Temporal dynamics of rhizosphere microbiome assembly and carbon-phosphorus coupling in poplar-medicinal plant intercropping systems.

BACKGROUND: Intercropping can reshape the rhizosphere microbiome, but how specific companion plants influence nutrient cycling and host growth remains unclear. We proposed that intercropping poplar with medicinal plants creates distinct rhizosphere niches that select for microbial communities with distinct functional potential, thereby improving tree nutrition. RESULTS: Intercropping significantly promoted poplar growth, with increases in diameter at breast height (DBH) of 15.33%, 14.3%, and 15.23% in systems with Anemarrhena asphodeloides, Belamcanda chinensis, and Saposhnikovia divaricata,

Radial branch hydraulics and cellular injury thresholds in poplar: insights from controlled dehydration and high-resolution dendrometry.

Branch water release and radial exchange influence drought responses. Although mechanistic models describe radial hydraulic and storage dynamics, experimental quantification of hydraulic capacitance (C) and radial resistance (Rr) across broad ranges of xylem water-potential (Ψx), particularly under severe dehydration, remains limited. We quantified how C and Rr vary with Ψx in Populus albaL., assessed the value of coupling controlled osmotic dehydration with high-resolution dendrometry, and evaluated cellular damage and bud viability across increasing dehydration levels. Branch segments were s

Microbial community biomass and structure in saline and non-saline soils associated with salt- and boron-tolerant poplar clones grown for the phytoremediation of selenium.

Poplar trees (Populus spp.) are often used in bioremediation strategies because of their ability to phytoextract potential toxic ions, e.g., selenium (Se) from poor quality soils. Soil microorganisms may play a vital role in sustaining health of soil and/or tolerance of these trees grown in poor quality soils by contributing to nutrient cycling, soil structure, overall soil quality, and plant survival. The effect of naturally occurring salts boron (B) and Se on soil microbial community composition associated with poplar trees is not known for bioremediation strategies. In this study, three Pop

Differential expression of a poplar copper chaperone gene in response to various abiotic stresses.

Copper chaperone (CCH) is upregulated during Arabidopsis (Arabidopsis thaliana L. Columbia) leaf senescence, suggesting that it mobilizes certain metal ions in leaves and transports them to other growing parts of the plants. The CCHs are also involved in defense mechanisms against oxidative stress in Arabidopsis and tomato (Lycopersicon esculentum Mill. cv. 'Ailsa Craig'). To elucidate the functions of CCH in poplar, we cloned a CCH gene (PoCCH) from Populus (Populus alba x P. tremula var. glandulosa) suspension cells and tested its expression in response to various treatments including heavy

PagDET2 promotes cambium cell division and xylem differentiation in poplar stem.

Secondary growth of the woody tree stem is governed by meristematic cell division and differentiation in the vascular cambium. Multiple hormonal signals and endogenous developmental programs regulate vascular cambium activity. Brassinosteroids (BRs) significantly promote secondary stem growth and wood formation in poplar trees. However, the underlying regulatory mechanisms of BRs within the vascular tissue remain unclear. Genetic and anatomical approaches were used here to elucidate the role of PagDET2, the rate-limiting enzyme for BRs biosynthesis, in regulating secondary vascular cambium act

Early responses to cadmium of two poplar clones that differ in stress tolerance.

Soil cadmium (Cd) contamination is becoming a matter of great global concern. The identification of plants differentially sensitive to Cd excess is of interest for the selection of genotype adaptive to grow and develop in polluted areas and capable of ameliorating or reducing the negative environmental effects of this toxic metal. The two poplar clones I-214 (Populus×canadensis) and Eridano (Populus deltoides×maximowiczii) are, respectively, tolerant and sensitive to ozone (O3) exposure. Because stress tolerance is mediated by an array of overlapping defence mechanisms, we tested the hypothesi

Conserved macromolecular architecture of poplar secondary cell walls revealed by ssNMR and atomistic modeling.

The macromolecular architecture of plant secondary cell walls governs wood's mechanical and biochemical properties, yet its natural intra-species variability remains poorly characterized. Here, we combined ¹³C solid-state NMR (ssNMR), multivariate statistical analysis, and molecular modeling to profile nanoscale structure across 13 genetically diverse Populus trichocarpa genotypes grown in ¹³C-enriched atmospheres. SsNMR-derived phenotypes spanning composition, structure, mobility, and inter-polymer proximities reveal a conserved architecture, with a subtle yet coordinated variation organizing

Alteration of cellulose accessibility, surface lignin area, and lignin hydrophobicity of Poplar Detritus through p-toluenesulfonic acid-assisting hydrothermal pretreatment.

In this work, altering cellulose accessibility, surface lignin area, and lignin hydrophobicity of Poplar Detritus (PD) was implemented through organic acid-assisting hydrothermal pretreatment, co-producing reducing sugars, xylo-oligosaccharides, furfural, and organic acids. After being treated under the optimal conditions [160 °C, 90 min, 1:10 solid-liquid ratio and 1 % p-toluenesulfonic acid (p-TsOH) concentration], the yield of reducing sugar increased by 8-folds, and the elimination of lignin and xylan were 63.7 % and 87.7 % respectively. Co-manufacturing of furfural, xylo-oligosaccharides,

Changes in clonal poplar leaf chemistry caused by stem galls alter herbivory and leaf litter decomposition.

Gall-inducing insects are highly specialized herbivores that modify the phenotype of their host plants. Beyond the direct manipulation of plant morphology and physiology in the immediate environment of the gall, there is also evidence of plant-mediated effects of gall-inducing insects on other species of the assemblages and ecosystem processes associated with the host plant. We analysed the impact of gall infestation by the aphid Pemphigus spirothecae on chemical leaf traits of clonal Lombardy poplars (Populus nigra var. italica) and the subsequent effects on intensity of herbivory and decompo

Improving carbon modeling in aspen-dominated forests: Lessons from CBM-CFS3 evaluation against long-term field data.

Boreal forests cover a substantial portion of Canada's land base and play a central role in the national carbon budget through carbon storage and fluxes across multiple above- and belowground pools. The Carbon Budget Model of the Canadian Forest Sector (CBM-CFS3) simulates and projects forest carbon dynamics under varying conditions and management scenarios, and ongoing model evaluation and calibration are essential to maintain accuracy in carbon monitoring, accounting, and reporting. This study assesses CBM-CFS3 performance using independent ground plot data from 64 aspen-dominated stands tha

Impact of benzene-ethanol extractives on deep eutectic solvent-mediated lignin extraction from poplar and eucalyptus.

Deep eutectic solvents (DES), an emerging class of green solvents, have demonstrated remarkable efficiency in the fractionation of lignocellulosic biomass. Benzene-ethanol extractives in woody biomass significantly affect the efficiency of DES-mediated fractionation. Therefore, investigating influence of benzene-ethanol extractives on DES-mediated pre-treatment is essential for optimizing of poplar and eucalyptus fractionation. Removing benzene-ethanol extractives significantly increased the polar component of the surface free energy (SFE) in both poplar and eucalyptus biomass. This prominentl

Microbial dyes: dyeing of poplar veneer with melanin secreted by Lasiodiplodia theobromae isolated from wood.

Dyeing methods that conform to environmental protection requirements and adapt to the development of industry are investigated using bleached poplar wood dyed with melanin secreted by Lasiodiplodia theobromae in this paper. Prolonging the dyeing time of coarse melanin can improve the dye uptake of veneers, and the brightness value increases, and the reflectivity declines with the increase of dyeing time. The stained surface of the vessel is gradually covered by coarse melanin dye, and more melanin particles can be clearly observed accumulating together on the stained surface with agar. Infrare

Can nutrient fertilization mitigate the effects of ozone exposure on an ozone-sensitive poplar clone?

We tested the independent and interactive effects of nitrogen (N; 0 and 80 kg ha-1), phosphorus (P; 0, 40 and 80 kg ha-1), and ozone (O3) application/exposure [ambient concentration (AA), 1.5 × AA and 2.0 × AA] for five consecutive months on biochemical traits of the O3-sensitive Oxford poplar clone. Plants exposed to O3 showed visible injury and an alteration of membrane integrity, as confirmed by the malondialdehyde by-product accumulation (+3 and +17% under 1.5 × AA and 2.0 × AA conditions, in comparison to AA). This was probably due to O3-induced oxidative damage, as documented by the prod

To each its own: differential response of specialist and generalist herbivores to plant defence in willows.

Plant-insect food webs tend to be dominated by interactions resulting from diffuse co-evolution between plants and multiple lineages of herbivores rather than by reciprocal co-evolution and co-cladogenesis. Plants therefore require defence strategies effective against a broad range of herbivore species. In one extreme, plants could develop a single universal defence effective against all herbivorous insects, or tailor-made strategies for each herbivore species. The evolution and ecology of plant defence has to be studied with entire insect assemblages, rather than small subsets of pairwise int

Interactive effects of supplemental UV-B and temperature in European aspen seedlings: Implications for growth, leaf traits, phenolic defense and associated organisms.

Past studies reveal opposite effects of elevated UV-B and temperature on plant growth and concentrations of UV-B absorbing compounds, yet few studies have dealt with the combined and interactive effects of these two climate change factors on woody dioecious plants. We investigated the interactive effects of UV-B and temperature treatments on growth, leaf traits and phenolic concentrations in Populus tremula L. (European aspen) seedlings. We also considered the consequences of these effects on their associated organisms: herbivorous insects, rust pathogens, the presence of endophytic fungi and

Climate-resilient agroforestry: physiological responses to climate change and engineering of crassulacean acid metabolism (CAM) as a mitigation strategy.

Global climate change threatens the sustainability of agriculture and agroforestry worldwide through increased heat, drought, surface evaporation and associated soil drying. Exposure of crops and forests to warmer and drier environments will increase leaf:air water vapour-pressure deficits (VPD), and will result in increased drought susceptibility and reduced productivity, not only in arid regions but also in tropical regions with seasonal dry periods. Fast-growing, short-rotation forestry (SRF) bioenergy crops such as poplar (Populus spp.) and willow (Salix spp.) are particularly susceptible

Phytoremediation of chlorinated ethenes in seepline sediments: tree selection.

Phytoremediation of chlorinated ethene (CE)-contaminated water was investigated at the Savannah River Site in Aiken, SC, USA. Perchloroethylene (PCE) and trichloroethylene (TCE) are present where CE-contaminated groundwater currently outcrops in seepline soils. Results of constructed and planted test cells, filled with soil from a noncontaminated seepline area and supplied with CE-contaminated groundwater (48 ppb) in the field for one season are presented. These test cells were planted with loblolly pines, hybrid poplars, coyote willow, and sweet gum. Cis-dichloroethylene (cDCE), a byproduct f

Functional study of the AfRAP2 gene in Amorpha fruticosa L. tolerance to saline-alkali and drought stress.

BACKGROUND: Amorpha fruticosa L. is a leguminous shrub with high tolerance to drought, poor soil, and saline-alkali stress conditions. As a member of the family of transcription factors in higher plants, the ethylene response factor AP2/ERF plays a crucial role in both plant adaptation to abiotic stress and in growth and development. In this study, based on genes identified from the transcriptomic sequencing of Amorpha fruticosa L. under drought stress, the upregulated gene AfRAP2 was isolated from its seedlings, with the aim of elucidating its stress-response function using molecular biologic
