Knee osteoarthritis and time-to all-cause mortality in six community-based cohorts: an international meta-analysis of individual participant-level data.

BACKGROUND: Osteoarthritis (OA) is a chronic joint disease, with increasing global burden of disability and healthcare utilisation. Recent meta-analyses have shown a range of effects of OA on mortality, reflecting different OA definitions and study methods. We seek to overcome limitations introduced when using aggregate results by gathering individual participant-level data (IPD) from international observational studies and standardising methods to determine the association of knee OA with mortality in the general population. METHODS: Seven community-based cohorts were identified containing k

Unsaturated Fatty Acids Affect Quorum Sensing Communication System and Inhibit Motility and Biofilm Formation of Acinetobacter baumannii.

The increasing threat of Acinetobacter baumannii as a nosocomial pathogen is mainly due to the occurrence of multidrug-resistant strains that are associated with the real problem of its eradication from hospital wards. The particular ability of this pathogen to form biofilms contributes to its persistence, increases antibiotic resistance, and promotes persistent/device-related infections. We previously demonstrated that virstatin, which is a small organic compound known to decrease virulence of Vibrio cholera via an inhibition of T4-pili expression, displayed very promising activity to prevent

medicalcoder: a unified and longitudinally aware framework for International Classification of Diseases code-based comorbidity assessment in R.

OBJECTIVES: Comorbidity algorithms derived from International Classification of Diseases (ICD) codes are central to risk adjustment and cohort characterization, but existing software inconsistently handles mixed ICD-9/10 data and typically operates at the encounter level. We developed medicalcoder to address these limitations. MATERIALS AND METHODS: medicalcoder is an R package with an internal ICD database and unified interface for applying variants of the Charlson, Elixhauser, and Pediatric Complex Chronic Conditions (PCCC) algorithms. It supports full and compact ICD codes, mixed ICD versi

Tyrosine hydroxylase in the olfactory system, forebrain and pituitary of the Indian major carp, Cirrhinus cirrhosus: organisation and interaction with neuropeptide Y in the preoptic area.

Dopamine (DA) inhibits, whereas gonadotrophin-releasing hormone (GnRH) stimulates, luteinisiing (LH) cells in the pituitary of some but not all teleosts. A reduction in the hypophysiotropic dopaminergic tone is necessary for the stimulatory effect of GnRH on LH cells. Neuropeptide Y (NPY) has emerged as one of the potent, endogenous agent that modulates LH secretion directly or indirectly via GnRH. Involvement of NPY in the regulation of hypophysiotropic DA neurones, however, is not known, but there is good evidence suggesting an interaction in the mammalian hypothalamus. DA neurones, identifi

CCR1 and CCR5 chemokine receptors are involved in fever induced by LPS (E. coli) and RANTES in rats.

This study, besides examining the involvement of CCR1 and CCR5 receptors in the LPS-induced fever (lipopolysaccharide, Escherichia coli) in male Wistar rats, evaluated if RANTES (regulated on activation, normal T cells expressed and secreted) injected into the preoptic area of the anterior hypothalamus (AH/POA) would promote an integrated febrile response via these receptors. Moreover, the effects of selective and non-selective cyclooxygenase blockers on both fever and the level of prostaglandin (PG)E(2) in the cerebrospinal fluid (CSF) after injection of RANTES into the AH/POA were also inves

Does the detoxification of penicillin side-chain precursors depend on microsomal monooxygenase and glutathione S-transferase in Penicillium chrysogenum?

The glutathione (GSH) S-conjugation of 1,2-epoxy-3-(4'-nitrophenoxy)propane was catalysed predominantly by microsomal glutathione S-transferase (mGST) in Penicillium chrysogenum. The specific mGST activity unlike the cytosolic GST (cGST) activity increased substantially when the penicillin side-chain precursor phenoxyacetic acid (POA) was included in the culture medium. Therefore, a microsomal monooxygenase (causing possible release of epoxide intermediates) and mGST-dependent detoxification pathway may exist for the side-chain precursors as an alternative to microsomal activation to acyl-CoA

Stress ensemble in the BNST mediates comorbid anxiety and sleep disruption.

Anxiety and insomnia are highly comorbid; however, the underlying brain-circuit mechanisms remain inadequately explored. Here, we identify a dual-functional ensemble within the bed nucleus of the stria terminalis (BNST) that modulates both anxiety-like behaviors and insomnia through distinct neural circuits. Specifically, a subset of BNST neurons exhibits enhanced activity in response to anxiety-related cues and during wakefulness. Activation of these ensemble neurons exacerbates anxiety-like behaviors and reduces non-rapid eye movement (NREM) sleep in chronic restraint stress (CRS) mice. Conv

Natural product P57 induces hypothermia through targeting pyridoxal kinase.

Induction of hypothermia during hibernation/torpor enables certain mammals to survive under extreme environmental conditions. However, pharmacological induction of hypothermia in most mammals remains a huge challenge. Here we show that a natural product P57 promptly induces hypothermia and decreases energy expenditure in mice. Mechanistically, P57 inhibits the kinase activity of pyridoxal kinase (PDXK), a key metabolic enzyme of vitamin B6 catalyzing phosphorylation of pyridoxal (PL), resulting in the accumulation of PL in hypothalamus to cause hypothermia. The hypothermia induced by P57 is si

Effects of deoxycorticosterone treatment on beta-subunit mRNA for (Na + K)ATPase in brain regions determined by in situ hybridization.

1. We have used in situ hybridization techniques to determine the mRNA for (Na + K)ATPase in 20 brain regions from control rats and rats treated with high doses of deoxycorticosterone (DOC). 2. DOC-treated rats developed a salt appetite following the second hormone administration on alternate days and were used after the fourth DOC administration. 3. DOC treatment did not change the number of silver grains/cell deposited in cells from Ca1, CA2, CA3, and CA4 hippocampal subfields, dentate gyrus, cerebral cortex, medial preoptic area (POA), substantia nigra, and periventricular gray matter. 4. Comparative outcomes of total knee arthroplasty in patients with posttraumatic versus primary knee osteoarthritis.

INTRODUCTION: Posttraumatic osteoarthritis (ptOA) of the knee accounts for 12.5% of all total knee arthroplasties (TKA) and is associated with significantly higher complication and revision rates than in TKA for primary osteoarthritis (pOA). However, the impact of ptOA on patient-reported outcomes is less clear. The purpose of this study is to analyse longitudinal clinical and patient-reported outcomes in these patient groups. MATERIALS AND METHODS: A retrospective analysis of prospectively collected data from an institutional database was performed (time frame 2006-2023). Patients with a his

Thyrotropin-releasing hormone (TRH) in the brain and pituitary of the teleost, Clarias batrachus and its role in regulation of hypophysiotropic dopamine neurons.

Thyrotropin-releasing hormone (TRH) regulates the hypothalamic-pituitary-thyroid axis in mammals and also regulates prolactin secretion, directly or indirectly via tuberoinfundibular dopamine neurons. Although TRH is abundantly expressed in teleost brain and believed to mediate neuronal communication, empirical evidence is lacking. We analyzed pro-TRH-mRNA expression, mapped TRH-immunoreactive elements in the brain and pituitary, and explored its role in regulation of hypophysiotropic dopamine (DA) neurons in the catfish, Clarias batrachus. Partial pro-TRH transcript from C. batrachus transcri

Glutamine synthetase genes family mediates nitrogen metabolism and disease responses in Kentucky bluegrass.

BACKGROUND: Kentucky bluegrass (Poa pratensis L.), an important cool-season turfgrass species, requires adequate nitrogen (N) fertilization for normal growth and development. It is also susceptible to infection by pathogens. The glutamine synthetase (GS) gene family, known to play central roles in plant N metabolism and disease responses across higher plants, has functions that remain uncharacterized in Kentucky bluegrass. RESULTS: We identified and characterized four GS genes (PpGS1.1, PpGS1.2, PpGS1.3, and PpGS2) in Kentucky bluegrass. Phylogenetic analysis revealed a close relationship wit

Changing leaf litter feedbacks on plant production across contrasting sub-arctic peatland species and growth forms.

Plant species and growth forms differ widely in litter chemistry, which affects decay and may have important consequences for plant growth via e.g. the release of nutrients and growth-inhibitory compounds. We investigated the overall short-term (9.5 months) and medium-term (21.5 months) feedback effects of leaf litter quality and quantity on plant production, and tested whether growth forms can be used to generalise differences among litter species. Leaf litter effects of 21 sub-arctic vascular peatland species on Poa alpina test plants changed clearly with time. Across all growth forms, litte

Allelopathic, Phytotoxic, and Insecticidal Effects of Thymus proximus Serg. Essential Oil and Its Major Constituents.

The chemical profile of Thymus proximus essential oil (EO) and its allelopathic, phytotoxic, and insecticidal activity was evaluated. Carvacrol, p-cymene, and γ-terpinene were detected as the major components of the EO, representing 85.9% of the total oil. About 50 g fresh plant material of T. proximus in a 1.5-L air tight container completely inhibited the seed germination of Amaranthus retroflexus and Poa anuua. Meanwhile, the EO exhibited potent phytotoxic activity, which resulted in 100% germination failure of both the test species when 2 mg/ml (for A. retroflexus) and 5 mg/ml (for Poa ann

Endocrine correlates of partner preference behavior in rams.

We studied a unique group of rams that would not mate with estrous ewes during extensive testing for sexual behavior. The same rams courted males in preference to females in 30-min sexual preference tests and were classified as male-oriented (n = 6). We compared the following endocrine profiles: systemic steroid concentrations, the capacity of the testes to biosynthesize 17 alpha-hydroxyprogesterone and testosterone from 3H-progesterone in vitro, and the levels of brain aromatase activity (AA) in male-oriented rams vs. rams that were proven breeders and designated as female-oriented (n = 7). Layered confinement of protein in synthetic fluorinated mica via stepwise polyamine exchange.

We have developed a process to incorporate the model protein, bovine serum albumin (BSA), into the layered spacing of swelled mica. By a stepwise intercalation, the sodium form of synthetic fluorinated mica (Mica) was first exchanged with the poly(oxyalkylene)-diamine salts (POA-amine) through an ionic exchange reaction and then the BSA embedment. The first step of the Mica space expansion from the pristine 12 A to 18-93 A was affected by hydrophobic POP-amines (POP2000 of 2000 g/mol and POP4000 of 4000 g/mol M(w)) and the hydrophilic POE2000 that intercalated Na+-Mica to afford different basa

Palmitoleic acid promoted by BMPR2 signaling primes CD169 macrophages and alleviates liver fibrosis.

Bone morphogenetic proteins (BMPs) participate in the energy metabolism. BMP receptor type 2 (BMPR2) is expressed in the liver. Whether BMPR2 is involved in the pathophysiology of nonalcoholic fatty liver disease (NAFLD) has not been studied. In this study, we analyzed the RNA-sequence data of several patient cohorts and found that BMPR2 expression decreases at the stages of advanced fibrosis. A mouse model featuring BMPR2 knockout demonstrate that BMPR2 knockout in the liver produces profibrotic effect upon long-term high-fat and high-fructose (HFF) diet challenge. BMP signaling promotes bios

Neurological basis of AMP-dependent thermoregulation and its relevance to central and peripheral hyperthermia.

Therapeutic hypothermia is of relevance to treatment of increased body temperature and brain injury, but drugs inducing selective, rapid, and safe cooling in humans are not available. Here, we show that injections of adenosine 5'-monophosphate (AMP), an endogenous nucleotide, promptly triggers hypothermia in mice by directly activating adenosine A1 receptors (A1R) within the preoptic area (POA) of the hypothalamus. Inhibition of constitutive degradation of brain extracellular AMP by targeting ecto 5'-nucleotidase, also suffices to prompt hypothermia in rodents. Accordingly, sensitivity of mice

Effects of chronic growth hormone overexpression on appetite-regulating brain gene expression in coho salmon.

Organisms must carefully regulate energy intake and expenditure to balance growth and trade-offs with other physiological processes. This regulation is influenced by key pathways controlling appetite, feeding behaviour and energy homeostasis. Growth hormone (GH) transgenesis provides a model where food intake can be elevated, and is associated with dramatic modifications of growth, metabolism, and feeding behaviour, particularly in fish. RNA-Seq and qPCR analyses were used to compare the expression of multiple genes important in appetite regulation within brain regions and the pituitary gland
