocytes and neutrophils from patients hospitalized for pneumonia and age- and sex-matched noninfectious control volunteers. We annotated 521 and 706 lipids in monocytes and neutrophils, respectively, which were normalized to an extensive set of internal standards per lipid class. The cellular lipidomes were profoundly altered in patients, with both common and distinct cha

Causal relationship between lipidome and acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS), with high morbidity and mortality, is a common clinical syndrome of acute respiratory failure caused by diffuse lung inflammation and edema. ARDS can precipitate in various ways. The complex pathophysiology of ARDS involves the activation and dysregulation of multiple metabolisms and immune responses. Using summary-level data from a genome-wide association study (GWAS), a two-sample Mendelian randomization (MR) analysis of 179 genetically predicted lipid species and ARDS (375 cases, 406,518 controls) was performed and validated in plasma and pulmonar

Early pregnancy serum PFAS are associated with alterations in the maternal lipidome.

Per- and polyfluoroalkyl substances (PFAS) have been detected in the blood of humans and animals worldwide. Exposure to some PFAS are associated with multiple adverse pregnancy outcomes. Existing literature has identified a strong association with PFAS exposure and metabolic dysfunction in humans, including modification of lipid metabolism. Using a subset of the Michigan Mother-Infant Pairs cohort (n = 95), this study investigated associations between first trimester plasma levels of PFAS and maternal lipids and metabolites in the first trimester (T1), at the time of delivery (T3), and in the

A large multisite lipidomic investigation of parity and aging in dairy cows.

Efforts to optimize the longevity of dairy cows are hindered by the increased risk of adverse health events, culling, or dying on farm with increased parity. Lipidomics provides a platform to help identify important biomarkers and biological pathways associated with increased parity and associated aging. A large multisite (15 pasture-based, 15 TMR farms) cross-sectional study collected plasma samples from nonlactating, late pregnant, dry cows (n = 696, ∼27 d prepartum) and peak milk cows (n = 796, ∼58 DIM) in a disproportionate stratified random sampling frame (parity: 0, 1, 2, >2 for dry cows

Lipidomic signatures in patients with early-onset and late-onset Preeclampsia.

BACKGROUND: Preeclampsia is a pregnancy-specific clinical syndrome and can be subdivided into early-onset preeclampsia (EOPE) and late-onset preeclampsia (LOPE) according to the gestational age of delivery. Patients with preeclampsia have aberrant lipid metabolism. This study aims to compare serum lipid profiles of normal pregnant women with EOPE or LOPE and screening potential biomarkers to diagnose EOPE or LOPE. METHODS: Twenty normal pregnant controls (NC), 19 EOPE, and 19 LOPE were recruited in this study. Untargeted lipidomics based on ultra-performance liquid chromatography-tandem mass

Male infertility risk and plasma lipidome: a Mendelian randomization study.

BACKGROUND: In recent years, the decline in sperm quality in men has become a global trend. There is a close relationship between sperm quality and pregnancy outcome. There is a large body of literature supporting the role of plasma lipidome in male infertility, while the complex mechanisms between them and male infertility are still less clear. Systematic study of the causal relationship between plasma lipidome and MI can help to provide new therapeutic ideas and targets for male infertility. METHODS: In this study, we used a two-sample Mendelian randomization analysis based on Genome-wide a

Effect of biological sex on human circulating lipidome: An overview of the literature.

Cardiovascular diseases (CVD) are the leading cause of death worldwide for both men and women, but their prevalence and burden show marked sex differences. The existing knowledge gaps in research, prevention, and treatment for women emphasize the need for understanding the biological mechanisms contributing to the sex differences in CVD. Sex differences in the plasma lipids that are well-known risk factors and predictors of CVD events have been recognized and are believed to contribute to the known disparities in CVD manifestations in men and women. However, the current understanding of sex di

A Comprehensive Insight and Mechanistic Understanding of the Lipidomic Alterations Associated With DCM.

Dilated cardiomyopathy (DCM) is one of the major causes of heart failure characterized by the enlargement of the left ventricular cavity and contractile dysfunction of the myocardium. Lipids are the major sources of energy for the myocardium. Impairment of lipid homeostasis has a potential role in the pathogenesis of DCM. In this review, we have summarized the role of different lipids in the progression of DCM that can be considered as potential biomarkers. Further, we have also explained the mechanistic pathways followed by the lipid molecules in disease progression along with the cardioprote

Lipidomic alterations in human saliva from cystic fibrosis patients.

Cystic fibrosis is a hereditary metabolic disorder characterized by impaired traffic of chloride ions and water through membranes of the respiratory and gastrointestinal, that causes inadequate hydration of airway surfaces, dehydrated mucous secretions and a high-sodium chloride sweat. Although the classical presentation of the condition is well known, a better characterization of metabolic alterations related is need. In particular, the metabolic composition alterations of biological fluids may be influence by the disease state and could be captured as putative signature to set targeted thera

Plasma lipidome profiling of newborns with antenatal exposure to Zika virus.

The 2015-2016 Zika virus (ZIKV) outbreak in Brazil was remarkably linked to the incidence of microcephaly and other deleterious clinical manifestations, including eye abnormalities, in newborns. It is known that ZIKV targets the placenta, triggering an inflammatory profile that may cause placental insufficiency. Transplacental lipid transport is delicately regulated during pregnancy and deficiency on the delivery of lipids such as arachidonic and docosahexaenoic acids may lead to deficits in both brain and retina during fetal development. Here, plasma lipidome profiles of ZIKV exposed microcep
