The effects of endothelin-1 on the cardiorespiratory physiology of the freshwater trout (Oncorhynchus mykiss) and the marine dogfish (Squalus acanthias).

The aim of the present study was to evaluate the effects of endothelin-l-elicited cardiovascular events on respiratory gas transfer in the freshwater rainbow trout (Oncorhynchus mykiss) and the marine dogfish (Squalus acanthias). In both species, endothelin-1 (666 pmol kg(-1)) caused a rapid (within 4 min) reduction (ca. 30-50 mmHg) in arterial blood partial pressure of O2. The effects of endothelin-1 on arterial blood partial pressure of CO2 were not synchronised with the changes in O2 partial pressure and the responses were markedly different in trout and dogfish. In trout, arterial CO2 part

Molecular Cloning and Characterization of a cDNA Encoding the Retinal Arylalkylamine N-Acetyltransferase of the Rainbow Trout, Oncorhynchus mykiss.

Melatonin synthesis in the retina as well as in the pineal gland exhibits daily variations with higher levels during the dark phase of light-dark cycles. To analyze the molecular mechanism of melatonin synthesis in the retina, we have cloned, sequenced and characterized a putative cDNA for arylalkylamine N-acetyltransferase (AANAT; EC 2.3.1.87), a rate-limiting enzyme in melatonin production, from the retina of the rainbow trout (Oncorhynchus mykiss). The trout AANAT cDNA (1,585 bp) contains an open reading frame encoding 240 amino acid protein (predicted molecular weight, 27,420) that is 51-6

Ontogenic development of salmon GnRH and chicken GnRH-II systems in the brain of masu salmon (Oncorhynchus masou).

Ontogenic development of salmon gonadotropin-releasing hormone (GnRH) and chicken GnRH-II systems in masu salmon (Oncorhynchus masou) was examined. Salmon GnRH was first detected by radioimmunoassay in the embryo on day 36 after fertilization. Salmon GnRH-immunoreactive fibers were detected initially by immunocytochemistry in the vicinity of the olfactory placode of the embryo (day 36) and were distributed widely in the brain as well as in the pituitary gland of fish just after hatching (day 80). Salmon GnRH-immunoreactive neuronal somata were first detected about 6 months after fertilization

Effects of chronic low-level copper exposure on ultrastructure of the olfactory system in rainbow trout (Oncorhynchus mykiss).

This study investigated the effects of a chronic exposure to a low level of copper on cell populations of the olfactory system in yearling rainbow trout. Fish were sacrificed after 15, 30 and 60 days of copper exposure. Transmission electron microscopy was used to describe the sequence of subcellular changes occurring in three tissues, the sensory epithelium, the olfactory nerve and the olfactory bulb. Data show that a 15-day exposure to 20 micrograms/l of copper causes specific degeneration of all mature receptor cells as well as numerous immature neurons. Moreover, degenerating receptor cell

Physiological and structural impacts of oxytetracycline exposure in rainbow trout (Oncorhynchus mykiss): implications for fish health and welfare.

The present study evaluated the pathophysiological and tissue alterations induced by dietary oxytetracycline (OTC) in rainbow trout (Oncorhynchus mykiss). Fish were exposed to three different doses (T1 = 40 mg OTC/kg body weight, T2 = 80 mg OTC/kg body weight, T3 = 120 mg OTC/kg body weight) alongside a control group (T0) for a period of 30 days. Hematological, immunological, and biochemical parameters were monitored on days 0, 15, and 30 days of OTC exposure. Higher OTC doses caused a significant reduction in total erythrocyte count (TEC), total leucocyte count (TLC), hemoglobin, packed cell

Unraveling the Systemic and Local Immune Response of Rainbow Trout (Oncorhynchus mykiss) to the Viral Hemorrhagic Septicemic Virus.

Viral outbreaks have caused significant mortality and economic losses in aquaculture, highlighting the urgent need for effective therapies and a deeper understanding of antiviral and immune mechanisms in key species. This study investigates the constitutive and virus-induced antiviral responses in juvenile rainbow trout (Oncorhynchus mykiss) following infection with viral hemorrhagic septicemia virus (VHSV). Trout (30 g) were infected by immersion with VHSV (TCID50 = 105 mL-1) for two hours. Samples were collected at 24, 72, and 120 h post-infection to assess hematology, innate immunity, viral

The effects of dietary exposure to Magnéli phase titanium suboxide and titanium dioxide on rainbow trout (Oncorhynchus mykiss).

Magnéli phase titanium suboxides (Magnéli TiOx) are promising, novel materials with superior properties compared to TiO2, they are substoichiometric titanium oxides with the chemical formula TinO2n-1 (where n ≥ 1). In this study, for the first time, subchronic effects of dietary intake of Magnéli TiOx were evaluated and compared with TiO2 particles of similar size, in concentrations 0.1% and 0.01% of feed. The experiment consisted of 38 d of an exposition period and 14 d of a depuration period. Minor effects on plasma biochemical profile and morphological parameters were recorded. A reduced co

Brain and gills as internal and external ammonia sensing organs for ventilatory control in rainbow trout, Oncorhynchus mykiss.

Ammonia is both a respiratory gas and a toxicant in teleost fish. Hyperventilation is a well-known response to elevations of both external and internal ammonia levels. Branchial neuroepithelial cells (NECs) are thought to serve as internal sensors of plasma ammonia (peripheral chemoreceptors), but little is known about other possible ammonia-sensors. Here, we investigated whether trout possess external sensors and/or internal central chemoreceptors for ammonia. For external sensors, we analyzed the time course of ventilatory changes at the start of exposure to high environmental ammonia (HEA,

Distribution of two isoforms of tryptophan hydroxylase in the brain of rainbow trout (Oncorhynchus mykiss). An in situ hybridization study.

Serotonin (5-HT) is one of the principal neurotransmitters in the nervous system of vertebrates. It is initially synthesized by hydroxylation of tryptophan (Trp) by means of tryptophan hydroxylase or TPH which is the rate-limiting enzyme in the production of 5-HT. In most vertebrates, there are two isoforms of TPH present, TPH1 and TPH2, which exhibit different catalytic or substrate specificity as well as different expression domains. Studies carried out in mammals show that only tph2 is expressed in the brain whereas tph1-mRNA is primarily localized in the enterochromaffin cells and pineal g

The expression of insulin and insulin receptor mRNAs is regulated by nutritional state and glucose in rainbow trout (Oncorhynchus mykiss).

Many species of fish, including rainbow trout, possess multiple INS- and IR-encoding mRNAs. In this study, rainbow trout (Oncorhynchus mykiss) were used as a model to study the regulation of INS (INS1, INS2) and IR (IR1, IR2, IR3, and IR4) mRNA expression by nutritional state and glucose. In the nutritional state study, fish were either fed continuously, fasted (4 or 6 weeks), or fasted 4 weeks, then refed for 2 weeks. Nutritional state regulated INS and IR mRNA expression in a subtype- and tissue-specific manner. A 4-week fast reduced INS1 expression in endocrine pancreas (Brockmann body) and

Subclinical effects of saxitoxin and domoic acid on aggressive behaviour and monoaminergic turnover in rainbow trout (Oncorhynchus mykiss).

The algal produced neurotoxins saxitoxin and domoic acid may have serious effects on marine life and can be responsible for the intoxication of for instance sea mammals, sea birds and fish. Given that farmed fish cannot escape algal blooms, they may be more susceptible to intoxication than wild stocks. In the present study, subclinical effects of saxitoxin and domoic on aggressive behaviour and monoaminergic systems in the brain of the rainbow trout (Oncorhynchus mykiss) were investigated. The resident-intruder test was used to measure aggression where only the resident fish were subjected to

The distribution of proliferating cell nuclear antigen-immunoreactive cells in the pineal organ of the rainbow trout Oncorhynchus mykiss.

Cell proliferation in the pineal organ of the immature rainbow trout Oncorhynchus mykiss was investigated by immunocytochemical demonstration of the proliferating cell nuclear antigen (PCNA) together with photoreceptor-specific opsin. Numerous PCNA-immunoreactive cells were found throughout the pineal end-vesicle and stalk. Two types of PCNA-immuno-reactive cells were distinguished: intensely stained, large ovoid and round cells, and mildly stained, slender fusiform cells. The ovoid type of the former cell was found often in the apical region and the round type in the basal region of the epith

The in vitro effects of some pesticides on carbonic anhydrase activity of Oncorhynchus mykiss and Cyprinus carpio carpio fish.

Systemic carbonic anhydrase (CA) inhibitors are among the most powerful agents to lower intraocular pressure. Unfortunately, their use is frequently accompanied by undesired side effects. Some are due to the relatively large amounts of drug that have to be systematically administered to inhibit the CA in the ciliary processes. The aim of the present work was to study in vitro effects of some pesticides on CA enzyme obtained from blood of fish, which play a key role in salt- and osmoregulation and acid-base balance in the fish, Oncorhynchus mykiss and Cyprinus carpio carpio living in freshwater

Biotransformation of pentachlorophenol, aniline and biphenyl in isolated rainbow trout (Oncorhynchus mykiss) hepatocytes: comparison with in vivo metabolism.

1. The biotransformation of pentachlorophenol (PCP), aniline and biphenyl in rainbow trout (Oncorhynchus mykiss) isolated liver cells was investigated to examine if fish hepatocytes represent a suitable alternative to the in vivo approach for studying the biotransformation of chemicals. Each compound was incubated at two concentrations (10 and 60 microM) for 2 h. For comparison, the metabolic profile of these xenobiotics was also studied in urine and bile of trout orally exposed to 1.8-4.0 mg/kg wet wt of each compound. 2. In vitro as in vivo, PCP glucuronide and to a lesser extent PCP sulphat

A nested polymerase chain reaction for the detection of genomic DNA of Myxobolus cerebralis in rainbow trout Oncorhynchus mykiss.

A nested polymerase chain reaction (PCR) test was developed to amplify a segment of the 18S rRNA gene from Myxobolus cerebralis, the agent causing whirling disease in salmonid fish. The PCR amplifies a 415 bp amplicon that was identified by dideoxynucleotide terminated sequencing to be identical to the known 18S rDNA sequence of M. cerebralis. There was no amplification of genomic DNA from 4 other myxosporean parasites of salmonid fish from the genus Myxobolus including M. arcticus, M. insidiosus, M. neurobius, and M. squamalis. The efficacy of the PCR test to detect early infections was demon

Postembryonic changes in the structure of the olfactory bulb of the chinook salmon (Oncorhynchus tshawytscha) across its life history.

Juvenile chinook salmon (Oncorhynchus tshawytscha) memorize odors characteristic of their natal stream, then use these imprinted olfactory cues to return to that same stream years later as sexually mature adults. In a preliminary effort to identify neuroanatomical changes in the salmon olfactory system that may underlie these behavioral capabilities, the structure of the olfactory bulb (OB) was studied at three developmental periods across the life history: in early juvenile development (0.1-4 months of age), in late juvenile development (11-16 months), and as spawning adults (48 months). Usin

Ontogenetic development of gonadotropin-releasing hormone-like immunoreactive neurons in the brain of the chum salmon, Oncorhynchus keta.

In the chum salmon, gonadotropin-releasing hormone-immunoreactive (GnRH-ir) cells were first detected in the olfactory placode of embryos at 16 days after fertilization, and then appeared sequentially in the olfactory nerve and the distal part of the forebrain by hatching. Four months after hatching, the terminal nerve was differentiated along the olfactory nerve, and GnRH-ir fibers extended to the hypothalamus and hypophysis. GnRH-ir cells occurred later in the preoptic area in about 1-year-old juveniles. These data suggest that GnRH neurons originate in the olfactory placode and then migrate

Early development of the pineal photoreceptors prior to the retinal differentiation in the embryonic rainbow trout, Oncorhynchus mykiss (Teleostei).

The development of the pineal and retinal photoreceptors in the embryonic rainbow trout, Oncorhynchus mykiss was investigated by means of light and electron microscopy. The pineal photoreceptors endowed with photoreceptive outer segments and signal-transmitting synapses appeared first 15 days after fertilization, and well developed by 21 days. In contrast, the retinal photoreceptors appeared as late as 27 days after fertilization (1 day before hatching). This temporal disparity in photoreceptor development has been compared with the photoperiodic formation of otolith increments, which is initi
