The need for novel antibacterial strategies and the awareness of the importance of quorum sensing (QS) in bacterial infections have stimulated research aimed at identifying QS inhibitors (QSIs). However, clinical application of QSIs identified so far is still distant, likely due to their unsuitability for use in humans. A promising way to overcome this problem is searching for anti-QS side activity among the thousands of drugs approved for clinical use in the treatment of different diseases. Here, we applied this strategy to the search for QSIs, by screening a library of FDA-approved compounds

INTRODUCTION: Pseudomonas aeruginosa is a significant pathogen associated with acute and chronic infections, particularly in immunocompromised individuals. Its capacity for biofilm formation, combined with antibiotic resistance, plays a critical role in the persistence of these infections. Peptidyl-arginine deiminases (PADs), including PAD2 and PAD4 isoforms, are involved in neutrophil phagocytic killing of P. aeruginosa. This study aimed to investigate the impact of PAD enzymes on biofilm development and virulence factor production in P. aeruginosa, with focus on the multidrug resistant strai

Pseudomonas aeruginosa is a widely prevalent multidrug-resistant pathogen in clinical environments, capable of forming biofilms and evading antibiotic clearance, which poses a significant threat to public health. Targeting the multidrug resistance and biofilm formation mechanisms of P. aeruginosa, this study designed and synthesized a series of novel N-acyl homoserine lactone (AHL)-based quorum sensing inhibitors (QSIs). Through structure-activity relationship optimization and activity screening, compound 5-6 was identified as a lead molecule. By competitively binding to the LasR protein recep

Pseudomonas aeruginosa is an opportunistic human pathogen that causes a wide range of infections. The increasing multidrug-resistance of P. aeruginosa poses a critical challenge for medical care. P. aeruginosa employs virulence factors and biofilms to establish infections in humans and protect itself from environmental stress or antibiotics. These factors are regulated by a quorum sensing mechanism involving multiple regulatory systems that act interdependently through signaling molecules. Therefore, interference with quorum sensing systems can suppress the pathogenicity of P. aeruginosa. In t

In the opportunistic pathogen Pseudomonas aeruginosa, the nitrogen-related phosphotransferase system (PTSNtr) influences multiple virulence behaviors. The PTSNtr is comprised of three enzymes: first PtsP, then the PtsO phosphocarrier, and the final PtsN phosphoacceptor. We previously showed that ptsP inactivation increases LasI-LasR quorum sensing, a system by which P. aeruginosa regulates genes in response to population density. LasI synthesizes a diffusible autoinducer that binds and activates the LasR receptor, which activates a feedback loop by increasing lasI expression. In this study, we

Bacterial keratitis (BK) causes visual morbidity/blindness if not treated effectively. Here, ciprofloxacin (CIP)-loaded nanoparticles (NPs) using glycol chitosan (GC) and poly(lactic acid) (PLA) conjugate at three different ratios (CIP@GC(PLA) NPs (1:1,5,15)) were fabricated. CIP@GC(PLA) NPs (1:1) were more effective than other tested ratios, indicating the importance of optimal hydrophobic/hydrophilic balance for corneal penetration and preventing bacterial invasion. The CIP@GC(PLA) (NPs) (1:1) realized the highest association with human corneal epithelial cells, which were nonirritant to the

As the antibiotic resistance of pathogens becomes increasingly severe, it is becoming more feasible to use methods that suppress the virulence of pathogens rather than exerting selective pressure on their growth. Pseudomonas aeruginosa, a dangerous opportunistic pathogen, infects hosts by producing multiple virulence factors, which are regulated by quorum-sensing (QS) systems, including the las systems, rhl systems, and pqs systems. This study used the chromosome lacZ transcription fusion reporter model to screen the traditional Chinese medicine monomer library and found that bakuchiol can eff

The quorum sensing mechanism relies on the detection and response to chemical signals, termed autoinducers, which regulate the synthesis of virulence factors including toxins, enzymes, and biofilms. Emerging therapeutic strategies for infection control encompass approaches that attenuate quorum-sensing systems. In this study, we evaluated the antibacterial, anti-quorum sensing, and anti-biofilm activities of Psidium guajava L. methanolic leaf extracts (PGME). Minimum Inhibitory Concentrations (MICs) of PGME were determined as 500 μg/ml for C. violaceum and 1000 μg/ml for P. aeruginosa PAO1. Si

The beneficial associations between Arachis hypogaea L. (peanut) and fluorescent Pseudomonas species have been poorly explored despite their predominance in the peanut rhizosphere. The present study explores the mutually beneficial interactions between peanut roots and P. aeruginosa P4 (P4) in terms of their impact on plant growth, defence physiology and the root-rhizobacterial interface. The efficient phosphate solubilizer P4 exhibited biocontrol abilities, including the production of siderophores, pyocyanin, indole-3-acetic acid and hydrogen cyanide. Porphyromonas gingivalis, a main etiologic agent and key pathogen responsible for initiation and progression of chronic periodontitis requires heme as a source of iron and protoporphyrin IX for its survival and the ability to establish an infection. Porphyromonas gingivalis is able to accumulate a defensive cell-surface heme-containing pigment in the form of μ-oxo bisheme. The main sources of heme for P. gingivalis in vivo are hemoproteins present in saliva, gingival crevicular fluid, and erythrocytes. To acquire heme, P. gingivalis uses several mechanisms. Among them, the best characterized a

The redox-sensitive transcription factor SoxR in enteric bacteria senses and regulates the cellular response to superoxide and nitric oxide. In other bacterial groups, however, it may respond to redox-active small molecules, as demonstrated for pyocyanin sensing in pseudomonads. The antibiotic-producing soil bacterium Streptomyces coelicolor contains a gene for an SoxR homologue (SCO1697) whose DNA recognition helix is identical to that of Escherichia coli SoxR. Using the E. coli SoxR binding sequence, we predicted five candidate genes of the SoxR regulon and demonstrated that SoxR binds to th

Quorum sensing (QS) has a central role in biofilm lifestyle and antimicrobial resistance, and disrupting these signaling pathways is a promising strategy to control bacterial pathogenicity and virulence. In this study, the efficacy of three structurally related benzaldehydes (4-hydroxybenzaldehyde, 4-hydroxy-3-methoxybenzaldehyde (vanillin) and 4-hydroxy-3,5-dimethoxybenzaldehyde (syringaldehyde)) in disrupting the las and pqs systems of Pseudomonas aeruginosa was investigated using bioreporter strains and computational simulations. Additionally, these benzaldehydes were combined with tobramyc

Development of drug hybrids with anti-quorum sensing (QS) and antivirulence properties has been widely regarded as a viable alternative for combating drug resistance in Pseudomonas aeruginosa. In this study, conjugates (1-3) were synthesized using three naturally-occurring phytochemicals: guaiacol, vanillin, and eugenol via pharmacophoric hybridization with catechol moiety. Among the synthesized conjugates that showed broad-spectrum antibacterial activity and iron-chelating potential, catechol-vanillin conjugate (2) demonstrated relatively higher degree of QS inhibition and significantly reduc

Pseudomonas aeruginosa is recognized as a critical-priority pathogen due to its major role in healthcare-associated infections (HAIs) and the rapid global rise of multidrug resistance. It is responsible for approximately 7.1-7.3% of HAIs worldwide and up to 23% of intensive care unit infections, specifically ventilator-associated pneumonia, where mortality can reach 32-42.8%. In chronic lung diseases, especially cystic fibrosis, P. aeruginosa commonly becomes the dominant pathogen in adulthood and contributes to progressive lung function decline. A major factor supporting long-term survival is

Rift valley fever virus (RVFV) is among the WHO list of priority diseases, yet no effective vaccine or treatment is currently available. Microbial pigments (MPs) represent a promising small-molecules library which can be exploited for the drug discovery of anti-RVFV compound. In this study, thirteen MPs were in silico screened to identify candidates with acceptable drug-likeness and possible ability to cross the blood brain barrier. Next, the binding interaction of the filtered molecules were compared against key RVFV proteins for the selection of the optimum inhibitor. Molecular dynamics simu

Phage therapy is increasingly recognised as a potential approach against antibiotic-resistant infections. Analogous to the rise of antibiotic resistance, bacteria can also evolve phage resistance, a process that frequently entails fitness costs. In this study, a new lytic Pseudomonas aeruginosa phage (Henu5) was isolated and characterised. Henu5 was classified within the Caudoviricetes class and displayed good biological characteristics. The Henu5 genome possesses 92,558 bp of linear dsDNA and a GC content of 49.36%. Additionally, three phage-resistant mutants, R3, R6 and R14, were isolated an

Biofilms are implicated in roughly 80% of chronic and 65% of acute bacterial infections. Therefore, targeting QS-regulated virulence and biofilm presents a potential strategy for the development of novel antimicrobials. In this investigation, AgNPs-capped Guar Gum-co-Polyacrylonitrile hybrid (GPAG) nanocomposites were synthesized biogenically by modifying guar gum (GG) chains chemically with polyacrylonitrile (PAN) chains, resulting in a copolymer matrix with suitable capping sites preventing the aggregation of Ag NPs during the synthesis of GPAG. Chemical and structural properties of the synt
