Selenate-dependent anaerobic arsenite oxidation by a bacterium from Mono Lake, California.

Arsenate was produced when anoxic Mono Lake water samples were amended with arsenite and either selenate or nitrate. Arsenite oxidation did not occur in killed control samples or live samples with no added terminal electron acceptor. Potential rates of anaerobic arsenite oxidation with selenate were comparable to those with nitrate ( approximately 12 to 15 mumol.liter(-1) h(-1)). A pure culture capable of selenate-dependent anaerobic arsenite oxidation (strain ML-SRAO) was isolated from Mono Lake water into a defined salts medium with selenate, arsenite, and yeast extract. This strain does not

Synergistic regulation of hydrogen sulfide and nitric oxide on biochemical components, exopolysaccharides, and nitrogen metabolism in nickel stressed rice field cyanobacteria.

The present study examined the regulatory mechanism of hydrogen sulfide (H2S) and nitric oxide (NO) in nickel (Ni) stressed cyanobacteria viz., Nostoc muscorum and Anabaena sp. by analyzing growth, photosynthetic pigments, biochemical components (protein and carbohydrate), exopolysaccharides (EPS), inorganic nitrogen content, and activity of enzymes comprised in nitrogen metabolism and Ni accumulation. The 1 µM Ni substantially diminished growth by 18% and 22% in N. muscorum and Anabaena sp. respectively, along with declining the pigment contents (Chl a/Car ratio and phycobiliproteins), and bi

[Peroxidase activity of octaheme nitrite reductases from bacteria of the Thioalkalivibrio genus].

Closely related penta- and octaheme nitrite reductases catalyze the reduction of nitrite, nitric oxide, and hydroxylamine to ammonium and of sulfite to sulfide. NrfA pentaheme nitrite reductase plays the key role in anaerobic nitrate respiration and the protection of bacterial cells from stresses caused by nitrogen oxides and hydrogen peroxide. Octaheme nitrite reductases from bacteria of the Thioalkalivibrio genus are less studied, and their function in the cell is unknown. In order to estimate the possible role of octaheme nitrite reductases in the cell resistance to oxidative stress, the pe

Biochemical parameters of plants as indicators of air pollution.

In the present study species like Mangifera indica, Linn., Cassia fistula, Linn., and Eucalyptus hybrid were exposed to different air pollution load for short duration (active biomonitoring). Variation in biochemical parameters like chlorophyll, protein, soluble sugar free amino acid, ascorbic acid, nitrate reductase, superoxide dismutase and peroxidase in the leaves were found to be pollution load dependent. These variations can be used as indicators of air pollution for early diagnosis of stress or as a marker for physiological damage to trees prior to the onset of visible injury symptoms. J

Isolation, genetic and functional characterization of novel soil nirK-type denitrifiers.

Denitrification, the reduction of nitrogen oxides (NO(3)(-) and NO(2)(-)) to N(2) via the intermediates NO and N(2)O, is crucial for nitrogen turnover in soils. Cultivation-independent approaches that applied nitrite reductase genes (nirK/nirS) as marker genes to detect denitrifiers showed a predominance of genes presumably derived from as yet uncultured organisms. However, the phylogenetic affiliation of these organisms remains unresolved since the ability to denitrify is widespread among phylogenetically unrelated organisms. In this study, denitrifiers were cultured using a strategy to gener

Roles, mechanism of action, and potential applications of sulfur-oxidizing bacteria for environmental bioremediation.

Sulfur (S) is a crucial component in the environment and living organisms. This work is the first attempt to provide an overview and critical discussion on the roles, mechanisms, and environmental applications of sulfur-oxidizing bacteria (SOB). The findings reveal that key enzymes of SOB embarked on oxidation of sulfide, sulfite, thiosulfate, and elemental S. Conversion of reduced S compounds was oxidatively catalyzed by various enzymes (e.g. sulfide: quinone oxidoreductase, flavocytochrome c-sulfide dehydrogenase, dissimilatory sulfite reductase, heterodisulfide reductase-like proteins). Sulfide-Induced Dissimilatory Nitrate Reduction to Ammonium Supports Anaerobic Ammonium Oxidation (Anammox) in an Open-Water Unit Process Wetland.

Open-water unit process wetlands host a benthic diatomaceous and bacterial assemblage capable of nitrate removal from treated municipal wastewater with unexpected contributions from anammox processes. In exploring mechanistic drivers of anammox, 16S rRNA gene sequencing profiles of the biomat revealed significant microbial community shifts along the flow path and with depth. Notably, there was an increasing abundance of sulfate reducers (Desulfococcus and other Deltaproteobacteria) and anammox microorganisms (Brocadiaceae) with depth. Pore water profiles demonstrated that nitrate and sulfate c

Ammonia fumigation combined with nitrogen inhibitor reduces root-knot nematode populations and N2O emission but not bacterial alpha diversity.

Aqueous ammonia can release high nitrogen inorganic substances of ammonia and has been used as soil fumigant. However, the information about how fumigation with ammonia affects the soil microbial community is still limited. In this study, a laboratory microcosm experiment was conducted to evaluated the effectiveness of ammonia fumigation in controlling root-knot nematodes (RKNs), its influence on soil microbial diversity, and its impact on nitrous oxide (N2O) emissions. The results indicated that Ammonia treatment increased N2O emissions and decreased the RKNs population by 75.73%. Adding the

Three bacterial strains efficiently reduce selenite to selenium nanoparticles in cell membranes.

Microbial synthesis of selenium nanoparticles (SeNPs) as a fertilizer can promote the development of selenium-rich agricultural products. However, most known selenium-reduction strains exhibit a tolerance to selenite of ≤ 100 mmol/L and possess relatively low reduction efficiency. In this study, three strains capable of tolerating selenite concentrations of > 300 mmol/L were screened from selenium-rich soil in Bama, Guangxi, China. Based on 16 S rRNA gene sequence analysis, the three strains were identified as Citrobacter sp.BM-1, Providencia sp.BM-2, and Brucella sp.BM-3. Notably, Brucella sp

Application of low-intensity ultrasound to enhance simultaneous nitrification/iron-based autotrophic denitrification.

Intermittent ultrasound with an intensity of 0.2 W/ml was applied during simultaneous nitrification/iron-based autotrophic denitrification to evaluate its impacts on total nitrogen (TN) removal efficiency and microbial characteristics during low carbon/nitrogen ratio (C/N) wastewater treatment. At an optimal dissolved oxygen (DO) concentration of 1.2 mg/L, the TN removal rate was 91 ± 4%, and the TN concentration in the effluent decreased by 31% owing to the ultrasound treatment. In addition, the number of iron-compounds that formed in the sludge and wastewater increased by 55% and 37%, respec

Controlling denitrification in closed artificial ecosystems.

Denitrification, the dissimilatory reduction of NO3- to N2O and N2, is found in a wide variety of organisms. In closed artificial systems, especially closed plant growth chambers, a significant loss of fixed-N occurs through denitrification, thereby decreasing the efficiency of the system and fouling the atmosphere with N2O. Denitrification is a form of anaerobic respiration. Whenever available, however, denitrifiers preferentially use O2 as their terminal electron acceptor. As a result, rates of denitrification and growth are a function of O2. Typically, in closed systems O2 consumption is gr

Nitrogen use efficiency in crops under salt stress: from molecular networks to intelligent breeding.

Soil salinization threatens global arable land and agricultural sustainability, severely reducing crop nitrogen use efficiency (NUE) by disrupting root ammonium and nitrate fluxes, impairing nitrogen-assimilation enzymes, and disrupting carbon-nitrogen (C-N) balance. This review synthesizes recent advances in the coordination of salt-stress signaling and nitrogen homeostasis in plants. Two mechanistically distinct regulatory axes have recently been proposed. In one, a nitrate transporter acts as a dual sensor for nitrate and abscisic acid (ABA); in the other, SOS kinase-mediated phosphorylatio

Synthesis and biological evaluation of stilbene derivatives coupled to NO donors as potential antidiabetic agents.

The work is focused on the design of drugs that prevent and treat diabetes and its complications. A novel class of stilbene derivatives were prepared by coupling NO donors of alkyl nitrate and were fully characterised by NMR and other techniques. These compounds were tested in vitro activity, including α-glucosidase inhibitory activity, aldose reductase (AR) inhibitory activity and advanced glycation end products (AGEs) formation inhibitory activity. A class of modified compounds could play a significant effect for treatment of diabetic complications. Target compounds 3e and 7c offered a poten

Hydrogen sulfide enhances rice tolerance to nickel through the prevention of chloroplast damage and the improvement of nitrogen metabolism under excessive nickel.

Hydrogen sulfide (H2S) modulates plant tolerance to abiotic stresses, but its regulatory effects on nitrogen metabolism and chloroplast protection under nickel (Ni) stress in crop plants remain elusive. Taking this into account, we investigated the potential roles of sodium hydrosulfide (NaHS), a H2S generator, in the improvement of growth performance of rice plants under Ni stress. Results showed that NaHS successfully reversed the adverse effects of Ni, as reflected in plant growth and biomass, and photosynthesis attributes including photosynthetic rates, stomatal conductance, transpiration

Sequential treatment of UV254 photodegradation and microalgal bioremediation in aquaculture effluent: Florfenicol toxicity mitigation and nutrient recovery.

Aquaculture effluent contains multiple pollutants, including the antibiotic florfenicol (FF) and nitrogen compounds, which pose challenges for simultaneous removal. This study investigated the effects of FF on Chlorella sorokiniana and evaluated UV254 irradiation as a pretreatment method for FF decomposition. C. sorokiniana exhibited strong tolerance to FF, with 54.9 % growth inhibition at 100 mg/L, but showed limited FF removal capacity. Furthermore, FF completely inhibited nitrate utilization by the C. sorokiniana. Molecular docking simulations, enzyme activity assays, and substrate removal

Spectroscopic and voltammetric characterisation of the bacterioferritin-associated ferredoxin of Escherichia coli.

The bacterioferritin-associated ferredoxin (Bfd) of Escherichia coli is a 64-residue polypeptide encoded by the bfd gene located upstream of the gene (bfr) encoding the iron-storage haemoprotein, bacterioferritin. The Bfd sequence resembles those of the approximately 60-residue domains found in NifU proteins (required for metallocluster assembly), nitrite reductases, and Klebsiella pneumoniae nitrate reductase. These related-domains contain four well-conserved cysteine residues, which are thought to function as ligands to a [2Fe-2S] cluster. The Bfd protein was over-produced, purified, and cha

Regulation of nitrogen metabolism in the nitrate-ammonifying soil bacterium Bacillus vireti and evidence for its ability to grow using N2 O as electron acceptor.

Bacillus vireti is a nitrate-ammonifying bacterium and a partial denitrifier, reducing NO3 (-) , NO2 (-) , NO and N2 O with NarG, NrfA, CbaA and NosZ respectively. Growth is optimized through successive use of the electron acceptors O2 and NO3 (-) , followed by NO2 (-) , NO and N2 O. Fermentation takes place simultaneously with anaerobic respiration. When grown in batch culture with 5 mM initial NO3 (-) , transcription of nrfA was high and most NO3 (-) was reduced to NH4 (+) . With 20 mM initial NO3 (-) , nrfA transcription was lower and more than 50% of the nitrate was recovered as NO, N2 O a
