Disease-specific iPS cell models in neuroscience.

Neurodegenerative diseases are a heterogeneous group of sporadic or familial disorders of the nervous system that mostly lead to a progressive loss of neural cells. A major challenge in studying the molecular pathomechanisms underlying these disorders is the limited experimental access to disease-affected human nervous system tissue. In addition, considering that the molecular disease initiation occurs years or decades before the symptomatic onset of a medical condition, these tissues mostly reflect only the final phase of the disease. To overcome these limitations, various model systems have

Neuroscience and eating disorders: the allocentric lock hypothesis.

Evidence from psychology and neuroscience indicates that our spatial experience, including the bodily one, involves the integration of different sensory inputs within two different reference frames egocentric (body as reference of first-person experience) and allocentric (body as object in the physical world). Even if functional relations between these two frames are usually limited, they influence each other during the interaction between long- and short-term memory processes in spatial cognition. If, for some reasons, this process is impaired, the egocentric sensory inputs are no more able t

Neuroscience insights improve neurorehabilitation of poststroke aphasia.

The treatment of aphasias-acquired language disorders-caused by stroke and other neurological conditions has benefitted from insights from neuroscience and neuropsychology. Hebbian mechanisms suggest that massed practice and exploitation of residual neurological capacities can aid neurorehabilitation of patients with poststroke aphasia, and progress in basic neuroscience research indicates that the language system of the human brain is functionally interwoven with perceptual and motor systems. Intensive speech and language therapies, including constraint-induced aphasia therapy, that activate

Clinical neuroscience attachments: a student's view of 'neurophobia'.

BACKGROUND: neurophobia has been described as a fear of clinical neuroscience, and is known to affect both medical students and junior doctors alike. There is some evidence that it may affect the practice of GPs, who may not feel confident enough to give advice to neurological patients. CONTEXT: I am a fourth year undergraduate at The University of Manchester, and have undertaken two four-week placements in neurology at the Greater Manchester Neuroscience Centre in Salford. The first was a student-selected component in movement disorders, and the second was part of the standard undergraduate

Neuroscience and heart-brain medicine: the year in review.

Important recent publications in the area of neuroscience and heart-brain medicine center largely around three topics: (1) mechanisms of cardiac sympathetic denervation in Parkinson disease, (2) cytoplasmic monoamine metabolites as autotoxins, and (3) the validity of power spectral analysis of heart rate variability to indicate cardiac sympathetic tone. Findings by Orimo et al support a centripetal, retrograde pathogenetic process involving alpha-synuclein deposition and degeneration of cardiac noradrenergic neurons in Parkinson disease. Several studies suggest that processes increasing cytopl

How neuroscience will change our view on consciousness.

Is there consciousness in machines? Or in animals? What happens to consciousness when we are asleep, or in vegetative state? These are just a few examples of the many questions about consciousness that are troubling scientists and laypersons alike. Moreover, these questions share a striking feature: They seem to have been around forever, yet neither science nor philosophy has been able to provide an answer. Why is that? In my view, the main reason is that the study of consciousness is dominated by what we know from introspection and behavior. This has fooled us into thinking that we know what

Neuroscience and the law: philosophical differences and practical constraints.

Controversies surrounding the value of neuroscience as forensic evidence are explored from the perspective of the philosophy of mind, as well as from a practical analysis of the state of the scientific research literature. At a fundamental philosophical level there are profound differences in how law and neuroscience view the issue of criminal responsibility along the continuum from free will to determinism. At a more practical level, significant limitations in the current state of neuroimaging research constrain its ability to inform legal decision-making. Scientifically supported and unsuppo

[Apraxia--neuroscience and clinical aspects. A literature synthesis].

Apraxic phenomena occur in various neurological conditions. Selective motion control is viewed as the basic capacity to make fine and precise, isolated or independent face or limb movements. Its deficit can indicate limb-kinetic apraxia if it is not explained by paresis, somatosensory deafferentation, or ataxia. The core deficit in ideomotor apraxia could be deficient movement representations, i.e. the combination of invariant features of intrinsic and extrinsic coding for a given movement, which are most important when movements have to be performed outside their typical context. Ideational a

The development of reading impairment: a cognitive neuroscience model.

This review discusses recent cognitive neuroscience investigations into the biological bases of developmental dyslexia, a common disorder impacting approximately 5 to 17 percent of the population. Our aim is to summarize central findings from several lines of evidence that converge on pivotal aspects of the brain bases of developmental dyslexia. We highlight ways in which the approaches and methodologies of developmental cognitive neuroscience that are addressed in this special issue-including neuroimaging, human genetics, refinement of cognitive and biological phenotypes, neural plasticity an

Mathematical neuroscience: from neurons to circuits to systems.

Applications of mathematics and computational techniques to our understanding of neuronal systems are provided. Reduction of membrane models to simplified canonical models demonstrates how neuronal spike-time statistics follow from simple properties of neurons. Averaging over space allows one to derive a simple model for the whisker barrel circuit and use this to explain and suggest several experiments. Spatio-temporal pattern formation methods are applied to explain the patterns seen in the early stages of drug-induced visual hallucinations.

The integration of neurology, psychiatry, and neuroscience in the 21st century.

OBJECTIVE: This article examines the historical basis for the divergence of neurology and psychiatry over the past century and discusses prospects for a rapprochement and potential convergence of the two specialties in the next century. METHOD: The author presents a brief historical overview of developments in neurology and psychiatry from the late 19th century. The histories of research and prevailing scientific opinion on two neuropsychiatric disorders, Alzheimer's disease and Tourette's syndrome, are compared to illustrate the effects of viewing a disease process from, respectively, the ne

Affective neuroscience and the development of social anxiety disorder.

Recent studies on the familial distribution and longitudinal outcome of SAD emphasize developmental aspects of the syndrome, consistent with the developmental psychopathology perspective. Key questions in this area concern factors that mediate familial transmission and that predict outcome. Prior studies provide incomplete answers to these questions. Recent studies in affective neuroscience suggest potential avenues for answering these questions. As reviewed in the current article, fMRI studies of face processing provide examples of such potentially informative research directions.

Methylated cytosine and the brain: a new base for neuroscience.

Methylation of genomic CpG residues is crucial for proper neuronal function. Rett syndrome, a common form of mental retardation, is associated with mutations in the gene encoding MeCP2, a methyl CpG binding protein linked to transcriptional repression. Gene knockouts of mouse Mecp2 have reproduced key aspects of the disease. A CNS-restricted knockout of Dnmt1, encoding the enzyme that maintains CpG methylation patterns, results in loss of mutant neurons and glia.

Neuroscience and appetitive behavior research: 25 years.

Neuroscience techniques have made major contributions to the understanding of appetitive behavior. Highlights in six areas are summarized to illustrate progress during the 25 years of the Columbia Appetitive Behavior Seminar: (1) discovery of angiotensin and aldosterone in the control of thirst and salt appetite; (2) electrophysiological decoding of chemoreceptive information in the brain; (3) a new foundation in the hypothalamus built on peptides, such as neuropeptide Y and galanin, interacting with monoamines and steroids in the control of appetite for macronutrients; (4) discovery of numero

Anatomic and physiologic aging: a behavioral neuroscience perspective.

Because hearing is accomplished by the brain (with neural input from the cochlea), presbyacusis can be ultimately accounted for by changes in brain activity that accompany aging. The anatomic and physiologic changes that accompany aging are of two basic types: the central effects of biological aging (CEBA) and the central effects of peripheral pathology (CEPP). Research using inbred mice and other animal models has provided insights into both CEPP and CEBA, and some implications of this research are reviewed, including the following. Age-related cochlear pathology results in changes in how fre
