Heparan sulfate in skeletal development, growth, and pathology: the case of hereditary multiple exostoses.

Heparan sulfate (HS) is an essential component of cell surface and matrix-associated proteoglycans. Due to their sulfation patterns, the HS chains interact with numerous signaling proteins and regulate their distribution and activity on target cells. Many of these proteins, including bone morphogenetic protein family members, are expressed in the growth plate of developing skeletal elements, and several skeletal phenotypes are caused by mutations in those proteins as well as in HS-synthesizing and modifying enzymes. The disease we discuss here is hereditary multiple exostoses (HME), a disorder

Biological maturation of youth athletes: assessment and implications.

The search for talent is pervasive in youth sports. Selection/exclusion in many sports follows a maturity-related gradient largely during the interval of puberty and growth spurt. As such, there is emphasis on methods for assessing maturation. Commonly used methods for assessing status (skeletal age, secondary sex characteristics) and estimating timing (ages at peak height velocity (PHV) and menarche) in youth athletes and two relatively recent anthropometric (non-invasive) methods (status-percentage of predicted near adult height attained at observation, timing-predicted maturity offset/age a

Do infections play a role in the development of chronic inflammatory arthritis? A 14-year follow-up study of patients with early arthritis.

BACKGROUND: The role of preceding infections in the development of reactive arthritis (ReA) is well known but is less studied in association with other inflammatory arthritides. Therefore, in 1979-80 we screened for infections in patients with early musculoskeletal symptoms who were referred for rheumatological consultation and assessed the role of infections and other clinical factors in the development of chronic disease in following 14 years. METHODS: A total of 104 consecutive patients with suspected inflammatory musculoskeletal symptoms with a duration < 6 months were examined and screen

Human Organ-on-a-Chip Microphysiological Systems to Model Musculoskeletal Pathologies and Accelerate Therapeutic Discovery.

Human Microphysiological Systems (hMPS), otherwise known as organ- and tissue-on-a-chip models, are an emerging technology with the potential to replace in vivo animal studies with in vitro models that emulate human physiology at basic levels. hMPS platforms are designed to overcome limitations of two-dimensional (2D) cell culture systems by mimicking 3D tissue organization and microenvironmental cues that are physiologically and clinically relevant. Unlike animal studies, hMPS models can be configured for high content or high throughput screening in preclinical drug development. Applications

Risk Factors for Musculoskeletal Symptoms Among Korean Broadcast Actors.

BACKGROUND: Musculoskeletal diseases (MSDs) are functional disabilities in the musculoskeletal area that occur when continuous damage to the muscles or tissues is caused by performing a repetitive task. These diseases are usually found in the waist, shoulder, neck, arm, and wrist. MSD is also referred to as cumulative trauma disorder, repetitive strain injury, occupational overuse syndrome, and visual display terminal, depending on the country. The condition is now commonly referred to as work-related musculoskeletal disorder. OBJECTIVES: The aim of this study was to develop a prevention plan

Multitechnological integration advances musculoskeletal regeneration: synergistic progress of organoids, 3D/4D bioprinting, single-cell omics and artificial intelligence.

Regenerative repair of injuries and degenerative diseases in the musculoskeletal system remains a paramount clinical challenge, as conventional single-modal technologies fail to recapitulate the intricate structural and functional complexity of native musculoskeletal tissues. In recent years, the rapid evolution of cutting-edge technologies-including organoids, 3D/4D bioprinting, single-cell omics and artificial intelligence (AI)-has unlocked novel opportunities for addressing this longstanding issue. This review aims to systematically elucidate how the integration of these four core technolog

Cell-cell junctions in developing and adult tendons.

Tendons connect muscles to bones to transfer the forces necessary for movement. Cell-cell junction proteins, cadherins and connexins, may play a role in tendon development and injury. In this review, we begin by highlighting current understanding of how cell-cell junctions may regulate embryonic tendon development and differentiation. We then examine cell-cell junctions in postnatal tendon, before summarizing the role of cadherins and connexins in adult tendons. More information exists regarding the role of cell-cell junctions in the formation and homeostasis of other musculoskeletal tissues,

Multivariate genetic analysis reveals three distinct pathological dimensions in musculoskeletal disorders.

The substantial overlap in symptoms and pathology across musculoskeletal disorders underscores the need to investigate their shared genetic mechanism. Using multivariate genetic modeling, we identify three distinct pathological factors, including a degenerative musculoskeletal disorder factor, a bone mass factor, and an autoimmune disorder factor, which collectively explain 48.27% of the total genetic variance in musculoskeletal disorders. Genome-wide association analyses of these factors identify 795 genomic risk loci, 136 of which are novel, along with 273 candidate genes, 20 of which intera

Gene expression analyses in malformed skeletal structures of gilthead sea bream (Sparus aurata).

The incidence of skeletal anomalies in reared fish has been translated for years in important economic losses for the aquaculture industry. In the present study, we have analysed the gene expression of extracellular matrix components and transcription factors involved in bone development in gilthead sea bream presenting different skeletal anomalies: lordosis (LD), lordosis-scoliosis-kyphosis (LSK) or opercular, dental or jaw malformations in comparison with control (CT) specimens. Results showed a possible link between the presence of LD and LSK and the significant downregulation of genes invo

Epidemiology of musculoskeletal disorders due to biomechanical overload.

The link between occupation and musculoskeletal disorders has been focused on in numerous research projects, ranging from those simply observing the different pathological findings reported among workers performing particular tasks, down to the latest studies actually quantifying the 'exposure' of workers to physical and psychosocial stimuli. For some disorders and certain tissues, it has been reported that specific types of work-related exposure are associated with the development of musculoskeletal pathologies, and that the relative risks for certain types of occupational exposure can be ext

Musculoskeletal senescence: a moving target ready to be eliminated.

Aging is the prime risk factor for the broad-based development of diseases. Frailty is a phenotypical hallmark of aging and is often used to assess whether the predicted benefits of a therapy outweigh the risks for older patients. Senescent cells form as a consequence of unresolved molecular damage and persistently secrete molecules that can impair tissue function. Recent evidence shows senescent cells can chronically interfere with stem cell function and drive aging of the musculoskeletal system. In addition, targeted apoptosis of senescent cells can restore tissue homeostasis in aged animals

Stronger Athlete Identity Is a Risk Factor for More Severe Depressive Symptoms After Musculoskeletal Injury in Pediatric Athletes: a Systematic Review.

PURPOSE OF REVIEW: Treatment for musculoskeletal sports injuries often neglects the psychological components of health and recovery. Pediatric patients require particular consideration of their psychosocial and cognitive development. This systematic review investigates the effects of musculoskeletal injury on mental health in pediatric athletes. RECENT FINDINGS: Athlete identity may increase in adolescence and is associated with worse mental health post-injury. Psychological models suggest loss of identity, uncertainty, and fear mediate the association between injury and symptoms of anxiety,

Protection Against Arthritis by the Parasitic Worm Product ES-62, and Its Drug-Like Small Molecule Analogues, Is Associated With Inhibition of Osteoclastogenesis.

The immunomodulatory actions of parasitic helminth excretory-secretory (ES) products that serendipitously protect against development of chronic inflammatory disorders are well established: however, knowledge of the interaction between ES products and the host musculoskeletal system in such diseases is limited. In this study, we have focused on ES-62, a glycoprotein secreted by the rodent filarial nematode Acanthocheilonema viteae that is immunomodulatory by virtue of covalently attached phosphorylcholine (PC) moieties, and also two synthetic drug-like PC-based small molecule analogues (SMAs)

MiR-214 is an important regulator of the musculoskeletal metabolism and disease.

MiR-214 belongs to a family of microRNA (small, highly conserved noncoding RNA molecules) precursors that play a pivotal role in biological functions, such as cellular function, tissue development, tissue homeostasis, and pathogenesis of diseases. Recently, miR-214 emerged as a critical regulator of musculoskeletal metabolism. Specifically, miR-214 can mediate skeletal muscle myogenesis and vascular smooth muscle cell proliferation, migration, and differentiation. MiR-214 also modulates osteoblast function by targeting specific molecular pathways and the expression of various osteoblast-relate

Krogh's principle for musculoskeletal physiology and pathology.

August Krogh was a comparative physiologist who used frogs, guinea pigs, cats, dogs, and horses in his research that led to his Nobel Prize on muscle physiology. His idea to choose the most relevant organism to study problems in physiology has become known as Krogh's principle. Indeed, many important discoveries in physiology have been made using naturally occurring animal models. However, the majority of research today utilizes laboratory mouse and rat models to study problems in physiology. This paper discusses how Krogh's principle can be invoked in musculoskeletal research as a complementa
