Microneedles Coated with Cow's Milk Proteins: Immunogenicity, Stability, and Safety Assessment.

Cow's milk allergy (CMA) is one of the most frequently occurring food allergies in children, especially in infants less than 3 years old. Mindful avoidance of CMA-triggering foods and prompt epinephrine injection to overcome anaphylaxis in the case of accidental ingestion are the only options currently available to allergic subjects. This study investigates the potential of coated microneedles for delivering CMA into the skin as a novel approach to allergen immunotherapy. Precise amounts of cow's milk proteins (CMP) were dip-coated onto stainless steel microneedle patches and reproducibly deli

Direct degradation and stabilization of proteins: New horizons in treatment of nonalcoholic steatohepatitis.

Nonalcoholic fatty liver disease (NAFLD) is featured with excessive hepatic lipid accumulation and its global prevalence is soaring. Nonalcoholic steatohepatitis (NASH), the severe systemic inflammatory subtype of NAFLD, is tightly associated with metabolic comorbidities, and the hepatocytes manifest severe inflammation and ballooning. Currently the therapeutic options for treating NASH are limited. Potent small molecules specifically intervene with the signaling pathways that promote pathogenesis of NASH. Nevertheless they have obvious adverse effects and show long-term ineffectiveness in cli

Drosophila Heterochromatin Stabilization Requires the Zinc-Finger Protein Small Ovary.

Heterochromatin-mediated repression is essential for controlling the expression of transposons and for coordinated cell type-specific gene regulation. The small ovary (sov) locus was identified in a screen for female-sterile mutations in Drosophila melanogaster, and mutants show dramatic ovarian morphogenesis defects. We show that the null sov phenotype is lethal and map the locus to the uncharacterized gene CG14438, which encodes a nuclear zinc-finger protein that colocalizes with the essential Heterochromatin Protein 1 (HP1a). We demonstrate Sov functions to repress inappropriate gene expres

Chaperones convert the energy from ATP into the nonequilibrium stabilization of native proteins.

During and after protein translation, molecular chaperones require ATP hydrolysis to favor the native folding of their substrates and, under stress, to avoid aggregation and revert misfolding. Why do some chaperones need ATP, and what are the consequences of the energy contributed by the ATPase cycle? Here, we used biochemical assays and physical modeling to show that the bacterial chaperones GroEL (Hsp60) and DnaK (Hsp70) both use part of the energy from ATP hydrolysis to restore the native state of their substrates, even under denaturing conditions in which the native state is thermodynamica

The effects of metal ions on the structure and stability of the DNA gyrase B protein.

The effects of mono- and divalent metal ions on the DNA gyrase B subunit, on its 43 kDa and 47 kDa domains, and on two mutants in the Toprim domain (D498A and D500C) were investigated by means of circular dichroism and protein melting experiments. Both types of metal ion, with the notable exception of Mn2+, did not affect the conformational properties of the enzyme subunit at room temperature, but were able to produce selective and differential effects on protein stability. In particular, monovalent (K+) ions increased the stability of the gyrase B structure, whereas destabilising effects were

Comparative evaluation of liposome and ergosterol-niosome systems for encapsulation of goat milk protein hydrolysates: Structural properties, in vitro digestive stability, and storage stability.

This study developed and compared liposome and niosome delivery systems to improve the stability and bioaccessibility of goat milk protein hydrolysates. The results showed that the niosome delivery system incorporating ergosterol exhibited superior encapsulation efficiency (96.5 ± 0.04%) and smaller average particle size (216 ± 4.0 nm) compared with the liposome system (p < 0.05). Atomic force microscope results showed that both delivery systems had blunt cone morphology. FTIR spectral analysis also confirmed the successful encapsulating of the enzyme hydrolysates within the delivery systems.

A mitochondrial outer membrane protein TOMM20 maintains protein stability of androgen receptor and regulates AR transcriptional activity in prostate cancer cells.

Prostate cancer (PCa) is an androgen-dependent malignancy, with HSP90 and HSP70 serving as classical molecular chaperones that maintain androgen receptor (AR) protein stability and regulate its transcriptional activation. Surprisingly, our study identified TOMM20, a mitochondrial outer membrane protein, as a potential molecular chaperone with similar roles to HSP90/HSP70. We found that TOMM20 expression is elevated in PCa tissues and cell lines and positively correlates with AR levels. RNA-seq analysis revealed that TOMM20 knockdown significantly reduced the mRNA levels of AR-regulated genes.

Identification of proline-rich protein 11 as a major regulator in mouse spermatogonia maintenance via an increase in BMI1 protein stability.

BACKGROUND: Spermatogenesis accompanied by self-renewal and differentiation of spermatogonia under complicated regulation is crucial for male fertility. Our previous study demonstrated that the loss of the B-lymphoma Mo-MLV insertion region 1 (BMI1) could cause male infertility and found a potential interaction between BMI1 and proline-rich protein 11 (PRR11); however, the specific co-regulatory effects of BMI1/PRR11 on spermatogonia maintenance remain unclear. METHODS AND RESULTS: The expression of PRR11 was downregulated in a mouse spermatogonia cell line (GC-1) via transfection with PRR11-

Dynamic unfolding of a regulatory subunit of cAMP-dependent protein kinase by capillary electrophoresis: Impact of cAMP dissociation on protein stability.

Characterization of the unfolding dynamics of a recombinant type IA regulatory subunit (RIalpha) of cyclic adenosine monophosphate (cAMP)-dependent protein kinase (cAPK) was examined by CE with UV detection. Electrophoretic separation of RIalpha by CE in a buffer devoid of cAMP resulted in rapid dissociation of the complex from the original sample due to the high negative mobility of the ligand relative to receptor. This process enabled in-capillary generation of cAMP-stripped RIalpha, which was used to estimate the apparent dissociation constant (Kd) of 0.6 +/- 0.2 microM. A comparison of RIa

Transition state complexes of the Klebsiella pneumoniae nitrogenase proteins. Spectroscopic properties of aluminium fluoride-stabilized and beryllium fluoride-stabilized MgADP complexes reveal conformational differences of the Fe protein.

Stable inactive 2 : 1 complexes of the Klebsiella pneumoniae nitrogenase components (Kp2/Kp1) were prepared with ADP or the fluorescent ADP analogue, 2'(3')-O-[N-methylanthraniloyl] ADP and AlF(4)(-) or BeF(3)(-) ions. By analogy with published crystallographic data [Schindelin et al. (1997) Nature 387, 370-376)], we suggest that the metal fluoride ions replaced phosphate at the two ATP-binding sites of the iron protein, Kp2. The beryllium (BeF(x)) and aluminium (AlF(4)(-)) containing complexes are proposed to correspond to the ATP-bound state and the hydrolytic transition states, respectively

A Fur family protein BosR is a novel RNA-binding protein that controls rpoS RNA stability in the Lyme disease pathogen.

The σ54-σS sigma factor cascade plays a central role in regulating differential gene expression during the enzootic cycle of Borreliella burgdorferi, the Lyme disease pathogen. In this pathway, the primary transcription of rpoS (which encodes σS) is under the control of σ54 which is activated by a bacterial enhancer-binding protein (EBP), Rrp2. The σ54-dependent activation in B. burgdorferi has long been thought to be unique, requiring an additional factor, BosR, a homologue of classical Fur/PerR repressor/activator. However, how BosR is involved in this σ54-dependent activation remains unclea

Expression of domains for protein-protein interaction of nucleotide excision repair proteins modifies cancer cell sensitivity to platinum derivatives and genomic stability.

Nucleotide excision repair (NER) is involved in the repair of DNA damage caused by platinum derivatives and has been shown to decrease the cytotoxic activity of these drugs. Because protein-protein interactions are essential for NER activity, we transfected human cancer cell lines (A549 and HCT116) with plasmids coding the amino acid sequences corresponding to the interacting domains between excision repair cross-complementation group 1 (ERCC1) and xeroderma pigmentosum, complementation group A (XPA), as well as ERCC1 and xeroderma pigmentosum, complementation group F (XPF), all NER proteins.

Influence of iron, calcium, and zinc on the oxidative stability of omega-3 fatty acids in pea protein-stabilized oil-in-water emulsions.

BACKGROUND: Plant-based emulsified foods fortified with omega-3 fatty acids and minerals are nutritionally attractive, but their formulation is challenging because alpha-linolenic acid (ALA) is highly susceptible to oxidation and some mineral salts may act as pro-oxidants. This study evaluated how iron, zinc, and calcium salts affect pea protein isolate (PPI) properties and ALA oxidative stability in PPI/kappa-carrageenan (KC) gelled emulsions. Treatments included PPI-mineral systems containing ferrous glycinate (FeGly), zinc gluconate (ZnG), calcium gluconate (CaG), calcium lactate (CaL), cal

High internal phase emulsions stabilized by polyphenol-mediated Moringa seed protein for β-carotene encapsulation: Enhanced stability and bioaccessibility.

In this study, Moringa oleifera seed protein (MOSP) served as the raw material, while three polyphenols-epicatechin (EC), epigallocatechin gallate (EGCG), and tannic acid (TA)-were incorporated for modification to produce a high internal phase emulsion (HIPE). For each of the three polyphenols, the optimal addition level was around 15 μg per 1 mg of MOSP. The inclusion of polyphenols resulted in the interfacial tension being at the oil-water interface was significantly diminished, decreasing from 10.22 mN/m to 5.43 mN/m, with the emulsifying activity index (EAI) of MOSP-TA reaching 72.50 m2/g.

Nrf2 exerts an unfavorable role in chemical hepatotoxicity in mice by maintaining CYP2E1 protein stability.

Nuclear factor erythroid 2-related factor 2 (Nrf2) is generally recognized for its protective role against chemical toxicity. However, in some cases the function of Nrf2 is not as expected. Using wild-type (WT) and Nrf2-knockout (Nrf2-KO) mice, we investigated the role of Nrf2 in hepatic injury induced by the typical hepatotoxicants, thioacetamide (TAA) and diethylnitrosamine (DEN). Both acute and chronic liver injury models were established. In the present study, Nrf2 deficiency was found to alleviate hepatic injury induced by TAA and DEN. Bioactivation of both TAA (reflected by its urinary o

The deubiquitinating enzyme UCHL3 promotes bladder cancer progression and glycolysis through enhancing PKM2 protein stability.

Pyruvate kinase M2 (PKM2) is a critical rate-limiting enzyme in glycolysis and is frequently upregulated in bladder cancer (BCa), contributing to the Warburg effect and tumor progression. However, the mechanism supporting abnormal PKM2 stabilization via deubiquitination in BCa remains to be characterized. Here, we identified ubiquitin C-terminal hydrolase L3 (UCHL3) as a bona fide deubiquitylase of PKM2 in BCa. UCHL3 stabilized PKM2 in a deubiquitylation activity-dependent manner. UCHL3 depletion significantly decreased PKM2 protein levels, suppressed glycolytic activity, and inhibited BCa cel

High hydrostatic pressure modulation of whey protein-malvidin derivative complexes: Mechanisms, structural changes, and stability.

This study investigated how high hydrostatic pressure (HHP) modulates the structural and functional properties of whey protein isolate (WPI) complexes with Malvidin-3-O-6-[acrylic acid-(2-hydroxy,4-carboxy-cyclohexanol) ester]-guaiacol (MV3ACEC). HHP at 350 MPa maximized the binding ratio (78.41 ± 0.94%) and thermal stability (denaturation temperature: 123.76 °C) of WPI-MV3ACEC, while reducing particle size (237 nm) and enhancing system stability (zeta potential: 12.7 mV). Systematic Characterization of Thermal Stability Assay Parameters and Application in Discovery of Peptide-Protein Interactions.

Thermal proteome profiling (TPP) and its higher-throughput derivative, the proteome integral solubility alteration (PISA) assay, measure changes in protein thermal stability upon ligand binding or other perturbations and have been widely adopted in drug discovery and biomedical research. Though the PISA workflow is straightforward, key parameters, including detergent concentration, methods for removing denatured aggregates, and temperature range selection, vary across studies and can markedly influence assay outcomes. Yet these factors have not been systematically evaluated, limiting rational
