Despite being a preventable and treatable disease, tuberculosis (TB) remained the second leading infectious cause of death globally in 2022, surpassed only by COVID-19. The death rate from TB is influenced by numerous factors that include antibiotic drug resistance, noncompliance with chemotherapy by patients, concurrent infection with the human immunodeficiency virus, delayed diagnosis, varying effectiveness of the Bacille-Calmette-Guerin vaccine, and other factors. Even with the recent advances in our knowledge of Mycobacterium tuberculosis and the accessibility of advanced genomic tools suc

Effective treatment options for Mycobacterium abscessus (MAB) pulmonary diseases (PD) are limited due to inadequate drug efficacy, rising drug resistance, and genetic mutations. New compounds are urgently needed to treat MAB-PD. The MAB Enoyl Acyl Carrier Protein (ACP) Reductase InhA (MAB-InhA) plays a crucial role in mycobacterial cell death and mycolic acid (MA) biosynthesis, making it a potential drug target for new lead identification. The purpose of this study was to identify new potential inhibitors of MAB-InhA in MAB-PD by using structure-based virtual screening, docking, molecular mech

Previous investigations demonstrated three taxonomic groups among 22 clinical isolates of Mycobacterium smegmatis. These studies were expanded to 71 clinical isolates, of which 35 (49%) (group 1) were identical to five ATCC reference strains including the type strain ATCC 19420T. Twenty-eight isolates (39%) were group 2, and eight isolates (11%) were group 3. Isolates of groups 2 and 3 were most often associated with post-traumatic or post-surgical wound infections including osteomyelitis, were susceptible to sulfamethoxazole, amikacin, imipenem and the tetracyclines, variably resistant to cla

Bacillus Calmette-Guérin (BCG) Tokyo 172 is a predominant World Health Organization Reference Reagent for the BCG vaccine. Recently, the BCG Tokyo 172 substrain was reported to consist of two subpopulations with different colony morphologies, smooth and rough. Smooth colonies had a characteristic 22-bp deletion in Rv3405c of the region of difference (RD) 16 (type I), and rough colonies were complete in this region (type II). We hypothesized that the morphological difference is related to lipid phenotype and affects their antigenicity. We determined the lipid compositions and biosynthesis of ty

OBJECTIVE/BACKGROUND: Isoniazid (INH) is one of the effective antituberculosis (TB) drugs used for TB treatment. However, most of the drug-resistant Mycobacterium tuberculosis (MTB) clinical strains are resistant to INH, a first-line antituberculous drug. Certain metabolic enzymes such as adenosylhomocysteinase (Rv3248c), universal stress protein (Rv2623), nicotinamide adenine dinucleotide (reduced)-dependent enoyl-acyl carrier protein reductase (Rv1484), oxidoreductase (Rv2971), dihydrofolate reductase (Rv2763c), pyrroline-5-carboxylate dehydrogenase (Rv1187) have been identified to bind INH-

To address intracellular mycobacterial infections, we developed a cocktail of four enzymes that catalytically attack three layers of the mycobacterial envelope. This cocktail is delivered to macrophages, through a targeted liposome presented here as ENTX_001. Endolytix Cocktail 1 (EC1) leverages mycobacteriophage lysin enzymes LysA and LysB, while also including α-amylase and isoamylase for degradation of the mycobacterial envelope from outside of the cell. The LysA family of proteins from mycobacteriophages has been shown to cleave the peptidoglycan layer, whereas LysB is an esterase that hyd

INTRODUCTION: The increasing resistance to anti-TB drugs has become a global issue, highlighting an urgent need to investigate new drug candidates for combating Mycobacterium tuberculosis (M.tb.). Asiatic Acid (AA), a major phytoconstituent of Centella asiatica, exhibits anti-microbial and anti-biofilm activities against several microbes. In the present investigation, AA was explored for prospective anti-biofilm activity against mycobacteria.
METHODS: We first used a computational approach to probe the in silico inhibitory potential of AA against selected target proteins of the FAS-II pathway

Mycobacterium abscessus is a non-tuberculous Mycobacterium (NTM) that is often associated with respiratory infections in persons with cystic fibrosis. High levels of antibiotic resistance make M. abscessus infections difficult to treat, highlighting the urgent need for alternative therapeutic strategies. Mycobacteriophage therapy-treatment of infections with viruses that infect Mycobacterium hosts-shows considerable promise, but the narrow host specificity of the current mycobacteriophage repertoire limits this approach to personalized therapies. Here, we characterize the anti-M. abscessus act

Mycobacterium abscessus, a rapidly growing nontuberculous mycobacterium, causes chronic pulmonary infections that are difficult to treat due to extensive intrinsic drug resistance. Through high-content screening of 786 FDA-approved drugs against intracellular M. abscessus in human THP-1 macrophages, we identified naftifine, an antifungal allylamine, as a novel antimycobacterial agent with dual-acting therapeutic mechanisms. Naftifine demonstrated potent activity against reference strains and multidrug-resistant clinical isolates. It showed enhanced efficacy in intracellular environments compar

Mycobacterium leprae (M. leprae) is the bacterium that causes leprosy. It is a public health problem in many regions, especially in developing countries. The situation is getting worse and worse as drug resistance spreads. InhA is one of the most important proteins for M. leprae's survival. It helps make mycolic acid, an important part of the bacterial cell envelope; hence, InhA is a good target for developing new anti-leprosy drugs. In this study, we focused on identifying natural plant-derived compounds (phytochemicals) capable of inhibiting InhA function using sophisticated computer-based t

Mycobacterium abscessus is one of the leading causes of pulmonary infections caused by non-tuberculous mycobacteria. The ability of M. abscessus to establish a chronic infection in the lung relies on a series of adaptive mutations impacting, in part, global regulators and cell envelope biosynthetic enzymes. One of the genes under strong evolutionary pressure during host adaptation is ubiA, which participates in the elaboration of the arabinan domains of two major cell envelope polysaccharides: arabinogalactan (AG) and lipoarabinomannan (LAM). We here show that patient-derived UbiA mutations no

BACKGROUND: Tuberculosis (TB) is still a leading cause of death worldwide. Almost a third of the world's population is infected with TB bacilli, and each year approximately 8 million people develop active TB and 2 million die as a result. Today's TB treatment, which dates back to the 1970s, is long and burdensome, requiring at least 6 mo of multidrug chemotherapy. The situation is further compounded by the emergence of multidrug-resistant TB (MDR-TB) and by the infection's lethal synergy with HIV/AIDS. Global health and philanthropic organizations are now pleading for new drug interventions th

The epidemiology of tuberculosis has been altered due to the increased use of immunosuppressive therapies (chemotherapy and biologic agents), the human immunodeficiency virus (HIV) epidemic, and the changing migration patterns. Extrapulmonary tuberculosis occurs most often in individuals with HIV infection. Osteoarticular tuberculosis is rare in Western countries, with its diagnosis being performed through culture and/or molecular detection, however muscular tuberculosis without coexistent skeletal involvement is rare, with only 30 cases reported in the literature. The authors present the case

BACKGROUND: Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a global health challenge, particularly with the emergence of multidrug-resistant and extensively drug-resistant strains. Mycolic acid biosynthesis is essential for the integrity of Mtb's cell wall, offering promising therapeutic intervention targets. The thioesterase domain of polyketide synthase 13, Pks13-TE, is at the heart of this pathway and, therefore, an attractive drug target.
METHODS: 1228 natural compound libraries were computationally screened to identify the most potentially active Pks13-TE inhibito

This study reports the synthesis of a library comprising three series of 21 phosphonium salt hybrids (1a-1 g, 2a-2 g, 3a-3 g) generated via a click reaction yielding 1, 2, 3-triazoles with potential antitubercular activity. The phosphonium salts were synthesized through a 1, 3-dipolar cycloaddition (click) reaction. The structures of all synthesized compounds were elucidated using 1H NMR, 13C NMR, AT-FTIR, and HR-LCMS spectroscopy. The compounds were screened for antitubercular activity against Mycobacterium tuberculosis H37Rv and multidrug-resistant TB (MDR-TB) with Rifampicin and Isoniazid,

Bromoepiandrosterone (BEA), a synthetic analog of the adrenal steroid DHEA, holds promise as a host-directed therapy for both active and latent tuberculosis (TB). Unlike DHEA, BEA lacks hormonal side effects yet retains potent immunomodulatory activity. It promotes a Th1-skewed immune response by enhancing interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α), critical cytokines for macrophage activation and intracellular control of Mycobacterium tuberculosis (Mtb), while suppressing Th2 cytokines such as IL-4. BEA also inhibits 11β-hydroxysteroid dehydrogenase-1, lowering intracellular cor

BACKGROUND: The Mycobacterium tuberculosis (MTB) uridine diphosphogalactofuranose (UDP)-galactopyranose mutase (UGM) is an essential flavoenzyme for mycobacterial viability and an important component of cell wall. It catalyzes the interconversion of UDP-galactopyranose into UDP-galactofuranose, a key building block for cell wall construction, essential for linking the peptidoglycan and mycolic acid cell wall layers in MTB through a 2-keto intermediate. Further, as this enzyme is not present in humans, it is an excellent therapeutic target for MTB. Thus, inhibition of this UGM enzyme is a good

It is a long held belief that the strong immunostimulatory activity of the Mycobacterium bovis bacillus Calmette-Guérin vaccine and Freund's complete adjuvant is due to specific mycobacterial cell envelope components, such as lipids and polysaccharides. Implicated mycobacterial lipids include, among others, the so-called cord factor or trehalose dimycolate, but limited information is available regarding the precise molecular nature of the stimulatory components responsible for the interaction with human APCs. In this regard, the majority of research aimed at identifying and characterizing indi

Mycobacterium tuberculosis (Mtb) owes its success as a pathogen in large measure to its ability to exist in a persistent state of 'dormancy' resulting in a lifelong latent tuberculosis (TB) infection. An understanding of bacterial adaptation during dormancy will help in devising approaches to counter latent TB infection. In vitro models have provided valuable insights into bacterial adaptation; however, they have limitations because they do not disclose the bacterial response to the intracellular environment wherein the bacteria are simultaneously exposed to multiple stresses. We describe the
