Precision Antimicrobial Therapy Against Fusobacterium nucleatum Using Bioengineered Probiotics Expressing Guided Antimicrobial Peptides (gAMPs).

Colorectal cancer (CRC) is a leading cause of cancer-related mortality, with Fusobacterium nucleatum (F. nucleatum) identified as a key contributor to its progression. This study explores a novel therapy that targets this pathogen by using a bioengineered probiotic that expresses guided antimicrobial peptides (gAMPs) to selectively inhibit F. nucleatum. Lactococcus lactis MG1363 was engineered to express gAMPs derived from Ovispirin and Cathelin-related peptide SCF, linked to a Statherin-derived guide peptide that binds specifically to the F. nucleatum membrane porin FomA. The bacteria express

Control of Listeria monocytogenes in Caramel Apples by Use of Sticks Pretreated with Potassium Sorbate.

A multistate listeriosis outbreak associated with caramel apples from 2014 to 2015 prompted research on the survival of Listeria monocytogenes in fresh apples and caramel apples. Research indicated that stem end-inoculated caramel apples with stick insertion allowed for the survival and growth of L. monocytogenes at both refrigeration and ambient temperatures. This study aimed to assess the effectiveness of chemical preservatives as pretreatments for the wooden stick component to reduce L. monocytogenes loads in stem end-inoculated caramel apples during storage. Wooden sticks were pretreated w

Natural Antimicrobial Activity of Nettle (Urtica dioica L.) Leaf Extract for Shelf-Life Extension of Mashed Potatoes.

The growing demand for minimally processed clean-label foods has intensified interest in natural antimicrobials as alternatives to synthetic preservatives. However, very little is known about the antimicrobial potential of several wild edible plants when incorporated into food matrices. This study evaluated the antimicrobial activity of nettle (Urtica dioica L.) leaf extract and as a clean-label preservative for extending the shelf life of fresh mashed potatoes. The extract exhibited strong antioxidant activity, with DPPH, ABTS, and FRAP values of 21.96 ± 0.76 μmol Trolox/mL, 17.51 ± 0.90 μmol

Contribution of the novel sulfur-producing adjunct Lactobacillus nodensis to flavor development in Gouda cheese.

We assessed the efficacy of Lactobacillus nodensis CSK964 as an adjunct culture in Gouda cheese under various industrial conditions. We set up 4 different systems: a direct vat inoculum with and without adjunct using the calf rennet Kalase, and an undefined bulk starter culture with and without adjunct using the microbial rennet Milase (both rennets from CSK Food Enrichment, Ede, the Netherlands). During ripening, we subjected the cheeses to the following analyses: viability of starter and adjunct cells, composition, proteolysis, and flavor development by detection of sulfur compounds and desc

Control of intramammary infections in goats: impact on somatic cell counts.

Udder-half infections were recorded throughout a lactation for 1,060 goats belonging to eight commercial herds. Bacteriological examination from aseptic milk samples and somatic cell counts (SCC) determined by Fossomatic cell counting were performed at the beginning, the middle, and the end of lactation. Coagulase-negative staphylococci (CNS) were the prevalent microorganisms isolated. Geometric means of SCC for uninfected halves or halves infected by CNS or major pathogens were 272 x 10(3) cells/mL, 932,000 x 10(3) cells/mL and 2,443,000 x 10(3) cells/mL, respectively. Two field trials were c

Production and targeting of the Brucella abortus antigen L7/L12 in Lactococcus lactis: a first step towards food-grade live vaccines against brucellosis.

Brucella abortus is a facultative intracellular gram-negative bacterial pathogen that infects humans and animals by entry mainly through the digestive tract. B. abortus causes abortion in pregnant cattle and undulant fever in humans. The immunogenic B. abortus ribosomal protein L7/L12 is a promising candidate antigen for the development of oral live vaccines against brucellosis, using food-grade lactic acid bacteria (LAB) as a carrier. The L7/L12 gene was expressed in Lactococcus lactis, the model LAB, under the nisin-inducible promoter. Using different signals, L7/L12 was produced in cytoplas

Emerging Biocontrol Strategies in Cheese Safety: A Critical Review of Bacteriophages, Antimicrobial Peptides, and Postbiotics.

Fresh and soft cheeses feature high moisture content, elevated water activity, and nutrient richness, creating favorable conditions for foodborne pathogens including Listeria monocytogenes, Salmonella spp., Shiga toxin-producing Escherichia coli, and Staphylococcus aureus, which can contaminate products via post-pasteurization biofilm formation on processing equipment. Because conventional thermal and chemical preservation methods can compromise organoleptic and nutritional quality, biological control strategies have emerged as promising alternatives to enhance microbiological safety. Staphylococcus aureus lipid factors modulate melanoma cell clustering and invasion.

PURPOSE: The zoonotic opportunistic pathogen Staphylococcus pseudintermedius 222 produces BacSp222 - an atypical peptide exhibiting the features of a bacteriocin, a virulence factor, and a molecule modulating the host inflammatory reaction. The peptide is secreted in an unmodified form and, additionally, two forms modified posttranslationally by succinylation. This study is a comprehensive report focusing on the proinflammatory properties of such molecules. METHODS: The study was performed on mouse monocyte/macrophage-like and endothelial cell lines as well as human neutrophils. The following

OBTAINING OF BACTERIOCINES FROM BACTERIA BACILLUS SUBTILIS ATCC 6633 STRAIN BY ORIGINAL METHODS.

In recent years, bacteriocins were in the centre of scientific discussions. Nowadays it is considered that these biologically active substances could be an adequate replacement for classic synthetic antibiotics. They play an important role in the syntheses of various antimicrobial matters, such as disinfectants, pesticides, bacteriophages and so on. Being a product of the vital activity of bacteria, in nature these substances protect bacteria from bacteria-competitors. According to numerous scientific studies, they demonstrate bacteriostatic and bactericidal effects in vivo and in vitro. Moreo

The two-component regulatory systems GraRS and SrrAB mediate Staphylococcus aureus susceptibility to Pep5 produced by clinical isolate of Staphylococcus epidermidis.

Staphylococcus aureus is a common bacterium on the skin and in the nose that sometimes causes severe illness. Bacteriocins, antimicrobial peptides, or proteins produced by bacteria are candidates for the treatment of S. aureus infection. In this study, we found that a clinical Staphylococcus epidermidis strain, KSE112, produced the lantibiotic Pep5, which showed anti-S. aureus activity. The complete nucleotide sequence of the Pep5-encoding plasmid was determined. Several S. aureus two-component regulatory systems (TCSs) are known to be involved in bacteriocin susceptibility. Therefore, suscept

Scintigraphic evaluation of colon targeting pectin-HPMC tablets in healthy volunteers.

The in vivo evaluation of colon-targeting tablets was conducted in six healthy male volunteers. A pectin-hydroxypropyl methylcellulose coating was compressed onto core tablets labelled with 4MBq (99m)Tc-DTPA. The tablets released in the colon in all subjects; three in the ascending colon (AC) and three in the transverse colon (TC). Tablets that released in the TC had reached the AC before or just after food (Group A). The other three tablets released immediately upon AC entry at least 1.5h post-meal (Group B). Release onset for Group B was earlier than Group A (343min vs 448min). Group B table

ABC and MFS efflux transporters mediate antibiotic resistance and bile salt tolerance in Enterococcus faecalis.

Bacteria that reside in the gastrointestinal tract must adapt to the antibacterial properties of bile salts (BSs) and other compounds. We investigated five putative transporters of the enteric-colonizing Enterococcus faecalis for their roles in adaptation to BS and antibiotic resistance. Transcript levels of ef2593 and ef0575 increased over fivefold, and ef0420 increased 12-fold when exposed to 0.02% and 0.1% BS concentrations. No change in the expression of ef2592 or ef1814 was seen. E. faecalis wild-type MMH594 overexpressing efflux pumps by plasmid constructs (EF2592, EF2593, EF0575, EF0420

Dissecting antibiotic effects on the cell envelope using bacterial cytological profiling: a phenotypic analysis starter kit.

Phenotypic analysis assays such as bacterial cytological profiling (BCP) have become increasingly popular for antibiotic mode of action analysis. A plethora of dyes, protein fusions, and reporter strains are available and have been used for this purpose, enabling both rapid mode of action categorization and in-depth analysis of antibiotic mechanisms. However, non-expert researchers may struggle choosing suitable assays and interpreting results. This is a particular problem for antibiotics that have multiple or complex targets, such as the bacterial cell envelope. Here, we set out to curate a m

The avermectin, emamectin benzoate, kills gram-positive bacteria and targets the cell envelope of Bacillus subtilis.

The avermectin family of anthelmintics is largely considered to lack antibacterial activity against gram-positive and gram-negative bacteria. Here, we screened six avermectins (ivermectin, eprinomectin, doramectin, abamectin, selamectin, and emamectin benzoate) and a structurally related milbemycin (moxidectin) for antibacterial activity against a panel of representative gram-positive and gram-negative bacteria. We report that emamectin benzoate exhibited activity against several species of gram-positive bacteria, whereas selamectin was active against Staphylococcus aureus and Staphylococcus e

Pre-treatment with chicken IL-17A secreted by bioengineered LAB vector protects chicken embryo fibroblasts against Influenza Type A Virus (IAV) infection.

The recent advances in our understanding of the host factors in orchestrating qualitatively different immune responses against influenza Type A virus (IAV) have changed the perception of conventional approaches for controlling avian influenza virus (AIV) infection in chickens. Given that infection-induced pathogenicity and replication of influenza virus largely rely on regulating host immune responses, immunoregulatory cytokine profiles often determine the disease outcomes. However, in contrast to the function of other inflammatory cytokines, interleukin-17A (IL-17A) has been described as a 'd
