Accumulation of Quinolinic Acid Modulates the Pulmonary Immune Response During Influenza Infection.

Influenza viruses cause a highly contagious, acute pulmonary disease that results in significant mortality each year. These infections trigger the production of interferons, known to induce the expression of the rate-limiting enzyme in the kynurenine degradation pathway in the lungs. As some kynurenine pathway metabolites are biologically active, we aimed to gain a better understanding of their role in influenza A virus infection. The expression of kynurenine pathway enzymes and the levels of their metabolites were quantified in the lungs of C57BL/6 mice 7 days after infection with an H3N2 inf

Multifunctional quinoline-based hydrazones as promising antidiabetic agents: experimental and computational insights.

A new library of quinoline-based hydrazone analogues (7a-7j) was designed and synthesized to evaluate their potential as multifunctional antidiabetic agents. In contrast to earlier quinoline derivatives primarily developed for single-enzyme inhibition, the present series incorporates a quinoline scaffold with a thiocarbohydrazide-derived hydrazone linker and diverse aryl/heteroaryl substituents to investigate a multitarget profile involving α-glucosidase, α-amylase, and PPAR-γ. Biological evaluation showed low-micromolar inhibitory activity against key carbohydrate-metabolizing enzymes. Compou

Design, synthesis, in vivo, and in silico evaluation of novel quinoline-based dihydrothiazoles as anticonvulsants.

The current study reports the synthesis of a novel series of quinoline-based dihydrothiazoles compounds, 3(a-j), and evaluates their potential as antiepileptic agents. Chemical synthesis, NMR, and SAR showed important insights and correlation with structure and biological efficacy. The in vivo anticonvulsant activity was assessed using an acute chemoconvulsant model, where seizures were induced by a single dose of pentylenetetrazole (PTZ 80 mg/kg). Electroencephalographic (EEG) recordings of treated animals showed a substantial reduction in epileptic activity compared to the PTZ-only group, in

Potent inhibition and limited reactivation of human cholinesterases by cholesterol- and quinoline-based oximes.

Current oxime antidotes exhibit poor blood-brain barrier (BBB) penetration due to their permanent positive charge, limiting the reactivation of organophosphorus (OP)-inhibited cholinesterases in the central nervous system. To address this limitation, the present study investigates nine lipophilic cholesterol- and quinoline-based oximes as potential reactivators of human acetylcholinesterase (hAChE) and butyrylcholinesterase (hBChE) inhibited by OP compounds. Several oximes exhibited strong reversible inhibition with pronounced potency toward hBChE (Ki as low as 4.6 nM). However, despite the fa

Design, synthesis, anticancer activity and molecular docking of quinoline-based dihydrazone derivatives.

Based on the biologically active heterocycle quinoline, we successfully synthesized a series of quinoline-based dihydrazone derivatives (3a-3d). 1H NMR, 13C NMR, ESI-HRMS, IR, element analysis, UV/Vis spectroscopy and fluorescence spectroscopy were performed to comprehensively characterize their chemical structures, spectral properties and stability. Nitrosamine impurities were not detected in 3a-3d, and the systemic toxicological assessment indicated that the toxicity of 3a-3d was lower. Furthermore, their anticancer activity was evaluated by MTT, AO/EB double staining, apoptosis detection an

Crystal structures of zinc(II) coordination complexes with iso-quinoline N-oxide.

The reaction of one equivalent of zinc(II) halide salts with two equivalents of iso-quinoline N-oxide (iQNO; C9H7NO) in methanol yields compounds of the general formula [ZnX 2(iQNO)2], with X = Cl- (I), Br- (II) and I- (III). However, starting with zinc(II) perchlorate or nitrate leads to the formation of complex ions with the compositions [Zn(iQNO)6](X)2 for X = ClO4 - (IV), and [Zn(iQNO)(H2O)5](iQNO)2 X 2 for X = NO3 - (V). Complexes (I), (II) and (III), namely di-chlorido-bis-(iso-quinoline N-oxide-κO)zinc(II) [ZnCl2(C9H7NO)2], di-bromido-bis-(iso-quinoline N-oxide-κO)zinc(II) [ZnBr2(C9H7NO

Design and synthesis of a novel quinoline thiazolidinedione hybrid as a potential antidiabetic PPARγ modulator.

Peroxisome proliferator-activated receptor γ (PPARγ) remains a critical target for antidiabetic drug development due to its role in glucose and lipid metabolism. However, the adverse effects associated with full agonists of the thiazolidinedione (TZD) class, such as weight gain and hepatotoxicity, limit their clinical utility. Herein, we report the design and synthesis of (Z)-5-benzylidene-3-((2-chloroquinolin-3-yl)methyl)thiazolidine-2,4-dione (compound 7), a novel TZD derivative that functions as a potential PPARγ modulator. Compound 7 reduced blood glucose level (BGL) by 22.33% after 15 day

Rational Optimization of Dihydropyrimidinone-Quinoline Hybrids as Plasmodium falciparum Glutathione Reductase Inhibitors.

A series of dihydropyrimidinone-based antimalarial compounds were designed and synthesised based on the previously identified amide-based quinoline hybrids which showed good resistance reversal ability against the resistant strain of Plasmodium falciparum. The aromatic ring on the dihydropyrimidinone of the original hits was exchanged for a methyl group to bring the molecular weights below 500 Da and also determine the effect of the aromatic ring count on the resistance reversal ability of the hybrids. Apart from the previously used amide bond, the hybrid linker was also extended to the triazo

Discovery and Optimization of Quinoline Analogues as Novel Potent Antivirals against Enterovirus D68.

Enterovirus D68 (EV-D68) is a nonpolio enterovirus that is mainly transmitted through respiratory routes and poses a potential threat for large-scale spread. EV-D68 infections mostly cause moderate to severe respiratory diseases in children and potentially induce neurological diseases. However, there are no specific antiviral drugs or vaccines against EV-D68. Herein, through virtual screening and rational design, a series of novel quinoline analogues as anti-EV-D68 agents targeting VP1 were identified. Particularly, 19 exhibited potent antiviral activity with an EC50 value ranging from 0.05 to

Quinoline based thiosemicarbazones as colorimetric chemosensors for fluoride and cyanide ions and DFT studies.

High toxicity and extensive accessibility of fluoride and cyanide ions in diverse environmental media encouraged attention for scheming well-organized probes for their detection. Keeping in mind we have designed and synthesized thiosemicarbazone-based chemosensors RB-1, RB-2 and RB-3 for the detection of fluoride and cyanide ions. The structural elucidation of the synthesized chemosensors is done by employing different analytical techniques including nuclear magnetic resonance and electronic absorption specrtoscopies. Medicinal chemistry updates on quinoline- and endoperoxide-based hybrids with potent antimalarial activity.

The resistance of conventional antimalarial drugs against the malarial parasite continues to pose a challenge to control the disease. The indiscriminate exploitation of the available antimalarials has resulted in increasing treatment failures, which urges on the search for novel lead molecules. Artemisinin-based combination therapy (ACT) is the current WHO-recommended first-line treatment for the majority of malaria cases. Hybrid molecules offer a newer strategy for the development of next-generation antimalarial drugs. These comprise molecules, each with an individual pharmacological activity
