Biodegradable polymeric microsphere-based vaccines and their applications in infectious diseases.

Vaccination, which provides effective, safe infectious disease protection, is among the most important recent public health and immunological achievements. However, infectious disease remains the leading cause of death in developing countries because several vaccines require repeated administrations and children are often incompletely immunized. Microsphere-based systems, providing controlled release delivery, can obviate the need for repeat immunizations. Here, we review the function of sustained and pulsatile release of biodegradable polymeric microspheres in parenteral and mucosal single-do

Treatment of periodontitis by local administration of minocycline microspheres: a controlled trial.

BACKGROUND: Periodontitis is an inflammatory condition of tooth-supporting tissues that is usually treated by mechanical removal of plaque and microorganisms that adhere to teeth. This treatment, known as scaling and root planing, is not optimally effective. Adjunctive therapy with locally delivered antimicrobials has resulted in improved clinical outcomes such as probing depth reduction. This article reports on the efficacy and safety of locally administered microencapsulated minocycline. METHODS: Seven hundred forty-eight (748) patients with moderate to advanced periodontitis were enrolled

Serological detection of Helicobacter pylori by a flow microsphere immunofluorescence assay.

A flow cytometric immunofluorescence assay (FMIA) for the detection of immunoglobulin G antibodies to Helicobacter pylori was developed. A multicomponent antigen was prepared and used to coat carboxylated polystyrene microspheres for reaction with patient sera followed by fluorescein isothiocyanate-labelled goat anti-human immunoglobulin G. The reacted microspheres were collected with a flow cytometer, and fluorescence was quantitated relative to the cutoff value provided by pooled sera from patients in whom H. pylori could not be demonstrated by culture or histology. Serum samples from 28 H.

Drainage of microspheres and RBCs from the vitreous of aphakic and phakic eyes.

The movements of plastic microspheres (7 to 10 mu in diameter) and autologous RBCs labeled with sodium chromate Cr 51 from the vitreous to the anterior chamber were studied in 24 rabbit eyes. Rabbits were made aphakic in one eye by intracapsular lens extraction of lensectomy via the pars plana, the fellow eyes serving as phakic controls. In phakic eyes the intravitreally injected microspheres and RBCs were retained inside the vitreous space, not gaining access to the anterior chamber. In aphakic eyes microspheres and RBCs moved to the anterior chamber and were usually seen on the corneal endot

Injectable liposome-microsphere composite system for synergistic therapy of periodontitis.

Periodontitis is characterized by a self-sustaining pathophysiological cycle in which bacterial infection propagates destructive inflammation and oxidative stress, compromises tissue repair capacities, and facilitates recurrent infection. Current monotherapeutic strategies exhibit limited efficacy against this multifactorial pathology. Although resveratrol (RSV) exhibits significant anti-inflammatory and antioxidant activities, its clinical application is limited by poor solubility and suboptimal bioavailability. Herein, we fabricate a kind of injectable composite microspheres (RSV@Lipo@PMS) t

Imaging properties of microsphere-assisted microscopy under transmitted illumination.

Microsphere-assisted microscopy (MAM) is a label-free super-resolution imaging technique based on traditional optical microscopy, but the current research usually chooses reflected illumination (RI). In this work, the effects of microsphere size, refractive index, and immersion environment on the imaging properties of MAM in imaging a 300-nm-period grating sample under transmitted illumination (TI) are systematically investigated. The experimental results reveal that under TI, microspheres with a smaller size exhibit higher resolution, and microspheres with a higher refractive index show highe

Sequential drug-release microsphere system for scarless wound healing.

The dynamic imbalance in skin wound healing is a key factor of hypertrophic scar (HS) formation. Traditional treatments often target only a certain stage of the wound healing process and fail to comprehensively address the dynamic imbalance, thus making it difficult to achieve scarless wound healing. This study developed a chronologically adaptive methacrylylated gelatin (GelMA)-based microsphere delivery system (G/E/T) that achieves comprehensive intervention in wound healing process through precisely orchestrated sequential release of epigallocatechin gallate (EGCG) and triamcinolone acetoni

Converting evanescent waves into propagating waves by hyper-hemi-microsphere.

Hyper-hemi-microspheres (HHMS) have shown promise in enhancing super-resolution imaging when combined with conventional optical microscopy. To offer actionable guidance for optimizing HHMS and hold broad applicability in the field of super-resolution imaging, the mechanism underpinning the enhanced imaging facilitated by HHMS is revealed by deriving the conversion and transmission conditions for evanescent waves. This is achieved by elucidating the intricate interplay between evanescent wave conversion and factors including refractive index, thickness, and surroundings of HHMS. Using the finit

Development of a microsphere-based immunoassay for the serological detection of glanders in equids.

Burkholderia mallei is the etiologic agent of glanders, an infectious disease of solipeds, with renewed scientific interest due to its increasing incidence in different parts of the world. More rapid, sensitive and specific assays are required by laboratories for confirmatory testing of this disease. A microsphere-based immunoassay consisting of beads coated with B. mallei recombinant proteins (BimA, GroEL, Hcp1, and TssB) has been developed for the serological diagnosis of glanders. The proteins' performance was compared with the OIE reference complement fixation test (CFT) and an indirect en

Microsphere controlled drug delivery for local control of tooth movement.

BACKGROUND: Because orthodontic tooth movement is dependent upon osteoclast-mediated resorption of alveolar bone adjacent to the pressure side of tooth roots, biologic mediators that regulate osteoclasts can be utilized to control tooth movement. OBJECTIVES: To develop a novel method to locally enhance orthodontic anchorage. METHODS: We encapsulated osteoprotegerin (OPG) in polymer microspheres and tested the effectiveness of microsphere encapsulated versus non-encapsulated OPG for enhancing orthodontic anchorage in a rodent model of tooth movement. A single injection of 1 mg/kg non-encapsul

Preparation and evaluation of tilmicosin microspheres and lung-targeting studies in rabbits.

Tilmicosin (TMS) is a macrolide used extensively for pulmonary infections in clinical veterinary medicine. However, TMS has frequent administration and short elimination half-life. Therefore, tilmicosin-gelatine microspheres (TMS-GMS) were prepared by an emulsion-chemical cross-linking technique as a sustained-release formulation to extend drug half-life. The particle size distribution, in-vitro sustained-release properties, stability, and physical characteristics, as well as pharmacokinetic (PK) characteristics, were evaluated in rabbits. TMS-GMS were spherical in shape and had a mean diamete

Design and characterization of chitosan-alginate microspheres for ocular delivery of azelastine.

The use of mucoadhesive biopolymers is one of the best approaches to prolong the drug residence inside the cul-de-sac, consequently increasing the bioavailability. Thus, the focus of this work was to develop mucoadhesive microspheres to overcome the limitations of ocular drug delivery. The chitosan-sodium alginate microspheres of azelastine hydrochloride were fabricated using modified ionotropic gelation technique. The particle size, zeta potential, entrapment efficiency and drug release kinetics were evaluated and characterized by SEM, FT-IR, DSC, in vitro mucoadhesion and in vivo study. The
