Ran Binding Protein 2 (RanBP2 or Nucleoporin358) is one of the main components of the cytoplasmic filaments of the nuclear pore complex. Mutations in the RANBP2 gene are associated with acute necrotizing encephalopathy type 1 (ANE1), a rare condition where patients experience a sharp rise in cytokine production in response to viral infection and undergo hyperinflammation, seizures, coma, and a high rate of mortality. Despite this, it remains unclear howRanBP2 and its ANE1-associated mutations contribute to pathology. Mounting evidence has shown that RanBP2 interacts with distinct viruses to re

Defects in microtubule-based transport are implicated in many neuropathologies. The filamentous fungi Aspergillus nidulans and Ustilago maydis are valuable models for studying transport owing to their yeast-like genetic and biochemical tractability and metazoan-like dependence on microtubule-based transport for cellular trafficking. In these organisms the role of microtubules in nuclear positioning is well studied, but recent work has expanded the range of cargos to include endosomes, messenger RNA, secretory vesicles, peroxisomes, and nuclear pore complexes, reflecting the diversity of metazo

A novel porous calcium silicate (PCS) material with unique pore structure prepared from coal fly ash (CFA) was reported. The microstructure was investigated through X-ray diffraction, Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, nuclear magnetic resonance cryoporometry, and Brunauer-Emmett-Teller method. Model describing the nanostructure of the prepared PCS was proposed in this work. Results show that the prepared PCS has open pores, a high specific surface area, and multi-peak pore size distributions (macro-, meso-, and micropores).

Low-field nuclear magnetic resonance has become one of the main methods to characterize static parameters and dynamic changes in unconventional reservoirs. The research focus of this paper is process simulation of coalbed methane (CBM) production. The dynamic variation of pore volume with different pore sizes during pressure drop, methane desorption-diffusion process, and methane-water interaction during migration is discussed. Moreover, the calculation principles of NMR single and multifractal models are systematically described, and the applicability of NMR fractal models within different re

Injecting steam into coal seam is an important means to accelerate gas desorption and improve gas extraction efficiency. However, the change law of pore-fracture structures of coal after high-temperature steam shock (thermal shock) is still unclear. Through this study, pore-fracture structures of coal samples before and after thermal shock were compared and analyzed based on the experimental methods of surface pore and fracture extraction, scanning electron microscopy (SEM), and nuclear magnetic resonance (NMR) spectroscopy. The results show that after thermal shock, the surface porosity, max

Hydrate-based CO2 storage (HBCS) has emerged as a promising approach within carbon capture and storage (CCS) technologies, offering a potential pathway for mitigating CO2 emissions. The permeability of hydrate-bearing sediments (HBSs) is crucial for predicting the feasibility of HBCS, yet the seepage mechanisms become quite complex during the formation or dissociation of hydrates. The objective of this paper is to investigate the hydrate pore habits and the evolution of key physical properties at both macroscopic and microscopic scales, utilizing an integrated set of methodologies such as CT s

UNLABELLED: The roles of nucleoplasm-residing nucleoporins (NUP) in solid tumors, including prostate cancer, remain unknown. In this study, we reveal the clinical significance and mechanistic role of the off-pore NUP, soluble POM121 (sPOM121), as a crucial transcriptional regulator that enhances the aggressiveness of metastatic prostate cancer. Using orthogonal methodologies in human samples, sPOM121 was identified as the predominantly expressed nucleoplasmic NUP in prostate cancer. Unbiased proteomic and epigenomic studies demonstrate that sPOM121, through its C-terminus, interacts with the c

The nuclear envelope (NE) is a complex intracellular structure unique to eukaryotic cells. The principal components identified by a variety of methods include the inner nuclear lamina comprised primarily of lamin proteins, the inner nuclear membrane (INM), the nuclear pore (NuP) complexes comprised of NuP proteins and the outer nuclear membrane (ONM). Dating to the earliest studies, the specific NE targets of human autoantibodies included: lamins A, B and C of the nuclear lamina, emerin, lamin B receptor (LBR), lamina-associated polypeptide 1 (LAP1) and lamina-associated polypeptide 2 (LAP2) o

INTRODUCTION: The diagnosis of primary biliary cholangitis (PBC) relies on the detection of specific antibodies, yet there is variability in the diagnostic accuracy and efficiency among the different diagnostic methods. We aimed to assess the individual diagnostic performance of these markers using different detection methods.
METHODS: A total of 112 participant-54 with PBC and 58 acting as disease controls-were enrolled. The levels of anti-M2-3E, anti-gp210, and anti-sp100 antibodies were measured using immunoblot tests from Euroimmun, flow cytometry-based multiplex bead immunoarray (MBIA [T

BACKGROUND: The nuclear pore complexes (NPCs) are built of about 30 different nucleoporins and act as key regulators of molecular traffic between the cytoplasm and the nucleus for sizeable proteins (> 40 kDa) which must enter the nucleus. Various nuclear transport receptors are involved in import and export processes of proteins through the nuclear pores. The most prominent nuclear export receptor is chromosome region maintenance 1 (CRM1), also known as exportin 1 (XPO1). One of its cargo proteins is the prolyl hydroxylase 2 (PHD2) which is involved in the initiation of the degradation of hypo

We determined that a mutation in the nucleoporin gene NUP170 leads to defects in chromosome transmission fidelity (ctf) and kinetochore integrity in Saccharomyces cerevisiae. A ctf mutant strain, termed s141, shows a transcription readthrough phenotype and stabilizes a dicentric chromosome fragment in two assays for kinetochore integrity. Previously, these assays led to the identification of two essential kinetochore components, Ctf13p and Ctf14p. Thus, s141 represents another ctf mutant involved in the maintenance of kinetochore integrity. We cloned and mapped the gene complementing the ctf m

Hereditary fibrosing poikiloderma (HFP) is a rare human dominant negative disorder caused by mutations in the FAM111B gene that encodes a nuclear trypsin-like serine protease. HFP patients present with symptoms including skin abnormalities, tendon contractures, myopathy and lung fibrosis. We characterized the cellular roles of human FAM111B using U2OS and MCF7 cell lines and report here that the protease interacts with components of the nuclear pore complex. Loss of FAM111B expression resulted in abnormal nuclear shape and reduced telomeric DNA content suggesting that FAM111B protease is requi

Protein homeostasis (proteostasis) refers to the balance of the cellular protein environment, tightly regulated by pathways governing protein synthesis, folding, trafficking, and degradation. Growing evidence supports the interconnection of these pathways to ensure the robustness of the proteo-stasis network. A recent study by Park et al. showed that, in macroautophagy/autophagy-deficient cells, the loss of proteasome or nuclear pore components causes synthetic lethality, as cytoplasmic proteins that accumulate under impaired autophagy are transported to the nucleus and degraded by nuclear pro

The antiviral factor CPSF6-358 interferes with the nuclear entry of human immunodeficiency virus type 1 (HIV-1). HIV-1 acquires resistance to CPSF6-358 through the N74D mutation of the capsid (CA), which alters its nuclear entry pathway. Here we show that compared to wild-type (WT) HIV-1, N74D HIV-1 is more sensitive to cyclosporine, has increased sensitivity to nevirapine, and is impaired in macrophage infection prior to reverse transcription. These phenotypes suggest a difference in the N74D reverse transcription complex that manifests early after infection and prior to interaction with the

Nuclear export of messenger RNAs (mRNAs) through nuclear pore complexes (NPCs) is a critical step in gene expression. Although N6-adenosine methylation (m6A) has been implicated in this process, the underlying mechanism remains obscure. Here we demonstrate, using single-molecule imaging, that m6A markedly accelerates the nuclear export of messenger ribonucleoproteins (mRNPs) by increasing export efficiency and shortening export time through NPCs. We further show that the m6A methyltransferase METTL3 localizes at NPCs and functionally associates with the nucleoporin NUP93 to promote the efficie

Nuclear envelope (NE) budding is a nuclear pore-independent nuclear export pathway, analogous to the egress of herpesviruses, and required for protein quality control, synapse development, and mitochondrial integrity. The physical formation of NE buds is dependent on the Wiskott-Aldrich Syndrome protein, Wash, its regulatory complex (SHRC), and Arp2/3, and requires Wash's actin nucleation activity. However, the machinery governing cargo recruitment and organization within the NE bud remains unknown. Here, we identify Pavarotti (Pav) and Tumbleweed (Tum) as new molecular components of NE buddin

Triple A syndrome is an autosomal recessive neurological disease, mimicking motor neuron disease, and is caused by mutant ALADIN, a nuclear-pore complex component. We recently discovered that the pathogenesis involved impaired nuclear import of DNA repair proteins, including DNA ligase I and the cerebellar ataxia causative protein aprataxin. Such impairment was overcome by fusing classical nuclear localization signal (NLS) and 137-aa downstream sequence of XRCC1, designated stretched NLS (stNLS). We report here that the minimum essential sequence of stNLS (mstNLS) is residues 239-276, downsize

We describe the route by which aldosterone-triggered macromolecules enter and exit the cell nucleus of Xenopus laevis oocyte. Oocytes were microinjected with 50 fmol aldosterone and then enucleated 2-30 min after injection. After isolation, nuclear envelope electrical resistance (NEER) was measured in the intact cell nuclei by using the nuclear hourglass technique. We observed three NEER stages: an early peak 2 min after injection, a sustained depression after 5-15 min, and a final late peak 20 min after injection. Because NEER reflects the passive electrical permeability of nuclear pores, we

An early step in the replication of the hepatitis B virus (HBV) genome is the transport of the viral DNA into the nucleus of the infected cell. So far only little is known about the events and mechanisms at the nuclear membrane required for entry of the viral genome into the nucleus. Using a hepatoblastoma cell line that constitutively produces hepatitis B virions and in so doing displays intracellular viral amplification, we showed that nonparticulated HBV core protein is associated with nuclear membrane pore complexes. Additionally, viral DNA has been detected firmly attached to the nuclear
