Molecular Epidemiology of Human Metapneumovirus in East Japan before and after COVID-19, 2017-2022.

Human metapneumovirus (hMPV) is genetically classified into two major subgroups, A and B, based on attachment glycoprotein (G protein) gene sequences. The A2 subgroup is further separated into three subdivisions, A2a, A2b (A2b1), and A2c (A2b2). Subgroup A2c viruses carrying 180- or 111-nucleotide duplications in the G gene (A2c 180nt-dup or A2c 111nt-dup ) have been reported in Japan and Spain. The coronavirus disease 2019 (COVID-19) pandemic disrupted the epidemiological kinetics of other respiratory viruses, including hMPV. In this study, we analyzed the sequences of hMPV isolates in Tokyo

First report of Avian metapneumovirus type B in Iraqi broiler flocks with swollen head syndrome.

BACKGROUND AND AIM: Swollen head syndrome (SHS) is a complex disease caused by various agents, including bacterial and viral pathogens, as well as environmental factors. Avian metapneumovirus (aMPV) is one of the most important causes of respiratory diseases and SHS in poultry and one of the most widespread viruses worldwide; however, it has not been recorded in Iraq. This study aimed at the molecular identification and subtyping of aMPV in poultry, with the objectives of investigating the prevalence of aMPV in infected broiler flocks with SHS and molecular typing using primers specific to the

Small Animal Models for Human Metapneumovirus: Cotton Rat is More Permissive than Hamster and Mouse.

Human metapneumovirus (hMPV) is the second most prevalent causative agent of pediatric respiratory infections worldwide. Currently, there are no vaccines or antiviral drugs against this virus. One of the major hurdles in hMPV research is the difficulty to identify a robust small animal model to accurately evaluate the efficacy and safety of vaccines and therapeutics. In this study, we compared the replication and pathogenesis of hMPV in BALB/c mice, Syrian golden hamsters, and cotton rats. It was found that BALB/c mice are not permissive for hMPV infection despite the use of a high dose (6.5 l

Epidemiological and phylogenic study of human metapneumovirus infections during three consecutive outbreaks in Normandy, France.

Human metapneumovirus (hMPV) is responsible for respiratory tract disease, particularly in the young and elderly population. An epidemiological and phylogenic study was performed on children admitted to hospital with an acute lower respiratory tract infection (LRI). Data were obtained and analyzed over three consecutive winters, from 2002-2003 to 2004-2005. Each year during the winter period, from November to March, 2,415 nasal swabs were tested by a direct immunofluorescence assay (DFA) for influenza viruses A and B, respiratory syncytial virus, parainfluenza viruses, and adenoviruses. A novel antiviral strategy targeting human metapneumovirus through pH modulation in human airway epithelial cells.

Human metapneumovirus (hMPV) is a major cause of respiratory infections, particularly in vulnerable populations, yet no approved vaccines or targeted antivirals are available. pH-regulated processes, including airway epithelial physiology, endosomal acidification and viral fusion mediated by the fusion (F) protein, are critical for hMPV infection. This study evaluates PHOH-001, an inhaled alkaline buffer, as a potential therapeutic strategy to modulate airway pH and inhibit hMPV infection. Using rhMPV-GFP (green fluorescent protein-expressing hMPV), viral replication was assessed in primary hu

Live human metapneumovirus vaccine candidates attenuated by temperature sensitivity mutations from human respiratory syncytial virus.

UNLABELLED: Human metapneumovirus (HMPV, genus Metapneumovirus) and respiratory syncytial virus (HRSV, genus Orthopneumovirus) are pediatric respiratory pathogens of the Pneumoviridae family. We previously developed live-intranasal HRSV vaccine candidates attenuated by genetically stabilized temperature sensitivity/attenuating (ts/att) mutations in the HRSV polymerase (L) ORF, including sites "1030s" (L codon changes S1313(AGC-to-TCA)/Y1321K(AAA), named after clone number 1030), and "Δ1313/I1314L," comprising a genetically stabilized deletion of L codon 1313. Unlocking antiviral potentials of traditional plants: A multi-method computational study against human metapneumovirus (HMPV).

BACKGROUND: Human metapneumovirus (HMPV) is a major cause of respiratory infections, especially in vulnerable populations. The absence of targeted antiviral therapies necessitates the exploration of novel drug candidates. Traditional medicinal plants offer a reservoir of bioactive compounds with potential antiviral properties. This study employs a multi-method computational approach to assess the antiviral potential of phytochemicals against HMPV. METHODS: A comprehensive in silico framework was employed, including virtual screening, molecular docking, molecular dynamics (MD) simulations, den

Rational design of uncleaved prefusion-closed trimer vaccines for human respiratory syncytial virus and metapneumovirus.

Respiratory syncytial virus (RSV) and human metapneumovirus (hMPV) cause human respiratory diseases and are major targets for vaccine development. In this study, we design uncleaved prefusion-closed (UFC) trimers for the fusion protein (F) of both viruses by examining mutations critical to F metastability. For RSV, we assess four previous prefusion F designs, including the first and second generations of DS-Cav1, SC-TM, and 847A. We then identify key mutations that can maintain prefusion F in a native-like, closed trimeric form (up to 76%) without introducing any interprotomer disulfide bond.

Phylogenetic analysis of human metapneumovirus among children with acute respiratory infections in Kuala Lumpur, Malaysia.

Human metapneumovirus (HMPV) is a recently discovered cause of viral respiratory infections. We describe clinical and molecular epidemiology of HMPV cases diagnosed in children with respiratory infection at University of Malaya Medical Centre, Kuala Lumpur, Malaysia. The prevalence rate of HMPV between 2010 and 2012 was 1.1%, and HMPV contributed 6.5% of confirmed viral respiratory infections. The HMPV patients had a median age of 1.6 years, and a median hospital admission of 4 days. The most common clinical presentations were fever, rhinitis, pneumonia, vomiting/diarrhoea, and bronchiolitis.

Human Metapneumovirus (HMPV): Advances in Diagnosis, Molecular Epidemiology, and Clinical Impact of an Underrecognized Respiratory Virus.

Human metapneumovirus or HMPV is an important respiratory pathogen of public health significance that primarily affects the immunocompromised, the very old, and young infants. However, recent studies have long since dispelled the idea that healthy adults are not at risk for serious sequelae, though it seems that HMPV has a particular affinity to infect children rather than adults. HMPV was first identified in 2001 and is implicated in a range of respiratory illnesses, from less severe upper respiratory infections to more severe pneumonia. This review compiles the recent literature on the epide

Repositioning antiviral phytoconstituents as broad-spectrum inhibitors of influenza A neuraminidase and human metapneumovirus fusion protein.

Viral respiratory infections still exist as a major global health concern. Influenza A viruses and human metapneumoviruses are among the main pathogens responsible for infections that can progress to severe acute respiratory forms. Despite the availability of certain therapeutic options, the need for effective treatment remains an issue. The exploration of innovative approaches based on medicinal plants, capable of simultaneously targeting several viruses, represents a promising strategic option for limiting the viral activity of these pathogens. In this work, an integrated computational workf

Human Metapneumovirus: an emerging respiratory pathogen and the urgent need for improved Diagnostics, surveillance, and vaccine development.

Human Metapneumovirus (HMPV) is a re-emerging respiratory pathogen causing significant morbidity and mortality, particularly among young children, the elderly, and immunocompromised individuals. First identified in 2001, HMPV has since been recognised as a leading cause of acute respiratory tract infections (ARTIs) worldwide. Its transmission occurs through droplets, direct contact, and surface contamination, with crowded spaces and healthcare facilities serving as key environmental amplifiers. HMPV's clinical manifestations, ranging from mild cold-like symptoms to severe pneumonia, often over
