The combination of morphogenic regulators BABY BOOM and GRF-GIF improves maize transformation efficiency and promotes leaf regeneration.

Transformation is an indispensable tool for plant genetics and functional genomics. Although stable transformation in maize is no longer a major obstacle, there remains a need for accessible and efficient methods for academic laboratories. Here, we present the GGB system, a rapid and efficient approach optimized for immature embryo transformation in B104 and other maize lines. This system combines two distinct morphogenetic regulators, the wheat GRF4-GIF1 chimera and the maize BABY BOOM (BBM) transcription factor (hence the name "GGB") with a modified QuickCorn protocol, enabling regeneration

MYB168 and WRKY20 transcription factors synergistically regulate lignin monomer synthesis during potato tuber wound healing.

Lignin is a critical component of the closing layer of the potato (Solanum tuberosum L.) tuber during healing; however, the molecular mechanism of its formation remains poorly understood. To elucidate the molecular mechanism of tuber healing, we screened the genes encoding transcription factors that regulate lignin synthesis(StMYB24/49/105/144/168, StWRKY19/20/22/23/34) and the key genes involved in lignin monomer synthesis (PHENYLALANINE AMMONIA LYASE 5 (StPAL5) and CINNAMYL ALCOHOL DEHYDROGENASE 14 (StCAD14)) for induced expression after wounding using transcriptome data. Manipulating the Biosynthesis of Bioactive Compound Alkaloids for Next-Generation Metabolic Engineering in Opium Poppy Using CRISPR-Cas 9 Genome Editing Technology.

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated9 (Cas9) endonuclease system is a powerful RNA-guided genome editing tool. CRISPR/Cas9 has been well studied in model plant species for targeted genome editing. However, few studies have been reported on plant species without whole genome sequence information. Currently, no study has been performed to manipulate metabolic pathways using CRISPR/Cas9. In this study, the type II CRISPR/SpCas9 system was used to knock out, via nonhomologous end-joining genome repair, the 4'OMT2 in opium poppy (Papaver somniferum L.

A Simple RUBY Reporter System in Nicotiana benthamiana for Studying Inositol Pyrophosphate Dynamics.

In recent years, there has been substantial progress in the development of methods to analyze inositol phosphates (InsPs) and inositol pyrophosphates (PP-InsPs). However, many of these techniques are labor- and cost-intensive and can usually only be carried out by laboratories specialized in InsPs/PP-InsPs analysis. In this chapter, we present a simple method that exploits the fact that phosphorylation and/or dephosphorylation of certain InsP/PP-InsP species induces the activation of promoters driving the expression of genes involved in phosphate starvation response (PSR). By linking PSR-induc

Transcription factor PbNAC71 regulates xylem and vessel development to control plant height.

Dwarfism is an important agronomic trait in fruit breeding programs. However, the germplasm resources required to generate dwarf pear (Pyrus spp.) varieties are limited. Moreover, the mechanisms underlying dwarfism remain unclear. In this study, "Yunnan" quince (Cydonia oblonga Mill.) had a dwarfing effect on "Zaosu" pear. Additionally, the dwarfism-related NAC transcription factor gene PbNAC71 was isolated from pear trees comprising "Zaosu" (scion) grafted onto "Yunnan" quince (rootstock). Transgenic Nicotiana benthamiana and pear OHF-333 (Pyrus communis) plants overexpressing PbNAC71 exhibit

Ghd7‑triggered transcriptional regulation for synergistically improving rice eating quality and yield.

Improving both eating quality and yield remains a major challenge in rice breeding. Here, we identify Ghd7 as the causal gene underlying the grain amylose content QTL qAC7 and reveal Ghd7-triggered transcriptional regulation that balances starch and protein to coordinate yield and quality. Ghd7 transcriptionally activates PCF3 and exhibits dominant epistasis over it. A natural TA/T variant in PCF3 determines its DNA-binding affinity: the PCF3‑T allele strongly represses OsNF-YC4, increasing starch and reducing protein in grain, whereas PCF3‑TA fails to exert this repressive effect. The transcription factor EIL1-MYB78 module negatively regulates cold tolerance in Actinidia chinensis.

Kiwifruit has become an important horticultural crop widely planted in temperate and subtropical regions. However, most commercially cultivated kiwifruit, particularly those of Actinidia chinensis, are highly sensitive to cold stress. To overcome this industrial bottleneck, it is crucial to reveal the molecular mechanisms behind cold stress responses, ultimately providing new insights into the breeding of cold-tolerant kiwifruit varieties. Here, we found that EIL1 negatively regulates cold tolerance of A. chinensis by directly binding to the promoters of CBF1 and CBF2 to inhibit their expressi

SmYABBY1, a Light-Inducible Transcription Factor, Positively Regulates Anthocyanin Biosynthesis in Eggplant (Solanum melongena L.).

Anthocyanin biosynthesis in eggplant (Solanum melongena L.) is highly light-dependent, and insufficient light severely impairs fruit coloration, which restricts the development of the eggplant industry. SmMYB75 is a key positive regulator of anthocyanin biosynthesis, but its regulatory partners remain unclear. In this study, seven SmYABBY genes were identified from the eggplant genome, all containing conserved zinc finger and YABBY domains. Expression analysis showed that SmYABBY1 was predominantly expressed in fruit peel and significantly induced by light, with a peak at 4 h after light expos

Functional characterization of the grape VvCAD15-like gene in enhancing cold tolerance via regulation of lignin biosynthesis.

Cinnamyl alcohol dehydrogenase (CAD) is a key enzyme in lignin biosynthesis, but its role in grapevine cold tolerance is not well understood. Here we identified 23 VvCAD members from the 'Pinot Noir' genome and focused on VvCAD15-like, a Group II member whose expression was induced by cold. Transient overexpression of this gene in grape calli and plantlets increased lignin content, antioxidant enzyme activities (CAT, POD, SOD), and proline level, while decreased MDA, H₂O₂, and membrane permeability. It also upregulated VvCBF1/3 and VvICE1a. Transcriptome data implied potential links between Vv

GhWRKY21 regulates ABA-mediated drought tolerance by fine-tuning the expression of GhHAB in cotton.

We report that GhWRKY21, a WRKY transcription factor, plays essential roles in regulating the intensity of the drought-induced ABA signalling pathway by facilitating the expression of GhHAB in cotton (Gossypium hirsutum). Abscisic acid (ABA) is one of the most important plant hormones in response to abiotic stress. However, activation of the ABA signalling pathway often leads to growth inhibition. The mechanisms that regulate the intensity of ABA signals are poorly understood. Here, we isolated and analysed the cotton group IId WRKY transcription factor (TF) gene GhWRKY21. Functional analysis

Utilization of Legume-Nodule Bacterial Symbiosis in Phytoremediation of Heavy Metal-Contaminated Soils.

With the increasing industrial activity of the growing human population, the accumulation of various contaminants in soil, including heavy metals, has increased rapidly. Heavy metals as non-biodegradable elements persist in the soil environment and may pollute crop plants, further accumulating in the human body causing serious conditions. Hence, phytoremediation of land contamination as an environmental restoration technology is desirable for both human health and broad-sense ecology. Legumes (Fabaceae), which play a special role in nitrogen cycling, are dominant plants in contaminated areas.

Transgenic pearl millet male fertility restorer line (ICMP451) and hybrid (ICMH451) expressing Brassica juncea Nonexpressor of pathogenesis related genes 1 (BjNPR1) exhibit resistance to downy mildew disease.

Brassica juncea Nonexpressor of pathogenesis-related genes 1 (BjNPR1) has been introduced into pearl millet male fertility restorer line ICMP451 by Agrobacterium tumefaciens-mediated genetic transformation. Transgenic pearl millet plants were regenerated from the phosphinothricin-resistant calli obtained after co-cultivation with A. tumefaciens strain LBA4404 harbouring Ti plasmid pSB111-bar-BjNPR1. Molecular analyses confirmed the stable integration and expression of BjNPR1 in transgenic pearl millet lines. Transgenes BjNPR1 and bar were stably inherited and disclosed co-segregation in subseq

Identification and functional characterization of GhCSLD3, a negative regulator of fruit branch elongation in cotton (Gossypium hirsutum L.).

The fruit branch is a critical determinant of plant architecture in cotton, and its length optimization is pivotal for high-density planting and mechanical harvesting. However, the genetic mechanisms governing fruit branch length (FBL) are not fully elucidated. In this study, an F2 population was generated from two contrasting upland cotton cultivars, Blanco3363 (BL, compact) and Sanjiang (SJ, loose). Using two different methodologies of bulked-segregant analysis (BSA)-seq, we identified three stable overlapping QTLs on chromosomes A08 (119.8-122.46 Mb), D01 (7.51-10.10 Mb), and D09 (16.33-18.

MdUGT88F1 enhances plant resistance to Fusarium proliferatum f.sp. malus domestica MR5 via root exudate-mediated assembly of disease-suppressive rhizosphere microbiota.

BACKGROUND: Apple replant disease (ARD) is a major threat to the sustainable development of China's apple industry. It is primarily caused by the accumulation of phloridzin and the pathogen Fusarium proliferatum f.sp. malus domestica MR5 (Fpmd MR5). MdUGT88F1-mediated phloridzin biosynthesis is known to enhance disease resistance, but its role in shaping the rhizosphere microbiome and conferring resistance against Fpmd MR5 remains unclear. In this study, we used wild-type (WT) and MdUGT88F1 transgenic apple lines to systematically investigate the mechanism by which MdUGT88F1 regulates the rhiz

Development of semi-dwarf Echinacea purpurea by inducing silencing of the endogenous brassinosteroid-biosynthetic gene DWF4.

Brassinosteroids (BRs) are plant steroidal hormones that are crucial for plant growth and development, with dwarfism being a hallmark of BR deficiency. DWARF4 (DWF4) is a key enzyme that catalyzes the rate-limiting C22 oxidation step in the BR biosynthetic pathway. Understanding its role in ornamental plants is crucial for developing dwarf varieties that can significantly enhance their aesthetics and broaden their applications. This study aimed to provide the functional analysis of the DWF4 gene isolated from Echinacea purpurea. We found that the EpDWF4 gene comprised a 1458 bp open reading fr

Functional characterization of CsSTK in floral organ development and its positive regulation by CsBPC5 in tea plants.

BACKGROUND: As an important economic crop, the development of floral organs in tea plants plays an essential role in tea yield, quality, and varietal improvement. Previous studies have shown significant downregulation of the CsSTK gene in the pistil-deficient mutant 'Yuncha Qirui' (CRQS) during flower development, suggesting its involvement in floral organ and pistil development. RESULTS: This study investigated CsSTK function and regulatory mechanisms, concurrently conducting a systematic identification of the BPC transcription factor family members in tea plants. The results showed that CsS

Antifungal Activity of Ribosome-Inactivating Proteins.

The control of crop diseases caused by fungi remains a major problem and there is a need to find effective fungicides that are environmentally friendly. Plants are an excellent source for this purpose because they have developed defense mechanisms to cope with fungal infections. Among the plant proteins that play a role in defense are ribosome-inactivating proteins (RIPs), enzymes obtained mainly from angiosperms that, in addition to inactivating ribosomes, have been studied as antiviral, fungicidal, and insecticidal proteins. In this review, we summarize and discuss the potential use of RIPs

Hydrogen sulfide antagonizes cytokinin to change root system architecture through persulfidation of CKX2 in Arabidopsis.

Hydrogen sulfide (H2S) is an endogenous gaseous signaling molecule, which has been shown to play an important role in plant growth and development by coupling with various phytohormones. However, the relationship between H2S and cytokinin (CTK) and the mechanisms by which H2S and CTK affect root growth remain poorly understood. Endogenous CTK was analyzed by UHPLC-ESI-MS/MS. Persulfidation of cytokinin oxidase/dehydrogenases (CKXs) was analyzed by mass spectrometry (MS). ckx2/CKX2wild-type (WT), OE CKX2 and ckx2/CKX2Cys(C)62alanine(A) transgenic lines were isolated with the ckx2 background. A novel OsGST gene encoding 9glutathione reductase negatively regulates cadmium accumulation in rice.

Cadmium (Cd) accumulates in rice and then moves up the food chain, causing serious health problems for humans. Glutathione S-transferase (GST) binds exogenous hazardous compounds to glutathione (GSH), which performs a variety of roles in plant responses to Cd stress. Here, Cd stimulated the transcripts of a novel OsGST gene, and the OsGST protein, which was localized in the nucleus and cytoplasm, was also induced by Cd. In OsGST deletion mutant lines generated by CRISPR/Cas9, more Cd was accumulated, and Cd hypersensitive phenotypes were observed, while transgenic lines overexpressing OsGST ex

Overexpression of CcFALDH from spider plant (Chlorophytum comosum) enhances the formaldehyde removing capacity of transgenic gloxinia (Sinningia speciosa)1.

Formaldehyde can cause leukemia and nasopharyngeal cancer in humans, and is a major indoor air pollutant. In this study, to improve the ability of flowering plants to purify formaldehyde, we cloned the CcFALDH gene encoding formaldehyde dehydrogenase (FALDH) from the spider plant (Chlorophytum comosum), which encodes 379 amino acids with the alcohol dehydrogenase (ADH) structural domain, and used it to transform the flowering plant gloxinia (Sinningia speciosa). The FALDH activity of transgenic gloxinia was 1.8-2.7 times that of wild-type (WT) with a considerable increase in formaldehyde stres
