Nitrogen Scavengers: History, Clinical Considerations and Future Prospects.

Nitrogen scavengers play a critical role in treating acute and chronic hyperammonemia, especially in urea cycle disorders (UCDs), where impaired ammonia detoxification leads to toxic nitrogen accumulation. These agents complement low-protein diets and urea cycle intermediates. Sodium benzoate and sodium phenylacetate are the main scavengers, conjugating with glycine and glutamine to form hippurate and phenylacetylglutamine, which are excreted in urine. This therapeutic approach, introduced in the 1980s, was based on early findings linking benzoate to reduced urea excretion. Nitrogen scavengers

Nitrogen-to-functionalized carbon atom transmutation of pyridine.

The targeted and selective replacement of a single atom in an aromatic system represents a powerful strategy for the rapid interconversion of molecular scaffolds. Herein, we report a pyridine-to-benzene transformation via nitrogen-to-carbon skeletal editing. This approach proceeds via a sequence of pyridine ring-opening, imine hydrolysis, olefination, electrocyclization, and aromatization to achieve the desired transmutation. The most notable features of this transformation are the ability to directly install a wide variety of versatile functional groups in the benzene scaffolding, including e

Biomineralized manganese oxide mediated nitrogen-contained wastewater treatment.

In recent years, manganese (Mn) has emerged as an accelerator for nitrogen metabolism. However, the bioactivity of manganese is limited by the restricted contact between microbes and manganese minerals in the solid phase and by the toxicity of manganese to microbes. To enhance the bioactivity of solid-phase manganese, biomineralized manganese oxide (MnOx) modified by Lactobacillus was introduced. Nitrogen removal performance have confirmed the effective role of biomineralized MnOx in accelerating the removal of total inorganic nitrogen (TIN). Metagenomic analysis has confirmed the enhancement

Developing nitrogen removal models for stormwater bioretention systems.

Bioretention systems have the potential of simultaneous runoff volume reduction and nitrogen removal. Internal water storage (IWS) layers and real-time control (RTC) strategies may further improve performance of bioretention systems. However, optimizing the design of these systems is limited by the lack of effective models to simulate nitrogen transformations under the influences of IWS design and environment conditions including soil moisture and temperature. In this study, nitrogen removal models (NRMs) are developed with two complexity levels of nitrogen cycling: the Single Nitrogen Pool (S

Enantioselective Bioreduction of Medicinally Relevant Nitrogen-Heteroaromatic Ketones.

We herein report an enantioselective bioreduction of ketones that bear the most frequently used nitrogen-heteroaromatics in FDA-approved drugs. Ten varieties of these nitrogen-containing heterocycles were systematically investigated. Eight categories were studied for the first time and seven types were tolerated, significantly expanding the substrate scope of plant-mediated reduction. By use of purple carrots in buffered aqueous media with a simplified reaction setup, this biocatalytic transformation was achieved within 48 h at ambient temperature, offering medicinal chemists a pragmatic and s

A review: Biological technologies for nitrogen monoxide abatement.

Nitrogen oxides (NOx), including nitrogen monoxide (NO) and nitrogen dioxide (NO2), are among the most important global atmospheric pollutants because they have a negative impact on human respiratory health, animals, and the environment through the greenhouse effect and ozone layer destruction. NOx compounds are predominantly generated by anthropogenic activities, which involve combustion processes such as energy production, transportation, and industrial activities. The most widely used alternatives for NOx abatement on an industrial scale are selective catalytic and non-catalytic reductions;

meso-Diazacorrphycenes: Neighboring Effect of Two Nitrogen Atoms.

The neighboring effect of two adjacent heteroatoms influences the structures and properties of heterocyclic compounds. Herein, we demonstrated that the introduction of two adjacent sp2 nitrogen atoms into a porphyrinic skeleton significantly enhanced its Brønsted basicity due to the repulsive interaction between two lone pairs on the nitrogen atoms. Palladium- and copper-templated ring closure of dichlorobis(dipyrrin) with hydrazine hydrate provided meso-diazacorrphycene palladium and copper complexes in good yields. The structural, magnetic, and electrochemical properties of the diazacorrphyc

Air-breathing and excretory nitrogen metabolism in fishes.

During water-land transition, ancient fishes acquired the ability to breathe air, but air-breathing engendered problems in nitrogenous waste excretion. Nitrogen is a fundamental component of amino acids, proteins, and nucleic acids, and the degradation of these nitrogen-containing compounds releases ammonia. Ammonia is toxic and must be removed. Fishes in water excrete ammonia as the major nitrogenous waste through gills, but gills of air-breathing fishes are modified for air-breathing or largely replaced by air-breathing organs. Notably, fishes emerged from water can no longer excrete ammonia

Mechanisms of Specific Embryonic Effects of Nitrogen Oxide.

The study of NO metabolism in chicken embryos showed that the intensity of oxidation of both endogenous and exogenous for the embryo NO donors to nitrate is determined by the presence or state of NO targets, rather than donor concentration. The mechanism of this oxidation and its physiological role are discussed. It was also shown that oxidation product nitrate is actively eliminated from the amnionic sac.

Nitrogen monoxide inhibits haem synthesis in mouse reticulocytes.

AI (anaemia of inflammation) often manifests in patients with chronic immune activation due to cancer, chronic infections, autoimmune disorders, rheumatoid arthritis and other diseases. The pathogenesis of AI is complex and involves cytokine-mediated inhibition of erythropoiesis, insufficient erythropoietin production and diminished sensitivity of erythroid progenitors to this hormone, and retention of iron in haemoglobin-processing macrophages. NO (nitric oxide) is a gaseous molecule produced by activated macrophages that has been identified as having numerous effects on iron metabolism. In t

Nitrogen removal with the anaerobic ammonium oxidation process.

Anaerobic ammonium-oxidizing (anammox) bacteria convert ammonium to N2 with nitrite as the terminal electron acceptor in the absence of O2. Nitritation-anammox bioreactors provide a cost-effective and environment-friendly alternative to conventional nitrification/denitrification nitrogen removal systems. Currently, this process is only applied for ammonium removal from wastewater with high ammonium load and temperature. Nevertheless, recent results obtained with laboratory-scale bioreactors suggest new possible routes of application of the Nitritation-anammox technology including (1) municipal

Nitrogen recovery from a stabilized municipal landfill leachate.

The present paper reports the results of an investigation aimed at evaluating the effectiveness of magnesium ammonium phosphate precipitation (MAP), commonly called struvite, for removing ammonia from a mature municipal landfill leachate. MAP precipitation was carried out at laboratory scale by adding phosphoric acid and magnesium oxide as external sources of phosphorus and magnesium, respectively, and regulating the pH at 9.0. The effect of Mg:NH(4):PO(3) ratio was studied. Due to the low solubility of MgO, a low ammonia removal efficiency (i.e. 67%), with a rather high residual concentration

RCM-mediated synthesis of trifluoromethyl-containing nitrogen heterocycles.

A ring-closing metathesis mediated pathway to trifluoromethyl-containing piperidines is detailed. This involves the development of a synthetic route to a new (trifluoromethyl)allylating reagent via a Diels-Alder/retro-Diels-Alder strategy, its application in the synthesis of a series of trifluormethyl-substituted diolefin precursors for ring-closing metathesis, and eventually the successful cyclization of these precursor molecules into the corresponding functionalized piperidines.

Chemical denitrification for nitrogen removal from landfill leachate.

A new system that removes nitrogen from landfill leachate and other waste waters with similar properties has been proposed with nitritation (i.e. oxidation of ammonium to nitrite) of half of the influent ammonium followed by chemical denitrification with a reaction between equal amounts of ammonium and nitrite to form nitrogen gas. Chemical denitrification occurs at high concentrations and the reactions were studied in combination with a concentration step. Studied concentration methods were freezing/thawing and evaporation/drying. Chemical denitrification is well-known in inorganic chemistry

Nitrogen requirements of the didelphid marsupial Caluromys philander.

The use of fruit-based diets by a small arboreal marsupial, the woolly opossum (Caluromys philander) was studied to elucidate the mechanisms used to compensate for a low dietary protein concentration. The passage of a liquid phase digesta marker (Cr-EDTA) through the gut was significantly faster when animals were fed a diet containing 0.45% N compared to that measured on a diet containing 0.90% N. The size of the gut of the two groups was similar except that the caecum of animals fed 0.45% N was significantly larger than in those animals fed 0.90% N. Animals fed a diet of 0.45% N ate significa

In vivo nitrogen metabolism in ornithine transcarbamylase deficiency.

We developed a new technique that monitors metabolic competency in female heterozygotes for ornithine transcarbamylase deficiency (OTCD). The method uses mass spectrometry to measure conversion of (15)NH4Cl to [15N]urea and [5-(15)N]glutamine following an oral load of (15)NH4Cl. We found that heterozygotes converted significantly less NH3 nitrogen to urea, with this difference being particularly obvious for symptomatic carriers, in whom the blood [15N]urea concentration (mM) was significantly less than control values at most time points. The blood concentration of [5-(15)N]-glutamine (microM)

Nitrogen dioxide-induced acute lung injury in sheep.

Lung mechanics, hemodynamics and blood chemistries were assessed in sheep (Ovis aries) before, and up to 24 h following, a 15-20 min exposure to either air (control) or approximately 500 ppm nitrogen dioxide (NO2). Histopathologic examinations of lung tissues were performed 24 h after exposure. Nose-only and lung-only routes of exposure were compared for effects on NO2 pathogenesis. Bronchoalveolar lavage fluids from air- and NO2-exposed sheep were analyzed for biochemical and cellular signs of NO2 insult. The influence of breathing pattern on NO2 dose was also assessed. Five hundred ppm NO2 e

Ammonia-constitutive nitrogen fixation mutants of Rhodobacter capsulatus.

Mutants of R. capsulatus that express nif genes constitutively with respect to ammonia were studied in order to define better the circuit that regulates nif gene transcription. One mutant class could be complemented in trans by a cosmid clone containing a wild-type gene (nifR5) defined by Tn5 inserts as being no longer than 1.6 kb. The nifR5 gene is unlinked to previously described nif genes. A second mutant class could not be complemented by the wild-type cosmid library. For one mutant in this class, a nifH::lac fusion was used to select further mutants that were Lac-. Only two of these could

Nitrogen photoreduction on desert sands under sterile conditions.

Sands from various geographic locations reduce N(2) from the air to NH(3) and traces of N(2)H(4) on exposure to sunlight. This N(2) photofixation occurs under sterile conditions on the surface of finely dispersed titanium minerals such as rutile, utilizing reducing equivalents generated through the photolysis of chemisorbed H(2)O. Abiological N(2) photofixation is suggested to be part of the nitrogen cycle in arid and semiarid regions. It is estimated that about 10 x 10(5) tons of N(2) is photoreduced on the total surface of the earth's deserts per year.
