Correlation of ELISA copro-antigen and oocysts count to the severity of cryptosporidiosis parvum in children.

A total of one thousand and fifty (1050) young children from Mansoura Pediatric Hospital, July 2005 to July 2006 were examined for cryptosporidiosis. Detailed history was taken from each participant, stool examination by direct smear, Formalin-ether sedimentation, Sheather's floatation, modified Ziehl-Nel-seen (MZN) stain, Cryptospordium oocysts count, coproantigen detection by ELISA. Other infections were excluded. Out of 1050 children examined, cryptosporidial oocysts were detected in 90 cases by modified Zeihl Nelseen (MZN) stain, and copro-antigen ELISA added another 10 cases. There was a

Protozoan predation as a mechanism for the removal of cryptosporidium oocysts from wastewaters in constructed wetlands.

The removal of the protozoan parasite, Cryptosporidium parvum, from wastewaters is becoming of increasing importance in the UK, especially since contamination of raw waters by sewage effluents has been implicated in major waterborne outbreaks of cryptosporidiosis in recent years. Compared to conventional wastewater-treatment processes, constructed wetlands have demonstrated favourable removal rates for Cryptosporidium oocysts. The removal mechanisms, however, remain unknown. Predation by free-living ciliated protozoa, which are commonly found in constructed wetlands, was investigated as a poss

Multiple Cryptosporidium serpentis oocyst isolates from captive snakes are not transmissible to amphibians.

Viable Cryptosporidium serpentis oocysts originating from 6 captive snakes were gastrically delivered to 12 Cryptosporidium-free African clawed frogs and 9 tadpoles and 3 recently metamorphosed adults of Cryptosporidium-free wood frogs. On days 7 and 14 postinoculation, no life-cycle stage of Cryptosporidium was observed in any of the histological sections of stomach, jejunum, ileum, cloaca, and cecum. However, viable inoculum-derived C. serpentis oocysts were recovered from the water in which the amphibians were kept. Amphibians may disseminate C. serpentis oocysts in the natural habitat.

Cryptosporidiosis in adults in Lusaka, Zambia, and its relationship to oocyst contamination of drinking water.

In Lusaka, where human immunodeficiency virus seroprevalence in young adults is approximately 25%, four townships were studied to establish the prevalence of persistent diarrhea in adults and the etiologic importance of cryptosporidiosis in adults with persistent diarrhea. Cryptosporidium parvum oocyst contamination of urban water supplies was measured and the results used to categorize these populations into high or low exposure. In total, 506 adults were reported as having had diarrhea in the 2 weeks prior to the survey; 101 of these episodes were persistent. Adults with persistent diarrhea

Comparison of oocyst shedding and the serum immune response to Cryptosporidium parvum in cattle and pigs.

A comparison was made between oocyst shedding and the presence of specific serum IgG antibodies to Cryptosporidium parvum in 108 bovines and 90 pigs. Oocysts were detected by a commercial immunofluorescence assay in feces from 26.8% of bovines and 34.4% of pigs, whereas positive titers as determined by an indirect fluorescent antibody method were found in sera from 12.9% and 48.9% of the respective animals. Infection was significantly most frequent in suckling calves (82.7%) and weaned piglets (87.5%). By contrast, the numbers of seropositives were highest in weaned calves (17.1%) and fattenin

Induction of folds or sutures on the walls of Cryptosporidium parvum oocysts and their importance as a diagnostic feature.

The proportion of oocysts of Cryptosporidium parvum showing a fold on oocyst walls when incubated with either fluorescent monoclonal antibody or a surface-reactive fluorescent dye was increased by incubating suspensions of oocysts with dimethyl sulfoxide, sucrose, or Hanks' balanced salt solution. Further incubation of sucrose-incubated oocysts with water showed this to be a reversible phenomenon. Oocysts demonstrating this fold after incubation in dimethyl sulfoxide were of the same viability as control oocysts and followed the same excystation dynamics. Despite this fold having been previous

Loss of TGME49_227100 (Glutaredoxin 5) Disrupts Oocyst Formation and Sporulation in Toxoplasma gondii.

Oocysts of Toxoplasma gondii exhibit remarkable resistance to environmental stressors and most conventional disinfectants. Despite its ability to infect a wide variety of host species, sexual reproduction and oocyst formation occur exclusively within felid definitive hosts. Despite the epidemiological significance of oocyst-mediated transmission, the molecular mechanisms governing oocyst production and sporulation remain incompletely understood. Glutaredoxin, serving as a central regulator of cellular redox homeostasis and multiple vital cellular processes in cells, is a potential regulator fo

Effect of a Blood Meal on Plasmodium Oocyst Growth Using the Enema Injection Method.

Objective: Anopheles mosquitoes transmit Plasmodium parasites through blood feeding. The oocyst stage in mosquitoes is crucial for Plasmodium transmission. Oocysts can form without a blood meal or the midgut passage and utilize nutrients from additional blood feeding. However, it remains unclear the impact of a blood meal during infection on oocysts. The present study evaluated how a blood meal during infection impacts oocyst growth to better understand oocyst development. Methods: We used a novel method for Plasmodium infection known as enema injection, which involves injecting Plasmodium ber

Development and optimization of an ELISA method to detect Toxoplasma gondii oocyst infection in cats.

Toxoplasmosis caused by Toxoplasma gondii (T. gondii) is a zoonotic disease with great medical and veterinary significance. Felines, the definitive hosts of T. gondii, play a crucial role in the transmission of toxoplasmosis. The booming pet industry has led to more cats and cat-owning families, increasing the risk of toxoplasmosis transmission from animals to humans. Monitoring feline T. gondii infection accurately is crucial for reducing transmission risks. However, existing diagnostic methods focus on detecting whether cats are infected with T. gondii but fail to trace whether feline toxopl

In Vivo Safety and Efficacy of Thiosemicarbazones in Experimental Mice Infected with Toxoplasma gondii Oocysts.

Background: Toxoplasma gondii is a globally widespread parasite responsible for toxoplasmosis, a zoonotic disease with significant impact on both human and animal health. The current lack of safe and effective treatments underscores the need for new drugs. Earlier, thiosemicarbazones (TSCs) and their metal complexes have shown promising activities against T. gondii. This study evaluated a gold (III) complex C3 and its TSC ligand C4 for safety in host immune cells and zebrafish embryos, followed by efficacy assessment in a murine model for chronic toxoplasmosis. Methods: The effects on viabilit

Plasmodium falciparum Development from Gametocyte to Oocyst: Insight from Functional Studies.

Malaria elimination may never succeed without the implementation of transmission-blocking strategies. The transmission of Plasmodium spp. parasites from the human host to the mosquito vector depends on circulating gametocytes in the peripheral blood of the vertebrate host. Once ingested by the mosquito during blood meals, these sexual forms undergo a series of radical morphological and metabolic changes to survive and progress from the gut to the salivary glands, where they will be waiting to be injected into the vertebrate host. The design of effective transmission-blocking strategies require

Use of Oxidative Stress Responses to Determine the Efficacy of Inactivation Treatments on Cryptosporidium Oocysts.

Cryptosporidium oocysts are known for being very robust, and their prolonged survival in the environment has resulted in outbreaks of cryptosporidiosis associated with the consumption of contaminated water or food. Although inactivation methods used for drinking water treatment, such as UV irradiation, can inactivate Cryptosporidium oocysts, they are not necessarily suitable for use with other environmental matrices, such as food. In order to identify alternative ways to inactivate Cryptosporidium oocysts, improved methods for viability assessment are needed. Here we describe a proof of concep

A chicken embryo model for the maintenance and amplification of Cryptosporidium parvum and Cryptosporidium baileyi oocysts.

Cryptosporidium is a genus of apicomplexan parasites that inhabit the respiratory and gastrointestinal tracts of vertebrates. Research of these parasites is limited by a lack of model hosts. This study aimed to determine the extent to which infection at the embryo stage can enhance the propagation of Cryptosporidium oocysts in chickens. Nine-day-old chicken embryos and one-day-old chickens were experimentally infected with different doses of Cryptosporidium baileyi and Cryptosporidium parvum oocysts. Post hatching, all chickens had demonstrable infections, and the infection dose had no effect
