Metabolomics Predicts Neuroimaging Characteristics of Transient Ischemic Attack Patients.

BACKGROUND: Neuroimaging is essential for the diagnosis and prognosis of transient ischemic attack (TIA). The discovery of a plasmatic biomarker related to neuroimaging findings is of enormous interest because, despite its relevance, magnetic resonance diffusion weighted imaging (DWI) is not always available in all hospitals that attend to TIA patients. METHODS: Metabolomic analyses were performed by liquid chromatography coupled to mass spectrometry in order to establish the metabolomic patterns of positive DWI, DWI patterns and acute ischemic lesion volumes. We used these methods with an in

Characterization and Discrimination of Ancient Grains: A Metabolomics Approach.

Hulled, or ancient, wheats were the earliest domesticated wheats by mankind and the ancestors of current wheats. Their cultivation drastically decreased during the 1960s; however, the increasing demand for a healthy and equilibrated diet led to rediscovering these grains. Our aim was to use a non-targeted metabolomic approach to discriminate and characterize similarities and differences between ancient Triticum varieties. For this purpose, 77 hulled wheat samples from three different varieties were collected: Garfagnana T. turgidum var. dicoccum L. (emmer), ID331 T. monococcum L. (einkorn) and

Targeted Metabolomics for Homocysteine-Related Metabolites in Primary Hepatocytes.

Liquid chromatography-tandem mass spectrometry has become the most convenient method to identify and quantify low molecular weight metabolites from various sources. Metabolomics studies of hepatocytes hold promise for the identification of the mechanisms of toxicant-related disease processes. In this chapter, we present a rapid and sensitive liquid chromatography-tandem mass spectrometry method for the quantification of intracellular concentrations of nine homocysteine-based metabolites, namely homocysteine, methionine, cysteine, dimethylglycine, cystathionine, S-adenosylmethionine, S-adenosyl

Ultrasound: a subexploited tool for sample preparation in metabolomics.

Metabolomics, one of the most recently emerged "omics", has taken advantage of ultrasound (US) to improve sample preparation (SP) steps. The metabolomics-US assisted SP step binomial has experienced a dissimilar development that has depended on the area (vegetal or animal) and the SP step. Thus, vegetal metabolomics and US assisted leaching has received the greater attention (encompassing subdisciplines such as metallomics, xenometabolomics and, mainly, lipidomics), but also liquid-liquid extraction and (bio)chemical reactions in metabolomics have taken advantage of US energy. Also clinical an

Identification of metabolites with anticancer properties by computational metabolomics.

BACKGROUND: Certain endogenous metabolites can influence the rate of cancer cell growth. For example, diacylglycerol, ceramides and sphingosine, NAD+ and arginine exert this effect by acting as signaling molecules, while carrying out other important cellular functions. Metabolites can also be involved in the control of cell proliferation by directly regulating gene expression in ways that are signaling pathway-independent, e.g. by direct activation of transcription factors or by inducing epigenetic processes. The fact that metabolites can affect the cancer process on so many levels suggests th

Untargeted serum metabolomics and air pollution in Parkinson's disease.

Mechanisms linking air pollution to Parkinson's disease (PD) remain poorly understood despite consistent toxicological and epidemiological evidence. We investigated metabolic disturbances associated with air pollution in PD using untargeted metabolomics. Serum samples were collected from 616 PD patients and 271 controls residing in central California, a region with high air pollution. Exposures to particulate matter (PM2.5), carbon monoxide, and nitrogen dioxide were estimated for the 1-, 3-, and 5-year periods prior to serum collection using residential addresses and three modeling approaches

Metabolomics Methods for Detecting Acetaminophen Poisoning: A Systematic Review.

INTRODUCTION: Detecting toxicity patterns in metabolic changes is a key indicator of potential threats. Metabolomics may provide an alternative way to find biomarkers for drug overdose and toxicity. In an acetaminophen overdose, glutathione scavenging causes mitochondrial malfunction and hepatocyte death. Metabolomics methods have enabled the discovery of novel mechanisms of liver injury in acetaminophen (APAP) overdose. METHODS: We conducted a peer-reviewed scientific review to assess the studies on the application of metabolomics in APAP poisoning. We searched PubMed and Scopus databases un

Integrated genomics and metabolomics survey of steroids from myxobacteria.

Steroids from myxobacteria remain largely underexplored compared to other natural product classes, leaving a significant gap in our understanding of their biosynthetic diversity, evolutionary trajectories, and pharmacological potential. To address this, we conducted a systematic genomic survey of myxobacterial genomes to delineate the distribution and phylogenetic context of sterol biosynthetic genes, which we found to be organized in a dispersed manner across the chromosome rather than clustered into canonical biosynthetic gene clusters. While the sterol biosynthetic machinery is broadly dist

Longitudinal untargeted metabolomics of cognitive impairment in Parkinson's disease.

INTRODUCTION: Cognitive impairment is a common non-motor symptom of Parkinson's disease (PD), yet the mechanism remains unclear. METHODS: Longitudinal serum samples from PD patients in a community-based case-control study were profiled, with cognition assessed by the Mini-Mental State Examination at baseline and follow-up. We performed three metabolome-wide association studies: a baseline cross-sectional analysis (n = 632), a time-to-event analysis (n = 412), and a repeated measures analysis (n = 265), followed by "Mummichog" pathway analysis. RESULTS: Of the 4762 metabolomic features, four

Antibiotic resistance of Pseudomonas aeruginosa strains-intracellular metabolomics analysis.

OBJECTIVES: Pseudomonas aeruginosa (Pa) stands as a dangerous multidrug-resistant human pathogen, and the associated antibiotic resistance mechanisms, governed by diverse molecular processes, present a persistent challenge in eradicating bacterial infections, especially in patients with cystic fibrosis. The search for novel antibiotic resistance markers has led researchers to explore the field of metabolomics. This study aims to discern alterations in metabolite levels resulting from varied antibiotic resistance profiles in Pa strains isolated from clinical patients. METHODS: In-depth analysi

Metabolomics in Mild Traumatic Brain Injury: A Scoping Review.

Mild traumatic brain injury (mTBI) accounts for around 80-90% of TBI cases, is increasing in incidence, and represents a major public health concern. Despite its high incidence, mTBI lacks reliable molecular biomarkers for diagnosis or prognosis. Metabolomics, the profiling of small-molecule metabolites (≤1 kDa), has emerged as a promising approach to characterize acute neurometabolic perturbations in mTBI and identify candidate biomarkers. However, metabolomic changes in mTBI have not yet been comprehensively characterized. Following the Preferred Reporting Items for Systematic Reviews and Me

Nontargeted plasma metabolomics associated with sow lifetime productivity traits.

The current study was conducted to characterize metabolomic plasma profiles among sows differing in their lifetime born alive and lifetime weaned after four parities. Plasma samples were collected at harvest between 12 and 15 d (luteal phase) following their fourth parity post-weaning estrus from 120 dams with consistent born alive and weaned at every farrowing event. Categories were derived as follows for average lifetime born alive (ba): High (H; 61ba), Mid (M; 50ba), and Low (L; 39ba) and raised (wn): High (H; 50wn) and Low (L; 34wn) generating 6 categories with 20 dams in each: HH, HL, MH,

Choice of cardioplegia influences metabolomics of human cardiac tissue.

Cardioplegia is often used prior to acquisition of human cardiac tissue to minimize warm ischemia time, which can severely confound studies of cardiac metabolism. However, there are several choices of cardioplegia solutions, and whether these solutions differentially impact tissue metabolism or metabolomic studies is not known. Here, we perform untargeted metabolomics, using both liquid chromatography-mass spectrometry and gas chromatography-mass spectrometry, on a large cohort of hearts transplanted for cardiomyopathy or from gift-of-life donors, and who have received different cardioplegia s

BACKGROUND: Hemorrhagic shock (HS) is a major risk factor for mortality and complications after severe trauma. Yet, complex mechanisms on a cellular and physiological level are still not fully understood, Metabolomics offers a powerful tool to unravel these complex biochemical alterations following HS and trauma (T). However, no systematic synthesis of metabolomic findings in in standardized translational animal models of HS and HS + T exists to date. OBJECTIVES: To systematically review and compare metabolomic alterations in animal models of isolated hemorrhagic shock and hemorrhagic shock wi

Characterization of trimethylamine metabolic pathways using pseudo-targeted metabolomics.

Trimethylamine (TMA) metabolism comprises choline-containing compounds' metabolization, TMA production and trimethylamine N-oxide (TMAO) generation. However, the presence of numerous compounds in the carnitine and phosphatidylcholine (PC) pool compositions complicates profiling work significantly. This study is aimed at developing an efficient method for profiling TMA metabolic pathways, including quantifying known compounds and semi-quantifying the differential metabolites in the carnitine and PC pool compositions. Pseudo-targeted metabolomics is applicable for characterization. Firstly, mult
