Multi-immobilization of viscozyme, alcalase and flavourzyme by nanomagnetic combi-CLEAs method for oil and protein hydrolysates extraction from rice bran in aqueous phase.

Edible oil isolation from oil seeds by hexane faces pivotal challenges for human health and the environment. The aqueous enzymatic extraction procedure has been remarked as a preferable technique for concurrent oil and protein extraction. A one-pot combined cross-linked enzyme aggregates of Viscozyme, Alcalase and Flavourzyme in nanoscale for coinciding isolation of oil and protein hydrolysates is presented. The ideal parameters for immobilization were 1:1:1 ratio of enzymes in ethanol as aggregator, 0.75:1 ratio of lysine/enzyme mixture, 15 mM glutaraldehyde at temp = 2-3 °C, ultrasonic waves

Discovery of a novel dual-functional peptide from oyster protein hydrolysates with antioxidant and osteogenic properties: in vitro, in silico, and zebrafish model evaluation.

Bioactive peptides are emerging as promising agents for promoting osteogenesis and alleviating oxidative stress - both critical factors in osteoporosis development. In this study, we aimed to discover peptides from oyster protein hydrolysates with dual antioxidant and osteogenic activities. Hydrolysates were prepared from oyster mantle, non-mantle, and whole-body tissues, among which the <1 kDa fraction of the mantle hydrolysate showed the highest bioactivity. Peptides in this fraction were identified by LC-MS/MS and screened via in silico bioactivity prediction and molecular docking with BMPR

Production of Fish Protein Hydrolysates from Scyliorhinus canicula Discards with Antihypertensive and Antioxidant Activities by Enzymatic Hydrolysis and Mathematical Optimization Using Response Surface Methodology.

Fish discards are of major concern in new EU policies. Alternatives for the management of the new biomass that has to be landed is compulsory. The production of bioactive compounds from fish protein hydrolysates (FPH) has been explored in recent years. However, the viability of Scyliorhinus canicula discards, which might account for up to 90-100% of captures in mixed trawler, gillnet, and longline industrial fisheries, to produce FPH from the muscle with bioactivities has still not been studied in terms of the optimization of the experimental conditions to enhance its production. The effect of

Dietary egg white protein hydrolysate improves orotic acid-induced fatty liver in rats by promoting hepatic phospholipid synthesis and microsomal triglyceride transfer protein expression.

We investigated the effects of egg white protein hydrolysates (EWH) on orotic acid (OA)-induced nonalcoholic fatty liver (NAFL) in rats. Effects of the egg white protein (EWP) and EWH were also compared. Four groups of male Sprague-Dawley rats were separately fed AIN-76-based diets, supplemented with 20% casein for control, or with 1% OA, together with either 20% casein (OA), 20% EWP, or 20% EWH, respectively, for 3 d (developing stage) and 14 d (developed stage). In both feeding periods, animals from the OA group showed higher accumulation hepatic triacylglycerol (TAG) compared with those fro

A collagen hydrolysate/milk protein-blend stimulates muscle anabolism equivalently to an isoenergetic milk protein-blend containing a greater quantity of essential amino acids in older men.

BACKGROUND & AIMS: Nutritional composition is key for skeletal muscle maintenance into older age. Yet the acute effects of collagen protein blended with other protein sources, in relation to skeletal muscle anabolism, are ill-defined. We investigated human muscle protein synthesis (MPS) responses to a 20 g blend of collagen protein hydrolysate + milk protein (CP+MP, 125 ml) oral nutritional supplement (ONS) vs. 20 g non-blended milk protein source (MP, 200 ml) ONS, in older adults. METHODS: Healthy older men (N = 8, 71±1 y, BMI: 27±1 kg·m-2) underwent a randomized trial of 20 g protein, from

Peptide identification from a Porphyra dioica protein hydrolysate with antioxidant, angiotensin converting enzyme and dipeptidyl peptidase IV inhibitory activities.

A Porphyra dioica protein extract was enzymatically hydrolysed and then fractionated using semi-preparative reverse-phase high performance chromatography. The hydrolysate and its fractions were tested for their oxygen radical absorbance capacity (ORAC) along with their angiotensin converting enzyme (ACE) and dipeptidyl peptidase IV (DPP-IV) inhibitory activities. The most potent fraction was analysed by liquid chromatography mass spectrometry. Eight peptide sequences were selected for synthesis based on their structure-activity criteria for bioactivity. Asp-Tyr-Tyr-Lys-Arg showed the highest O

Herring Milt Protein Hydrolysate Improves Insulin Resistance in High-Fat-Diet-Induced Obese Male C57BL/6J Mice.

Protein consumption influences glucose homeostasis, but the effect depends on the type and origin of proteins ingested. The present study was designed to determine the effect of herring milt protein hydrolysate (HPH) on insulin function and glucose metabolism in a mouse model of diet-induced obesity. Male C57BL/6J mice were pretreated with a low-fat diet or a high-fat diet for 6 weeks. Mice on the high-fat diet were divided into four groups where one group continued on the high-fat diet and the other three groups were fed a modified high-fat diet where 15%, 35%, and 70%, respectively, of casei

Improvement of Ultrasound-Assisted Enzymatic Treatment and Exogenous Amino Acids on Structural, Antioxidant, and Instrumental Taste Profile of Maillard Reaction Products from Potato Protein Hydrolysates.

Potato protein hydrolysates (PPHs) produced by ultrasound-assisted enzymatic hydrolysis exhibit strong antioxidant activity. However, their inherent bitterness limits their application in food sectors. Therefore, this research investigated the effects of energy-divergent ultrasound (EDU)- and energy-gathered ultrasound (EGU)-assisted enzymatic treatment and exogenous amino acids (alanine (Ala), phenylalanine (Phe), and valine (Val)) on the peptide structure, antioxidant activity, and instrumental taste profile of Maillard reaction products (MRPs) prepared from PPHs. It was found that EDU and E

Machine learning-driven discovery of bioactive peptides from duckweed (Lemnaceae) protein hydrolysates: Identification and experimental validation of 20 novel antihypertensive, antidiabetic, and/or antioxidant peptides.

Duckweed, a sustainable, protein-rich aquatic plant, has recently emerged as a promising source of bioactive peptides. However, their identification remains limited and challenging in such complex mixtures. Following duckweed hydrolysis with pepsin, chymotrypsin, trypsin and papain, and a centrifugation step producing two fractions: supernatant (DS) and pellet (DP), interesting half-maximal inhibitory concentration (IC50) for dipeptidyl peptidase (DPP)-IV and angiotensin-converting enzyme (ACE) inhibition were obtained for DS fractions, especially with pepsin (IC50 = 0.7 and 0.07 mg/mL, respec

Digestive tract morphology and enzyme activities of juvenile diploid and triploid Atlantic salmon (Salmo salar) fed fishmeal-based diets with or without fish protein hydrolysates.

Triploid, sterile Atlantic salmon (Salmo salar) could make a contribution to the development of the farming industry, but uncertainties about the performance and welfare of triploids have limited their adoption by farmers. In this study, we compared the ontogeny of digestive tract morphology and enzyme activities (pepsin, trypsin, chymotrypsin, alkaline phosphatase and aminopeptidase) of diploid and triploid Atlantic salmon. Fish were fed diets based on fishmeal (STD) or a mix of fishmeal and hydrolysed fish proteins (HFM) whilst being reared at low temperature from start-feeding to completion

Hypolipidemic effects of protein hydrolysates from Trachinotus ovatus and identification of peptides implied in bile acid-binding activity using LC-ESI-Q-TOF-MS/MS.

In the present work, analysis of the hypolipidemic properties of Trachinotus ovatus protein hydrolysates (TOPHs) and identification of peptides with bile acid-binding activity were performed. Hydrolysates prepared by trypsin digestion exhibited the highest in vitro bile acid-binding capacities compared with hydrolysates prepared with the other four proteases and were mainly composed of small peptides and amino acids with molecular weights <3 kDa, accounting for 77.30%. Among the five ultra-filtration fractions of TOPHs, TOPHs-5 (<3 kDa) exhibited the highest in vitro bile acid-binding capacity

Potato protein hydrolysate (PP902) inhibits dexamethasone-induced muscle atrophy by regulating MuRF1 and MAFbx expression in C2C12 myotubes in vitro.

BACKGROUND: Skeletal muscle atrophy occurs when muscle mass and strength decrease due to aging, starvation, cancer, and cachectic diseases. Recently, an increasing focus has been on investigating potato protein's nutritional and functional properties and its wide-ranging applications. Steroids, especially glucocorticoids, have been extensively examined and found to cause muscle atrophy. Recently, we found that PP902 prevents hyperglycemia-induced muscle atrophy in cultured myotubes, although its impact on steroid-induced muscle atrophy remains unclear. Therefore, this study aimed to investigat

A Casein Hydrolysate Does Not Enhance Ileal Endogenous Protein Flows Compared With the Parent Intact Casein When Fed to Growing Pigs.

BACKGROUND: The form of dietary nitrogen (free peptides or intact proteins) may influence the amount of endogenous amino acids found at the terminal ileum of the pig, and it has been speculated that hydrolyzed dietary protein may lead to increased endogenous amino acids. OBJECTIVE: To compare the effect of dietary free peptides on ileal endogenous nitrogen and amino acid flows [ileal endogenous nitrogen flow (ENFL), ileal endogenous amino acid flow (EAAFL)] with that of peptides released naturally from dietary protein during digestion, from the same intact parent protein source. METHODS: Six
