Integration of carbon and nitrogen signalling: A central framework for plant metabolism and development.

Carbon (C) and nitrogen (N) availability must be continuously integrated to sustain plant growth and development, yet the molecular mechanisms underlying this integration have long remained elusive. In this review, we highlight emerging evidence that C and N signaling converge on a regulatory network involving SnRK1, NLP7, TOR, and potentially nitric oxide. The clearest conceptual advance is the demonstration that SnRK1 directly phosphorylates the nitrate sensor and transcriptional regulator NLP7, providing an integration node through which energy limitation and C deficiency can suppress the p

Pan-species genome-wide evolution of the RGF peptide gene family and its impact on adventitious root regeneration in apple.

Root meristem growth factors (RGFs) are pivotal regulators of root growth in plants and are critically involved in the regulation of root development in fruit trees. In this study, we constructed a comprehensive small secreted peptide library from 64 plant species. Using a structure-based screening strategy, we identified a total of 495 RGF genes. Phylogenetic analysis revealed that the modern RGF gene family originated in ferns and exhibited an evolutionary pattern of initial contraction and subsequent expansion, consistent with the evolutionary emergence and diversification of plant root sys

Chimeric mutations in grapevine ENHANCED DISEASE RESISTANCE1 improve resistance to powdery mildew without growth penalty.

Grapevine (Vitis vinifera L.) incurs severe quality degradation and yield loss from powdery mildew, a major fungal disease caused by Erysiphe necator. ENHANCED DISEASE RESISTANCE1 (EDR1), a Raf-like mitogen-activated protein kinase kinase kinase, negatively regulates defense responses against powdery mildew in Arabidopsis (Arabidopsis thaliana). However, little is known about the role of the putatively orthologous EDR1 gene in grapevine. In this study, we obtained grapevine VviEDR1-edited lines using CRISPR/Cas9. Plantlets containing homozygous and bi-allelic indels in VviEDR1 developed leaf l

Genome editing beyond transgenes: DNA-free technologies in crop improvement.

Conventional and transgenic approaches to crop improvement are increasingly constrained by biosafety concerns, stringent regulatory frameworks, and limited public acceptance, necessitating new precision breeding methods. DNA-free genome editing using the CRISPR/Cas9 approach has emerged as a transformative approach that enables precise, targeted genetic modifications without the stable integration of a foreign gene (transgene). This strategy transiently introduces CRISPR/Cas9 ribonucleoprotein (RNP) complexes, or in vitro transcribed mRNA, into plant cells. This review focuses primarily on CRI

Pairwise inoculation of plant growth-promoting rhizobacteria unveils predominant strain-specific effects in root growth and hormone-responsive promoter activity in Arabidopsis thaliana.

Plant growth-promoting rhizobacteria are promising tools for agricultural management. We previously identified Achromobacter sp. 5B1, Bacillus sp., Micrococcus luteus, and Pseudomonas brassicae strains that influence the growth of Arabidopsis seedlings in highly contrasting manners. Here, pairwise combinations of these four bacterial isolates were tested to identify single or additive traits influenced by cocultivation with Arabidopsis seedlings in vitro. Root architectural modifications, such as primary root growth and lateral root formation, as well as root and shoot biomass production were

AGAMOUS Coordinates Flower Development Beyond Organ Identity: AGAMOUS Coordinates Flower Development in Space and Time.

AGAMOUS (AG) is a C-class Mcm1, Agamous, Deficiens, and Serum response factor (MADS)-box transcription factor classically defined by its roles in specifying stamen and carpel identity and enforcing floral meristem determinacy. Accumulating evidence now indicates that AG also coordinates a series of late developmental programs whose outputs are rewired over time based on tissue context, chromatin state, and phytohormone-mediated inter-organ communication. In the gynoecium, AG promotes regional patterning and tissue differentiation by regulating downstream target genes, thereby supporting ovule-

Identification of circRNAs in industrial hemp (Cannabis sativa L.) and prediction of regulatory networks underlying female differentiation.

INTRODUCTION: Circular RNAs (circRNAs) are important regulators of plant development and stress responses, but their roles in 6-benzylaminopurine (6-BA)-induced female flower differentiation in industrial hemp remain unclear. METHODS: Industrial hemp plants were treated with 60 mg·L-1 6-BA, and female flower samples were collected at different developmental stages. Genome-wide high-throughput sequencing was used to identify circRNAs. Selected circRNAs and miRNAs were validated by qRT-PCR. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were performed, and putative circRNA-m

Breaking the bloom: Exploring the genetic pathways in crop plants from primordia formation to seed development.

Genetic regulation of fertility and sterility is a key determinant of reproductive success, seed formation, and hybrid breeding efficiency in crop plants. Modern genetics has encouraged us to analyze the genetic and molecular pathways of plant crops and discuss the respective roles of genes. The concept of modern genetics is so vast, and it has opened new doors in scientific research, like the introduction and analysis of several genes that direct plant developmental processes toward either fertility or sterility. Normal plant development depends on coordinated genetic regulation of reproducti

Dynamic Transcriptomic Networks Underlying Early Bolting in Non-Heading Chinese Cabbage.

Bolting time is a pivotal agronomic trait that determines the yield and commercial quality of Brassica rapa ssp. chinensis var. utilis. To investigate the molecular basis of early bolting, an early-bolting line 'm662' and a late-bolting line 't151' were used in this study. Phenotypic evaluation combined with shoot apical meristem (SAM) observation showed that 10 days of low-temperature vernalization markedly accelerated bolting in 't151'. Subsequently, SAM samples from 'm662', non-vernalized 't151', and 10-day vernalized 'V10-t151' were collected at five developmental stages (7, 10, 13, 16, an

Magnetic Resonance Imaging-Based Monitoring of the Accumulation of Polyethylene Terephthalate Nanoplastics.

Polyethylene terephthalate (PET) is one of the most produced plastic materials in the world. The emergence of microplastics and nanoplastics (MPs/NPs) as a significant environmental contaminant has become a matter of increasing concern. While the toxicological effects of PET NPs have been widely researched, there is a lack of methodologies for studying their accumulation. The present study introduces a novel method to monitor the distribution of PET NPs in germinating wheat (Triticum aestivum L.) seeds. This involves the functionalization of superparamagnetic iron oxide nanoparticles (SPIONs)

Common garden experiment reveals altered nutritional values and DNA methylation profiles in micropropagated three elite Ghanaian sweet potato genotypes.

Micronutrient deficiency is the cause of multiple diseases in developing countries. Staple crop biofortification is an efficient means to combat such deficiencies in the diets of local consumers. Biofortified lines of sweet potato (Ipomoea batata L. Lam) with enhanced beta-carotene content have been developed in Ghana to alleviate Vitamin A Deficiency. These genotypes are propagated using meristem micropropagation to ensure the generation of virus-free propagules. In vitro culture exposes micropropagated plants to conditions that can lead to the accumulation of somaclonal variation with the po

Mediation of Flower Induction by Gibberellin and its Inhibitor Paclobutrazol: mRNA and miRNA Integration Comprises Complex Regulatory Cross-Talk in Apple.

Guaranteeing successful flowering is very important in economic plant species, especially apple (Malus domestica Borkh.), which is difficult to induce to flower. However, the gene expression and networks involved in flowering have not been totally characterized. Here, we employed mRNA and microRNA (miRNA) sequencing to understand the different responses to gibberellin- and its inhibitor paclobutrazol- (PAC) mediated flower induction. Significant opposite cytological and morphological changes were observed in treated terminal buds, which led to a reduced flowering rate under gibberellin and an

Identification of O-acetylserine(thiol)lyase (OASTL) genes in sorghum (Sorghum bicolor) and gene expression analysis under cadmium stress.

Cysteine (Cys) is the first identified molecule in plant metabolism which includes both sulfur and nitrogen. It can be synthesized in three cellular compartments, containing chloroplast, cytoplasm and mitochondrion. The final step of cysteine biosynthesis is catalyzed by the O-acetylserine(thiol)lyase enzyme (OASTL, E.C. 4.2.99). In the present study, seven members of the OASTL gene family in the sorghum (Sorghum bicolor) genome were identified at a genome-wide scale and comparative bioinformatics analyses were performed between sorghum and Arabidopsis OASTLs. In all OASTL proteins, a pyridoxa

Spatial genetic structure reflects extensive clonality, low genotypic diversity and habitat fragmentation in Grevillea renwickiana (Proteaceae), a rare, sterile shrub from south-eastern Australia.

BACKGROUND AND AIMS: The association of clonality, polyploidy and reduced fecundity has been identified as an extinction risk for clonal plants. Compromised sexual reproduction limits both their ability to adapt to new conditions and their capacity to disperse to more favourable environments. Grevillea renwickiana is a prostrate, putatively sterile shrub reliant on asexual reproduction. Dispersal is most likely limited by the rate of clonal expansion via rhizomes. The nine localized populations constituting this species provide an opportunity to examine the extent of clonality and spatial geno

Integration of Multidimensional Challenges by Roots in the Life of a Plant.

Roots of vascular plants continuously explore the soil environment and optimize their growth behavior by stimulating or suppressing local growth depending on the spatially diverse environmental conditions of the rooting substrate. The developmental toolbox of roots comprises activity control of apical meristems, initiation and growth of root hairs, lateral and adventitious root formation and cambial growth and, cum grano salis, is well understood. The underlying molecular mechanisms have been described in some detail, in particular with respect to single environmental variables. In a converse

Comprehensive Profiling of the Rice OsEPF/EPFL Gene Family Under Biotic and Abiotic Stresses Reveals the Involvement of OsEPF2 in Disease Resistance.

Members of the EPIDERMAL PATTERNING FACTOR (EPF) and EPF-Like (EPFL) families perform diverse regulatory functions in plant tissue morphogenesis, controlling the development of stomata, awns, shoot apical meristems (SAMs), and inflorescences. Nevertheless, the biological functions of OsEPF/EPFL family members in mediating responses to biotic/abiotic stresses are not yet widely characterized. Here, we demonstrated abundant cis-acting elements in the putative promoters of OsEPF/EPFL genes, including those associated with dehydration-, MeJA-, MYB binding site for drought, stress-, and ABA-respons

Developmental stages and episode-specific regulatory genes in andromonoecious melon flower development.

BACKGROUND AND AIMS: Given the lack of specific studies on floral development in melon (Cucumis melo L.), we carried out an extensive study involving morphological and transcriptomic analyses to characterize floral development in this species. METHODS: Using an andromonoecious line, we analysed the development of floral buds in male and hermaphrodite flowers with both light microscopy and scanning electron microscopy. Based on flower lengths, we established a correlation between the developmental stages and four main episodes of floral development and conducted an extensive RNA sequencing ana

An LTR retrotransposon insertion inside CsERECTA for an LRR receptor-like serine/threonine-protein kinase results in compact (cp) plant architecture in cucumber.

The compact (cp) phenotype in cucumber (Cucumis sativus L.) is an important plant architecture-related trait with a great potential for cucumber improvement. In this study, we conducted map-based cloning of the cp locus, identified and functionally characterized the candidate gene. Comparative microscopic analysis suggested that the short internode in the cp mutant is due to fewer cell numbers. Fine genetic mapping delimited cp into an 8.8-kb region on chromosome 4 harboring only one gene, CsERECTA (CsER) that encodes a leucine-rich repeat receptor-like kinase. A 5.5-kb insertion of a long ter
