GABAergic Interneurons in the Insular Cortex Alleviate Acute Stress-Induced Gastric Injury by Suppressing the IC-NTS Glutamatergic Pathway.

Acute stress induces gastric mucosal injury, but the central mechanisms remain unclear. We investigated whether GABAergic interneurons, particularly somatostatin (SST) neurons, in the insular cortex (IC) are involved using restraint water-immersion stress (RWIS) in mice. RWIS upregulated GAD65 and SST but downregulated parvalbumin (PV) in the IC. Chemogenetic inhibition of IC GABAergic or SST neurons exacerbated gastric mucosal injury and reduced gastric tight junction proteins (Occludin, Claudins) and PCNA, whereas activation of SST neurons attenuated injury and increased these proteins. Remimazolam alleviates blood-brain barrier damage by regulating the PI3K/AKT signaling pathway in sepsis-associated encephalopathy mice.

Sepsis-associated encephalopathy (SAE) is a common neurological complication in critically ill patients. However, therapeutic strategies for SAE remain limited. Increasing evidence suggests that impairment of the blood-brain barrier (BBB) plays a crucial role in the progression of SAE. Remimazolam has been demonstrated in previous studies to exert neuroprotective effects through the modulation of neuroinflammatory responses. This study aimed to determine whether remimazolam alleviates BBB disruption in a murine model of SAE and to elucidate the underlying signaling mechanisms. A murine model o

Alterations of retinal vasculature in cystathionine-β-synthase heterozygous mice: a model of mild to moderate hyperhomocysteinemia.

Mild to moderate hyperhomocysteinemia is prevalent in humans and is implicated in neurovascular diseases, including recently in certain retinal diseases. Herein, we used hyperhomocysteinemic mice deficient in the Cbs gene encoding cystathionine-β-synthase (Cbs(+/-)) to evaluate retinal vascular integrity. The Cbs(+/+) (wild type) and Cbs(+/-) (heterozygous) mice (aged 16 to 52 weeks) were subjected to fluorescein angiography and optical coherence tomography to assess vasculature in vivo. Retinas harvested for cryosectioning or flat mount preparations were subjected to immunofluorescence micros

Effect of tryptophan supplementation on diet-induced non-alcoholic fatty liver disease in mice.

Intestinal serotonin (5-hydroxytrypamine, 5-HT) metabolism is thought to play a role in gut functions by regulating motility, permeability and other functions of the intestine. In the present study, we investigated the effect of tryptophan (TRP), the precursor of 5-HT, supplementation on intestinal barrier functions and non-alcoholic fatty liver disease (NAFLD). An established mouse model of NAFLD induced by feeding a fructose-rich diet (N group) was used in the present study. TRP was administered orally for 8 weeks to C57BL/6J control or NAFLD mice. NAFLD-related liver parameters (hepatic TAG

Protein kinase Cα inhibitor protects against downregulation of claudin-1 during epithelial-mesenchymal transition of pancreatic cancer.

Protein kinase Cα (PKCα) is highly expressed in pancreatic cancer. However, the effects of PKCα on Snail and claudin-1, which play crucial roles in epithelial cell polarity during epithelial-mesenchymal transition (EMT), remain unclear. In this study, we investigated the mechanisms of regulation of Snail and claudin-1 via a PKCα signal pathway during EMT in pancreatic cancer cells and in normal human pancreatic duct epithelial cells (HPDEs). By immunostaining, overexpression of PKCα and downregulation of claudin-1 were observed in poorly differentiated human pancreatic cancer tissues and the p

Expression of the Lactobacillus plantarum surface layer MIMP protein protected NCM460 epithelial cells from enteroinvasive Escherichia coli infection.

Источник пометил публикацию как отозванную

Our previous studies demonstrated that the micro integral membrane protein (MIMP) located within the integral membrane protein of Lactobacillus plantarum CGMCC 1258 protected the intestinal barrier from injury. To further analyze the protective effects conferred to intestinal epithelial cells by MIMP, we established transient MIMP-expressing NCM460 cells (NCM460/MIMP) as a means of assessing their susceptibility to infection. We constructed a recombinant eukaryotic expression vector using pcDNA3.1(-) and the MIMP gene. The recombinant vector was then transduced into NCM460 cells and the anti-i

Xyloglucan protects the intestinal barrier and reduces bacterial translocation in experimental cirrhosis - A promising non-antibiotic strategy.

BACKGROUND: cirrhosis alters the intestinal barrier, increasing permeability and promoting bacterial translocation (BT). Norfloxacin (NF) is currently the only effective strategy to reduce BT, but the rise of multidrug-resistant bacteria highlights the need for new approaches. AIMS: to evaluate the effect of xyloglucan, alone or with norfloxacin, on the intestinal barrier in cirrhotic rats with ascites. METHODS: decompensated cirrhosis with ascites was induced in 32 rats using CCl4. Subsequently, xyloglucan (XG), NF, XG + NF (XG+NF), or water (control) were administered for one week. Paramet

Increasing interleukin-17 induced by β-Amyloid deposition is involved in early cognitive decline after sevoflurane anesthesia.

Postoperative delirium is a major concern in elderly patients and is associated with early cognitive decline after anesthesia exposure. Sevoflurane anesthesia increases amyloid-β (Aβ) production, leading to neuroinflammation, blood-brain barrier (BBB) disruption, and postoperative delirium. Interleukin-17 (IL-17) is implicated in BBB breakdown, and Aβ deposits induce elevated expression. Hence, we explored the relationships between early postoperative cognitive alterations and Aβ deposits, IL-17 expression in the hippocampus, and BBB damage in aged rats after fracture surgery under sevoflurane

Localization and pneumococcal alteration of junction proteins in the human alveolar-capillary compartment.

Loss of alveolar barrier function with subsequent respiratory failure is a hallmark of severe pneumonia. Although junctions between endo- and epithelial cells regulate paracellular fluid flux, little is known about their composition and regulation in the human alveolar compartment. High autofluorescence of human lung tissue in particular complicates the determination of subcellular protein localization. By comparing conventional channel mode confocal imaging with spectral imaging and linear unmixing, we demonstrate that background fluorescent spectra and fluorophore signals could be rigorously

Mechanisms involved in the blood-brain barrier increased permeability induced by Phoneutria nigriventer spider venom in rats.

We have recently demonstrated by electron microscopy, using lanthanum nitrate as an extracellular tracer, that the intravenous injection of Phoneutria nigriventer spider venom (PNV) induces blood-brain barrier (BBB) breakdown in rat hippocampus. One and nine days after PNV injection, tracer was found in pinocytic vesicles crossing the endothelium and in the interendothelial cleft, suggesting that BBB breakdown had occurred through enhanced transendothelial transport and/or tight-junction opening. In the present work, we investigated the mechanisms by which PNV (850 microg/kg, i.v.) increased t

H5N1 infection impairs the alveolar epithelial barrier through intercellular junction proteins via Itch-mediated proteasomal degradation.

The H5N1 subtype of the avian influenza virus causes sporadic but fatal infections in humans. H5N1 virus infection leads to the disruption of the alveolar epithelial barrier, a pathologic change that often progresses into acute respiratory distress syndrome (ARDS) and pneumonia. The mechanisms underlying this remain poorly understood. Here we report that H5N1 viruses downregulate the expression of intercellular junction proteins (E-cadherin, occludin, claudin-1, and ZO-1) in several cell lines and the lungs of H5N1 virus-infected mice. H5N1 virus infection activates TGF-β-activated kinase 1 (T
