Endogenous opioid peptides in intrapartum uterine blood.

Endogenous opioid peptides have consistently been reported to exert significant influence on several parameters of reproductive physiology, including tolerance to pain, release of prolactin, pleasurable behavior patterns, and appetite satiety. Immunoreactive endorphins measured in several hundred samples of postpartum uterine blood had a mean value of 190 pg/ml and a range of 60 to 520 pg/ml. Individual values in 100 mothers who were delivered at the Northwest Hospital failed to correlate with doses of postpartum pain medications, infant feeding patterns, and recorded estimates of maternal bon

Gastrointestinal peptides--rôle in pathophysiology and disease.

Progress in gut hormone research has considerably increased our knowledge in gastrointestinal physiology. However, this knowledge has not yet helped the understanding of common gastrointestinal diseases. A pathophysiological role of gut hormones has been established only for rare conditions This is because the clinical significance of the gut hormones is difficult to evaluate. Morphological and biochemical methods used in classical endocrinology can rarely be applied to gastrointestinal endocrinology because of the special design of the gut hormone system. Also gut hormones and autonomous nerv

Studies on opioid peptides as endogenous anticonvulsants.

In both normal and hypophysectomized rats, electroconvulsive shock (ECS) produced a significant postictal rise in seizure threshold (S.T.) to flurothyl, a volatile convulsant. This ECS-induced increase in S.T. was markedly attenuated by naloxone (10 mg/kg s.c.), which itself did not alter basal S.T. A dose of bicuculline (0.1 mg/kg i.p.) which was slightly proconvulsant in the flurothyl test did not significantly alter the postictal rise in S.T. produced by ECS. With hypophysectomized rats, there was a 24% increase in basal S.T. to flurothyl challenge. Hypophysectomy had no influence on the po

Alzheimer's disease against peptides products of enzymatic cleavage APP protein: Biological, pathobiological and physico-chemical properties of fibrillating peptides.

Various peptides products of enzymatic cleavage of key for Alzheimer's disease Amyloid Precursor Protein (APP) are well known, but still are matter of scientific debate. The Aβ type products are especially challenging for experimental and medical research. This paper outlines several, still poorly known, biological and medical processes such as peptides biology, i.e., formation, biodistribution, translocation, transport and finally removal from brain compartments and body fluids like Intracellular Fluid (ICF), Cerebrospinal Fluid (CSF), Interstitial Fluid (ISF), blood serum or urine. Alzheimer's disease against peptides products of enzymatic cleavage of APP protein. Forming and variety of fibrillating peptides - some aspects.

Various and different peptides products resulting from enzymatic protein cleavage of Amyloid Precursor Proteins (APP) are the main agents in the pathophysiology of Alzheimer's disease (AD). Although relatively well-known, they still arouse interest leading to further intense and wide-ranging research. Their biology and physico-chemical properties still are challenging for basic, experimental research and are matter of scientific debate. The APP itself and its functions are still somewhat enigmatic and therefore it is also called the All Purpose Protein. Apart from well known amyloidogenic and

Brain delivery of proteins with shuttle peptides.

Therapeutic proteins have emerged as potential candidates for the management of neurodegenerative disorders affecting the central nervous system (CNS). However, their effective delivery to the CNS remains a major challenge, primarily due to the presence of the blood-brain barrier (BBB). To overcome this challenge, several brain-targeted delivery strategies based on peptides have been developed. These peptides harness endogenous BBB transport pathways to enhance the delivery of therapeutic proteins to the CNS. In order to develop new potential protein-based therapeutic candidates, considerable

Antimicrobial effects of novel Hermetia illucens peptides.

Antimicrobial resistance represents a significant global health challenge, implicated in nearly 5 million deaths per year. This study investigates the antimicrobial properties of selected peptides derived from the Black Soldier Fly (BSF), identified in silico. Ten synthesized peptides were evaluated in vitro against Gram-positive bacteria (Staphylococcus aureus, Bacillus cereus), Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa), and fungi (Aspergillus fumigatus, Candida albicans ). Cytotoxicity was assessed using human lung fibroblasts and red blood cells. Among the peptides,

Brain shuttle peptides derived from phage display.

Vascular beds within different tissues, including the brain, exhibit diverse molecular heterogeneity. Screening of peptide libraries using in vivo phage display exploits this heterogeneity, enabling the discovery of peptide ligands which selectively home to specific tissues. A prime example is identification of the iron-mimicking peptide CRTIGPSVC which undergoes receptor-mediated transport across the blood-brain barrier (BBB) by binding to the transferrin/transferrin receptor (TfR) complex. Various other brain-homing and BBB-crossing peptides have been discovered by in vivo phage display as w

De novo Folding Mechanisms of Lasso Peptides.

Lasso peptides adopt a distinctive [1]rotaxane conformation, yet the principles governing the folding of this kinetically trapped structure have remained elusive. Here, we integrated extensive molecular dynamics simulations and deep learning to elucidate the de novo folding mechanism of 20 lasso peptides lacking secondary post-translational modifications. We constructed Multi-Ensemble Markov Models for each lasso peptide and uncovered a universal uphill folding landscape with spontaneous folding probabilities consistently below 0.8%. Loop stability strongly correlated with folding propensity,

Structured peptides as vaccine antigens against infections.

The onset of a new infection by a pathogen requires a rapid and prompt response from the body. Here, we present a simple and effective strategy to stimulate antibody production by grafting immunogenic peptides onto a beta-strand peptide scaffold. Analysis of the method using a known immunogenic region, the disulfide loop of the PilA protein of the Pseudomonas aeruginosa pilus, shows that the molecule folds properly in solution and, upon vaccination of mice, provides significant protection against intravenous challenge with the P. aeruginosa PAK strain.

Extracellular defense of bacteria against antimicrobial peptides.

Antimicrobial peptides (AMPs) are short chains of amino acids naturally produced by all kingdoms of life, which exhibit broad-spectrum activity against bacteria, fungi, and viruses and thus play a crucial defense role in organisms. Unlike conventional antibiotics, AMPs are less prone to induce bacterial resistance since they can act on multiple targets, mainly affecting cell membranes. Thus, AMPs are considered promising antibiotic agents for medical applications. However, bacteria have developed different mechanisms to resist the action of AMPs, which operate at the extracellular, surface, an

Lasso Peptides-A New Weapon Against Superbugs.

The emergence of multi-drug-resistant bacteria (known as superbugs) represents one of the greatest challenges for human health and modern medicine. Due to their remarkable ability to rapidly develop resistance to currently used antibiotics, new molecular targets for bacteria and substances capable of effectively combating related infections are still being sought. Lasso (known also as lariat) peptides are an unusual subclass of ribosomally synthesized and post-translationally modified peptides (RiPPs) with a structurally constrained knotted fold resembling a lasso. They are synthesized by cert

Hypoxia in myocardial infarction and natriuretic peptides.

BACKGROUND: Mechanical stress on the heart is commonly considered the sole stimulus explaining the synthesis and release of circulating natriuretic peptides and their derivatives. While one of the most critical paradigms in cardiology is that mechanical load increases oxygen consumption, clinical studies on these peptides have neglected the relationship between mechanical stress and oxygen metabolism. At the cellular level, cardiac myocytes have a ubiquitous oxygen-sensing pathway mediated by a nuclear transcription factor, the hypoxia-inducible factor (HIF). Published studies indicate that th

Proteins and peptides responsible for bone remodeling.

Bones are living tissues that are periodically renewed through the bone remodeling process. It starts when monocyte-macrophage-type cells begin to proliferate and recruit into the bone tissue due to the action of various proteins. The proteins are generated by the body in response to a lack of calcium in the blood or a damaged bone tissue. Then, other chemical signals promote the differentiation of monocyte-macrophage-type cells into osteoclasts. Finally, osteoclasts carry out a process known as bone resorption in which they degrade bone tissue. After, bone resorption a process known as osteog

2024 FDA TIDES (Peptides and Oligonucleotides) Harvest.

In 2024, the FDA approved fifty novel drugs, including four peptides and oligonucleotides (TIDEs) (two pepTIDEs and two oligonucleoTIDEs), highlighting their increasing importance as effective alternatives to traditional drug classes. TIDEs provide essential therapies for complex diseases, such as genetic disorders, rather than merely addressing symptoms. In addition to oligonucleotide therapeutics for various genetic conditions, peptides became the first approved treatment for Rett Syndrome in 2023 and were also used to treat Niemann-Pick disease type C (NPC) in 2024. Interestingly, among the

Renalase peptides reduce pancreatitis severity in mice.

Acute pancreatitis, an acute inflammatory injury of the pancreas, lacks a specific treatment. The circulatory protein renalase is produced by the kidney and other tissues and has potent anti-inflammatory and prosurvival properties. Recombinant renalase can reduce the severity of mild cerulein pancreatitis; the activity is contained in a conserved 20 aa renalase site (RP220). Here, we investigated the therapeutic effects of renalase on pancreatitis using two clinically relevant models of acute pancreatitis. The ability of peptides containing the RP220 site to reduce injury in a 1-day post-endos

Antimicrobial peptides: Defending the mucosal epithelial barrier.

The recent epidemic caused by aerosolized SARS-CoV-2 virus illustrates the importance and vulnerability of the mucosal epithelial barrier against infection. Antimicrobial proteins and peptides (AMPs) are key to the epithelial barrier, providing immunity against microbes. In primitive life forms, AMPs protect the integument and the gut against pathogenic microbes. AMPs have also evolved in humans and other mammals to enhance newer, complex innate and adaptive immunity to favor the persistence of commensals over pathogenic microbes. The canonical AMPs are helictical peptides that form lethal por

Strategies to identify and develop antiviral peptides.

The emergence and re-emergence of viral pathogens capable of causing epidemics or pandemics pose a serious healthcare burden. Small molecule antivirals used in conventional therapy have given rise to the severe problem of viral resistance against them. Peptides are generally considered safe, effective and are less likely to induce viral resistance. Antiviral peptides can be identified from screening of phage display of combinational peptide libraries, peptide array libraries or designed against viral targets. Limitations of peptides such as bioavailability can be improved with chemical modific
