alpha-, beta-, and gamma-Tubulin polymerization in response to DNA damage.

Microtubules provide structural support for a cell and play key roles in cell motility, mitosis, and meiosis. They are also the targets of several anticancer agents, indicating their importance in maintaining cell viability. We have investigated the possibility that alterations in microtubule structure and tubulin polymerization may be part of the cellular response to DNA damage. In this report, we find that gamma-radiation stimulates the production and polymerization of alpha-, beta-, and gamma- tubulin in hematopoeitic cell lines (Ramos, DP16), leading to visible changes in microtubule struc

ActA and human zyxin harbour Arp2/3-independent actin-polymerization activity.

The actin cytoskeleton is a dynamic network that is composed of a variety of F-actin structures. To understand how these structures are produced, we tested the capacity of proteins to direct actin polymerization in a bead assay in vitro and in a mitochondrial-targeting assay in cells. We found that human zyxin and the related protein ActA of Listeria monocytogenes can generate new actin structures in a vasodilator-stimulated phosphoprotein-dependent (VASP) manner, but independently of the Arp2/3 complex. These results are consistent with the concept that there are multiple actin-polymerization

A WASp-VASP complex regulates actin polymerization at the plasma membrane.

Proteins of the Wiskott-Aldrich syndrome and Ena/VASP families both play essential functions in the regulation of actin dynamics at the cell leading edge. However, possibilities of functional interplay between members of these two families have not been addressed. Here we show that, in hemopoietic cells, recruitment of the C-terminal VCA (Verprolin homology, Cofilin homology, Acidic) domain of WASp at the plasma membrane by a ligand technique using rapamycin as an intermediate is not sufficient to elicit efficient Arp2/3 complex-mediated actin polymerization. Other domains of WASp, in particul

Hemoglobin polymerization in sickle cells studied by circular polarized light scattering.

We have studied intracellular polymerization of hemoglobin S in suspensions of small populations of sickle cells using circular polarized light scattering. We argue that the preferential scattering of right circular polarized light (as expressed by measurements of the S14 Mueller scattering matrix element) directly reflects the amount of polymer inside cells. This technique has made it possible to investigate the effect of oxygen tension, cell density and osmotic stress on intracellular hemoglobin polymerization. Using S14 to determine hemoglobin polymer, we show that the polymer increases wit

Concentration and degree of polymerization of hyaluronate in equine synovial fluid.

In addition to its well-known physicochemical properties, hyaluronate (HA) has recently been shown to have important biological and pathophysiologic regulatory effects on granulocytes, monocytes, fibroblasts, and endothelial cells, as well as on the healing of wounds and various joint disorders. Many of these effects depend on or are reflected in the concentration and degree of polymerization of HA. Therefore, high-performance liquid chromatography with size-exclusion column was used to characterize the concentration and degree of polymerization of HA in equine synovial fluid (SF). The mean (+

Stereochemical course of polymerization catalyzed by avian myeloblastosis virus reverse transcriptase.

The Sp diastereomer of thymidine 5'-O-(1-thiotriphosphate) was polymerized by avian myeloblastosis virus reverse transcriptase using poly(A) . d(pT)10 as template-primer. Degradation of the template poly(A) by alkaline hydrolysis and isolation by gel chromatography gave a single-stranded poly(d(p(S)T)), a polymer of thymidine 5'-phosphorothioate. To determine the configuration of the phosphorothioate internucleotide linkage, this material was degraded by snake venom phosphodiesterase. Comparison of the rates of degradation by snake venom phosphodiesterase of poly(d(p(S)T)) prepared by reverse

Rapid "one-pot" preparation of polymeric monolith via photo-initiated thiol-acrylate polymerization for capillary liquid chromatography.

A facile approach was exploited for fast preparation of polymer-based monoliths in UV-transparent fused-silica capillaries via "one-pot" photo-initiated thiol-acrylate polymerization reaction of dipentaerythritolpenta-/hexaacrylate (DPEPA) and 1-octadecanethiol (ODT) in the presence of porogenic solvents (1-butanol and ethylene glycol). Due to relative insensitivity of oxygen inhibition in thiol-ene free-radical polymerization, the polymerization could be performed within 5 min. The effects of composition of prepolymerization solution on the morphology and permeability of poly(ODT-co-DPEPA) mo

Polymeric Network Hierarchically Organized on Carbon Nano-onions: Block Polymerization as a Tool for the Controlled Formation of Specific Pore Diameters.

The organization of specific pores in carbonaceous three-dimensional networks is crucial for efficient electrocatalytic processes and electrochemical performance. Therefore, the synthesis of porous materials with ordered and well-defined pores is required in this field. The incorporation of carbon nanostructures into polymers can create material structures that are more ordered in comparison to those of the pristine polymers. In this study we applied polymer-templated methods of carbon material preparation, in which outer blocks of the star copolymers form the carbon skeleton, while the core p

Electrochemical ultrasensitive detection of miRNA-21 via RAFT/ROMP dual polymerization amplification.

The ultrasensitive detection of microRNAs in bodily fluids is crucial for early diagnosis of cancer. Herein, we design an electrochemical biosensor based on a dual-signal amplification strategy utilizing reversible addition-fragmentation chain transfer (RAFT) and ring-opening metathesis polymerization (ROMP) to achieve ultra-sensitive quantitative detection of miRNA-21. The sensing interface starts from a gold electrode carrying a thiol-immobilized peptide nucleic acid (PNA) capture probe. Once miRNA-21 hybridizes specifically with the PNA, we anchor the RAFT chain-transfer agent 2-(dodecylthi

Non-Stoichiometric Click Polymerization for Size-Controlled and Mild Encapsulation of Cells.

Artificial cell shells can achieve protection, identification, measurement, and manipulation of cells, but most cell encapsulation methods fail to control shell thickness, directly limiting the shell function in practical applications. The in-situ polymerization method has the potential to control the encapsulation shell but lacks chemical driving forces to enable shell thickness from nanoscale to micrometer-scale. The micrometer-scale shell can enhance the functionality in cell encapsulation and extend its applications to broader fields. In this work, we utilized the efficient and mild thiol-

Beyond hemoglobin polymerization: Mitochondrial pathobiology and therapeutic frontiers in sickle cell disease.

Sickle cell disease (SCD) is a global health issue that affects millions of individuals and is caused by a mutation in the β-globin gene (HBB), leading to the formation of sickle-shaped erythrocytes. Traditionally, the primary focus in SCD has been vaso-occlusion, hemoglobin polymerization, and hemolysis; however, recent studies have started to unravel the role of mitochondrial dysfunction in SCD pathophysiology. The current review aims to present the role of mitochondrial dysfunction in SCD-related clinical features and associated outcomes. It uniquely integrates mitochondrial dynamics, inter

Accelerated Charge Recombination in MOF Cocrystals Enables Efficient NIR-Driven Photothermal Polymerization.

Understanding charge transfer across donor-acceptor interfaces is essential for designing high-performance optoelectronic materials. Herein, we construct host-guest metal-organic framework (MOF) cocrystals employing a calcium-based naphthalenediimide (NDI) host and electron-rich aromatic guests to investigate photoinduced charge transfer at well-defined crystalline donor-acceptor interfaces. Challenging the conventional view that a high concentration of radical anions is typically associated with high photothermal conversion efficiency, our study uses electron paramagnetic resonance, surface p

Mechanism of ultrasound-modulated gluten-inulin interactions: Impact of inulin polymerization degree.

This study investigated how ultrasound treatment (0-40 s) modulated the interaction between wheat gluten and inulin of different polymerization degrees (long-chain, FXL; short-chain, FS). The results revealed a distinct chain-length-dependent mechanism. Under optimal sonication (20-30 s), FXL promoted hydrophobic interactions, increased β-sheet content (up to 34.4%) and disulfide bonds (up to 34.9%), and facilitated the formation of a continuous, elastic protein network. This led to enhanced thermal stability (denaturation temperature up to 100.60 °C) and stable viscoelasticity (lower tan δ).

Tuning the Length of Filomicelles Prepared Via Templated Polymerization-Induced Self-Assembly.

High-aspect-ratio polymeric micelles (filomicelles, FMs) are attractive anisotropic nanoparticles for a wide range of applications, however, their formation is typically confined to narrow hydrophilic/hydrophobic polymer block ratios, limiting systematic structure-property investigations. Consequently, independent control over micelle width, length, and stiffness remains challenging. A supramolecular polymerization-induced self-assembly (PISA) strategy using a bis-urea core template was recently shown to robustly yield wormlike micelles across a wide range of core block lengths; however, the r

Enhanced volumetric additive manufacturing via Reversible Addition-Fragmentation Chain Transfer (RAFT) polymerization.

Computed Axial Lithography (CAL), a Volumetric Additive Manufacturing (VAM) technology, enables the rapid, full body i.e. not layer-by-layer, fabrication of freeform geometries within seconds through the superposition of projected light patterns. However, as conventional CAL relies on free radical polymerization (FRP), it is an intrinsically exothermic process (ΔT > 60 °C) that can trigger auto-acceleration, so compromising print fidelity and limiting scalability. By regulating polymer chain length during propagation through reversible chain transfer, Reversible Addition-Fragmentation Chain Tr

Exploring a Ugi-Mediated Polymerization-Induced Self-Assembly Approach to Peptoid Particles.

This work investigates the synthesis of polypeptoid nanoparticles through Ugi-mediated polymerization-induced self-assembly (U-PISA), combining aqueous Ugi four-component polycondensation with PEG-based stabilization. Polypeptoid nano-objects were produced directly from amino acid-derived building blocks via a one-pot process under mild, open-air conditions. The effects of PEG molar mass and feed composition on particle formation and morphology were systematically examined, providing access to tunable spherical and worm-like nanostructures. The in situ formation of amphiphilic PEG-polypeptoid

Generation of Diversely Functionalized DNA Libraries Using Ligase-Catalyzed Oligonucleotide Polymerization (LOOPER).

Ligase-catalyzed oligonucleotide polymerization (LOOPER) enables the synthesis of highly modified nucleic acid libraries, which can be used as pools to evolve functional nucleic acids, such as aptamers. Here, we describe the process to generate a LOOPER-derived ssDNA library comprising 16 different chemical modifications installed in a sequence-defined manner. The preparation of the building blocks, their templated polymerization, and strand separation to generate the modified ssDNA library are outlined.

Spontaneous, Catalyst-Free Co-Polymerization of Ionic Liquids With CO2 Toward Polycarbonate.

The conversion of carbon dioxide (CO2) into value-added polymer is a cornerstone of sustainable chemistry, yet current strategies typically require high pressures, metal catalysts, and yield materials with limited functionality. Herein, we report a spontaneous, catalyst-free polymerization that directly converts CO2 and hydroxyl-functionalized ionic liquids (ILs) into multifunctional ionic polycarbonates under ambient conditions (0.1 MPa, 60°C). Mechanism studies, combining spectroscopic analysis and density functional calculations, elucidate a synergistic process wherein IL cations function a

Design and Synthesis of Combretastatin A4-Chalcone Hybrids as Tubulin Polymerization Inhibitors.

A new series of combretastatin A4-chalcone hybrids were designed and synthesized to explore their potential as cytotoxic agents. The compounds were evaluated for cytotoxic activity, tubulin inhibition, and anti-angiogenic properties. The designed hybrids exhibited significant cytotoxic effects across all the tested cancer cell lines. Among them, two compounds, 5d and 5o, exhibited potent cytotoxicity, with IC50 values of 0.02 and 0.03 μM, respectively, against the prostate cancer-3 cell line. Further biological investigations suggested that treatment with 5d and 5o induces apoptosis, as eviden

Paraspeckle condensation is controlled via TDP-43 polymerization and linked to neuroprotection.

The paraspeckle is a disease-relevant biomolecular condensate assembled from long non-coding RNA (lncRNA) NEAT1_2 ribonucleoprotein particles. Paraspeckle biogenesis is suppressed in normal tissues, yet it can be rapidly upregulated under stress. Here we demonstrate that a neurodegeneration-linked RNA-binding protein TDP-43 inhibits NEAT1_2 ribonucleoprotein particle condensation into the paraspeckle, in a concentration-dependent manner, which requires its intact polymerization and RNA binding. This effect is counterbalanced by core paraspeckle proteins such as FUS. Below disruptive concentrat
