Comparative histopathological and immunological analysis of LRV2-positive and LRV2-negative Leishmania tropica in a BALB/c mouse cutaneous leishmaniasis model.

Leishmaniasis is a vector-borne disease endemic in more than 99 countries and manifests primarily as cutaneous, mucocutaneous, or visceral forms. This study aimed to assess the impact of Leishmania RNA virus 2 (LRV2) on immune responses and disease pathology in mice infected with Leishmania tropica. BALB/c mice were inoculated subcutaneously in the right hind footpad with L. tropica either lacking or carrying LRV2 (LRV2- and LRV2 + groups, respectively) and monitored for 24 weeks. Footpad and liver tissues were collected post-sacrifice, and parasite presence was determined using microscopy, cu

The immunological effects of intradermal particle-based vaccine delivery using a novel microinjection needle studied in a human skin explant model.

For intradermal (ID) immunisation, novel needle-based delivery systems have been proposed as a better alternative to the Mantoux method. However, the penetration depth of needles in the human skin and its effect on immune cells residing in the different layers of the skin has not been analyzed. A novel and user-friendly silicon microinjection needle (Bella-muTM) has been developed, which allows for a perpendicular injection due to its short needle length (1.4-1.8 mm) and ultrashort bevel. We aimed to characterize the performance of this microinjection needle in the context of the delivery of a

Early life stress, air pollution, inflammation, and disease: An integrative review and immunologic model of social-environmental adversity and lifespan health.

Socially disadvantaged individuals are at greater risk for simultaneously being exposed to adverse social and environmental conditions. Although the mechanisms underlying joint effects remain unclear, one hypothesis is that toxic social and environmental exposures have synergistic effects on inflammatory processes that underlie the development of chronic diseases, including cardiovascular disease, diabetes, depression, and certain types of cancer. In the present review, we examine how exposure to two risk factors that commonly occur with social disadvantage-early life stress and air pollution-

Modelling the immunological response to a tetravalent dengue vaccine from multiple phase-2 trials in Latin America and South East Asia.

BACKGROUND: The most advanced dengue vaccine candidate is a live-attenuated recombinant vaccine containing the four dengue viruses on the yellow fever vaccine backbone (CYD-TDV) developed by Sanofi Pasteur. Several analyses have been published on the safety and immunogenicity of the CYD-TDV vaccine from single trials but none modelled the heterogeneity observed in the antibody responses elicited by the vaccine. METHODS: We analyse the immunogenicity data collected in five phase-2 trials of the CYD-TDV vaccine. We provide a descriptive analysis of the aggregated datasets and fit the observed p

The behavioral and immunological effect of GM-1 ganglioside on nerve root regeneration following C5 nerve root avulsion in a rat model.

Preganglionic nerve root avulsion precludes sensory return, but motor regeneration is possible with sparing of motoneurons. The effect of GM-1 ganglioside treatment was studied with parallel evaluation of the autoimmune response. Rats (N=64) received injections of either GM-1 ganglioside or saline for 30 days following either C5 root avulsion or a hemilaminectomy control. The Bertelli grooming test assessed functional return. Before sacrifice at 5 months, serum was collected for enzyme-linked immunoabsorbent assay testing. Only 44% of the rats treated with ganglioside had a good functional out

Transplantation of neural stem cells in a rat model of stroke: assessment of short-term graft survival and acute host immunological response.

The use of progenitors and stem cells for neural grafting is promising, as these not only have the potential to be maintained in vitro until use, but may also prove less likely to evoke an immunogenic response in the host, when compared to primary (fetal) grafts. We investigated whether the short-term survival of a grafted conditionally immortalised murine neuroepithelial stem cell line (MHP36) (2 weeks post-implantation, 4 weeks post-ischaemia) is influenced by: (i) immunosuppression (cyclosporin A (CSA) vs. no CSA), (ii) the local (intact vs. lesioned hemisphere), or (iii) global (lesioned v

Exercise-induced extracellular vesicles delay tumor development by igniting inflammation in an immunologically cold triple-negative breast cancer mouse model.

BACKGROUND: Preclinical studies demonstrate that exercise reduces tumor incidence and growth. Rapid release of extracellular vesicles (EVs) during exercise suggests their potential role as mediators of exercise-induced systemic effects and physiological adaptation. This study investigated the impact of exercise-induced plasma EVs on tumor growth and immune tumor microenvironment in murine models of triple-negative breast cancer (TNBC): EO771 (a C57BL/6-derived TNBC cell line) and 4T1 (a BALB/c-derived TNBC cell line). METHODS: Size exclusion chromatography was used to isolate exercise-induced

Clinical and immunological parameters in patients infected with Cutaneous Leishmaniasis: Case control study- investigation of fractal fractional mathematical model in Lebesgue space.

Cutaneous Leishmaniasis (CL) is a skin disease that causes plaques, ulcers, and nodules on various body parts. The first aim of this study is to investigate the effect of CL on hematological and immunological markers. A total of 180 individuals (80 cases and 100 controls) examined. All patients received parasitological confirmation by a dermatologist and stained smears, and blood samples had been collected for the analysis of hematological parameters using a Coulter count machine. IL-17 and TGF levels were measured using the ELISA technique. A high rate of CL was observed among males, rural re

Early life exposure to broccoli sprouts confers stronger protection against enterocolitis development in an immunological mouse model of inflammatory bowel disease.

To our knowledge, IL-10-KO mice have not previously been used to investigate the interactions of host, microbiota, and broccoli, broccoli sprouts, or broccoli bioactives in resolving symptoms of CD. We showed that a diet containing 10% raw broccoli sprouts increased the plasma concentration of the anti-inflammatory compound sulforaphane and protected mice to varying degrees against disease symptoms, including weight loss or stagnation, fecal blood, and diarrhea. Younger mice responded more strongly to the diet, further reducing symptoms, as well as increased gut bacterial richness, increased b

Effects of artichoke extracts and probiotic on maternal immunological activation, valproic acid-induced oxidative stress, and gut leakiness in a rat model of autism.

Maternal health during pregnancy is a leading factor influencing offspring risk. This study investigated whether postnatal dietary supplementation could reduce oxidative stress and gut leakiness in a rat model of autism spectrum disorder (ASD). Male rat pups were prenatally exposed to valproic acid (VPA) or lipopolysaccharide (LPS) to induce ASD-like conditions. A total of 54 offspring of Wistar albino rats were divided into nine groups to evaluate various postnatal treatments, including an artichoke-based prebiotic (AR), probiotics (Pro), and omega-3 fatty acids (ω3). Early prediction of immunological non-responders in people living with HIV using machine learning: Model development and validation in multicenter cohorts in China.

BACKGROUND: Despite antiretroviral therapy (ART), 10-40 % of people living with HIV (PLWH) fail to normalize CD4+ T cells, known as immune non-responders (INR), associated with poor clinical outcomes. Due to the complex pathogenesis and the absence of effective treatments, early prediction and intervention of INR are critical. With the rapid advancements in artificial intelligence, particularly in machine learning (ML), developing an interpretable ML model to identify individuals at high risk of INR can facilitate personalized treatment strategies and improve clinical management. METHODS: Thi

Early life exposure to broccoli sprouts confers stronger protection against enterocolitis development in an immunological mouse model of inflammatory bowel disease.

Crohn's Disease (CD) is a presentation of Inflammatory Bowel Disease (IBD) that manifests in childhood and adolescence, and involves chronic and severe enterocolitis, immune and gut microbiome dysregulation, and other complications. Diet and gut-microbiota-produced metabolites are sources of anti-inflammatories which could ameliorate symptoms. However, questions remain on how IBD influences biogeographic patterns of microbial location and function in the gut, how early life transitional gut communities are affected by IBD and diet interventions, and how disruption to biogeography alters diseas

Development of a novel chimeric PA-LF antigen of Bacillus anthracis, its immunological characterization and evaluation as a future vaccine candidate in mouse model.

Tremendous efforts are being made to develop an anthrax vaccine with long term protection. The main component of traditional anthrax vaccine is protective antigen (PA) with the trace amount of other proteins and bacterial components. In this study, we developed a recombinant PA-LF chimera antigen of Bacillus anthracis by fusing the PA domain 2-4 with lethal factor (LF) domain 1 and evaluated its protective potential against B. anthracis in mouse model. The anti-PA-LF chimera serum reacted with both PA and LF antigen, individually. The chimera elicited a strong antibody titer in mice with predo

Immunological basis for protection in a murine model of tick-borne encephalitis by a recombinant adenovirus carrying the gene encoding the NS1 non-structural protein.

The humoral immune response to flaviviruses is mainly directed to the major envelope protein, E, and a glycosylated non-structural protein, NS1. Cell-mediated immune responses, however, appear to be directed mainly against non-structural proteins. Experiments described here show that a defective recombinant adenovirus (Rad51) containing the gene encoding the NS1 protein of tick-borne encephalitis virus can induce a strong protective immune response against several pathogenic tick-borne flaviviruses in an experimental animal model, and can enhance the efficacy of conventional vaccine preparatio
