OBJECTIVE: Several genome-wide association studies (GWAS) have associated variants in late-onset Alzheimer disease (LOAD) susceptibility genes; however, these single nucleotide polymorphisms (SNPs) have very modest effects, suggesting that single SNP approaches may be inadequate to identify genetic risks. An alternative approach is the use of multilocus genotype patterns (MLGPs) that combine SNPs at different susceptibility genes.
METHODS: Using data from 1,365 subjects in the National Institute on Aging Late-Onset Alzheimer's Disease Family Study, we conducted a family-based association stud

Transient global amnesia (TGA) is characterized by a profound but transient deficit of episodic memory. The study of cerebral blood flow and oxygen metabolism with positron emission tomography (PET) provides relevant pathophysiologic data, but only three patients have been reported so far and in only one was concomitant neuropsychological testing performed. We report here the concomitant neuropsychological and PET assessment of two additional patients. Episodic disturbance was characterized by a storage disturbance for one case and an incapacity to learn episodic associations in the other, ill

OBJECTIVES: Five-factor model (FFM) personality traits are associated with concurrent memory function and risk of incident dementia but are less consistently associated with the change in episodic memory. The present research analyzes multiple large-scale studies with a consistent analytic approach to evaluate the association between personality and change in episodic memory over time.
METHOD: Across nine public longitudinal data sets, 120,640 participants provided 471,821 memory assessments over up to 26 years (age range 18-108). FFM traits were tested as predictors of the average level (int

OBJECTIVE: Subtle decline in memory is thought to arise in the preclinical phase of Alzheimer's disease (AD). However, detecting these initial cognitive difficulties cross-sectionally has been challenging, and the exact nature of the decline is still debated. Accelerated long-term forgetting (ALF) has been recently suggested as one of the earliest and most sensitive indicators of memory dysfunction in subjects at risk of developing AD. The objective of this study was to design and validate the 1-week memory battery (1WMB) for assessing episodic memory and ALF in cognitively unimpaired individu

The amygdala is considered crucial to the formation of emotional episodic memories, yet causal evidence in humans is limited due to challenges in noninvasive neuromodulation of deep brain structures. In a double-blind, sham-controlled, repeated-measures study, we tested whether transcranial low-intensity focused ultrasound (tFUS) targeting the left amygdala before encoding emotional and neutral pictures impacted memory for these pictures 24 hr later (n = 23). Using a putative inhibitory tFUS protocol previously shown to attenuate amygdala BOLD-dependent signal, we hypothesized that preencoding

BackgroundMemory consolidation problems are often prototypical in Alzheimer's disease (AD). However, it remains undetermined whether episodic memory presents similarly in early-onset Alzheimer's disease (EOAD) relative to the more commonly occurring late-onset Alzheimer's disease (LOAD).ObjectiveThis study examined episodic memory and its neuroanatomical correlates in amnestic early-onset (aEOAD) relative to amnestic late-onset AD (aLOAD).MethodsUsing our single center data set obtained from a memory clinic setting (N = 180), we examined group differences in multiple markers of episodic memory

INTRODUCTION: In an increasingly aging society, testing hippocampal-dependent cognition in a quick and low resource manner will be crucial in: assessing the potential benefits of lifestyle choices and interventions affecting cognitive ageing (such as those involving exercise, diet, and sleep); detecting pathological aging, such as in Alzheimer's disease, where hippocampal degeneration occurs relatively early on.
METHODS: Over 300 participants aged 18-89 completed three cognitive tests, namely the Addenbrooke's Cognitive Examination-III (ACE-III), The Four Mountains Task (4MT), and a new task

Episodic memory, the ability to rapidly learn and explicitly remember past events and experiences, plays a critical role in children's academic learning and knowledge acquisition. The formation of lasting memories relies on the brain's ability to dynamically organize its activity. How these neural configurations unfold moment-by-moment across encoding and offline phases remains poorly understood. To probe these dynamics, we applied a novel Bayesian Switching Dynamic Systems approach, a hidden Markov model with automatic state detection, to fMRI data from children performing scene encoding foll

BACKGROUND: Intraneuronal accumulation of hyperphosphorylated tau is a hallmark of Alzheimer's disease (AD). Given the significant correlation between tau pathology and memory loss in AD patients, identifying vulnerable brain regions, particularly susceptible neuron types in these regions, will advance our understanding of AD onset and shed light on therapeutic strategies to manage its progression.
METHODS: Immunofluorescent staining was employed to identify the brain regions and neuron types vulnerable to tau pathology in AD. A combination of chemogenetics, electrophysiological recording, in

We reviewed the performance of global episodic amnesic patient H.M. Although he was affected by severe anterograde and retrograde amnesia (i.e. global amnesia), he occasionally showed some "islands" of residual intact memory. Therefore, we also searched for the presence of islands of memory in other global amnesic patients with the aim of comparing their performance with that of H.M. We sustain that islands of memory might be guided by residual brain structures and memory mechanisms that are not affected by lesions causing global episodic amnesia. Finally, we considered some possible cues for

BACKGROUND: Cognitive disorders associated with addictive disorders are well established in the literature for numerous substances and behaviours. Very few studies have examined the effect of polydrug use on cognitive functioning. These studies have focused on the cognitive effect of one substance among others in very small samples. They did not take into account HCV/HIV infections, which are highly prevalent among polydrug users due to frequent injections and sharing of materials and whose impact on cognition is well documented. In our study, we aim to investigate two critical cognitive funct

The advanced evolution of the human cerebral cortex forms the basis for our high-level cognitive functions. Through a comparative analysis of single-nucleus transcriptome data from the human neocortex and that of chimpanzees, macaques, and marmosets, we discovered 20 subgroups of cell types unique to the human brain, which include 11 types of excitatory neurons. Many of these human-unique cell clusters exhibit significant overexpression of genes regulated by human-specific enhancers. Notably, these specific cell clusters also express genes associated with disease risk, particularly those relat

INTRODUCTION: Arranging Pictures is a new episodic memory test based on the NIH Toolbox (NIHTB) Picture Sequence Memory measure and optimized for self-administration on a personal smartphone within the Mobile Toolbox (MTB). We describe evidence from three distinct validation studies.
METHOD: In Study 1, 92 participants self-administered Arranging Pictures on study-provided smartphones in the lab and were administered external measures of similar and dissimilar constructs by trained examiners to assess validity under controlled circumstances. In Study 2, 1,021 participants completed the extern

BACKGROUND: Schizophrenia is associated with impairments in verbal episodic memory. Strategy for Semantic Association Memory (SESAME) training represents a promising cognitive remediation program to improve verbal episodic memory. Virtual reality (VR) may be a novel tool to increase the ecological validity and transfer of learned skills of traditional cognitive remediation programs. The present proof-of-concept study aimed to assess the feasibility, acceptability, and preliminary efficacy of a VR-based cognitive remediation module inspired by SESAME principles to improve the use of verbal epis

OBJECTIVE: Allelic variation in the brain-derived neurotrophic factor (BDNF) Val66Met polymorphism has been shown to moderate rates of cognitive decline in preclinical sporadic Alzheimer's disease (AD; i.e., Aβ + older adults), and pre-symptomatic autosomal dominant Alzheimer's disease (ADAD). In ADAD, Met66 was also associated with greater increases in CSF levels of total-tau (t-tau) and phosphorylated tau (p-tau181). This study sought to determine the extent to which BDNF Val66Met is associated with changes in episodic memory and CSF t-tau and p-tau181 in Aβ + older adults in early-stage spo

It is unclear whether metacognitive judgments are made on the basis of domain-generality or domain-specificity. In the current study, we compared both behavioral and event-related potential (ERP) correlates of retrospective (retrospective confidence judgments: RCJs), and prospective (feeling of knowing: FOK) metacognitive judgments during episodic and semantic memory tasks in 82 participants. Behavioral results indicated that FOK judgments reflect a domain-specific process, while RCJ reflect a domain-general process. RCJ and FOK judgments produced similar ERP waveforms within the memory tasks,

It is clear evidence that individuals diagnosed with obsessive-compulsive disorder (OCD) lack confidence in their memory and have low metamemory performance (judgment and accuracy). However, it is still unclear whether low metamemory performance is specific to first, domain general or domain specific, and second, to stimulus domain. To address these issues, we compared individuals diagnosed with OCD and healthy controls (HCs) on recognition, retrospective (judgments of learning [JOL]) and prospective (feeling of knowing [FOK]) metamemory judgments and under three different episodic memory task

It is increasingly being recognized that new declarative, consciously accessible information can be learned in anterograde amnesia, but it is not clear whether this learning is supported by episodic or semantic memory. We report a case of a 55-year-old man who experienced severe amnesia after limited damage to the medial temporal lobe following neurosurgical complications. His general cognitive performance and knowledge of new French words and public events that occurred before and after the onset of amnesia were assessed. Performance remained satisfactory on post-morbid vocabulary and public

Episodic memory requires associating items with temporal context, a process for which the medial temporal lobe (MTL) is critical. This study uses recordings from 27 human subjects who were undergoing surgical intervention for intractable epilepsy. These same data were also utilized in Umbach et al. (2020). We identify 103 memory-sensitive neurons in the hippocampus and entorhinal cortex, whose firing rates predicted successful episodic memory encoding as subjects performed a verbal free recall task. These neurons exhibit important properties. First, as predicted from the temporal context model

Repetitive transcranial magnetic stimulation (rTMS) of the inferior parietal cortex (IPC) increases resting-state functional connectivity (rsFC) of the hippocampus with the precuneus and other posterior cortical areas and causes proportional improvement of episodic memory. The anatomical pathway(s) responsible for the propagation of these effects from the IPC is unknown and may not be direct. In order to assess the relative contributions of candidate pathways from the IPC to the MTL via the parahippocampal cortex and precuneus, to the effects of rTMS on rsFC and memory improvement, we used dif
