Cholesterol-lowering probiotics as potential biotherapeutics for metabolic diseases.

Cardiovascular diseases are one of the major causes of deaths in adults in the western world. Elevated levels of certain blood lipids have been reported to be the principal cause of cardiovascular disease and other disabilities in developed countries. Several animal and clinical trials have shown a positive association between cholesterol levels and the risks of coronary heart disease. Current dietary strategies for the prevention of cardiovascular disease advocate adherence to low-fat/low-saturated-fat diets. Although there is no doubt that, in experimental conditions, low-fat diets offer an

Probiotics for the prevention of pediatric antibiotic-associated diarrhea.

BACKGROUND: Antibiotics alter the microbial balance within the gastrointestinal tract. Probiotics may prevent antibiotic-associated diarrhea (AAD) via restoration of the gut microflora. Antibiotics are prescribed frequently in children and AAD is common in this population. OBJECTIVES: The primary objectives were to assess the efficacy and safety of probiotics (any specified strain or dose) used for the prevention of AAD in children. SEARCH METHODS: MEDLINE, EMBASE, CENTRAL, CINAHL, AMED, and the Web of Science (inception to May 2010) were searched along with specialized registers including t

Probiotics for prevention of necrotizing enterocolitis in preterm infants.

BACKGROUND: Necrotizing enterocolitis (NEC) and nosocomial sepsis are associated with increased morbidity and mortality in preterm infants. Through prevention of bacterial migration across the mucosa, competitive exclusion of pathogenic bacteria, and enhancing the immune responses of the host, prophylactic enteral probiotics (live microbial supplements) may play a role in reducing NEC and associated morbidity. OBJECTIVES: To compare the efficacy and safety of prophylactic enteral probiotics administration versus placebo or no treatment in the prevention of severe NEC and/or sepsis in preterm

Use of probiotics in gastrointestinal disorders: what to recommend?

Perturbation of bacterial microflora of the gastrointestinal (GI) tract may play an important role in the pathophysiology of some GI disorders. Probiotics have been used as a treatment modality for over a century. They may restore normal bacterial microflora and effect the functioning of the GI tract by a variety of mechanisms. Probiotics are not currently regulated and only few randomized controlled trials exist investigating their efficacy in different GI disorders. They are available in a variety of formulations and delivery systems making interpretation and comparison of studies even more

Review article: antibiotics and probiotics in inflammatory bowel disease.

Treatment with antibiotics in inflammatory bowel disease has a long tradition and is widely used. The indications for antibiotic therapy are wide ranging, from specific situations such as abscesses or fistulae, to patients with severe disease (as an unspecific 'protective' measure), and to address the hypothesis that the enteric flora as a whole, or specific microorganisms such as mycobacteria, are involved in the pathogenesis of inflammatory bowel disease. The best-studied single antibiotic compound is metronidazole. However, overall, the scientific basis for the use of antibiotics is limited

The Use of Probiotics in Head and Neck Infections.

Recurrent episodes of infections in the upper respiratory tract are very common. The pathogenesis behind these recurrences is still not well understood, but much attention has been paid to the adherence of the microorganisms to epithelial structures and to the protective function of the normal bacterial flora (bacterial interference). Thus, in vitro as well as in vivo studies have shown that both aerobic and anaerobic bacteria of the normal flora in the upper respiratory tract can hinder the growth of pathogens and the establishment of a renewed infection. Studies have shown that lack of inter

Leveraging intratumoral probiotics for pancreatic cancer immunotherapy via xenophagy.

Although pancreatic ductal adenocarcinoma (PDAC) tumors harbor unique intracellular bacteria, therapeutic strategies to harness them and overcome immunotherapy resistance remain elusive. Through 16S rDNA sequencing of PDAC tumors from 40 patients and screening of probiotic strains, we identified Bifidobacterium longum (B. longum) as capable of colonizing tumors and invading PDAC cells. In a murine orthotopic tumor model, B. longum's anti-tumor effects are exclusively driven by rapamycin-induced xenophagy. This xenophagy-mediated probiotic therapy, termed XenoPro, significantly suppresses tumor

Engineered probiotics block arsenite absorption in the gastrointestinal tract.

Chronic exposure to arsenite in food and water is a major global health concern, yet no practical strategies exist to prevent ingested arsenite from entering the body through the gastrointestinal tract. Here, we engineered Escherichia coli Nissle 1917 (EcN) to sense and sequester arsenite in situ, creating a probiotic-based approach to reduce host absorption of arsenite. The system involved an arsenite-responsive genetic toggle switch that activated chelator expression upon exposure; after arsenite was removed, it sustained output under biostatic conditions but shut off during active cell divi

Probiotics in Alleviating Constipation: Mechanisms, Strain Screening, and Applications.

Constipation is a prevalent gastrointestinal disorder with limited effective therapeutic options. Probiotics have emerged as promising microecological interventions, yet marked strain-specific efficacy and incomplete mechanistic understanding hinder clinical translation. This review systematically synthesizes evidence from human randomized controlled trials, animal studies, and mechanistic investigations on probiotic interventions for constipation. We catalog major anti-constipation strains across Bifidobacterium, Lactobacillus, Bacillus, and other genera, and summarize key findings from publi

Engineered Probiotics Protect against Pseudomonas aeruginosa Post Antibiotic Exposure.

Broad-spectrum antibiotics profoundly disrupt the commensal microbiota and compromise mucosal immunity, creating a vulnerable postantibiotic window that predisposes hosts to opportunistic infections such as Pseudomonas aeruginosa. Strategies for restoring host homeostasis and stimulating mucosal immunity during this period remain limited. Here, we developed a modular and programmable probiotic strategy based on engineered Escherichia coli Nissle 1917 (EcN) that integrates metabolite replenishment with antigen-specific mucosal immunization. The butyrate-overproducing strain restored immune home

Probiotics as a Treatment of Chronic Stress Associated Abnormalities.

Chronic stress is a widespread problem that significantly affects both physical and mental health, leading to numerous complications such as mood disorders, cognitive impairments, gastrointestinal issues, and chronic diseases. The dysregulation of the hypothalamic pituitary adrenal (HPA) axis and the gut-brain axis underlies several stress related disorders, leading to systemic inflammation, neuroinflammation, dysbiosis, and impaired gut barrier integrity. This review emphasizes the growing significance of probiotics as a potential treatment strategy for addressing chronic stress. Probiotics a

Redefining acne treatment: emerging role of probiotics in dermatology.

Acne is a common dermatological condition that requires novel therapeutic strategies. Probiotics, live microorganisms that confer health benefits, have emerged as a promising option for acne management. This review summarizes current evidence on the role of probiotics, focusing on their ability to modulate the skin microbiota, reduce inflammation, and influence the skin-gut axis. Both oral and topical probiotic applications have shown potential-oral formulations primarily act through gut-skin immune modulation, whereas topical preparations directly target the cutaneous microbiota and local inf

Probiotics mitigates ovarian angiogenic disturbance in letrozole-induced PCOS.

BACKGROUND: Among metabolic/endocrine diseases affecting women in the reproductive years, polycystic ovarian syndrome (PCOS) is well documented as the leading cause of female infertility and several complications including dyslipidemia, cardiomorbidities, among others. Vascular endothelial growth factor (VEGF) is a proangiogenic marker which plays crucial roles in endothelial dysfunction, including ovarian dysfunction. Probiotics are gut microbiome that regulate metabolic health via epigenetic modulation of histone. Nevertheless, the present study was designed to investigate the beneficial eff

Engineered Probiotics Mitigate Gut Barrier Dysfunction Induced by Nanoplastics.

Micro- and nanoplastics, particularly those derived from food-contact materials like polyethylene terephthalate (PET), can damage gut barriers, making the gastrointestinal system more vulnerable to inflammation and infections. Here, a probiotic-based drug delivery system (EcNT@L) is devised to mitigate nanoplastics-induced gut barrier dysfunction by modulating gut immunity and microbiota. Escherichia coli Nissle 1917 (EcN) is genetically engineered to produce transforming growth factor-β (TGF-β) and then modified with an Eudragit L100-55 coating. This engineered probiotic acts as an in vivo "d

Probiotics in the management of radiation-induced oral mucositis.

Oral mucositis is a common and debilitating oral complication in head and neck cancer patients undergoing radiotherapy, resulting in diminished quality of life and potential treatment disruptions. Oral microbiota has long been recognized as a contributing factor in the initiation and progression of radiation-induced oral mucositis (RIOM). Numerous studies have indicated that the radiation-induced oral microbial dysbiosis promotes the occurrence and severity of oral mucositis. Therefore, approaches that modulate oral microbial ecology are promising for the management of RIOM. Probiotics as a re
