Ventromedial prefrontal cortex/anterior cingulate cortex Glx, glutamate, and GABA levels in medication-free major depressive disorder.

Glutamate (Glu) and gamma-aminobutyric acid (GABA) are implicated in the pathophysiology of major depressive disorder (MDD). GABA levels or GABAergic interneuron numbers are generally low in MDD, potentially disinhibiting Glu release. It is unclear whether Glu release or turnover is increased in depression. Conversely, a meta-analysis of prefrontal proton magnetic resonance spectroscopy (1H MRS) studies in MDD finds low Glx (combination of glutamate and glutamine) in medicated MDD. We hypothesize that elevated Glx or Glu may be a marker of more severe, untreated MDD. We examined ventromedial p

Multisynaptic projections from the ventrolateral prefrontal cortex to hand and mouth representations of the monkey primary motor cortex.

Different sectors of the prefrontal cortex have distinct neuronal connections with higher-order sensory areas and/or limbic structures and are related to diverse aspects of cognitive functions, such as visual working memory and reward-based decision-making. Recent studies have revealed that the prefrontal cortex (PF), especially the lateral PF, is also involved in motor control. Hence, different sectors of the PF may contribute to motor behaviors with distinct body parts. To test this hypothesis anatomically, we examined the patterns of multisynaptic projections from the PF to regions of the p

Cell adhesion molecule expression in the dorsolateral prefrontal cortex and anterior cingulate cortex in major depression in the elderly.

BACKGROUND: Neuroimaging studies have demonstrated changes in the dorsolateral prefrontal cortex (DLPFC) and anterior cingulate cortex (ACC) in major depression. AIMS: We investigated the expression of cell adhesion molecules (CAMs) in the prefrontal cortex in depression. METHOD: Immunohistochemistry to localise CAMs in post-mortem tissue from 20 subjects with major depression and 20 controls, and image analysis to quantify their expression. RESULTS: We found significant increases in CAMs in the grey matter of the DLPFC in the depression group but no comparable differences in the ACC or occ

Elucidating the underlying components of metacognitive systematic bias in the human dorsolateral prefrontal cortex and inferior parietal cortex.

Metacognitive systematic bias impairs human learning efficiency, which is characterized by the inconsistency between predicted and actual memory performance. However, the underlying mechanism of metacognitive systematic bias remains unclear in existing studies. In this study, we utilized judgments of learning task in human participants to compare the neural mechanism difference in metacognitive systematic bias. Participants encoded words in fMRI sessions that would be tested later. Immediately after encoding each item, participants predicted how likely they would remember it. Multivariate anal

Dopamine receptor transcript expression in striatum and prefrontal and occipital cortex. Focal abnormalities in orbitofrontal cortex in schizophrenia.

BACKGROUND: The identification of novel subtypes of the dopamine receptors has renewed interest in the involvement of dopaminergic mechanisms in schizophrenia. We determined the expression of transcripts encoding the dopamine receptors in the brains of schizophrenic patients. METHODS: The levels of the messenger RNA molecules encoding the 5 dopamine receptors were quantified in postmortem brain samples from 16 schizophrenic patients and 9 control subjects. Samples from multiple regions of the prefrontal cortex, primary visual cortex, and striatum were subjected to in situ hybridization follow

The functional connectivity between dorsal-medial prefrontal cortex and middle cingulate cortex links trait rumination and depressive symptoms.

Task-based functional neuroimaging has revealed that rumination is associated with functional activations in the default mode network (DMN). The present study aimed to examine whether resting state functional connectivity (FC) within the DMN is associated with individual differences in trait rumination. Using the seed-based functional connectivity analysis in a relatively large sample of late adolescents, this study investigated the neural correlates of trait rumination and their associations with depressive symptoms. Results showed that higher functional connectivity between the left dorsal m

The Theoretical Evolution of Social Memory and the Core Contribution of the Medial Prefrontal Cortex.

Social interaction is a process in which two or more conspecific individual information and engage with one another, mutually influencing each other's behavior. Within this framework, social cognition and social memory constitute fundamental components that enable adaptive social behaviors. Deficits in social cognition and memory have been implicated in various psychiatric disorders since social memory itself represents a core expression of episodic memory. In this review, we summarize the theoretical evolution of understanding the social cognitive memory from the grandmother cell hypothesis t

The representation of facial emotion expands from sensory to prefrontal cortex with development.

Facial expression recognition develops rapidly during infancy and improves from childhood to adulthood. As a critical component of social communication, this skill enables individuals to interpret others' emotions and intentions. However, the brain mechanisms driving the development of this skill remain largely unclear due to the difficulty of obtaining data with both high spatial and temporal resolution from young children. By analyzing intracranial EEG data collected from childhood (5-10 years old) and post-childhood groups (13-55 years old), we find differential involvement of high-level br

Neuronal dynamics of cerebellum and medial prefrontal cortex in adaptive motor timing.

Precise temporal control of sensorimotor coordination and adaptation is a fundamental basis of animal behavior. How different brain regions are involved in regulating the flexible temporal adaptation remains elusive. Here, we investigated the neuronal dynamics of the cerebellar interposed nucleus (IpN) and the medial prefrontal cortex (mPFC) neurons during temporal adaptation between delay eyeblink conditioning (DEC) and trace eyeblink conditioning (TEC). When mice were trained for either DEC or TEC and subsequently subjected to a new paradigm, their conditioned responses (CRs) adapted virtual

The Dorsolateral Prefrontal Cortex: A High-level Hub for Allostatic Cardiovascular Control.

The dorsolateral prefrontal cortex (dlPFC) is increasingly targeted by various noninvasive transcranial magnetic stimulation or transcranial current stimulation protocols in a range of neuropsychiatric and other brain disorders. The rationale for this therapeutic modulation remains elusive. A model is proposed, and up-to-date evidence is discussed, suggesting that the dlPFC is a high-level cortical centre where uncertainty management, movement facilitation, and cardiovascular control processes are intertwined and integrated to deliver optimal behavioural responses in particular environmental o

Intracranial directed connectivity links subregions of the prefrontal cortex to major depression.

Research on the neural basis of major depressive disorder suggests that it is fundamentally a disease of cortical disinhibition, where breakdowns of inhibitory neuronal systems lead to diminished emotion regulation and intrusive rumination. Subregions of the prefrontal cortex are thought to be sources of this disinhibition. However, due to limited opportunities for intracranial recordings from humans with major depression, this hypothesis has not been directly tested. Here, we use intracranial recordings from the dorsolateral prefrontal, orbitofrontal, and anterior cingulate cortices from pati

Prefrontal cortex hypoactivation in response to sleep-related pictures in shift workers.

Shift work can lead to mental health issues such as sleep disturbances and cognitive impairment. Neural activation in response to external sleep-related stimuli may vary according to shift work patterns. In this study, we investigated the differences in brain activity in response to sleep-related stimuli between shift-worker (SW) nurses and healthy controls (HCs), and we also assessed the relationships between sleep-related problems and brain activity. The hypothesis was that shift workers would exhibit altered activation in the prefrontal cortex (PFC) when processing sleep-related stimuli, re

Acute alcohol in prefrontal cortex is characterized by enhanced inhibition that transitions to excitation.

Acute alcohol can have profound effects on the brain and behavior. Dorsal medial prefrontal cortex (dmPFC) function is especially sensitive to disruption from acute alcohol exposure. The acute, broadly inhibitory pharmacodynamics of alcohol in dmPFC have been well characterized in in vitro and ex vivo preparations, but seemingly contradict the stimulatory behavioral effects of alcohol at low-to-moderate doses. To determine the effects of acute alcohol on dmPFC activity, this study combined multiple routes of alcohol exposure across multiple rodent species and preparations and utilized modeling

Role of prefrontal cortex in auditory-verbal hallucinations: Actor or passive bystander?

In this paper we discuss the role of the prefrontal cortex (PFC) in schizophrenia and particularly in auditory verbal hallucinations (AVH). The paper starts with a review of the concept of hypofrontality in the 1970s and a view of PFC as a passive bystander unable to regulate or control internally generated sensory impulses. We further discuss the relationship between PFC and memory processes and the hypothesis that AVH experiences occur because of intrusions of negative emotional memories, and regulation of the emotional content of the voices. Similarly, AVH experiences can be the result of f

Prefrontal Cortex as a Central Hub of Structural Diaschisis Following Subcortical Stroke.

BACKGROUND: After a subcortical stroke, structural and functional alterations have often been observed in brain regions far from the lesion, a phenomenon suspected but not yet confirmed as diaschisis. This study investigated the existence and patterns of diaschisis following subcortical stroke. METHODS: This observational, single-center, cross-sectional study was conducted in China from August 2014 to September 2023. We recruited patients with subcortical stroke with hemiplegia and unilateral lesions in the basal ganglia or corona radiata. Healthy controls were recruited from the community us

Prefrontal cortex oxygenation during a mentally fatiguing task in normoxia and hypoxia.

Mental fatigue (MF) and hypoxia impair cognitive performance through changes in brain hemodynamics. We want to elucidate the role of prefrontal cortex (PFC)-oxygenation in MF. Twelve participants (22.9 ± 3.5 years) completed four experimental trials, (1) MF in (normobaric) hypoxia (MF_HYP) (3.800 m; 13.5%O2), (2) MF in normoxia (MF_NOR) (98 m; 21.0%O2), (3) Control task in HYP (CON_HYP), (4) Control in NOR (CON_NOR). Participants performed a 2-back task, Digit Symbol Substitution test and Psychomotor Vigilance task before and after a 60-min Stroop task or an emotionally neutral documentary. Br

Intracranial Directed Connectivity Links Subregions of the Prefrontal Cortex to Major Depression.

Understanding the neural basis of major depressive disorder (MDD) is vital to guiding neuromodulatory treatments. The available evidence supports the hypothesis that MDD is fundamentally a disease of cortical disinhibition, where breakdowns of inhibitory neural systems lead to diminished emotion regulation and intrusive ruminations. Recent research also points towards network changes in the brain, especially within the prefrontal cortex (PFC), as primary sources of MDD etiology. However, due to limitations in spatiotemporal resolution and clinical opportunities for intracranial recordings, thi

Unlocking the epigenome: Stress and exercise induced Bdnf regulation in the prefrontal cortex.

Aversive caregiving in early life is a risk factor for aberrant brain and behavioral development. This outcome is related to epigenetic dysregulation of the brain-derived neurotrophic factor (Bdnf) gene. The Bdnf gene encodes for BDNF, a neurotrophin involved in early brain development, neural plasticity, learning, and memory. Recent work suggests that exercise may be neuroprotective in part by supporting BDNF protein and gene expression, making it an exciting target for therapeutic interventions. To our knowledge, exercise has never been studied as a therapeutic intervention in preclinical ro

Intoxication due to Δ9-tetrahydrocannabinol is characterized by disrupted prefrontal cortex activity.

Neural states of impairment from intoxicating substances, including cannabis, are poorly understood. Cannabinoid 1 receptors, the main target of Δ9-tetrahydrocannabinol (THC), the primary intoxicating cannabinoid in cannabis, are densely localized within prefrontal cortex; therefore, prefrontal brain regions are key locations to examine brain changes that characterize acute intoxication. We conducted a double-blind, randomized, cross-over study in adults, aged 18-55 years, who use cannabis regularly, to determine the effects of acute intoxication on prefrontal cortex resting-state measures, as

Chronic stress disturbed the metabolism of homocysteine in mouse hippocampus and prefrontal cortex.

Stress is an independent risk factor for cognitive impairment, with elevated plasma homocysteine (HCY) levels playing a crucial role in stress-induced cognitive decline. While the rise in plasma HCY levels is linked to abnormal peripheral catabolism, the impact of stress on HCY catabolism in the brain remains unclear. This study investigated the effect of stress on HCY metabolism in the brain by analyzing HCY and its metabolic enzymes in the hippocampus and prefrontal cortex. The results showed a significant decrease in enzymes MS (methionine-synthase), CBS (cystathionineβ-synthase), and CSE (
