BACKGROUND: Neuroplasticity in the medial prefrontal cortex (mPFC) is critically implicated in major depressive disorder (MDD). Progranulin (PGRN), a multifunctional protein that is abundant in the cortex, regulates neuronal development and synaptic maintenance, positioning it as a candidate therapeutic target for MDD. However, current findings indicate the effects of PGRN in depression are contingent upon both brain region and cell type. Whether neuronal PGRN in the mPFC contributes to the pathogenesis of depression remains unknown.
METHODS: A chronic unpredictable mild stress (CUMS) mouse m

Inflammation, oxidative stress and apoptosis, in particular, play critical roles in the pathophysiology of myocardial infarction (MI). Isoproterenol (ISO) is a frequently used agent in experimental models to induce MI. This study aimed to determine the temporal changes in Spexin (SPX) and Progranulin (PRG) levels in an ISO-induced experimental MI model. The study consisted of control and ISO-treated groups (ISO-1, ISO-2, ISO-4, ISO-6 and ISO-24). In the ISO-induced MI model, serum CK-MB and troponin (I-T) levels increased, confirming cardiac damage. However, increases in oxidative parameters a

BACKGROUND: Diabetic keratopathy is characterized by delayed epithelial healing and reduced corneal nerve density, however effective treatment remains a significant challenge. Progranulin (PGRN), a secreted anti-inflammatory and neuroprotective factor, has been implicated in ocular homeostasis, but its role in the cornea has not been well defined.
OBJECTIVE: To investigate the function of PGRN in corneal repair under normal and diabetic conditions and to identify underlying mechanisms.
METHODS: PGRN expression in normal and diabetic mouse corneas was assessed by Western blot, ELISA, and immu

Accumulating evidence indicates that crosstalk between immune cells and intrinsic cochlear cells is critical for maintaining auditory homeostasis. However, the activation profile of cochlear microglia-like cells (MLCs) and their mechanistic role in age-related hearing loss (ARHL) remain poorly defined. Here, we show that MLCs are robustly activated in the cochlea of aged mice, and that pharmacologic depletion of cochlear MLCs significantly attenuates ARHL progression. To dissect the mechanisms underlying MLC-driven cochlear damage in ARHL, we leveraged the single-cell transcriptomic landscape

INTRODUCTION: Progranulin (PGRN), a secreted glycoprotein, plays a crucial role in several biological processes. However, its association with cognitive impairment in patients with coronary artery disease (CAD) remains unclear. This study aimed to evaluate the relationship between serum PGRN levels and cognitive impairment in patients with CAD.
METHODS: A total of 177 patients with CAD and 150 non-CAD controls were included, and serum PGRN concentrations and other CAD-related risk factors were measured. Cognitive function was assessed, and logistic regression analysis was performed to identif

In a previous study, we demonstrated that neutrophil elastase is activated in the brain parenchyma after cerebral ischemia, which enzyme cleaves progranulin (PGRN), an anti-inflammatory factor. In that study, we also found that sivelestat, a selective neutrophil elastase inhibitor, attenuates ischemia-induced inflammatory responses. However, it was not clear whether this anti-inflammatory effect was due to the direct effect of sivelestat. In this study, we evaluated the effects of sivelestat or recombinant PGRN (rPGRN) on cell injuries in cultured neurons, astrocytes, and microglia under oxyge

OBJECTIVES: Previously we discovered antibodies against progranulin (PGRN-abs) in a protein array-based screening of sera from various rheumatic diseases. Here we conducted a study to evaluate the prevalence of PGRN-abs in seropositive and seronegative rheumatoid arthritis (RA).
METHODS: PGRN-abs were determined in the sera from 257 RA patients being seropositive for RF-IgM and/or ACPA-IgG and from 224 seronegative RA patients who were prospectively included in this study (total RA cohort n=481). All serum samples from the included participants were tested for RF-IgM as well as for ACPA-IgG,

The enzyme-linked immunosorbent assay (ELISA) is a well-established methodology for detection of analytes in various biological fluids. The assay described herein has been validated for the detection of PGRN/GP88/GEP in blood (serum/ plasma), urine and cerebrospinal fluid (CSF), and synovial fluid and may also be used for breast milk, ductal lavage, nipple aspirates, and saliva. The ability to measure circulating levels of PGRN/GP88/GEP has proven to have clinical utility for several human diseases such as cancer where changes of PGRN/GP88/GEP can be determined as a mean to monitor disease sta

We examined progranulin (PGRN) levels in cerebrospinal fluid (CSF) samples during the acute phase in 15 patients with neuromyelitis optica spectrum disorders (NMOSD) and compared the results with those from 17 patients with multiple sclerosis (MS), 30 patients with other inflammatory neurological diseases (OIND), and 20 non-inflammatory controls (NIC). CSF PGRN levels of NMOSD patients were significantly higher than those of MS patients and NICs. These levels correlated with CSF interleukin-6 levels, CSF cell counts, CSF protein levels, improvements in the Expanded Disability Status Scale scor

Since progranulin (PGRN) is a natural ligand of TNF receptors, we assessed whether serum PGRN levels predict and/or reflect responsiveness of RA patients to TNF-antagonist therapy. TNF-antagonist-naïve RA patients (N = 35) were started on TNF-antagonist therapy. At baseline and at follow-up visits, DAS28-ESR, DAS28-CRP, and CDAI were calculated, and venous blood was collected for serum PGRN determination. Disease activity and clinical response were based on EULAR criteria. Baseline serum PGRN levels varied considerably and correlated with ESR and CRP. DAS28-ESR, DAS28-CRP, and CDAI were greate

Osteoarthritis (OA) is a prevalent condition that compromises and even jeopardizes the life quality of millions of people. Common symptoms in OA includes joint stiffness and soreness, and they are often associated with inflammations to various extend. Due to the avascular and aneural nature of articular hyaline cartilage, it has limited self-repair capabilities; especially under inflammatory conditions, damages inflicted on cartilage are often irreversible. Hence, treatment approaches focus on anti-inflammation or articular cartilage replacement. In this study, an engineered, dual-functional l

AIMS: The glycoprotein progranulin directly binds to TNF-receptors and thereby can antagonize the inflammatory effects of TNF-α. Here we analyzed the impact of both cytokines on cytotoxicity and viability of trophoblast cells.
METHODS: Isolated villous first trimester human trophoblast cells and the human choriocarcinoma cell line BeWo were treated with recombinant human progranulin and TNF-α. Analyses were performed by LDH- and MTT-assay and measurement of caspase-8-activity.
RESULTS: Progranulin treatment showed some cytoprotective effects on isolated trophoblast cells. However, TNF-α-indu

BACKGROUND: Elevated progranulin levels are associated with visceral obesity, elevated plasma glucose, and dyslipidemia. Progranulin has not been previously investigated as a biomarker of nonalcoholic fatty liver disease (NAFLD). We sought to determine whether serum progranulin levels are altered in patients with biopsy-proven NAFLD and if they are associated with their clinical, biochemical, and histological characteristics.
SUBJECTS AND METHODS: We measured serum progranulin levels in 95 patients with biopsy-proven NAFLD and 80 age- and sex-matched controls. The potential associations betwe

BACKGROUND: The progranulin gene (GRN) is mutated in 5-10% of patients with frontotemporal lobar degeneration (FTLD) and in about 20% of patients with familial FTLD. The most common mutation in GRN is Arg493X. We aimed to establish the contribution of this mutation to FTLD and related disorders.
METHODS: We measured the frequency of Arg493X in 3405 unrelated patients with various neurodegenerative diseases using Taqman single-nucleotide polymorphism (SNP) genotyping. Clinicopathological characterisation and shared haplotype analysis were done for 30 families with FTLD who carry Arg493X. To in

BACKGROUND: Cholesterol efflux is increasingly recognized as an important regulator of macrophage polarization and immunosuppressive features; however, the tumor-derived signals that govern this process in oral squamous cell carcinoma (OSCC) remain poorly defined.
METHODS: We performed integrated single-cell RNA sequencing (scRNA-seq) on primary OSCC tumors (n = 3) and validated the findings in 77 head and neck squamous cell carcinoma (HNSCC)-related samples across five public datasets. The role of tumor-derived progranulin (PGRN) in macrophage cholesterol efflux and associated phenotypic cha

OBJECTIVE: Progranulin (PGRN) is a secreted glycoprotein that can have pro-inflammatory or anti-inflammatory effects, depending on the disease. The serum and urinary levels and mechanism of action of PGRN in antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) are unclear.
METHODS: Thirty patients with active AAV were enrolled from May 2022 to May 2024; 28 were myeloperoxidase-positive, and two were proteinase 3-positive. A healthy control cohort consisted of five male and five female volunteers. Age, gender, AAV subtype, and laboratory data were collected. Using an enzyme-l
