Lesions in sheep following administration of a vaccine of a Freund's complete adjuvant nature used in the control of ovine paratuberculosis.

CASE HISTORIES: Occurrences of adverse reactions in seven sheep flocks in Australia following vaccination against paratuberculosis where veterinary attention was requested are reviewed. All cases occurred within the 3-year period following commencement of use of a vaccine of a Freund's complete adjuvant nature, at a time when approximately six million doses of vaccine had been administered. CLINICAL FINDINGS: In the first case, 26/58 (45%) Merino sheep vaccinated as adults had palpable tissue reactions at or near the site of vaccination; enlarged prescapular lymph nodes were palpated in 17 (2

Comparison of a commercial DNA probe test and three cultivation procedures for detection of Mycobacterium paratuberculosis in bovine feces.

Diagnosis of paratuberculosis using the IDEXX DNA probe test and 3 methods for cultivation of Mycobacterium paratuberculosis from fecal specimens were compared. Twenty-one of 170 fecal specimens were DNA probe test positive, whereas 35 specimens were positive by 1 or more of the cultivation methods evaluated. Four specimens were DNA probe test positive but were negative by fecal culture. The probe test detected M. paratuberculosis DNA in 62.9% of the specimens positive by a sedimentation culture method, in 56.6% of those positive by a centrifugation culture method, and in 65.4% of the specimen

Antimicrobial activity of rifabutin in combination with two and three other antimicrobial agents against strains of Mycobacterium paratuberculosis.

The inhibitory and bactericidal, synergistic, and antagonistic activities of rifabutin combined with ciprofloxacin, ethambutol, clofazimine, cefazolin, and amikacin in dual and triple combinations against various human and animal isolates of Mycobacterium paratuberculosis were determined. Synergism was observed when rifabutin was combined with either cefazolin or clofazimine in double combinations. The greatest amount of synergy occurred with the rifabutin-cefazolin combination in which bactericidal synergism was present with all strains. Of the triple combinations examined, only rifabutin in

Diagnostic value of antibody responses to Mycobacterium avium subsp. paratuberculosis-derived proteins PtpA and PtpB in rheumatoid arthritis.

Evidence suggests that Mycobacterium avium subspecies paratuberculosis (MAP) may contribute to autoimmune diseases such as rheumatoid arthritis (RA), partly through effector proteins-particularly the tyrosine phosphatases PtpA and PtpB-that modulate macrophage signaling and promote bacterial persistence. This study evaluated whether serum antibodies against these proteins serve as biomarkers of RA. Humoral responses to PtpA and PtpB were quantified in Mexican RA patients (n = 100) and healthy controls (n = 100) using in-house ELISAs. Associations with disease activity (DAS28), ROC performance,

Can Mycobacterium avium subsp. paratuberculosis Survive During Pecorino Cheesemaking Process? An Experimental Evaluation Through a Microbiological Challenge Test.

Paratuberculosis is an infectious disease mainly affecting domestic ruminants, caused by Mycobacterium avium subsp. paratuberculosis (MAP). Affected animals shed the pathogen through feces, also during the subclinical stages of the disease. Although there is still no conclusive evidence of MAP involvement in human diseases, particularly Crohn's disease and autoimmune disorders, exposure to MAP through the consumption of contaminated dairy products has become a public health concern. Therefore, in this study, the survival of MAP during the production of Pecorino, an Italian cheese made from raw

Subtle shifts in fecal microbiota diversity during early stages of Mycobacterium avium subsp. paratuberculosis pathogenesis in dairy cattle.

Mycobacterium avium subsp. paratuberculosis (MAP) in cattle causes an intestinal infection that can induce persistent inflammation or tissue remodeling. Several studies have reported differences in microbial diversity between MAP-infected and non-infected ruminants. Still, most of these studies have focused on animals in advanced stages of the disease, whether subclinical or clinical. The present study aimed to describe fecal microbiota composition in dairy cattle during early stages of MAP-associated pathological processes, which remain poorly characterized. A cross-sectional study was conduc

Major membrane protein of Mycobacterium avium subp. paratuberculosis activates immune and autophagic pathways in bovine monocyte-derived macrophages.

Mycobacterium avium subspecies paratuberculosis (Map), the etiological agent of Johne's disease in ruminants, poses challenges to veterinary health and food safety. Despite an immune response that partially controls early infection, Map persists in macrophages through mechanisms not well understood. Here, we explored how the Map major membrane protein (MMP) modulates immune pathways in bovine monocyte-derived macrophages (MoMΦs). MMP is a key component of the bacterial cell membrane recognized in cattle with Johne's disease, making it a critical antigenic target for immune studies. Using high-

Functional analysis of the intracellular survival of Mycobacterium avium subsp. paratuberculosis in THP-1 cells using CRISPR interference.

UNLABELLED: Mycobacterium avium subsp. paratuberculosis (MAP) is a causative agent of Johne's disease in ruminants and a potential zoonotic agent linked with Crohn's disease in humans. Despite the possible risk to public health, few studies have focused on the virulence of MAP against human macrophages. Therefore, a functional analysis of mycobacterial genes associated with virulence, especially the intracellular survival of MAP, was performed after infection of MAP CRISPR interference (CRISPRi) mutants in the human THP-1 macrophages. MAP mutants were targeted to four genes (mdh, pknG, MAP1981

Cross-species virulence strategies of Mycobacterium avium subsp. paratuberculosis: Gene expression and infection progression in sheep and guanacos.

Pathogenic bacteria adapt to their hosts modulating virulence-gene expression. This flexible and responsive virulence is frequently associated with the capacity of bacterial pathogens to infect a variety of host species, including wildlife and domestic animals. Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease and infects a broad range of ruminant-like herbivores. While its virulence has been well-studied in livestock, its behavior in wildlife hosts remains poorly understood. By studying infection progression and virulence-gene expression, we gain impo

Mycobacterium avium subspecies paratuberculosis (MAP) shedding and seropositivity in youngstock in MAP-infected dairy herds in Alberta, Canada.

Johne's disease (JD) control and eradication programs aim to reduce the prevalence of Mycobacterium avium subspecies paratuberculosis (MAP) infections and associated economic losses. Youngstock are particularly vulnerable to MAP infection-they can shed MAP in feces, develop antibody titers, and transmit the pathogen to pen mates. However, most JD control programs do not include youngstock in MAP testing strategies, potentially limiting their effectiveness by overlooking this transmission route. This study aimed to investigate the age at which youngstock ≤12 mo old in MAP-positive dairy herds s

An Escherichia coli carrier vaccine with surface-displayed protein MAP3061c elicits protective immunity against Mycobacterium paratuberculosis in mice.

Johne's disease, or paratuberculosis, is a chronic granulomatous enteritis of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). This disease occurs worldwide and results in considerable economic losses in the livestock industry. There are no effective treatments for Johne's disease, so there is an urgent need to develop an efficient, economical, and stable vaccine for MAP control. Here, a live Escherichia coli (E. coli) surface display vaccine harboring the MAP3061c gene was developed through an ice nucleation protein (INP) surface display system. The experimental data dem

In vivo kinetics of peripheral cellular immune responses in Mycobacterium avium subspecies paratuberculosis (MAP) infected and vaccinated goats.

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of paratuberculosis (ParaTB) also known as Johne's disease (JD) in ruminants, which is characterized by chronic intestinal inflammation. A similar counterpart has been observed in the form of Crohn's disease in humans. The present study is the first trail in goats to understand the peripheral cellular immune responses following experimental MAP infection and vaccination. Fifteen apparently healthy male kids (3-6 months old) of Barbari breed were included in this study. In the experimental study, 5 kids were infected w

WC1+ and WC1neg γδ T lymphocytes in intestinal mucosa of healthy and Mycobacterium avium subspecies paratuberculosis-infected calves.

Mucosal surfaces such as the gastrointestinal tract, and skin are the front line of host defence and immunity against many pathogens. Gamma delta (γδ) T lymphocytes preferentially localize to the mucosal surfaces in several species including cattle, and are thought to play crucial roles in immunosurveillance and host defence, particularly against mycobacteria. Many γδ T cells are present in young calves, which is the period when calves are thought to be initially exposed to Mycobacterium avium subspecies paratuberculosis (Map). The role of mucosal γδ T cells in cattle, especially during host-p

Characterization of Mycobacterium avium subspecies paratuberculosis disseminated infection in dairy cattle and its association with antemortem test results.

Mycobacterium avium subspecies paratuberculosis (MAP) disseminated infection in dairy cattle affects animal health and productivity and is also a potential public health concern. The study objectives were to characterize MAP disseminated infection in dairy cattle and to determine the role of antemortem tests in detecting cattle with disseminated infection. Forty culled dairy cows representing a variety of serum enzyme-linked immunosorbent assay (ELISA) results and body conditions were selected for the study. The physical condition of the cows was assessed via clinical examination prior to euth

A disease resembling paratuberculosis (Johne's disease) in roe deer (Capreolus capreolus L.). An aetiological and patho-anatomical study.

A disease in wild living roe deer (Capreolus capreolus L.) caused by acid-fast bacteria is described. The morphological and cultural properties of these bacteria agree closely with corresponding properties of Mycobact. johnei. Enlarged lymph nodes, especially mesenteric lymph nodes with greyish-yellow necroses, were the most prominent macroscopic lesions. No intestinal lesions were present. The histopathological picture of the lymph nodes resembled mostly lesions which can occur in paratuberculous sheep and goats. The epithelioid cells contained masses of acid-fast bacteria. Such rods were als

Comprehensive genome analysis of Mycobacterium avium subsp. paratuberculosis in camels from Saudi Arabia: Molecular epidemiology and antimicrobial resistance.

BACKGROUND AND AIM: Paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a significant infectious disease affecting livestock, including camels in Saudi Arabia, leading to severe economic losses. Despite its impact, limited genomic studies have been conducted to characterize MAP strains in camels and their antimicrobial resistance (AMR) profiles. This study aimed to (1) characterize the clinical and pathological findings of MAP infections in camels; (2) determine the seroprevalence of MAP in the eastern region of Saudi Arabia; (3) differentiate between MAP strains

Differential role of M cells in enteroid infection by Mycobacterium avium subsp. paratuberculosis and Salmonella enterica serovar Typhimurium.

Infection of ruminants such as cattle with Mycobacterium avium subsp. paratuberculosis (MAP) causes Johne's disease, a disease characterized by chronic inflammation of the small intestine and diarrhoea. Infection with MAP is acquired via the faecal-to-oral route and the pathogen initially invades the epithelial lining of the small intestine. In this study we used an in vitro 3D mouse enteroid model to determine the influence of M cells in infection of the gut epithelia by MAP, in comparison with another bacterial intestinal pathogen of veterinary importance, Salmonella enterica serovar Typhimu

Evaluation of Immune Exhaustion and Co-Inhibitory Receptor Expression in Mycobacterium avium Subspecies paratuberculosis (MAP) Seropositive Diarrhoeic Bovines.

Mycobacterium avium subspecies paratuberculosis (MAP) infection leads to chronic, persistent granulomatous enteritis, causing prolonged diarrhoea and emaciation. The disease is managed using medications such as antibiotics, live vaccines, mycobacteriophage therapies and other treatments; however, a notable proportion of affected animals do not show improvement with this approach. We hypothesise that immunoinhibitory receptors TIM-3 (T cell immunoglobulin mucin protein-3) and PD-1 (Programmed death receptor 1) may be upregulated on Peripheral blood mononuclear cells (PBMCs) of MAP-seropositive

Macrophage Proteome Analysis at Different Stages of Mycobacterium avium Subspecies paratuberculosis Infection Reveals a Mechanism of Pathogen Dissemination.

Johne's disease is a chronic and usually fatal enteric infection of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP) and is responsible for hundreds of millions of dollars in losses for the agricultural industry. Natural infection typically begins with bacterial uptake and translocation through the epithelium of the small intestine, followed by ingestion by tissue macrophages and dissemination via the lymphatic or blood system throughout the body. To gain insights into the host responses and adaptation of MAP within phagocytic cells, we utilized the previously develope
