Effect of zaldaride maleate, an antidiarrheal compound, on visceral pain reflex induced by small intestinal distention in anesthetized rats.

Using distention of the small intestine as a visceral pain model, we investigated the effect of zaldaride maleate (ZAL), a selective inhibitor of calmodulin, on the depressor response. In pentobarbital-anesthetized rats, small intestine distention was induced by rapid application of intraluminal pressures of 40 cmH2O causing a reflex fall in arterial blood pressure. The depressor response to intestinal distention was abolished by intraperitoneal administration of capsaicin (5 mg/rat), which depletes neuropeptides such as substance P from the sensory neurons, on the mesenteric stalk and by neon

[Pharmacological studies on proglumetacin maleate, a new non-steroidal anti-inflammatory drug. (2). Analgesic and antipyretic effects].

Analgesic and antipyretic effects of proglumetacin maleate (PGM), a new indomethacin (IND) derivative, were investigated in comparison with those of IND on an equimolar-dose basis. The suppression of phenylquinone-induced writhing in mice by PGM was about 0.8 and 2 times as potent as that by IND when given 1 and 4 hr before the phenylquinone injection, respectively. The analgesic activity of PGM in rat silver nitrate arthritis was about 1.5 times more potent than that of IND. PGM was slightly less active in rat adjuvant arthritic pain than IND. On the other hand, PGM provoked a dose-dependent

Preparation and Characterization of a Low-Molecular-Weight Chitosan Nanoparticle-Based Ocular Formulation for Enhanced Permeation of Timolol Maleate.

INTRODUCTION/OBJECTIVE: Timolol Maleate (TM) is primarily used to treat glaucoma by reducing intraocular pressure. However, its efficacy is limited by poor ocular bioavailability (<5%) and systemic side effects. This study aimed to develop and evaluate Low-Molecular-Weight Chitosan (LMWC)-based complexes in an eye formulation to sustain TM release and enhance its corneal permeability. METHODS: TM-LMWC Polyelectrolyte Complexes (PECs) were prepared by ionic interactions between TM and LMWC and characterized for particle size, polydispersity, surface charge, and physicochemical properties. TM L

Quantifying crystallinity of amlodipine maleate in amorphous solid dispersions produced by fluidized bed granulation using PAT tools.

One of the main concerns with formulations containing amorphous solid dispersions (ASDs) is their physical stability. Stability can be compromised if a formulation contains any residual crystallinity of an active pharmaceutical ingredient (API) that could act as seeds for further crystallisation. This study presents four methods for crystalline amlodipine maleate quantification in ASD, which were developed using one Raman and three NIR process analysers. A preliminary analysis revealed distinct differences between amorphous and non-amorphous forms of the API, both in the API alone and in the f

Edonerpic maleate enhances functional recovery from spinal cord injury with cortical reorganization in non-human primates.

While spinal cord injury (SCI) aggravates the quality of life in humans by severe paralysis, clinical intervention to promote functional recovery from SCI is limited. We recently identified a small compound, edonerpic maleate (edonerpic MA), which accelerates training-dependent motor functional recovery from brain damage in rodents (cryo-genic cortical injury) and non-human primates (internal capsule haemorrhage) by the facilitation of experience-dependent synaptic trafficking of glutamate α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors. In the present study, we investigated whe

Edonerpic maleate prevents epileptic seizure during recovery from brain damage by balancing excitatory and inhibitory inputs.

Functional recovery from brain damage, such as stroke, is a plastic process in the brain. The excitatory glutamate α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid receptor (AMPAR) plays a crucial role in neuronal functions, and the synaptic trafficking of AMPAR is a fundamental mechanism underlying synaptic plasticity. We recently identified a collapsin response mediator protein 2 (CRMP2)-binding compound, edonerpic maleate, which augments rehabilitative training-dependent functional recovery from brain damage by facilitating experience-driven synaptic delivery of AMPARs. In animals reco

Boiling water resistant fully bio-based adhesive made from maleated chitosan and glucose with excellent performance.

Biomass resources are widely considered potential alternatives to formaldehyde-based wood adhesives because of their abundance. In this study, an environmentally friendly biomass adhesive, carboxylated chitosan-glucose (CSC-G), was prepared using chitosan, maleic anhydride, and glucose. The structure and water resistance of the adhesive were analyzed in detail. Maleic anhydride act as a bridge connecting chitosan and glucose, giving the adhesive good water solubility and resistance. The improved water resistance of the CSC-G adhesive was attributed to the formation of covalent cross-linked str

Influence of commonly used excipients on the chemical degradation of enalapril maleate in its solid state: The role of condensed water.

The physicochemical stability of enalapril maleate was investigated in the presence of fourteen different excipients divided into four different classes. The extent of a drug-excipient interaction was investigated by following the chemical stability using HPLC. It was found that there is a certain order in the stability of enalapril maleate. Enalapril maleate remained most stable in the presence of: disaccharides > celluloses > starches > superdisintegrants. The amount of degradation can be related to the excipient characteristics. A material with a higher water sorption capacity and lower cry

The comparative ocular hypotensive effect of apraclonidine with timolol maleate in exfoliation versus primary open-angle glaucoma patients.

PURPOSE: To compare the effect of adding apraclonidine 0.5% to timolol maleate 0.5% in patients with exfoliation versus primary open-angle glaucoma. Since exfoliation glaucoma is known to demonstrate higher pressures than primary open-angle glaucoma on timolol maleate therapy alone, the authors wished to determine whether apraclonidine equalised the intraocular pressure (IOP) between these two glaucomas when added to timolol maleate. METHODS: We age-matched 30 consecutive exfoliation and 30 primary open-angle glaucoma patients who had an IOP > or = 22 mmHg on timolol maleate alone. Patients u

Blockade of N-methyl-D-aspartate receptors by MK-801 (dizocilpine maleate) rescues motoneurones in developing rats.

In rats following nerve injury at birth a large proportion of motoneurones to the soleus muscle dies. Blocking of N-methyl-D-aspartate (NMDA) receptors by MK-801 (dizocilpine maleate) for 12 days after nerve injury at birth leads to rescue of a proportion of motoneurones destined to die. Retrograde labelling of soleus motoneurones shows that 6-8 weeks after crushing the sciatic nerve in one hindlimb, only 10.9 +/- 2.3% of the motoneurones have survived. In animals treated with an NMDA receptor blocker MK-801 (2 mg/kg i.p., from birth to 12 days old) 50.6 +/- 3.8% of soleus motoneurones survive

Anti-inflammatory effects of proglumetacin maleate (PGM), a new indomethacin (IND) derivative, were compared with those of IND on an equimolar-dose basis. PGM produced a dose-dependent inhibition of vascular permeability and carrageenin edema. These inhibitory effects of PGM were greater when given 4 hr prior to phlogistic agents than when given 1 hr before. Moreover, these effects of PGM were long-acting. Inhibitory effects of PGM on kaolin edema and UV-erythema were slightly less active than those of IND. PGM markedly reduced the leukocyte migration in carrageenin pleurisy. Subacute anti-exu
