STUDY OBJECTIVE: To determine the effects of tobacco dust extract (TDE) on isolated guinea pig tracheal smooth muscle.
DESIGN: A controlled, in vitro smooth-muscle study of the effect of pharmacologic agents on TDE.
METHODS: The effect of TDE on isolated guinea pig tracheal smooth muscle was tested using water-soluble extracts of dust obtained from machines in a cigarette manufacturing plant. Mast cell infiltration of the arterial wall has been demonstrated in atherosclerotic vessels and implicated in coronary artery spasm. Spasm of vein bypass grafts has also been reported. In this study we performed vein bypass grafting of the carotid arteries in rabbits and examined the grafts for the presence of mast cells. The separation and detection of five antihistamine drugs commonly found within over-the-counter allergy and cold pharmaceutical products was performed by HPLC with chemiluminescence (CL) detection. Comparable detection limits at 5-10 pmol were found for the antihistamines by both UV at 214 nm and tris(2,2'-bipyridine) ruthenium(III) CL. However, urine samples were found not to generate as large an unretained peak by CL detection as compared to those peaks by UV detection at 214 and 254 nm. Multiple mediators are involved in the pathophysiology of allergic and inflammatory disorders. Antigen challenge of isolated perfused kidneys of sensitized guinea-pigs consistently released a large proportion of their histamine content, and caused a reduction in renal perfusate flow rate (RPFR) which could not simply be due to the released histamine, since much larger amounts of exogenous histamine were needed to produce a comparable effect. Reduction in RPFR by histamine was blocked by mepyramine but not by cimetidine, i.e. it is mediated by an H1-receptor mechanism.
