Evaluation of humoral immune response and milk antibody transfer in calves and lactating cows vaccinated with inactivated H5 avian influenza vaccine.

The detection of Highly Pathogenic Avian Influenza (HPAI) A(H5N1) in dairy cattle in the United States has raised concerns about human exposure. This study evaluated the efficacy of various doses of an inactivated H5 AI vaccine in cattle and assessed antibody transfer in milk against a recent bovine isolate of HPAI A(H5N1, clade 2.3.4.4b). Calves were inoculated with different vaccine doses, while lactating cows received the vaccine four weeks later. The humoral immune response was measured using the Hemagglutination Inhibition (HI) test and ELISA. Results showed a dose-dependent immune respon

African Swine Fever Vaccinology: The Biological Challenges from Immunological Perspectives.

African swine fever virus (ASFV), a nucleocytoplasmic large DNA virus (NCLDV), causes African swine fever (ASF), an acute hemorrhagic disease with mortality rates up to 100% in domestic pigs. ASF is currently epidemic or endemic in many countries and threatening the global swine industry. Extensive ASF vaccine research has been conducted since the 1920s. Like inactivated viruses of other NCLDVs, such as vaccinia virus, inactivated ASFV vaccine candidates did not induce protective immunity. However, inactivated lumpy skin disease virus (poxvirus) vaccines are protective in cattle. Unlike some e

Two proteins, one goal: ELISAs based on p32 and L1R for LSDV antibodies detection.

INTRODUCTION: Lumpy skin disease virus (LSDV), a member of the Capripoxvirus genus, poses a significant threat to livestock health and productivity in both endemic and newly affected regions. The disease is primarily transmitted by blood-feeding insects, leading to fever, cutaneous nodules, lymphadenopathy, and substantial economic losses. While vaccination remains the cornerstone of control efforts, effective surveillance-especially in high-risk areas-relies on robust and scalable diagnostic tools. Although the virus neutralization test is considered the reference standard among serological a

Relationship between burden of infection in ungulate populations and wildlife/livestock interfaces.

In southern African transfrontier conservation areas (TFCAs), people, livestock and wildlife share space and resources in semi-arid landscapes. One consequence of the coexistence of wild and domestic herbivores is the risk of pathogen transmission. This risk threatens local livelihoods relying on animal production, public health in the case of zoonoses, national economies in the context of transboundary animal diseases, and the success of integrated conservation and development initiatives. The level of interaction between sympatric wild and domestic hosts, defining different wildlife/livestoc

Insights into goatpox virus and sheeppox virus genomes from pangenome graphs.

The capripoxviruses comprise three species: goatpox virus (GTPV), sheeppox virus (SPPV) and lumpy skin disease virus (LSDV). They are large double-stranded DNA viruses with highly conserved core genomes and variable terminal regions. Previous studies have described variation in Capripoxvirus gene content, their broader population structure and the contribution of non-coding and structural variation remains opaque. This study investigated the genomic diversity and evolutionary history of GTPV and SPPV using phylogenetics, pangenome variation graphs (PVGs), and gene-specific analyses. We found c

Tick-borne diseases in Egypt: A one health perspective.

BACKGROUND: Ticks are important arthropod vectors that transmit pathogens to humans and animals. Owing to favourable climatic and environmental conditions, along with animal importation from neighbouring countries, ticks and tick-borne diseases (TBDs) are widespread in Egyptian localities. Here, we review the current knowledge on the epidemiology of TBDs in Egypt in light of the One Health paradigm. METHODS AND RESULTS: Five scientific databases, including "Web of Science", "Scopus", "PubMed", "Science Direct", and "Google Scholar", were searched for articles describing TBDs in Egypt. A total

Establishment of an Indirect ELISA for Bovine Infectious Rhinotracheitis Based on Eukaryotic Expression of gD Protein in CHO Cells.

To develop an indirect ELISA kit for detecting serum antibodies against bovine herpesvirus 1 (BoHV-1), the gD protein of BoHV-1 was expressed in CHO-S cells and purified for use as the coating antigen. The optimized conditions were as follows: a coating antigen concentration of 2 µg/mL; serum dilution at 1:40 with incubation at 37 °C for 60 min; enzyme-labeled antibody dilution at 1:5000 with incubation at 37 °C for 60 min; and color development with TMB substrate at 37 °C for 15 min. The absorbance was then measured with a microplate reader. The kit could detect BoHV-1-positive serum at a dil

Effectiveness and profitability of preventive veterinary interventions in controlling infectious diseases of ruminant livestock in sub-Saharan Africa: a scoping review.

Agriculture in general, and livestock production in particular, serve as a livelihood source for many people in sub-Saharan Africa (SSA). In many settings, lack of control of infectious diseases hampers livestock productivity, undermining the livelihood of rural populations. This scoping review sought to identify veterinary interventions previously evaluated as well as their relative effectiveness in controlling infectious livestock diseases. To be included, papers had to be written in English, German or French, and had to describe the effectiveness and/or profitability of preventive veterinar

In-silico characterization of LSDV132 protein divulged its BCL-2-like nature.

Lumpy skin disease virus (LSDV) belongs to Poxviridae family. This virus possesses various proteins which impart potential functions to it including assembly of newly synthesized viruses in the replication cycle and forming their structure. LSDV132 protein is also one of such proteins. Its key characteristics were unknown because, no any relevant study was reported about it. This study aimed to investigate its characteristic features and essential functions using several bioinformatics techniques. These analyses included physiochemical characterization and exploring the crucial functional and

Veterinarians' perception of livestock infectious disease: results from a five country cross-European survey (2024).

BACKGROUND: Livestock production accounts for nearly half of all agricultural output globally, but the long-term sustainability of the sector is threatened by a range of infectious diseases. Endemic diseases lead to a wide range of production losses and animal welfare issues, while transboundary and epidemic diseases can lead to widespread deaths, the implementation of stringent control measures, and a disruption of trade. Zoonotic diseases further threaten human health. It is therefore critical that livestock diseases are effectively controlled. When implementing disease control strategies an

The genome of swinepox virus.

Swinepox virus (SWPV), the sole member of the Suipoxvirus genus of the Poxviridae, is the etiologic agent of a worldwide disease specific for swine. Here we report the genomic sequence of SWPV. The 146-kbp SWPV genome consists of a central coding region bounded by identical 3.7-kbp inverted terminal repeats and contains 150 putative genes. Comparison of SWPV with chordopoxviruses reveals 146 conserved genes encoding proteins involved in basic replicative functions, viral virulence, host range, and immune evasion. Notably, these include genes with similarity to genes for gamma interferon (IFN-g

WildHealthNet: Supporting the development of sustainable wildlife health surveillance networks in Southeast Asia.

Wildlife and wildlife interfaces with people and livestock are essential surveillance targets to monitor emergent or endemic pathogens or new threats affecting wildlife, livestock, and human health. However, limitations of previous investments in scope and duration have resulted in a neglect of wildlife health surveillance (WHS) systems at national and global scales, particularly in lower and middle income countries (LMICs). Building on decades of wildlife health activities in LMICs, we demonstrate the implementation of a locally-driven multi-pronged One Health approach to establishing WHS in

Extended sequencing of vaccine and wild-type capripoxvirus isolates provides insights into genes modulating virulence and host range.

The genus Capripoxvirus in the subfamily Chordopoxvirinae, family Poxviridae, comprises sheeppox virus (SPPV), goatpox virus (GTPV) and lumpy skin disease virus (LSDV), which cause the eponymous diseases across parts of Africa, the Middle East and Asia. These diseases cause significant economic losses and can have a devastating impact on the livelihoods and food security of small farm holders. So far, only live classically attenuated SPPV, GTPV and LSDV vaccines are commercially available and the history, safety and efficacy of many have not been well established. Here, we report 13 new caprip
