Receptor-like kinase-mediated cell wall integrity signaling in plant environmental stress responses.

Receptor-like kinases (RLKs) serve as central signaling hubs that integrate extracellular signals with intracellular responses to maintain cell wall integrity and cellular homeostasis. Their structural and functional diversity enables precise perception of developmental, immune, and environmental cues. Recent advances in structural biology, phosphoproteomics, and genome engineering have substantially deepened our understanding of RLK function and regulation. Harnessing this knowledge offers promising opportunities to engineer crops with enhanced resilience to biotic and abiotic stresses, contr

Plant- and Algae-Derived Compounds Enhance the Anticancer Activity of Doxorubicin in Colorectal Cancer Cell Lines.

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide, and the efficacy of conventional chemotherapy is frequently limited by systemic toxicity, chemoresistance, and tumor recurrence. Natural products derived from marine algae and plants have attracted increasing interest as multitarget adjuvant agents capable of modulating apoptosis, oxidative stress, and tumor-associated signaling pathways. In the present study, we evaluated the anticancer activity of commercially available formulations enriched in fucoxanthin, fucoidan, tocotrienols, astaxanthin, an

Plant Cell Wall-Inspired Interfacial Bridging Enables Ultrastrong and Tough Carbon Nanotube Fibers.

Achieving simultaneous enhancement of strength and toughness in carbon nanotube fibers (CNTFs) remains a persistent challenge due to inefficient interfacial load transfer, low nanotube alignment, and high porosity. Herein, inspired by the hierarchical architecture and borate-mediated cross-linking of plant cell walls, we report a bioinspired interfacial bridging strategy to fabricate ultrastrong and tough CNTFs. This approach involves the sequential infusion of poly(pyridobisimidazole) (PIPD) nanofibers and chains into CNTF networks, followed by borate-induced covalent cross-linking and mechan

Comparative genomics approach to infer ancestral cell karyotypes and reconstruct the evolutionary trajectories of plant chromosomes.

Chromosomes are key structural and functional components of heredity. Reconstruction of ancestral cell karyotypes (ACKs) and evolutionary trajectories of chromosomes (CETs) can reveal how structural and functional changes in chromosomes have occurred during evolution. The whole-genome duplication integrated analysis toolkit implements a telomere-centric model on the basis of the comparative analysis of gene collinearity within and between plant genomes to reconstruct the ACKs and CETs of many angiosperm plants whose genomes have been complicated by repeated polyploidization and subsequent repa

Promoting plant protoplast fusion via membrane decoration with cell-penetrating peptide-conjugated lipids.

Protoplast fusion is a powerful plant cell engineering approach that enables the formation of new hybrids and facilitates genetic exchange across species barriers. However, conventional fusion techniques, such as polyethylene glycol (PEG)-induced fusion and electrofusion, often have low efficiency and poor reproducibility. In this study, we used Tat peptide-conjugated PEG-lipids (Tat-PEG-lipids) with different alkyl chain lengths (C9, C12, and C14) to examine the effect of PEG-lipid conjugates on fusion efficiency in rice (Oryza sativa L. cv. Nipponbare) protoplasts. Initially, we used rhodami

Targeted imaging of specialized plant cell walls by improved cryo-CLEM and cryo-electron tomography.

Cryo-focused ion beam scanning electron microscopy (Cryo-FIBSEM) has become essential for preparing cryo-lamellae. Here, we present a series of improvements that speed up and enhance the efficiency of the Serial Lift-Out and SOLIST (Serialized On-grid Lift-In Sectioning for Tomography) procedures. We extend the cryo-FIBSEM session to 5 days and eliminate the need of copper or gold block intermediates and reduce curtaining effects. Finally, we report a routine to target lamellae with a precision of approximately 1 µm in X/Y/Z. We demonstrate the power of our improvements by targeting Casparian

PACER: a novel 3D plant cell wall model for the analysis of non-catalytic and enzymatic responses.

BACKGROUND: Substrate accessibility remains a key limitation to the efficient enzymatic deconstruction of lignocellulosic biomass. Limited substrate accessibility is often addressed by increasing enzyme loading, which increases process and product costs. Alternatively, considerable efforts are underway world-wide to identify amorphogenesis-inducing proteins and protein domains that increase the accessibility of carbohydrate-active enzymes to targeted lignocellulose components. RESULTS: We established a three-dimensional assay, PACER (plant cell wall model for the analysis of non-catalytic and

The antifungal plant defensin HsAFP1 induces autophagy, vacuolar dysfunction and cell cycle impairment in yeast.

The plant defensin HsAFP1 is characterized by broad-spectrum antifungal activity and induces apoptosis in Candida albicans. In this study, we performed a transcriptome analysis on C. albicans cultures treated with HsAFP1 to gain further insight in the antifungal mode of action of HsAFP1. Various genes coding for cell surface proteins, like glycosylphosphatidylinositol (GPI)-anchored proteins, and proteins involved in cation homeostasis, autophagy and in cell cycle were differentially expressed upon HsAFP1 treatment. The biological validation of these findings was performed in the model yeast S

Comprehensive Analyses of Nitric Oxide-Induced Plant Stem Cell-Related Genes in Arabidopsis thaliana.

Plant stem cells are pluripotent cells that have diverse applications in regenerative biology and medicine. However, their roles in plant growth and disease resistance are often overlooked. Using high-throughput RNA-seq data, we identified approximately 20 stem cell-related differentially expressed genes (DEGs) that were responsive to the nitric oxide (NO) donor S-nitrosocysteine (CySNO) after six hours of infiltration. Among these DEGs, the highest number of positive correlations (R ≥ 0.8) was observed for CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE) 12. Gene ontology (GO) terms for molec

Human ortholog of a plant salicylic acid receptor found in SK-N-SH cell line.

Our previous studies have described the purification and characterization of a novel plant NAD(P)-reductase like protein (RL) from the thermogenic appendix of the Sauromatum guttatum inflorescence. RL is mainly located in cytoplasm of thermogenic plants and it can act like a bistable switch. It adopts a compact conformation during heat-production and a more expanded conformation when heat is not generated. Addition of salicylic acid, a natural thermogenic inducer, at picomolar concentration to a solution of purified RL induced a discontinuous volume phase transition in which the volume of RL i

Interaction of plant cell signaling molecules, salicylic acid and jasmonic acid, with the mitochondria of Helicoverpa armigera.

The cotton bollworm, Helicoverpa armigera is a polyphagous pest in Asia, Africa, and the Mediterranean Europe. Salicylic acid (SA) and jasmonic acid (JA) are the cell signaling molecules produced in response to insect attack in plants. The effect of these signaling molecules was investigated on the oxidative phosphorylation and oxidative stress of H. armigera. SA significantly inhibited the state III and state IV respiration, respiratory control index (RCI), respiratory complexes I and II, induced mitochondrial swelling, and cytochrome c release in vitro. Under in vivo conditions, SA induced s

Producing cell-free culture broth of rhamnolipids as a cost-effective fungicide against plant pathogens.

Biosurfactants of rhamnolipids have been enthusiastically investigated for substitutes of synthetic agrochemicals against plant pathogens. However, all such studies have been conducted on rhamnolipids with high purity which have limitations due to high costs. This paper focused on the applicability of rhamnolipid-containing cell-free culture broth. It was found that rhamnolipids in cell-free culture broth of Pseudomonas aeruginosa ZJU211 were largely composed of di-rhamnolipid and mono-rhamnolipid with the ratio varying with culture time. After 96 h of fermentation, the mass ratio of di-rhamno

Closer to the native state: Sandwich freezing and freeze-substitution of microorganisms, cultured cells, and animal and plant tissues for electron microscopy.

Since the invention of the electron microscope, morphologists have developed numerous techniques to visualise the native ultrastructure of biological cells and tissues. Although conventional chemical fixation followed by room temperature dehydration remains widely used, it often fails to preserve native structures, leading to distortion and leakage of cellular components. In contrast, cryofixation combined with freeze substitution offers a superior alternative, producing micrographs that more accurately reflect native ultrastructure. We have developed a 'sandwich freezing method' for rapid cry

[The role of reactive oxygen species and calcium ions in the implementation of the stress-protective effect of brassinosteroids on plant cells].

The effect of the brassinosteroids (BS) 24-epibrassiniolide and 24-epicastasterone on the thermoresistance of wheat coleoptile (Triticum aestivum L.) and their generation of the superoxide anion radical and antioxidant enzymes activity were investigated. The treatment of coleoptiles with 10 nM solutions of BS caused a transient increase in O2·- generation and a subsequent increase in the activity of superoxide dismutase and catalase and an improvement in heat resistance. Pretreatment of coleoptiles with the NADPH oxidase inhibitor imidazole leveled the increase in production of the superoxide

Suppression of inhibitor formation against FVIII in a murine model of hemophilia A by oral delivery of antigens bioencapsulated in plant cells.

Hemophilia A is the X-linked bleeding disorder caused by deficiency of coagulation factor VIII (FVIII). To address serious complications of inhibitory antibody formation in current replacement therapy, we created tobacco transplastomic lines expressing FVIII antigens, heavy chain (HC) and C2, fused with the transmucosal carrier, cholera toxin B subunit. Cholera toxin B-HC and cholera toxin B-C2 fusion proteins expressed up to 80 or 370 µg/g in fresh leaves, assembled into pentameric forms, and bound to GM1 receptors. Protection of FVIII antigen through bioencapsulation in plant cells and oral
