Mechanical strain increases protein tyrosine phosphorylation in airway smooth muscle cells.

Mechanical stress contributes to normal structure and function of the lung as well as pathology in such diseases as bronchopulmonary dysplasia and adult respiratory distress syndrome. Stress-related increases in airway smooth muscle (ASM) quantity are reflected in vitro where cultured ASM cells respond to cyclic deformational strain with increased proliferation, cell reorientation, protein production, stress fibers, and focal adhesions. To understand the mechanisms of mechanical signaling in ASM cells, we investigated whether strain increased tyrosine phosphorylation of focal adhesion-related

Prostaglandins increase matrix metalloproteinase release from human ciliary smooth muscle cells.

PURPOSE: To identify matrix metalloproteinases (MMPs) released by ciliary smooth muscle cells in vitro and to determine whether MMP release is altered by exposure to prostaglandins (PGs). METHODS: Human ciliary smooth muscle cells were grown to confluence in monolayer cultures and treated with PGF2 alpha, 11-deoxy-PGE1, or PhXA85 (the nonesterified analogue of PhXA41) for 12 to 72 hours. The activity of MMP in the medium was assayed using gelatin and casein zymography. Identification of the specific MMP associated with each band was made by Western blot analysis. Band intensity, which reflect

Muscarinic effects on cellular functions in cultured human ciliary muscle cells.

PURPOSE: To characterize the pharmacology of the carbachol-induced changes of phospholipase C (PLC) activity and intracellular calcium concentration ([Ca2+]i) in cultured human ciliary muscle cells. METHODS: Changes in PLC activity of cultured human ciliary muscle cells were determined by production of inositol phosphates. Single-cell dynamic fluorescence ratio imaging was used to determine [Ca2+]i. RESULTS: Carbachol, oxotremorine-M, aceclidine, and pilocarpine stimulated PLC with mean EC50s of 20, 8, 17, and 2 microM, respectively. The effect of carbachol on PLC was partially suppressed by

Measurement of intracellular pH in isolated human detrusor smooth muscle cells.

Intracellular pH, pHi, was measured in isolated human detrusor smooth muscle cells by epifluorescence microscopy. The mean value was 7.07 +/- 0.19 when superfused with a CO2/HCO3- solution. Alteration of superfusate PCO2 also changed pHi whereas NaHCO3 changes were without effect, suggesting that the cell membrane is permeable to CO2 but impermeable to HCO3-. Nonbicarbonate H+ buffering power of the cell was calculated as 20.3 mequiv. x 1(-1) x pH unit-1. Addition of NH4Cl to the superfusate induced an alkalosis and, upon removal, a transient acidosis. The recovery from the acidosis allowed ca

Dexamethasone enhances active cation transport in cultured aortic smooth muscle cells.

Previous studies have indicated that ouabain-sensitive cation transport is increased in arteries obtained from rats with glucocorticoid-induced hypertension. However, it remained unclear whether this effect reflected direct glucocorticoid activation or rather depended on the elevated arterial pressure per se. In the present study, we examined the effect of dexamethasone on cultured rat aortic smooth muscle cells. Dexamethasone (10(-9) mol/L) increased ouabain-sensitive 86Rb uptake by about 50% (control: 16.5 +/- 1.6 pmol/mg protein/10 min; dexamethasone 10(-9) mol/L: 24.4 +/- 1.3 pmol/mg prote

Regulation of functional muscarinic receptor expression in tracheal smooth muscle cells.

Recent studies have shown that cultured tracheal smooth muscle cells (TSMCs) do not respond to muscarinic agonists with a significant increase in intracellular Ca2+ concentration. This may be due to a downregulation of muscarinic receptors (mAChRs) in TSMCs. We report here that the individual component of growth factors or hormones at the concentration used is not sufficient to stimulate growth of TSMCs in the primary culture with 1% fetal bovine serum (FBS). In the presence of 1% FBS, TSMCs withdraw from the cell cycle and express high levels of cell surface mAChRs. Furthermore, insulin-like

Methylxanthine treatment of smooth muscle cells differentially modulates adenylate cyclase responsiveness.

The DDT1 MF-2 smooth muscle cell line was used to study regulation of the A1 and A2 adenosine receptor (AR)-adenylate cyclase system by two different methylxanthines. 3-isobutyl 1-methylxanthine (IBMX) is both an AR antagonist and a phosphodiesterase inhibitor, while xanthine amine congener is an AR antagonist without phosphodiesterase activity. Incubation of cells for 18 hr with 100 microM IBMX produced a significant (P less than .05) decrease in the basal, isoproterenol- and sodium fluoride-stimulated adenylate cyclase activity. This generalized decrease in adenylate cyclase activity was ass

Measurement and simulation of noninactivating Ca current in smooth muscle cells.

An attempt was made to obtain electrophysiological evidence for continuous influx of Ca ion through voltage-dependent Ca channel (VDCC) in smooth muscle during long depolarization, for example in high K solution. Noninactivated Ca current [ICa(ni)] remaining after the accomplishment of voltage-dependent inactivation by prolonged depolarization for approximately 1 min was detected by three means under whole cell voltage clamp in several types of smooth muscle cells. The measurement of ICa(ni) was performed by micropuff application of Cd2+ or Ca2+ in the presence or absence of 5 mM extracellular

Membrane properties and innervation of smooth muscle cells in Hirschsprung's disease.

Mechanical and membrane properties of smooth muscle cells and/or neuroeffector transmission in the aganglionic segment of the large intestine (Hirschsprung's disease) were compared with findings in the ganglionic segment. Tension-recording, microelectrode, and double sucrose gap methods were used. There was no difference in resting membrane potential of the longitudinal or circular muscle cell in these two segments, which were obtained at biopsy in Japanese children. In the ganglionic preparations, generations of regular prepotentials, with or without the spike, correlated well to the rhythmic

Effect of stretch and contraction on caveolae of smooth muscle cells.

The number of caveolae present at the surface of smooth muscle cells of guinea-pig taenia coli and visualized by freeze-fracture is about 35 per micron2. (By comparison, endothelial cells of intramuscular capillaries have about 73 caveolae per micron2). The packing density of smooth muscle caveolae is not significantly different in muscle strips isotonically contracted with carbachol or stretched and relaxed in a calcium-free solution, under a range of loads varying from 1 to 15 g. Also the diameter of the fractured necks of the caveolae appears unchanged in all the experimental conditions tes

Studies on the ionic permeability of muscle cells and their models.

We studied the effect an alkali-metal ion exercised on the rate of entry of another alkali-metal ion into frog sartorius muscle cells and their models (i.e., ion exchange resin and sheep's wool). In the case of frog muscle, it was shown that the interaction fell into one of four categories; competition, facilitation, and two types of indifference. The observed pK value (4.6 to 4.7) of the surface anionic groups that combine with the alkali-metal ions suggests that they are beta- or gamma-carboxyl groups of proteins on the cell surface. The results were compared with four theoretical models whi

Engineering Functional Skeletal Muscle Tissues from Human Induced Pluripotent Stem Cells.

In vitro cell culture models of human skeletal muscle offer a valuable platform for studies of pathology and discovery of new therapeutics. However, traditional two-dimensional cell culture models suffer from limited durability and challenges with assessment of skeletal muscle contractile function. Furthermore, finite proliferation potential and scarce availability of patient-derived primary myoblasts limit experimental throughput when using these cells. Here, we describe a method for using human induced pluripotent stem cells (hiPSCs) to generate functional three-dimensional skeletal muscle t

PCSK9 expression in human atherosclerotic lesions and vascular smooth muscle cells.

AIMS: Proprotein convertase subtilisin/kexin type 9 (PCSK9) is the third gene involved in autosomal dominant hypercholesterolemia, an atherosclerosis major cause. Its role in regulating plasma cholesterol through hepatocyte functions has been widely reported, and anti-PCSK9 drugs have been developed. However, direct interactions between PCSK9 and arterial pathology are not fully elucidated. As vascular smooth muscle cells (vSMC) play a major role in arterial wall pathophysiology, we describe PCSK9's presence in human atherosclerotic lesions and its interaction with vSMC. METHODS: Healthy and

Quantitative Evaluation of Exon Skipping in Immortalized Muscle Cells in Vitro.

Exon skipping through the use of antisense oligonucleotides (AOs) is currently one of the most promising approaches for treating Duchenne muscular dystrophy (DMD). While we now have a number of AO drug candidates in clinical trials, we are still faced with issues of poor or controversial efficacy in some of these drugs. This is the case with eteplirsen, an exon 51-skipping AO that is the first and only FDA-approved drug for DMD to date. Effective procedures must, therefore, be set up for the in vitro screening of potential AOs for DMD treatment. Here, we describe one such procedure using immor

Defective autophagy in vascular smooth muscle cells promote uremic accelerated atherosclerosis.

Individuals with chronic kidney disease (CKD) exhibit a significantly elevated risk of premature mortality, predominantly attributable to cardiovascular etiologies. Atherosclerosis is a multifactorial vascular pathology driven by dysregulated lipid metabolism. Notably, patients with CKD exhibit an accelerated progression of atherosclerosis, a condition referred to as uremia-accelerated atherosclerosis (UAAS). Autophagy represents an evolutionarily conserved cellular process that plays a pivotal role in maintaining intracellular homeostasis across cardiovascular cell types. However, the involve

Causal role of immune cells in muscle atrophy: mendelian randomization study.

Immune system and inflammation had a great influence on the progression of muscle atrophy. However, the causal relationship with specific immune cell traits remained uncertain. The aim of this study was to elucidate the genetic influences on these associations, providing insights into the underlying mechanisms of muscle atrophy. A bidirectional two-sample Mendelian randomization (MR) analysis was conducted to investigate the causal relationship between immune cell phenotypes and muscle atrophy. Data on immune cell phenotypes were obtained from a research cohort containing data on 731 immune ce
