Effects of Colonization of the Roots of Domestic Rice (Oryza sativa L. cv. Amaroo) by Burkholderia pseudomallei.

Burkholderia pseudomallei is a saprophytic bacterium that causes melioidosis and is often isolated from rice fields in Southeast Asia, where the infection incidence is high among rice field workers. The aim of this study was to investigate the relationship between this bacterium and rice through growth experiments where the effect of colonization of domestic rice (Oryza sativa L. cv Amaroo) roots by B. pseudomallei could be observed. When B. pseudomallei was exposed to surface-sterilized seeds, the growth of both the root and the aerosphere was retarded compared to that in controls. The organi

OsMOGS is required for N-glycan formation and auxin-mediated root development in rice (Oryza sativa L.).

N-glycosylation is a major modification of glycoproteins in eukaryotic cells. In Arabidopsis, great progress has been made in functional analysis of N-glycan production, however there are few studies in monocotyledons. Here, we characterized a rice (Oryza sativa L.) osmogs mutant with shortened roots and isolated a gene that coded a putative mannosyl-oligosaccharide glucosidase (OsMOGS), an ortholog of α-glucosidase I in Arabidopsis, which trims the terminal glucosyl residue of the oligosaccharide chain of nascent peptides in the endoplasmic reticulum (ER). OsMOGS is strongly expressed in rapi

Characterization of mercury species in brown and white rice (Oryza sativa L.) grown in water-saving paddies.

In China, total Hg (HgT) and methylmercury (MeHg) were quantified in rice grain grown in three sites using water-saving rice cultivation methods, and in one Hg-contaminated site, where rice was grown under flooded conditions. Polished white rice concentrations of HgT (water-saving: 3.3±1.6 ng/g; flooded: 110±9.2 ng/g) and MeHg (water-saving 1.3±0.56 ng/g; flooded: 12±2.4 ng/g) were positively correlated with root-soil HgT and MeHg contents (HgT: r2=0.97, MeHg: r2=0.87, p<0.05 for both), which suggested a portion of Hg species in rice grain was derived from the soil, and translocation of Hg spe

Role of salicylic acid in alleviating oxidative damage in rice roots (Oryza sativa) subjected to cadmium stress.

Time-dependent changes in enzymatic and non-enzymatic antioxidants, and lipid peroxidation were investigated in roots of rice (Oryza sativa) grown hydroponically with Cd, with or without pretreatment of salicylic acid (SA). Exposure to 50 microM Cd significantly decreased root growth, and activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD), but increased the concentrations of H(2)O(2), malondialdehyde (MDA), ascorbic acid (AsA), glutathione (GSH) and non-protein thiols (NPT). However, pretreatment with 10 microM SA enhanced the activities of antioxidant enzymes and th

Exogenous anti-MoPMK1 microRNA and precursor formats reduce Magnaporthe oryzae infection in rice.

Exogenous RNA application is emerging as a useful strategy to investigate plant-pathogen interactions. Most studies against fungal pathogens have focused on long double-stranded RNAs, whereas the potential of artificial miRNA and miRNA-precursor formats remains less explored. Here, we targeted the Magnaporthe oryzae MAP kinase gene MoPMK1, a central regulator of appressorium-mediated penetration, invasive growth and host tissue colonization, using three exogenous RNA formats: chemically synthesized mature miRNA duplexes, in planta-produced anti-MoPMK1-containing small RNA fractions and in vitr

Isolation of a deuA-deficient strain of Aspergillus oryzae and its application to soy sauce fermentation.

The thermostable protease DeuA from Aspergillus oryzae contributes to undesirable proteolysis in processed foods prepared with heated soy sauce. To obtain a deuA-deficient A. oryzae strain without recombinant DNA techniques or genome editing, we developed a high-throughput screening method utilizing the bacteriostatic effect of protamine on Latilactobacillus sakei. Loss of DeuA activity reduced protamine degradation, thereby suppressing bacterial acid production and enabling rapid identification of mutants. Screening of approximately 20,000 UV-mutagenized colonies from the industrial strain A.

Evaluation of activity and stability of immobilized thermostable Aspergillus oryzae lipase on cotton cloth fibrils.

In this study, a thermostable lipase from Aspergillus oryzae (AOL) was immobilized on cotton cloth fibrils using a multilayer approach involving polyethyleneimine (PEI) and glutaraldehyde (GA). Prior to immobilization onto the support, PEI-lipase complexes were formed in solution, yielding a protein precipitation efficiency of 78% at an optimal PEI concentration of 5 mg/mL without adversely affecting enzyme activity. Cross-linking with 0.625% GA enhanced structural stability, allowing the immobilized lipase to remain firmly attached to the support after extensive washing. The immobilized bioca

Photocatalytic performance of CuI/Cu2+ -TiO2 for controlling Magnaporthe oryzae infection in rice plants.

This study presents the synthesis, characterisation, and photocatalytic antifungal performance of a Cu+/Cu2+-TiO2 nanocomposite designed to control rice blast disease caused by Magnaporthe oryzae. The heterostructure was fabricated through a chemical precipitation process in which TiO2 was first doped with Cu2+ ions, followed by partial reduction to Cu+ to form CuI. X-ray Photoelectron Spectroscopy (XPS) confirmed the coexistence of both Cu+ and Cu2+ oxidation states. A notable feature of this material is its Cu+/Cu2+ redox cycling, which enhances charge separation during photocatalysis and su

MoSun4 Contributes to Pathogenicity and Is Associated with Altered Expression of Virulence-Related Genes in Magnaporthe oryzae.

The SUN family protein MoSun4 in Magnaporthe oryzae has been previously implicated in mitophagy and has potential as a target for reducing rice blast, but its role as a secreted protein remains poorly understood. In this study, signal peptide prediction and yeast secretion assays confirmed MoSun4 signal peptide function, and co-localization revealed the localization of the extra-invasive hyphal membrane (EIHM)-associated apoplastic compartment or matrix, establishing MoSun4 as a secreted protein. In addition, deletion of the MoSUN4 gene reduced the hyphal growth, conidiation and virulence of M

Targeting a cryptic Magnaporthe oryzae-specific pocket in succinate dehydrogenase to combat rice blast.

The global efficacy of succinate dehydrogenase inhibitors (SDHIs), a leading class of fungicides, is increasingly threatened by resistance, driven largely by their shared binding mode at the conserved ubiquinone-binding (Q) site. This challenge is particularly acute for rice blast, as no SDHI is registered for this disease and available compounds offer limited protection while remaining vulnerable to Q-site mutations. In this study, we identified a previously unrecognized, pathogen-adaptive pocket within the SDH complex of Magnaporthe oryzae, which is structurally distinct from the canonical Q

Divergent volatile metabolomes and flavor attributes in rice fermented by Aspergillus oryzae and Aspergillus flavus.

INTRODUCTION: The generally recognized as safe (GRAS) fungus Aspergillus oryzae has been used for millennia in the production of traditional Asian fermented foods and beverages. Domestication has led to genomic and phenotypic adaptations that distinguish A. oryzae from its wild relative, Aspergillus flavus. While differences between these species have been partially characterized, their comparative production of volatile compounds during food fermentation remains poorly understood. METHODS: We evaluated alpha-amylase activity, aflatoxin production, taste attributes using an electronic tongue,

Molecular immune responses of male locusts Locusta migratoria to an Aspergillus oryzae XJ1 infection.

BACKGROUND: Many fungal pathogens infect and kill host insects successfully through the suppression of host immune systems, making them potential biological control agents. Aspergillus oryzae is a new fungal pathogen of locust Locusta migratoria, which is an important insect pest worldwide; consequently, information on the molecular mechanisms that it uses to suppress the host's immune system is limited. Here, transcriptomic analyses of male locusts during an A. oryzae infection were used to characterize the temporal variations in the host's molecular immune responses. RESULTS: Two key immune

Global population genomics redefines domestication and clinical diversity in the Aspergillus flavus-oryzae complex.

Aspergillus flavus is a globally important human pathogen and agricultural contaminant, while its domesticated relative A. oryzae is widely used in food fermentation and biotechnology. Despite their importance, the evolutionary relationship, population structure and domestication history of these fungi remain unresolved. Here, we present the first global population genomic analysis of 639 A. flavus and A. oryzae isolates from clinical, environmental and food-fermentation sources across multiple continents. Our analyses reveal a complex evolutionary landscape comprising well-separated clades in

Structure-activity relationship studies of tanzawaic acid A, an antifungal agent against Rhizopus oryzae.

Through structure-activity relationship studies of tanzawaic acid A (TZAA), this research aims to contribute the development of potent antifungal drugs for treating mucormycosis infections. To this end, new TZAA analogs bearing chemical modifications in the side chain and/or C8-methyl group moieties were synthesized. These analogs are not accessible from natural TZAA and therefore required total synthesis. As a result, four of the newly synthesized TZAA analogs exhibited more potent and selective inhibitory activity against R. oryzae compared to TZAA.

Morphological aspects and the effectiveness of photodynamic inactivation against Rhizopus oryzae in different life cycles.

Mucormycosis is an extremely aggressive fungal disease with a high mortality rate, especially in people with compromised immune systems. Most cases of mucormycosis are caused by the fungus Rhizopus oryzae. The treatments used are based on high doses of antifungals, associated with surgical resections, when it is possible. However, even with this aggressive treatment, the estimated attributable mortality rate is high. There is therefore a need to develop adjuvant treatments. Photodynamic Inactivation (PDI) may be an auxiliary therapeutic option for mucormycosis. Due to the lack of reports in th

A Comprehensive in vitro and in silico Assessment on Inhibition of CYP51B and Ergosterol Biosynthesis by Eugenol in Rhizopus oryzae.

Mucormycosis, also known as Zygomycosis, is a disease caused by invasive fungi, predominantly Rhizopus species belonging to the Order of Mucorales. Seeing from the chemistry perspective, heterocyclic compounds with an "azole" moiety are widely employed as antifungal agent for minimising the effect of mucormycosis as a prescribed treatment. These azoles serve as non-competitive inhibitors of fungal CYP51B by predominantly binding to its heme moiety, rendering its inhibition. However, long-term usage and abuse of azoles as antifungal medicines has resulted in drug resistance among certain fungal

Endocytic protein Pal1 regulates appressorium formation and is required for full virulence of Magnaporthe oryzae.

Endocytosis plays key roles during infection of plant-pathogenic fungi, but its regulatory mechanisms are still largely unknown. Here, we identified a putative endocytosis-related gene, PAL1, which was highly expressed in appressorium of Magnaporthe oryzae, and was found to be important for appressorium formation and maturation. Deletion of PAL1 significantly reduced the virulence of M. oryzae due to defects in appressorial penetration and invasive growth in host cells. The Pal1 protein interacted and colocalized with the endocytosis protein Sla1, suggesting it is involved in endocytosis. The

Piper Essential Oils Inhibit Rhizopus oryzae Growth, Biofilm Formation, and Rhizopuspepsin Activity.

Piper is the largest genus of the Piperaceae family. The species of this genus have diverse biological activities and are used in pharmacopeia throughout the world. They are also used in folk medicine for treatment of many diseases in several countries including Brazil, China, India, Jamaica, and Mexico. In Brazil, Piper species are distributed throughout the national territory, making this genus a good candidate for biological activity screening. During our studies with Piper essential oils, we evaluated its activity against Rhizopus oryzae, the main agent of mucormycosis. The main compounds

Proteomic analysis of salicylic acid-induced resistance to Magnaporthe oryzae in susceptible and resistant rice.

To probe salicylic acid (SA)-induced sequential events at translational level and factors associated with SA response, we conducted virulence assays and proteomic profiling analysis on rice resistant and susceptible cultivars against Magnaporthe oryzae at various time points after SA treatment. The results showed that SA significantly enhanced rice resistance against M. oryzae. Proteomic analysis of SA-treated leaves unveiled 36 differentially expressed proteins implicated in various functions, including defense, antioxidative enzymes, and signal transduction. Majority of these proteins were i

Basilar artery occlusion after multifactor coagulopathy including Rhizopus oryzae infection in burns.

The authors report a case of a 29-year-old male patient with severe burn injuries (54% TBSA) and inhalation injury. He developed a candidemia and a cutaneous zygomycotic superinfection with Rhizopus oryzae while he received burn intensive care. Despite aggressive surgery, sepsis persisted, and therapy was limited by uncontrollable coagulopathy and catecholamine refractory shock after 15 days. Autopsy revealed a thromboembolic occlusion of the basilar artery that resulted in liquefactive necrosis of the basal brain tissue and the brain stem.
