Neuronal lipofuscinosis caused by Kufs disease/CLN4 DNAJC5 mutations but not by a CSPα/DNAJC5 deficiency.

Kufs disease/CLN4 is an autosomal dominant neurodegenerative disorder caused by unknown mechanisms through Leu115Arg and Leu116Δ mutations in the DNAJC5 gene that encodes the synaptic vesicle co-chaperone cysteine string protein α (CSPα/DNAJC5). To investigate the disease mechanisms in vivo, we generated three independent mouse lines overexpressing different versions of CSPα/DNAJC5 under the neuron-specific Thy1 promoter: wild-type (WT), Leu115Arg, and Leu116Δ. Mice expressing mutant Leu115Arg CSPα/DNAJC5 are viable but develop motor deficits. As described in patients with Kufs disease, we obs

Neuroprotective role of Bacopa monniera extract against aluminium-induced oxidative stress in the hippocampus of rat brain.

Bacopa monniera is a nerve tonic used extensively in traditional Indian medicinal system "Ayurveda". Reports regarding its various antioxidative, adaptogenic and memory enhancing roles have already appeared in the last few decades. In the present study, aluminium chloride (AlCl(3)) was used to generate neurotoxicity. We have investigated the neuroprotective effect of Bacopa extract against aluminium-induced changes in peroxidative products, such as thio-barbituric acid-reactive substance (TBA-RS) and protein carbonyl contents and superoxide dismutase (SOD) activity. Effect on lipofuscin (age p

[Amiodarone pigmentation. A rare, but typical drug side-effect].

Amiodarone is one of the most effective antiarrhythmic and anti-anginose drugs. A rare but typical side-effect with an incidence of 1:100 is sunburn, and of 1:1,000 a black-violet to slaty pigmentation of the sun-exposed skin areas. According to histochemical and ultrastructural findings the skin pigment deposits are considered to be lipofuscin.

Apocrine chromhidrosis involving the areolae in a 15-year-old amateur figure skater.

Apocrine chromhidrosis is a rare disease frequently localized to the face or axillae, and rarely has been reported to occur around the breasts. We report a 15-year-old amateur figure skater who displayed localized chromhidrosis around her areolae. The most common precipitating event was exercise. She was treated topically with capsaicin cream 0.025% with a subsequent decrease in symptomatology.

Lysosomal abnormalities in degenerating neurons link neuronal compromise to senile plaque development in Alzheimer disease.

Antibodies to the lysosomal hydrolases, cathepsins B and D and beta-hexosaminidase A, revealed alterations of the endosomal-lysosomal system in neurons of the Alzheimer disease brain, which preceded evident degenerative changes and became marked as atrophy, neurofibrillary pathology, or chromatolysis developed. At the earliest stages of cell atrophy, hydrolase-positive lysosomes accumulated at the basal pole and then massively throughout the perikarya and proximal and proximal dendrites of affected pyramidal neurons in Alzheimer prefrontal cortex and hippocampus, far exceeding the changes of n

Autofluorescent granular cells in oral mucosal hyperplasias.

Four hyperplastic lesions of the hard palate are described. They were characterized by the presence of an atrophic surface epithelium, an oedematous matrix and the presence of large numbers of autofluorescent granular cells. In addition there were numerous mast cells. Histochemical and electronmicroscopical studies suggest that the granules are composed of lipofuscin. The pathogenesis of these lesions is discussed.

[Melanosis of choroid plexus papilloma of the lateral ventricle: a case report].

A case of pigmented, well differentiated, choroid plexus papilloma is reported. A 45-year-old man came to our hospital complaining of attacks of vertigo and nausea. Skull X-ray films and CT scan demonstrated a calcified tumor in the left middle fossa. Tumor removal was done, and it was found that the tumor occupied the inferior horn of the left lateral ventricle. Histological examination revealed well differentiated choroid plexus papilloma containing various shaped pigment granules in the cytoplasm. The pigment granules in the tumor cells were stained black by Fontana-Masson method for melani

Photoperiod-induced apoptosis in the male genital tract epithelia of the golden hamster.

The aim of this study was to identify some details of the changes induced by a short-day light regime (8:16 light:dark) on the male genital tract and accessory sex glands of the golden hamster Mesocricetus auratus. We principally examined the presence of apoptotic cells in the epithelium from different regions of the epididymis, seminal vesicles, prostate and coagulating gland. We detected an increase in the percentage of apoptotic cells in situ using the TUNEL technique in animals that were maintained for 6, 8 or 12 weeks in a short photoperiod. That those cells were indeed undergoing apoptos

Small bowel intussusception and brown bowel syndrome in association with severe malnutrition.

Brown bowel syndrome is a rare condition characterized by deposition of lipofuscin in the smooth muscle cells of the gastrointestinal tract. The number of reported cases is small, but all are associated with malabsorptive states. Despite these small numbers, there is considerable evidence that vitamin E deficiency is important etiologically. We report here the case of a severely malnourished [body mass index 11.7 kg/m (2): normal range 20-25 kg/m (2)] 31-yr-old black male with a longstanding history of alcohol abuse, who was on anti-tuberculosis therapy. The patient presented with an acute abd

Mucopolysaccharidosis type V. (Scheie syndrome). A postmortem study by multidisciplinary techniques with emphasis on the brain.

Multidisciplinary studies were conducted on the brain and other tissues of patients who died with the antemortem diagnosis of mucopolysaccharidosis (MPS) of one of the following types; type V, Scheie disease (MPS-V); type I, Hurler disease (MPS-I): and type II, Hunter disease (MPS-II). The principal new finding in the brain of the patient with MPS-V is the presence of lesions in the periadventitial mesenchymal tissue of the white matter, similar to those of MPS-I, while the nerve cells in MPS-V are histologically normal, in contradistinction to MPS-I, in which the neuronal abnormality is sever

Increased iron levels correlate with the selective nigral dopaminergic neuron degeneration in Parkinson's disease.

The staging of Lewy-related pathology in sporadic Parkinson's disease (PD) reveals that many brain nuclei are affected in PD during different stages, except the ventral tegmental area (VTA), which is close related to the substantia nigra (SN) and enriched in dopamine (DA) neurons. Why DA neurons are selectively degenerated in the SN of PD is far from known. In the present study, we observed that the number of tyrosine hydroxylase immunoreactive neurons decreased and iron-staining positive cells increased in the SN, but not in the VTA, in the chronic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine

Liver abnormalities associated with chronic mercury accumulation in stranded Atlantic bottlenose dolphins.

Eighteen stranded Atlantic bottlenose dolphins (Tursiops truncatus) examined postmortem were sampled for histologic study. All cases were examined for ferric ion and lipofuscin. Ages were determined from tooth growth layers. Electron microscopic (EM) examination and X-ray spectroscopy (EDAX) were performed. Chemical analysis for mercury was conducted on 12 of the animals by atomic absorption spectrophotometry. Nine animals were found to have excessive lipofuscin in both liver and kidney. Four of these nine animals also exhibited active liver disease (fat globules, central necrosis, lymphocytic

Stargardt macular dystrophy.

Stargardt macular dystrophy (STGD1) is the most prevalent inherited macular dystrophy, characterized by progressive central vision loss and decline of visual acuity. It is caused by homozygous or compound heterozygous variants in the ABCA4 gene, with a large genetic heterogeneity that poses challenges in establishing genotype-phenotype correlations. Null variants are generally associated with earlier and more severe disease onset, whereas missense and deep intronic variants have been identified as contributors to late-onset disease with milder phenotypes. Pathophysiologic mechanisms involve im

Senescence as a novel mechanism involved in β-adrenergic receptor mediated cardiac hypertrophy.

Pathological cardiac hypertrophy used to be elucidated by biomechanical, stretch-sensitive or neurohumoral mechanisms. However, a series of hints have indicated that hypertrophy process simulates senescence program. However, further evidence need to be pursued. To verify this hypothesis and examine whether cardiac senescence is a novel mechanism of hypertrophy induced by isoproterenol, 2-month-old male Sprague Dawley rats were subjected to isoproterenol infusion (0.25mg/kg/day) for 7 days by subcutaneous injection). Key characteristics of senescence (senescence-associated β-galactosidase activ

Hemosiderin deposits in chronic graft-vs.-host disease related myopathy.

Chronic graft-vs.-host disease (cGVHD) occurs in 20-50% of patients who survive for at least 100 d after allogeneic stem cell transplantation (SCT). cGVHD includes scleroderma-like skin changes, chronic cholangitis, obstructive lung disease and general wasting syndrome. Polymyositis or myopathy are rare manifestations of cGVHD with approximately 40 reported cases. Polymyositis accompanied by hemosiderin deposits in cGVHD has been reported only once, and there are no reports on lipofuscin deposits in skeletal muscle cells in cGVHD. We report here on a 56-yr-old male who underwent allogeneic SCT

Effect of hormone replacement therapy in normalizing age related neuronal markers in different age groups of naturally menopausal rats.

Aging of the normal brain is accompanied by changes in its structure, function, and metabolism. There are significant gender differences in aging brain. Most of these changes increase during menopausal condition in females when the level of estradiol and progesterone are decreased. The objective of this study was to determine the effect of estradiol and progesterone (separate as well as combined) hormones in neuronal tissues from naturally menopausal rats of different age groups. Results show decreased activity of Acetylcholine esterase (AChE) whereas the level of lipid peroxidation increased
