Synapse formation in the mouse olfactory bulb. I. Quantitative studies.

A quantitative study of synapse formation in the mouse olfactory bulb has been carried out using serial sections. Volumetric synaptic density as well as absolute number of synapses per olfactory bulb for eight distinct synaptic types have been determined at 15 different ages, from the beginning of synapse formation at embryonic day 14 (E14) to postnatal day 44 (P44). Synapses are first found in appreciable numbers at E15 when both axo-dendritic and a few dendro-dendritic synapses occur in the presumptive glomerular layer. Initial synapse formation correlates closely with the reorientation of m

Short-axon cells in the olfactory bulb: dendrodendritic synaptic interactions.

1. In the rabbit olfactory bulb, analysis has been carried out of extracellular unitary responses in the glomerular layer to olfactory nerve volleys. 2. Units in the glomerular layer responded to single volleys with single, double, triple or longer repetitive spike discharges. The shortest initial latencies are consistent with monosynaptic excitation from the olfactory nerves; longer latencies may reflect longer nerve pathways or polysynaptic connexions in the glomerular layer. 3. Like mitral and tufted cells, some glomerular layer units gave evidence of activation by discrete nerve bundles. T

Changes in olfactory bulb volume and olfactory sulcus depth in COVID-19 infection: an autopsy study.

PURPOSE: The pathophysiology of COVID-19 remains unclear. Olfactory dysfunction is one of the neurological symptoms of the disease. There is no evidence to evaluate whether there is a morphometric change in the olfactory pathways. This study aimed to examine the effect of COVID-19 on olfactory center sections through morphometric measurements obtained from autopsy cases. METHODS: Our study was conducted on 63 autopsy cases (COVID-19 [n = 37], control (non-COVID) [n = 26]) between the ages of 18-80 years who came to Tokat Forensic Medicine Institute. The anatomical structures of the olfactory

Olfactory rehabilitation and olfactory bulb volume changes in patients after total laryngectomy: a prospective randomized study.

INTRODUCTION: After total laryngectomy, decreased olfactory function and olfactory bulb volume shrinkage have been reported to occur due to olfactory deprivation caused by nasal airflow interruption. There is evidence that the olfactory system can be modulated by repeated exposure to odors in a procedure called olfactory training. However, it is not known whether any recovery of the lost olfactory bulb volume is possible by eliminating olfactory deprivation via olfactory rehabilitation long after laryngectomy. OBJECTIVE: This study examined the recovery of olfactory function and the change in

3D atlas describing the ontogenic evolution of the primary olfactory projections in the olfactory bulb of Xenopus laevis.

The adult Xenopus presents the unique capability to smell odors both in water and air thanks to two different olfactory pathways. Nevertheless, the tadpole can initially perceive only water-borne odorants, as the olfactory receptor neurons (ORN) that will detect air-borne odorants develop later. Such a phenomenon requires major reorganization processes. Here we focused on the precise description of the neuroanatomical modifications occurring in the olfactory bulb (OB) of the tadpole throughout metamorphosis. Using both carbocyanine dyes and lectin staining, we investigated the evolution of ORN

The topographic organization of associational fibers of the olfactory system in the rat, including centrifugal fibers to the olfactory bulb.

This study analyzed the topographic organization of the associational fibers within the olfactory cortex of the rat, by using the autoradiographic method. Small injections of 3H-leucine were placed in all of the subdivisions of the olfactory cortex, to label selectively the fibers arising in each area. Intracortical fibers were identified from all of the olfactory cortical areas except the olfactory tubercle and were classified into two major systems (the layer Ib system and the layer II-deep Ib system) on the basis of their laminar pattern of termination (see Luskin and Price, '83). The layer

Evoked potential and single unit responses to olfactory nerve volleys in the isolated turtle olfactory bulb.

An in vitro preparation of the whole olfactory bulb of the turtle has been investigated using extracellular evoked potentials and unitary spikes. The compound action potential has a slow conduction velocity (0.15 mm/msec), brief relatively refractory period (10 msec) and prolonged supernormality (several hundred msec). The evoked potentials undergo prolonged periods of partial and complete suppression, as tested by the responses to paired volleys in the olfactory nerves. The suppression has a complex temporal pattern, and lasts more than 4 sec. Similar periods were revealed by the unitary spik

Regulation of spike timing-dependent plasticity of olfactory inputs in mitral cells in the rat olfactory bulb.

The recent history of activity input onto granule cells (GCs) in the main olfactory bulb can affect the strength of lateral inhibition, which functions to generate contrast enhancement. However, at the plasticity level, it is unknown whether and how the prior modification of lateral inhibition modulates the subsequent induction of long-lasting changes of the excitatory olfactory nerve (ON) inputs to mitral cells (MCs). Here we found that the repetitive stimulation of two distinct excitatory inputs to the GCs induced a persistent modification of lateral inhibition in MCs in opposing directions.

Morphological study of the effects of intranasal zinc sulfate irrigation on the mouse olfactory epithelium and olfactory bulb.

The effects of intranasal zinc sulfate (ZnSO4) irrigation on the morphology of the olfactory epithelium and olfactory bulb were studied in mice with short survival times (as early as 1 day) and with long survival times (up to 593 days) after the irrigation procedure. As in several previous studies, the olfactory epithelium was completely destroyed within a few days after the ZnSO4 treatment. Within 2-4 days, the septum and turbinates were covered by a new, cuboidal epithelium, the cells of which differed significantly from any cells normally seen in the olfactory epithelium. Slowly, over sever

Differential localization of G proteins, Gi and Go, in the olfactory epithelium and the main olfactory bulb of the rat.

The immunohistochemical localization of proteins Gi1 (plus Gi3). Gi2 and Go was studied in the olfactory epithelium and the main olfactory bulb of rats, using purified antibodies to the respective alpha subunits and beta gamma subunits of these G proteins. In the olfactory epithelium, only a restricted population of olfactory cells was immunopositive for Gi2 alpha, but others were not. The immunoreactivity for Gi1 alpha/Gi3 alpha was not observed. The olfactory epithelium was immunopositive for both Go alpha and beta gamma, but its apical surface was immunopositive only for beta gamma. In the

Quantitative observations on the nasal epithelia and olfactory innervation in bats. Suggested design mechanisms for the olfactory bulb.

The nasal epithelia of two species of bats were quantified with respect to relative surface areas and olfactory epithelial volumes. In the macrosmatic Aribeus jamaicensis 55.9% of the nasal cavity surface was covered by olfactory epithelium (232.4 mm2), in contrast to only 28.9% in the microsmatic Myotis lucifugus (36.4 mm2). The roles of the various nasal epithelia have been discussed as they may relate to olfaction, respiration and echolocation. In the olfactory bulbs of both species, the estimated concentration of mitral cells approximated at 2,500/mm2 compared to an olfactory nerve concent

Glomerulus-Specific Inhomogeneity of the Basal Activity Map in the Olfactory Bulb.

Glomeruli are signal-processing units of the olfactory bulb (OB) that play a key role in many OB computations, including contrast enhancement, gain control, and odorant-selective habituation. In awake mice, we unveil an extremely stable, inhomogeneous map of basal glomerulus-specific activity that serves as the background against which olfactory signal processing occurs. This activity is strongly driven by centrifugal cholinergic inputs; endogenous and airflow-evoked spiking of olfactory sensory neurons; and, to a minor extent, by the odor environment. Moreover, it is brain-state dependent and

Neuropathological assessment of the olfactory bulb and tract in individuals with COVID-19.

The majority of patients with Parkinson disease (PD) experience a loss in their sense of smell and accumulate insoluble α-synuclein aggregates in their olfactory bulbs (OB). Subjects affected by a SARS-CoV-2-linked illness (COVID-19) also frequently experience hyposmia. We previously postulated that microglial activation as well as α-synuclein and tau misprocessing can occur during host responses following microbial encounters. Using semiquantitative measurements of immunohistochemical signals, we examined OB and olfactory tract specimens collected serially at autopsies between 2020 and 2023.

Induction of smell through transethmoid electrical stimulation of the olfactory bulb.

BACKGROUND: Anosmia has an estimated prevalence of 5% of the general population. Outside of inflammatory causes, therapeutic options are limited despite research advances. Bypassing peripheral neuronal damage through central stimulation is a potential therapeutic option that has shown success in other sensory systems, most notably with hearing. We performed a pilot study to determine the feasibility of inducing smell through artificial electrical stimulation of the olfactory bulbs in humans. METHODS: Subjects with a history of sinus surgery, including total ethmoidectomy, with intact ability

Activity Patterns Elicited by Airflow in the Olfactory Bulb and Their Possible Functions.

Olfactory sensory neurons (OSNs) can sense both odorants and airflows. In the olfactory bulb (OB), the coding of odor information has been well studied, but the coding of mechanical stimulation is rarely investigated. Unlike odor-sensing functions of OSNs, the airflow-sensing functions of OSNs are also largely unknown. Here, the activity patterns elicited by mechanical airflow in male rat OBs were mapped using fMRI and correlated with local field potential recordings. In an attempt to reveal possible functions of airflow sensing, the relationship between airflow patterns and physiological para

Effects of Pinealectomy and Gonadectomy on Olfactory Bulb Dopaminergic Neurons in Rats.

BACKGROUND: Olfactory disorder is an early manifestation of Parkinson's disease (PD), likely to be associated with abnormalities of the dopaminergic neurons in the olfactory bulb (OB); however, the causes of olfactory disorder in PD are not entirely clear. Some studies showed that melatonin (MT) and androgens (mainly testosterone, T) might participate in the pathogenesis of PD. The research aimed to investigate effects of MT or T deficiency on OB dopaminergic neurons in rats. METHODS: One hundred and twenty normal male Wistar rats were randomly divided into the control, sham operation pineale

Histological Properties of Main and Accessory Olfactory Bulbs in the Common Hippopotamus.

The olfactory system of mammals comprises a main olfactory system that detects hundreds of odorants and a vomeronasal system that detects specific chemicals such as pheromones. The main (MOB) and accessory (AOB) olfactory bulbs are the respective primary centers of the main olfactory and vomeronasal systems. Most mammals including artiodactyls possess a large MOB and a comparatively small AOB, whereas most cetaceans lack olfactory bulbs. The common hippopotamus (Hippopotamus amphibius) is semiaquatic and belongs to the order Cetartiodactyla, family Hippopotamidae, which seems to be the closest
