Detection of amphetamine and methamphetamine following administration of benzphetamine.

Interpretation of urine drug-testing results is a challenging endeavor for several reasons. Effects of pH, dilution, legitimate and illicit sources of the drugs, and, perhaps the most challenging, the possibility of the methamphetamine and/or amphetamine being the result of the use of some other drug. Although it is known that 14 different compounds are metabolized to methamphetamine or amphetamine or both, there is little information on the metabolic profile of many of these compounds, making interpretation of results difficult. Benzphetamine, administered as a single Didrex tablet, was given

Integrating treatments for methamphetamine abuse: a psychosocial perspective.

The recent West Coast experience with increased methamphetamine use is showing signs of spreading to other parts of the US. The risk of corresponding medical and psychosocial problems has led to a call to action at the highest levels of government. The next few years will likely witness a substantial increase in treatment research on methamphetamine abuse/dependence, with particular emphasis on the development and application of novel pharmacotherapies. The evaluation of these agents presupposes that we understand the clinical syndrome resulting from chronic methamphetamine use. To establish a

Noradrenergic activity and spontaneous recurrence of methamphetamine psychosis.

The process that triggers flashbacks due to previous methamphetamine (MAP) psychosis was studied in 28 flashbackers, along with 84 non-flashbackers with a history of MAP psychosis. We measured plasma monoamine metabolite levels in 12 of the flashbackers and eight of the non-flashbackers, along with 28 normal controls. Most flashbackers had undergone frightening experiences during previous MAP use. The dominant triggering factor was a mild fear of other people. Plasma norepinephrine (NE) levels in the 12 flashbackers were increased during flashbacks. Thus, MAP use associated with threatening ex

[Methamphetamine-associated pulmonary arterial hypertension: a case report].

Here we reported a 41-year-old male patient who presented with shortness of breath after activity. He had a history of methamphetamine snorting, and echocardiography showed a generalized hypokinesia of the right ventricular wall. Right-heart catheterization suggested precapillary pulmonary arterial hypertension, with the possibility of pulmonary hypertension caused by left heart diseases. After receiving a subcutaneous infusion of treprostinil, his condition improved, and his exercise tolerance was better than before.

Mechanisms underlying methamphetamine-induced disruption of testicular integrity.

Methamphetamine (METH) abuse is a global health concern with serious repercussions, including potential disruption of the male reproductive system. Testicular integrity is particularly vulnerable to the toxic effects of METH. This review aims to comprehensively examine the current literature to elucidate the key mechanisms by which METH induces testicular damage, focusing on the cellular and molecular pathways involved. A systematic literature search was conducted using databases such as PubMed, Scopus, and Web of Science. Keywords included "methamphetamine," "testicular toxicity," "reproducti

Addressing Methamphetamine Use in Primary Care: Provider Perspectives.

INTRODUCTION: The opioid epidemic has evolved into a combined stimulant epidemic, with escalating stimulant and fentanyl-related overdose deaths. Primary care providers are on the frontlines grappling with patients' methamphetamine use. Although effective models exist for treating opioid use disorder in primary care, little is known about current clinical practices for methamphetamine use. METHODS: Six semistructured group interviews were conducted with 38 primary care providers. Interviews focused on provider perceptions of patients with methamphetamine use problems and their care. Data were

Natural Products in Modulating Methamphetamine-Induced Neuronal Apoptosis.

Methamphetamine (METH), an amphetamine-type psychostimulant, is highly abused worldwide. Chronic abuse of METH causes neurodegenerative changes in central dopaminergic neurons with numerous neuropsychiatric consequences. Neuronal apoptosis plays a critical role in METH-induced neurotoxicity and may provide promising pharmacological targets for preventing and treating METH addiction. In recent years, accumulating evidence has revealed that natural products may possess significant potentials to inhibit METH-evoked neuronal apoptosis. In this review, we summarized and analyzed the improvement eff

Multiple Biventricular Thrombi Associated With Methamphetamine-Associated Cardiomyopathy.

Background: As methamphetamine use has increased around the world, cardiovascular mortality has also increased. Methamphetamine-associated cardiomyopathy (MACM) is one of the serious cardiovascular complications of methamphetamine use. Limited evidence has been published regarding the increased risk of thrombogenicity in the setting of methamphetamine use. We propose that increased thrombogenicity presents a risk factor for intracardiac thrombi. Case Report: A 48-year-old female with a history of MACM was admitted to the hospital with acute decompensated heart failure. Transthoracic echocardio

Spontaneous Atraumatic Subdural Hematoma Related to Methamphetamine Use.

There are multiple risk factors associated with spontaneous subdural hematoma (SDH), including substance abuse, hypertension, vascular abnormalities, and neoplasms. The illicit drugs typically cited as risk factors for spontaneous SDH are alcohol and cocaine. We report a rare case of spontaneous, significant SDH associated exclusively with methamphetamine. Although it is unclear whether the underlying pathophysiology involves vasculitis, sympathomimetic-induced hypertension, or a combination of both, this case further illustrates the risks of methamphetamine abuse.

Methamphetamine-related postmortem cases in Jeddah, Saudi Arabia.

A more than 500% increase in the number of deaths involving methamphetamine occurred between 2016 and 2018 in Jeddah, Saudi Arabia. As such, this report employed a validated liquid chromatography tandem mass spectrometry method to quantify methamphetamine and its metabolites in bodily fluids from 47 postmortem cases in which methamphetamine was involved. The mean age of the deceased was 33 years old (median: 30, range: 16-63), and 94% were male. Methamphetamine was co-ingested with another drug in 32 of the cases (68%); however, the deaths were only due to the combined toxicity of methamphetam

Neurocognitive decision-making processes of casual methamphetamine users.

Neuroadaptations caused by chronic methamphetamine (MA) use are likely major contributors to high relapse rate following treatment. Thus, focusing intervention efforts at pre-empting addiction in vulnerable populations, thereby preventing MA-use-induced neurological changes that make recovery so challenging, may prove more effective than targeting chronic users. One approach is studying casual/recreational users, not diagnosed with substance use disorder. This group may be at high risk for addiction due to their experience with MA. On the other hand, they may be resilient against addiction sin

Adenosine 2A receptors modulate reward behaviours for methamphetamine.

Addiction to methamphetamine (METH) is a global health problem for which there are no approved pharmacotherapies. The adenosine 2A (A2 A ) receptor presents a potential therapeutic target for METH abuse due to its modulatory effects on striatal dopamine and glutamate transmission. Notably, A2 A receptor signalling has been implicated in the rewarding effects of alcohol, cocaine and opiates; yet, the role of this receptor in METH consumption and seeking is essentially unknown. Therefore, the current study used A2 A knockout (KO) mice to assess the role of A2 A in behaviours relevant to METH add

Mirtazapine alters cue-associated methamphetamine seeking in rats.

BACKGROUND: Methamphetamine (METH) is a potent psychostimulant, repeated use of which can result in a substance abuse disorder. Withdrawn individuals are highly prone to relapse, which may be driven, at least in part, by a hyperresponsivity to METH-associated cues that can prompt METH-seeking. Clinically efficacious pharmacotherapies for METH abuse are critically needed. Mirtazapine (Remeron) is an atypical antidepressant that antagonizes activated norepinephrine(α)₂, histamine₁ serotonin (5-HT)₂(A/C), and 5-HT₃ receptors. This pharmacologic profile prompted our interest in its potential for p

Methamphetamine causes sustained depression in cerebral blood flow.

The use prevalence of the highly addictive psychostimulant methamphetamine (MA) has been steadily increasing over the past decade. MA abuse has been associated with both transient and permanent alterations in cerebral blood flow (CBF), hemorrhage, cerebrovascular accidents and death. To understand MA-induced changes in CBF, we exposed C56BL/6 mice to an acute bolus of MA (5mg/kg MA, delivered IP). This elicited a biphasic CBF response, characterized by an initial transient increase (~ 5 minutes) followed by a prolonged decrease (~ 30 minutes) of approximately 25% relative to baseline CBF--as m

Methamphetamine use among gay men across the UK.

BACKGROUND: Concerns about gay men's methamphetamine use reflect its physical and psychological harms, and association with sexual-risk behaviour and HIV infection. Existing research suggests gay men's use in the UK is low and may be largely confined to London. METHODS: Cross-sectional survey of convenience sample of UK gay men. RESULTS: Methamphetamine was used by a significant minority of gay men in London but also in other regions. Overall, use was highest among those men aged 30-49, HIV-positive and reporting sexual-risk behaviours. CONCLUSION: Methamphetamine was used by a significant

Differences between Ecstasy-using and nonusing methamphetamine users.

There have been recent alarming increases in Ecstasy use and growing evidence of possible long-term neurological and cognitive effects. This article considers Ecstasy use within its common polydrug context (specifically with methamphetamine [MA]), examining differences between Ecstasy-using and nonusing subgroups of clients treated for MA use, and exploring the relationship of Ecstasy use to selected treatment outcomes. Multivariate logistic regression showed Ecstasy+MA users differing from MA users who have not used Ecstasy primarily in terms of sociodemographics (higher income, fewer childre

Recurrent corneal ulcerations associated with smokeable methamphetamine abuse.

PURPOSE: We studied a case of chronic, recurrent, bilateral, corneal ulcerations associated with smokeable methamphetamine abuse, commonly known as "ice," in an otherwise healthy 31-year-old woman. METHODS: Every few months the patient had recurrent corneal ulcerations. Each time, she was hospitalized and treated successfully with topical antibiotics. RESULTS: Even though she had undergone numerous formal attempts at drug rehabilitation, she continued to have relapses, and ulceration recurred only during periods of smokeable methamphetamine abuse. CONCLUSION: Illicit use of smokeable metham

The effects of acute oral naltrexone pretreatment on the abuse potential of intranasal methamphetamine, and the relationship between reward/punishment sensitivity and methamphetamine's effects.

One potential medication for treating methamphetamine use disorder is the opioid antagonist naltrexone (NLTX). Despite encouraging preclinical findings, the results of clinical studies have been mixed. The primary aim of the current trial was to examine the effects of acute NLTX pretreatment on the subjective and reinforcing effects of intranasal methamphetamine. Nonmedical psychostimulant users completed outpatient testing sessions in which they received oral placebo (0 mg) or NLTX (50 mg) before intranasal methamphetamine (30 mg/70 kg). Primary outcome measures were peak positive subjective

Association between kratom (Mitragyna speciosa Korth.) use and methamphetamine craving in males who used methamphetamine with and without heroin use history.

Field and preclinical studies suggest that kratom (Mitragyna speciosa) may have therapeutic potential in moderating methamphetamine (METH) craving. However, there is a lack of clinical studies to affirm this claim. This study explored patterns and motivations of kratom use among people who used METH with and without a history of heroin use, and determined the associations of kratom use patterns with METH craving. A treatment sample of 403 male respondents who had used METH, with and without kratom and with and without a history of heroin use was recruited for this cross-sectional study. A semi

Stereoisomeric identification of norephedrine derived from methamphetamine or amphetamine: urinalysis results of 33 methamphetamine abusers and 1 amphetamine abuser in Japan.

Stereoisomeric identification of norephedrine (NE) derived from methamphetamine (MA) or amphetamine (AM) was investigated by SIM-GC/MS assay using the urine of 33 MA abusers and 1 AM abuser. The assay simultaneously identified TFA-derivatized MA and AM metabolites, including AM, p-hydroxyl-MA (p-HMA), and p-hydroxyl-AM (p-HAM). The analysis lasted approximately 43 min, with a signal-to-noise ratio of >or=3 and a detection limit of 50 ng/mL. Among 12 urine samples from different subjects, only the S (+) form of MA and its metabolites (AM, p-HMA, p-HAM) was detected, however, a (1R,2S)-(-)-NE st
