We have studied the transport characteristics of cyclosporine A (CSA) in isolated rabbit renal proximal tubular cells (PTC). The uptake as well as efflux was very rapid and dependent on temperature. PTC accumulated CSA by several fold above the incubation medium concentration. Kinetic analysis yielded an apparent Km and Vmax values of 5.1 microM and 47 Pmoles/10(6) cells/min respectively. Calcium channel blockers verapamil or diltiazem, at concentrations (0.5-1.0 mM) that inhibited calcium uptake, reduced CSA uptake significantly. Other calcium transport modulators A23187 (5 microM), trifluoro

Zinc is an essential micronutrient that is crucial for many vital cellular functions such as DNA and protein synthesis, metabolism, and intracellular signaling. Therefore, the intracellular zinc concentration is tightly regulated by zinc transporters and zinc-binding proteins. The members of the SCL39 transporter family transport zinc into the cytosol. The SLC39A2 (hZIP2) protein is highly expressed in prostate epithelial cells and was found to be involved in prostate cancer development. Thus far, there is no specific modulator available for the SLC39 transporters. The aim of this study was to

Despite its established role as a cornerstone of chemotherapy for solid tumors, 5-Fluorouracil (5-FU) clinical efficacy remains limited by chemoresistance and heterogeneous drug response. Traditional explanations have focused on intracellular metabolism and genetic determinants; however, increasing evidence identifies the plasma membrane as a critical regulatory interface controlling drug availability, signaling integration, and cell fate. Here, we propose a membrane-centered framework in which compartmentalized cAMP/PKA signaling, modulated by repurposed vasoregulatory agents-levosimendan, mi

Glycinergic inhibitory neurons of the spinal dorsal horn exert critical control over the conduction of nociceptive signals to higher brain areas. The neuronal glycine transporter 2 (GlyT2) is involved in the recycling of synaptic glycine from the inhibitory synaptic cleft and its activity modulates intra and extracellular glycine concentrations. In this report we show that the stimulation of P2X purinergic receptors with βγ-methylene adenosine 5'-triphosphate induces the up-regulation of GlyT2 transport activity by increasing total and plasma membrane expression and reducing transporter ubiqui

The N-Myc Downregulated-Like (NDL) protein family in Arabidopsis was identified as an interacting partner of components of the G-protein complex. NDL's role has been established in regulating auxin transport through modulation of auxin transport facilitators in an Arabidopsis G-protein β subunit (AGB1)-dependent manner. NDL proteins are modulators of abiotic stress response, such as high salt and drought, and may serve as predictors of plant morphophysiological responses based on their gene expression patterns. The domain organization of NDL proteins predicts that they contain esterases/lipase

GABA is the major inhibitory amino acid in the central nervous system (CNS), and its homeostasis is crucial for inhibitory/excitatory balance. Its reuptake is mediated by GABA transporters, including type 1 (GAT-1), a high-affinity GABA plasma membrane transporter. Ethanol (EtOH) consumption enhances GABA inhibitory effects; however, the impact of acute EtOH exposure on GABAergic function during development remains to be understood. Therefore, we aimed to evaluate whether acute EtOH exposure modulates GABA transport in chicken retina, a well-established model for investigating neurochemical st

Purine derivatives, especially adenosine and ATP, play an important role in the function of many organs, being released either as purinergic transmitters or as local modulators in a non-specific way from all tissues. Adenosine and adenine nucleotides act physiologically by interfering with specific cell membrane receptors (P1 and P2 respectively) that are associated with several signal transduction systems. The adenosine system seems to act in complex situations in which cells and tissues must deal with high energy consumption, maintaining or restoring an adequate balance between metabolic req

Rising ocean temperatures due to global warming pose severe threats to marine organism survival. Membrane proteins perform important biological functions and serve as sensitive environmental stress sensors. However, the characterization of membrane protein content in response to temperature adaptation remains poorly understood in marine organisms. Here, we performed reciprocal transplantation experiments between the northern/cold and southern/warm natural habitats of two allopatric but closely related oyster species, Crassostrea gigas and Crassostrea angulata. Comparative membrane proteomics c

To bypass the enduring limitations intrinsic to traditional size-sieving separation, alternative materials for CO2/H2 separation need to be developed. Here, we program a permeation inversion in CO2/H2 transport by constructing a nanoconfined ionic liquid membrane through the synergistic engineering of graphene oxide nanosheets and ionic liquids. A substrate-assisted size-selection strategy enables the preferential incorporation of large nanosheets, which form ordered laminates, while short-chain CO2-philic ionic liquids are precisely confined within the interlayer galleries. The resulting memb

Abnormal fetal growth is associated with perinatal complications and adult disease. The placental mechanistic target of rapamycin (mTOR) signaling activity is positively correlated with placental nutrient transport and fetal growth. However, if this association represents a mechanistic link, it remains unknown. We hypothesized that trophoblast-specific Mtor knockdown in late pregnant mice decreases trophoblast nutrient transport and inhibits fetal growth. PiggyBac transposase-enhanced pronuclear injection was performed to generate transgenic mice containing a trophoblast-specific Cyp19I.1 prom

Desalination based on carbon nanomembranes offers high water permeance and salt rejection, making them promising for addressing global freshwater shortages and energy demands in reverse osmosis (RO) desalination. Enhancing ion rejection by modulating the energy barrier for ion transport through wide carbon nanotubes (CNTs) is a critical challenge for highly efficient desalination. We perform molecular dynamics simulations on water desalination using CNTs membranes, highlighting the key role of nanoconfinement coupled with an electric field. The results show that the electric field extends the

Light-responsive control over supramolecular self-assembly provides a route to dynamically regulate nanoscale transport processes. Here we demonstrate that photoisomerization of a membrane-bound chalcone amphiphile enables light-driven reprogramming of catanionic self-assembly, producing composition-dependent morphological transitions that directly govern drug-release kinetics. Mixtures of a 2-hydroxychalcone-derived cationic amphiphile (C6NCh) with the biocompatible anionic surfactant sodium N-lauroyl sarcosinate spontaneously form stable unilamellar vesicles (hydrodynamic diameter ≈ 130-200

Resveratrol (Rsv) is a natural polyphenol with neuroprotective properties that modulates several pathways implicated in Alzheimer's disease (AD). Cholesterol homeostasis is disrupted in AD patients, and this imbalance plays a key role in amyloid precursor protein (APP) processing, β-amyloid aggregation and membrane stability. The effect of Rsv on the cerebellum, an emerging structure in cognitive networks and AD pathology due to its high connectivity with other brain regions, remains largely unexplored. This study aims to characterize the effects of Rsv on the cerebellum of SAMP8 mice, an anim

The electrocatalytic CO2 reduction reaction (CO2RR) in membrane electrode assemblies (MEAs) represents a pivotal technology for carbon neutrality, yet its industrial deployment is severely restricted by the carbonate dilemma. Traditional alkali-cation systems (e.g., K+ and Cs+) suffer from salting deposition and carbonate crossover, which lead to gas diffusion electrode flooding and abrupt mass-transport failure. This review provides a comprehensive and timely analysis of non-alkali cationic systems, including inorganic ammonium salts, quaternary ammonium salts, and cationic polyelectrolytes,

Bile acids (BAs) are amphipathic molecules important for metabolism of cholesterol, absorption of lipids and lipid soluble vitamins, bile flow, and regulation of gut microbiome. There are over 30 different BA species known to exist in humans and mice, which are endogenous modulators of at least 6 different membrane or nuclear receptors. This diversity of ligands and receptors play important roles in health and disease; however, the full functions of each individual BA in vivo remain unclear. We generated a mouse model lacking the initiating enzymes, CYP7A1 and CYP27A1, in the two main pathways

Beneficial effects of the microbiota-derived metabolite butyrate at the colonic level are well established, particularly through its relevance in colorectal cancer (CRC) and inflammatory bowel disease (IBD), two major intestinal pathologies. Therefore, the mechanisms involved in butyrate transport across colonic epithelial cell membranes (uptake transporters: monocarboxylate transporter 1 (MCT1) and sodium-coupled monocarboxylate transporter 1 (SMCT1); efflux transporters: breast cancer resistance protein (BCRP) and MCT1/monocarboxylate transporter 4 (MCT4)), which are determinant for its intr

UNLABELLED: The cell envelope of many bacteria possesses components that play central roles in bacterial pathogenesis. Unique to mycobacteria is the presence of a lipid-rich cell wall which is critical for the virulence of pathogenic mycobacteria such as Mycobacterium tuberculosis ( Mtb ). The biosynthesis of mycobacterial cell wall lipids has been well characterized, but the mechanisms of lipid transport still remain largely unknown. MmpL (mycobacterial membrane protein large) proteins have been implicated in the biosynthesis and/or transport of mycobacterial cell wall lipids, but due to thei

The discovery of ClC proteins at the beginning of the 1990s was important for the development of the Cl- transport research field. ClCs form a large family of proteins that mediate voltage-dependent transport of Cl- ions across cell membranes. They are expressed in both plasma and intracellular membranes of cells from almost all living organisms. ClC proteins form transmembrane dimers, in which each monomer displays independent ion conductance. Eukaryotic members also possess a large cytoplasmic domain containing two CBS domains, which are involved in transport modulation. ClC proteins functio

Presynaptic serotonin (5-hydroxytryptamine, 5-HT) transporters (SERTs) mediate antidepressant-sensitive clearance of 5-HT following release. Although we have been aware for decades that SERT-mediated 5-HT clearance can be modulated by exogenous agents including serotonin-selective reuptake inhibitors, amphetamines, and cocaine, we have had little reason to speculate that SERT activity was actively controlled through endogenous pathways. Recent studies indicate that SERTs are likely to be trafficked to specific plasma membrane subdomains to achieve localized clearance of 5-HT, and that the numb

BACKGROUND: Induction or inhibition of drug transporting proteins by concomitantly administered drugs can cause serious drug-drug interactions (DDIs). However, in vitro assays currently available are mostly for studying the inhibitory potential of drugs on intestinal transporter proteins, rather than induction. Therefore, this study investigated the suitability of the frequently used intestinal Caco-2 cell line to predict transporter-mediated DDIs as caused by induction via activation of nuclear receptors.
METHODS: TaqMan® low density arrays and LC-MS/MS based targeted proteomics were used to
