Evaluating physical and nutritional stress during mycelial growth as inducers of tolerance to heat and UV-B radiation in Metarhizium anisopliae conidia.

Elevated tolerance to UV-B radiation and heat may be induced in conidia produced on fungi exposed during mycelial growth to sublethal stresses other than heat or UV-B. This is due to a phenomenon referred to as 'cross-protection'. Several mechanisms are associated with this increased conidial tolerance, one of which is the accumulation of trehalose and mannitol within conidia. In the present study, conidia of the insect-pathogenic fungus Metarhizium anisopliae var. anisopliae were produced on mycelium subjected to nutritive, heat-shock, osmotic, or oxidative stress. The tolerance levels to UV-

Stimulatory effect of growth in the presence of alcohols on biotransformation of penicillin G into cephalosporin-type antibiotics by resting cells of Streptomyces clavuligerus NP1.

Growth of Streptomyces clavuligerus NP1 in the presence of methanol or ethanol resulted in a marked increase in production of cephalosporin(s) from penicillin G by resting cells. The mycelium produced in alcohol-supplemented medium was fragmented and dispersed as compared with growth in control medium. HPLC analysis showed that at least two products were present in the biotransformation supernatant fluid after 1 h incubation. One of them has been identified as deacetoxycephalosporin G (DAOG).

[Etiology of the first African case of systemic blastomycosis].

Several authors have suggested that african and north american forms of Blastomyces dermatitidis appeared separate taxa. Morphologic and pathogenic differences not only prevail, but also the failing to mate between them. African forms lose their mycelium-yeast conversion capability in old cultures. So I propose to create the complementary appellation Blastomyces dermatitidis tunisiens nov.subsp. from the african forms, in memory of the first case of systemic blastomycosis in this continent, discovered in Tunisia.

In Vitro Culture Systems to Study the Functions of Common Mycorrhizal Network in Arbuscular Mycorrhizal Symbioses.

In vitro culture systems provide a controlled platform for studying common mycorrhizal networks (CMNs) in arbuscular mycorrhizal fungi (AMF), overcoming the difficulties of pot- or field-based approaches. These systems make it possible to study with precision plant-to-plant communication through the exchange of nutrients and signaling compounds. CMNs also open up new avenues for exploring the hyphobiome by shedding light on the microbial communities living on the surface of the hyphae connecting plants. Here, we present standardized, compartmented systems establishing contaminant-free CMNs for

Development and growth of fruit bodies and crops of the button mushroom, Agaricus bisporus.

We studied the appearance of fruit body primordia, the growth of individual fruit bodies and the development of the consecutive flushes of the crop. Relative growth, measured as cap expansion, was not constant. It started extremely rapidly, and slowed down to an exponential rate with diameter doubling of 1.7 d until fruit bodies showed maturation by veil breaking. Initially many outgrowing primordia were arrested, indicating nutritional competition. After reaching 10 mm diameter, no growth arrest occurred; all growing individuals, whether relatively large or small, showed an exponential increa

Production and secretion of 5-n-alkylresorcinols by Fusarium culmorum.

Fusarium culmorum F1 was found to produce and secrete into the culture medium several of 5-n-alkylresorcinols. The amount of resorcinolic lipids was 5.3 microg/g and 0.9 microg/l in mycelium and in post-culture liquid, respectively. First of all F. culmorum F1 produces saturated homologues with C15 to C25 side chains. The extract from the medium contained only homologues with shorter carbon chains (C13 to C17).

An evaluation of delayed hypersensitivity in guinea pigs to various trichophytin preparations.

Guinea pigs immunized with mycelium from Trichophyton mentagrophytes in Freund's complete adjuvant were tested intradermally with the following trichophytin preparations: TrI extracted by ethylene glycol, TrII extracted by phenol and TrII, a commercially obtained preparation fromSächsisches Serumverk KG. In our experience, TrI was found to be superior to the other preparations. It showed signifiicant delayed skin reactions in all sensitized animals and none in the controls. Lymphocyte stimulation test with this preparation (TrI) gave positive results.

Unravelling the Potential of Fungal Division of Labour in the Laccase Producer Coriolopsis trogii MUT3379 Through Protoplast Formation and Regeneration.

The white-rot fungus Coriolopsis trogii MUT3379 produces Lac3379-1 laccase at high yields due to the previous development of a robust fermentation process. Throughout the extended use of this strain, we observed the occurrence of substrate-specific guaiacol and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) oxidizing enzymes other than Lac3379-1 Since we did not succeed in producing these enzymes in significant amounts by conventional strain selection and fermentation tools, we developed an approach based on protoplast preparation and regeneration to isolate stable producers of

The antigenic and catalytically active formamidase of Paracoccidioides brasiliensis: protein characterization, cDNA and gene cloning, heterologous expression and functional analysis of the recombinant protein.

Paracoccidioides brasiliensis is a well-characterized pathogen of humans. To identify proteins involved in the fungus-host interaction, P. brasiliensis yeast proteins were separated by liquid isoelectric focusing, and fractions were subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot analysis. Immunoreactive bands were detected with pooled sera of patients with P. brasiliensis infection. A protein species with a molecular mass of 45 kDa was subsequently purified to homogeneity by preparative gel electrophoresis. The amino acid sequence of four endoproteinase

Nocardia veterana sp. nov., isolated from human bronchial lavage.

A nocardioform bacterium was isolated from the bronchoscopic lavage of a 78-year-old man with a past history of tuberculous pleurisy, who presented with bilateral upper lobe lesions at Austin and Repatriation Medical Centre, Heidelberg, Australia. The strain was aerobic, Gram-positive, produced beige substrate mycelium and scant white aerial mycelium. It showed chemotaxonomic markers which were consistent with the classification of Nocardia: i.e. meso-diaminopimelic acid, N-glycolylmuramic acid, arabinose and galactose as diagnostic sugars; phospholipids phosphatidylinositol mannosides, phosph

The interface of mycology and endocrinology.

Message molecules such as hormones or pheromones have been described in non-pathogenic fungi and largely control mating. In one instance, a fungal hormone has a sequence homologous with a mammalian hormone, binds to a mammalian receptor and produces a functional response in a mammalian cell. Some mammalian hormones have also been shown to inhibit or stimulate pathogenic and non-pathogenic fungi, although until recently fungal binders (receptors) had not been demonstrated and the concentrations studied were not physiologic. We studied Paracoccidioides brasiliensis, for human sex hormone binding

The effect of 5,5-diethylbarbituric acid on the biosynthesis of anthracyclines in Streptomyces galilaeus.

5,5-Diethylbarbituric acid (barbital) stimulates the production of anthracycline antibiotics called galirubins in Streptomyces galilaeus in dependence on the strain, concentration and cultivation conditions. The stimulation is more pronounced (up to 300%) in the low-producing strain than in the production mutant. Under conditions of limited aeration the effect of barbital is increased in both strains. In the production strain barbital narrows the spectrum of metabolites produced. Higher barbital concentrations inhibit growth of the mycelium of both strains and decrease the formation of free an

The mutualistic fungi of the bark beetle Pityokteines vorontzowi are nutrient-rich and efficiently deplete their medium of fir chemical defenses.

Conifers are a challenging host for herbivores since their tissues are very low in essential nutrients but high in chemical defenses. For herbivorous insects, such as phloem-colonizing bark beetles, mutualistic fungi may improve their diet by providing a nutritious mycelium. A recent study revealed that two filamentous fungi are mutualists of the European fir engraver beetle Pityokteines vorontzowi, but a potential nutritional contribution of the fungi, as well as their capability to degrade plant antiherbivore defenses remains unknown. We analyzed the nutrient content of the fungal mutualists

Extracellular Biosynthesis, Characterization and Antimicrobial Activity of Silver Nanoparticles Synthesized by Filamentous Fungi.

The green synthesis of metal nanoparticles has received substantial attention due to their applications in various domains. The aim of the study was to obtain silver nanoparticles (AgNPs) by green synthesis with filamentous fungi, such as Cladosporium cladosporoides, Penicillium chrysogenum, and Purpureocillium lilacinum. Fungal species were grown on nutrient media and aqueous mycelium extracts were used to reduce Ag+ to Ag (0). The silver nanoparticles were analyzed by various techniques, such as UV-Visible spectroscopy (UV-Vis), scanning electron microscopy (SEM), Fourier transform infrared

X-ray fluorescence and XANES spectroscopy revealed diverse potassium chemistries and colocalization with phosphorus in the ectomycorrhizal fungus Paxillus ammoniavirescens.

Ectomycorrhizal (ECM) fungi play a major role in forest ecosystems and managed tree plantations. Particularly, they facilitate mineral weathering and nutrient transfer towards colonized roots. Among nutrients provided by these fungi, potassium (K) has been understudied compared to phosphorus (P) or nitrogen (N). The ECM fungus Paxillus ammoniavirescens is a generalist species that interacts with the root of many trees and can directly transfer K to them, including loblolly pine. However, the forms of K that ECM fungi can store is still unknown. Here, we used synchrotron potassium X-ray fluores

Antifungal Activity of Alpha-Mangostin against Colletotrichum gloeosporioides In Vitro and In Vivo.

We investigated alpha-mangostin (α-mangostin, α-MG), a xanthone natural product extracted from the pericarp of mangosteen (Garcinia mangostana), for its antifungal activities and possible mechanism against Colletotrichum gloeosporioides, which causes mango anthracnose. The results demonstrated that α-MG had a relatively high in vitro inhibitory activity against C. gloeosporioides among 20 plant pathogenic fungi. The median effective concentration (EC50) values of α-MG against mycelial growth were nearly 10 times higher than those of spore germination inhibition for both strains of C. gloeospor

[Morphogenesis of experimental primary pulmonary coccidioidal mycosis].

Morphogenesis of experimental primary pulmonary coccidioidomycosis in white mice was studied. The aspirated particles of the mycelium by the 5th day formed sporilating sporangiums, the latter being the main cause of a specific inflammatory process. Three stages in the development of the infectious process were singled out: incubation, in the course of which accumulation of the pathogene in the lungs took place (the first seven days), initiation and progressing of pneumonia, culminated with the animals' perish in the course of 12-14 days.

Constitutive transcriptomic reprogramming underlies differential resistance of Flammulina velutipes to Pseudomonas tolaasii.

Pseudomonas tolaasii, the causal agent of brown blotch disease, impairs not only fruiting body quality but also early mycelial development in Flammulina velutipes. However, the molecular basis underlying strain-dependent resistance at the vegetative stage remains poorly understood. In this study, we compared a resistant strain (S2595) and a susceptible strain (S2506) using dual-culture confrontation assays, scanning electron microscopy (SEM), and comparative transcriptome profiling with three biological replicates per condition. Under mock conditions, both strains exhibited comparable mycelial

Rhizopus arrhizus in Italy as the causative agent of primary cerebral zygomycosis in a drug addict.

A rare case of primary cerebral zygomycosis in an Italian drug addict is described. The diagnosis was based on the histologic detection of broad, aseptate, hyaline mycelium in fluid aspirated from a brain mass detected by computerized axial tomography. The zygomycete isolated from the clinical specimen was identified as Rhizopus arrhizus var arrhizus. The patient had no known predisposing condition that would have suppressed his immunological defenses. Once the diagnosis was established, treatment with amphotericin B was initiated, but the patient died during the first day of treatment.

[Pseudomycetomatous subcutaneous phaeohyphomycosis].

The authors report the first Nigerian case of cutaneous and sub-cutaneous phaeohyphomycosis. It was characterized by its original macroscopy: the lesion was pseudo-tumoral and provided with fistulae. So it looked like a mycetoma. The diagnosis was finally made by histology: it showed the typically pigmented mycelium. Cultures have not been made. Etiological agents of phaeohyphomycosis being opportunistic, this disease occurs often among immunosuppressed patients. The knowledge of unusual clinical forms may allow to suspect the diagnosis and to cultivate the exudate: this provides the only spec
