Antibody-drug conjugates for non-muscle invasive bladder cancer: current status and future development.

PURPOSE OF REVIEW: Antibody-drug conjugates (ADCs) have recently emerged as a promising targeted therapeutic strategy in urothelial carcinoma, and their application in nonmuscle invasive bladder cancer (NMIBC) is an evolving frontier. This review is timely as it synthesizes recent advances in ADC development for NMIBC, addressing critical unmet needs among both Bacillus Calmette-Guérin (BCG) naïve high-risk and BCG-unresponsive patients. RECENT FINDINGS: The literature reveals that ADCs that target HER2, Trop-2, Nectin-4 are under active investigation. Several retrospective and prospective st

FOXK2 in skeletal muscle development: a new pathogenic gene for congenital myopathy with ptosis.

Congenital ptosis, a genetic disorder involving levator palpebrae muscle dysfunction, is often associated with congenital myopathy. The genetic causes of this condition remain poorly understood. In this study, we identified FOXK2 mutations in five pedigrees with congenital myopathy and ptosis through whole exome sequencing and Sanger sequencing. Zebrafish with foxk2 deficiency exhibited underdeveloped skeletal muscles and reduced mobility, while mice with Foxk2 deletion in skeletal muscle stem cells (MuSCs) showed generalized skeletal muscle abnormalities. Further analysis revealed that FOXK2

Extensive Sensorimotor Training Predetermines Central Pain Changes During the Development of Prolonged Muscle Pain.

Repetitive movements (RM) are a main risk factor for musculoskeletal pain, which is partly explained by the overloading of musculoskeletal structures. However, RM may also drive brain plasticity, leading to maladaptive changes in sensorimotor areas and altered pain processing. This study aimed to understand whether individuals performing extensive RM (musicians) exhibit altered brain processing to prolonged experimental muscle pain. Nineteen healthy musicians and 20 healthy nontrained controls attended 3 sessions (Day 1-Day 3-Day 8). In each session, event-related potentials (ERPs) to non-noci

Inhibition of Activin/Myostatin signalling induces skeletal muscle hypertrophy but impairs mouse testicular development.

Numerous approaches are being developed to promote post-natal muscle growth based on attenuating Myostatin/Activin signalling for clinical uses such as the treatment neuromuscular diseases, cancer cachexia and sarcopenia. However there have been concerns about the effects of inhibiting Activin on tissues other than skeletal muscle. We intraperitoneally injected mice with the Activin ligand trap, sActRIIB, in young, adult and a progeric mouse model. Treatment at any stage in the life of the mouse rapidly increased muscle mass. However at all stages of life the treatment decreased the weights of

Tiny Regulators of Massive Tissue: MicroRNAs in Skeletal Muscle Development, Myopathies, and Cancer Cachexia.

Skeletal muscles are the largest tissues in our body and the physiological function of muscle is essential to every aspect of life. The regulation of development, homeostasis, and metabolism is critical for the proper functioning of skeletal muscle. Consequently, understanding the processes involved in the regulation of myogenesis is of great interest. Non-coding RNAs especially microRNAs (miRNAs) are important regulators of gene expression and function. MiRNAs are small (~22 nucleotides long) noncoding RNAs known to negatively regulate target gene expression post-transcriptionally and are abu

Loss of sonic hedgehog gene leads to muscle development disorder and megaesophagus in mice.

Sonic hedgehog ( Shh) is crucial for organogenesis in the foregut. This study investigated the function of Shh at the late-gestational stage; during which, the esophagus continues to differentiate. We established cytokeratin 14 ( CK14)-Cre;Shhfl/fl mice in which the down-regulation of Shh in the epithelium occurred at approximately the same time as esophageal muscle conversion. Hematoxylin and eosin and immunohistochemical staining, with antibodies against keratin 14, Shh, patched 1 (Ptch1), Gli1, proliferating cell nuclear antigen (PCNA), α-smooth muscle actin (αSMA), high-molecular-weight ca

Ephrin-A3 promotes and maintains slow muscle fiber identity during postnatal development and reinnervation.

Each adult mammalian skeletal muscle has a unique complement of fast and slow myofibers, reflecting patterns established during development and reinforced via their innervation by fast and slow motor neurons. Existing data support a model of postnatal "matching" whereby predetermined myofiber type identity promotes pruning of inappropriate motor axons, but no molecular mechanism has yet been identified. We present evidence that fiber type-specific repulsive interactions inhibit innervation of slow myofibers by fast motor axons during both postnatal maturation of the neuromuscular junction and

The Role of Skeletal Muscle in External Ear Development: A Mouse Model Histomorphometric Study.

BACKGROUND: Mechanical stimuli imparted by skeletal muscles play an important role during embryonic development in vertebrates. Little is known whether skeletal muscles are required for normal external ear development. METHODS: We used Myf5-/-:MyoD-/- (double-mutant) mouse embryos that completely lack skeletal musculature and analyzed the development of the external ear. We concentrated on the external ear because several studies have suggested a muscular cause to various congenital auricular deformities, and middle and inner ear development was previously reported using the same mouse model.

Decrease in muscle contraction time complements neural maturation in the development of dynamic manipulation.

Developmental improvements in dynamic manipulation abilities are typically attributed to neural maturation, such as myelination of corticospinal pathways, neuronal pruning, and synaptogenesis. However the contributions from changes in the peripheral motor system are less well understood. Here we investigated whether there are developmental changes in muscle activation-contraction dynamics and whether these changes contribute to improvements in dynamic manipulation in humans. We compared pinch strength, dynamic manipulation ability, and contraction time of the first dorsal interosseous muscle i

Development aggravates the severity of skeletal muscle catabolism induced by endotoxemia in neonatal pigs.

Accretion rates of muscle protein are elevated in normal neonates, but this anabolic drive decreases with maturation. As this change occurs, it is not known whether development also influences muscle protein catabolism induced by sepsis. We hypothesize that protein degradation in skeletal muscle induced by endotoxemia becomes more severe as the neonate develops. Fasted 7- and 26-day-old pigs were infused for 8 h with LPS (0 and 10 μg·kg(-1)·h(-1)), while plasma amino acids (AA), 3-methylhistidine (3-MH), and α-actin concentrations and muscle protein degradation signal activation were determine

Is ribosomal capacity a potential metabolic marker of muscle development? Measurement by muscular biopsy.

Metabolic markers of muscle metabolism could help geneticists and nutritionists predict the breast meat development of chickens. The aim of the current study was to test the ribosomal capacity (CS), a potential metabolic marker, and to evaluate a simple biopsy method on the pectoralis major muscle. Ribosomal capacity was measured in three commercial meat chicken genotypes differing in their growth rate. Fast-, medium-, and slow-growing male chickens were fed using three commercial dietary programs of increasing energy and protein concentration (nine treatments). Biopsy was performed at 4 wk of

Experimental development of an electrically stimulated biological skeletal muscle ventricle for chronic aortic counterpulsation.

OBJECTIVE: The chronic shortage of donor organs for cardiac transplantation and the high costs for mechanical assist devices demand the development of alternative cardiac assist devices for the treatment of severe heart failure. Cardiac assistance by stimulated skeletal muscles is currently investigated as such a possible alternative. The goal of the presented study was to construct a newly designed biological skeletal muscle ventricle and to evaluate its possible hemodynamic efficacy in an acute sheep model. METHODS: A total of 14 adult sheep were used for acute experiments. The entire thora

Humoral factors trophic for vascular smooth muscle during the development of hypertension in rats.

There is evidence that a humoral factor or factors in rats with one-kidney, one-clip (1K1C) hypertension increase growth of cultured vascular smooth muscle cells. Such humoral trophic factors may contribute to the abnormal growth of arterial muscle in hypertension. To further study the longitudinal expression of this trophic factor or factors, we prepared rats with 1K1C hypertension of different durations. To determine if the factor or factors are also expressed in other forms of experimental hypertension, we additionally prepared rats with two-kidney, one-clip (2K1C) hypertension and paired t

Effect of maternal alcohol consumption on pre- and post-natal muscle development of mice.

The effect of maternal alcohol consumption during pregnancy on muscle development of the offspring was investigated in 28 mice (14 males, 14 females) at 12 weeks of age. Both the control and alcohol-fed groups received food ad-libitum throughout the period of study. The control group received water ad-libitum also throughout the period of study. The alcohol-fed group, on the other hand, received only water up to six weeks of age. Subsequently, between 6 and 8 weeks of age they received 10% ethanol (v/v) in water and 20% ethanol (v/v) from 8 to 11 weeks age. Both groups were bred at 11 weeks of

Characteristics of Ca2+- and Mg2+-induced tension development in chemically skinned smooth muscle fibers.

Chemically skinned fibers from guinea pig taenia caecum were prepared by saponin treatment to study the smooth muscle contractile system in a state as close to the living state as posible. The skinned fibers showed tension development with an increase of Ca2+ in the solution, the threshold tension occurring as 5 X 10(-7) M Ca2+. The maximal tension induced with 10(-4) M Ca2+ was as large and rapid as the potassium-induced contracture in the intact fibers. The slope of the pCa tension curve was less steep than that of skeletal muscle fibers and shifted in the direction of lower pCa with an incr

Detection of Muscle Fiber Orientation During Human Tongue Development: Analysis Using Diffusion Tensor Imaging.

The tongue consists of intrinsic and extrinsic muscles that enable complex movements. Previous studies, based on gross and histological observations, were limited in their ability to fully distinguish between them, which may have resulted in inaccurate differentiation timelines for intrinsic and extrinsic muscles. This study analyzed tongue muscle fiber orientation during embryonic development using diffusion tensor imaging (DTI). Human embryonic and fetal specimens (n = 29) were used in this study. A 7-Tesla magnetic resonance imaging (MRI) system was used to acquire T1-weighted images and DT

Changes in gut microbiota affect DNA methylation levels and development of chicken muscle tissue.

The intestinal microbiome is essential in regulating host muscle growth and development. Antibiotic treatment is commonly used to model dysbiosis of the intestinal microbiota, yet limited research addresses the relationship between gut microbes and muscle growth in yellow-feathered broilers. In this study, Xinghua chickens were administered broad-spectrum antibiotics for eight weeks to induce gut microbiome suppression. We investigated the relationships between the gut microbiome and muscle growth using 16S rRNA sequencing and transcriptomic analysis. Results indicated that antibiotic treatmen

Development of 3D Muscle Cell Culture-Based Screening System for Metabolic Syndrome Drug Research.

Developing effective drug screening methods for type 2 diabetes requires physiologically relevant models. Traditional 2D cell cultures have limitations in replicating in vivo conditions, leading to challenges in assessing drug efficacy. To overcome these issues, we developed a 3D artificial muscle model that induces insulin resistance, a hallmark of type 2 diabetes. Using C2C12 myoblasts cultured in a scaffold of 1% alginate and 1 mg/mL collagen type 1, we optimized conditions for differentiation and structural stability. Insulin resistance was induced using palmitic acid (PA), and glucose upt

Rbm24-mediated post-transcriptional regulation of skeletal and cardiac muscle development, function and regeneration.

RNA-binding proteins are critically involved in the post-transcriptional control of gene expression during embryonic development and in adult life, contributing to regulating cell differentiation and maintaining tissue homeostasis. Compared to the relatively well documented functions of transcription factors, the regulatory roles of RNA-binding proteins in muscle development and function remain largely elusive. However, deficiency of many RNA-binding proteins has been associated with muscular defects, neuromuscular disorders and heart diseases, such as myotonic dystrophy, amyotrophic lateral s
