Morinda citrifolia, commonly known as noni, has a long history of utilization throughout much of tropical Polynesia and is considered to be the second most important medicinal plant in the Hawaiian Islands. Recently, the use of noni as a dietary supplement in the United States has greatly increased. Bioassay-guided fractionation of a dichloromethane-soluble partition of a MeOH extract of noni fruits has led to the isolation of an extremely potent quinone reductase inducer, 2-methoxy-1,3,6-trihydroxyanthraquinone (1). This new anthraquinone (1) was nearly 40 times more potent than a positive co

Xenobiotics and antioxidants induce expression of detoxifying enzymes including NAD(P)H: quinone oxidoreductase (NQO1), NRH:quinone oxidoreductase (NQO2), and glutathione S-transferase Ya (GST Ya), presumably to provide protection to cells against electrophilic and oxidative stress. Antioxidant response elements (AREs) have been found in the promoter regions of the various detoxifying enzyme genes. An ARE is required for basal expression and induction of the various detoxifying enzyme genes in response to xenobiotics and antioxidants. In this study, we demonstrated that exposure of cells to xe

Eleven strains of eugonic, nonoxidative, gram-negative rods isolated from clinical specimens formed a distinct group that was designated CDC group IIg. Five of the 11 isolates were from wounds. The phenotypic characteristics of CDC group IIg were most similar to those of Weeksella species, with the major difference being that CDC group IIg strains grew on MacConkey agar in 1 to 2 days, did not hydrolyze gelatin, and did not produce urease. All 11 strains of CDC group IIg possessed a distinct fatty acid profile that was characterized by large amounts (19 to 29%) of 18:1 omega 7c, 16:0, and 16:1

Disruption of sex pheromone activity in female Rocky Mountain wood ticks, Dermacentor andersoni Stiles, and American dog ticks, Dermacentor variabilis (Say), was achieved by treatment with the quinones para-benzoquinone and menadione. The most effective treatments were those administered by inoculation to unfed, mature, adult females prior to feeding. Inoculation of p-benzoquinone was also effective when administered to feeding ticks, from 2 to 5 days after they had attached, though not to the same extent as when it was given to unfed ticks. Decreases in sex pheromone activity were also observ

Quinones are among the most important components in natural organic matter (NOM) for redox reactions; however, no quinones in complex environmental media have been identified. To aid the identification of quinone-containing molecules in ultracomplex environmental samples, we developed a chemical tagging method that makes use of a Michael addition reaction between quinones and thiols (-SH) in cysteine (Cys) and cysteine-contained peptides (CCP). After the tagging, candidates of quinones in representative aqueous environmental samples (water extractions of biochar) were identified through high-r

Aromatic amine antioxidants (AAs) and p-phenylenediamine quinones (PPD-Qs) are emerging environmental contaminants, yet little is known about their associations with human thyroid function, particularly among the elderly. We quantified 19 AAs and 6 PPD-Qs in urine samples from 479 Chinese older adults and assessed their associations with thyroid hormones and sensitivity indices using traditional linear models, mixed-exposure models (WQS), explainable machine learning (XGBoost-SHAP), and molecular docking. Individual exposure to (cyclohexylamino)diphenylamine (CPPD) was associated with elevated

Chagas disease is caused by the protozoan Trypanosoma cruzi, and its current treatment is limited to the use of two nitroderivatives, benznidazole (Bz) and nifurtimox; however, their toxicity often leads to discontinuation, justifying the search for new therapeutic options. The biological activity of quinones has long shown efficacy towards pathogenic microorganisms. In our previous investigations, two naphthoquinones combining ortho- and para-quinoidal moieties exhibited remarkable trypanocidal activity and presented low toxicity to host cells. Here, these two active compounds were further as

6-PPD quinone (6-PPDQ) has been recognized as a typical emergent contaminant with the potential to cause multiple aspects of damage to organisms. Creatine is an important metabolite that mediates energy homeostasis. In Caenorhabditis elegans, creatine content was reduced by 0.1-10 μg/L of 6-PPDQ, which was accompanied by a decrease in the expression of argk-1 encoding a creatine kinase and snf-5 encoding a potential creatine transporter. Creatine content could be reduced by RNAi of argk-1 and snf-5. Moreover, RNAi of argk-1 and snf-5 aggravated 6-PPDQ-induced immunosuppression, reflected by de

Pyrroloquinoline quinone (PQQ) is a redox cofactor derived from prokaryotes that participates in various biological processes involving dehydrogenase enzymes. Previous field trials identified a PQQ-producing endophyte, Burkholderia seminalis 869T2, which enhances banana growth and reduces Fusarium wilt incidence from 24.5% to 3.4%. While more recent studies have confirmed its agricultural benefits across multiple plant species, the underlying molecular mechanisms remain unclear. Here, integrated omics and imaging mass spectrometry were employed to investigate the role of PQQ in planta. Our res

Ram spermatozoa are highly susceptible to cold-induced oxidative damage due to the abundance of polyunsaturated fatty acids (PUFAs) in their plasma membrane, which ultimately compromises fertilizing capacity. Adding antioxidants to semen extenders is a known strategy to protect sperm quality during cooling. This study evaluated the effects of different concentrations of pyrroloquinoline quinone (PQQ) in ram semen extender during chilled storage at 4°C. Semen samples were collected, diluted, and split into five groups: control (nothing added) and four groups with 10, 100, 1000, or 10000 nM PQQ.

An alcohol-derived quinone monoacetal (QMA)-enabled strategy for the direct synthesis of sulfonamidated benzoquinones via a nucleophilic addition-elimination sequence using N-methoxy sulfonamides as reagents is reported. QMA serves as a uniquely competent surrogate of benzoquinone. The protocol exhibits broad substrate scope, excellent functional group tolerance, and gram-scale applicability. Mechanistic studies, supported by DFT calculations, reveal that acid-promoted acetal hydrolysis followed by tautomerization-assisted methanol elimination is the key step governing product formation.

Quinones are privileged redox-active scaffolds in medicinal chemistry, yet their clinical utility is often limited by poor solubility and nonspecific toxicity. To address this, we developed a stereodivergent, one-pot strategy for the synthesis of quinone-triazole [3 + 2] adducts as structurally defined polyol-based glycohybrids. The tandem process involves nucleophilic addition of 5-azido-2,3-dideoxy glycols to electrophilic 1,4-quinones, followed by intramolecular cycloaddition and proton transfer to furnish the fused products with an acyclic polyol chain. The protocol delivers a broad range

The underlying pathological mechanism of ischemic stroke is complex, with oxidative stress and inflammation being two key factors that are intertwined and mutually influential. They also serve as important potential targets for the intervention of cerebral ischemia. Pyrroloquinoline quinone (PQQ) is known for its neuroprotective properties and the ability to modulate immune system function. Previous studies have demonstrated that PQQ mitigates brain infarction in rodent models of cerebral ischemia; however, the neuroprotective mechanisms underlying PQQ's effects against ischemic brain injury a

6PPD-quinone (N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone), a transformation product of the antiozonant 6PPD (N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine) is a likely causative agent of coho salmon (Oncorhynchus kisutch) pre-spawn mortality. Stormwater runoff transports 6PPD-quinone into freshwater streams, rapidly leading to neurobehavioral, respiratory distress, and rapid mortality in laboratory-exposed coho salmon, but causing no mortality in many laboratory-tested species. Given this identified hazard, and potential for environmental exposure, we evaluated a set of U.

Real-time monitoring of critical tumor microenvironment parameters, such as intracellular pH, remains challenging yet essential for understanding cancer progression and therapeutic resistance. We recently observed that the trityl radical OX063 undergoes consecutive oxidation to a diamagnetic quinone methide (QM) under superoxide-generating conditions in culture media with high thiol concentrations. This is followed by thiol-mediated reduction to OX063-OH: a structurally modified, pH-sensitive trityl radical in which one carboxyl group is replaced by a hydroxyl moiety. In this study, we develop

Neuraminidases (NAs) are critical virulence factors in pathogens. In viruses such as influenza A, neuraminidase facilitates the release of virions, thereby enabling infection propagation. In pathogenic bacteria, NA activity has been linked to the pathogenicity of species such as S. pneumoniae, P. aeruginosa, and V. cholerae. Studies suggest that bacterial NAs play roles in mucus degradation, exposing host epitopes to enhance bacterial adhesion, biofilm formation, and bacterial survival. However, the specific mechanisms by which bacterial NAs contribute to pathogenesis remain poorly understood

Single cell protein (SCP) is an important supplement to animal protein feed, and pyrroloquinoline quinone (PQQ) is frequently used as a functional food and animal feed ingredient. In this study, we aimed to coproduce SCP and PQQ through the fermentation of molasses and biogas slurry by Hyphomicrobium denitrificans SSWJ1 (H. denitrificans SSWJ1). We evaluated the tolerance of H. denitrificans SSWJ1 to alkaline conditions, revealing optimal growth at pH 8, and compared the SCP amounts synthesized in molasses and biogas slurry media (2.0 g/L) and in methanol and ammonium sulfate media (0.7 g/L).

Methylmercury in paddy soils poses threats to food security and thus human health. Redox-active phenolic and quinone moieties of natural organic matter (NOM) mediate electron transfer between microbes and mercury during mercury reduction. However, their role in mercury methylation remains elusive. Here, artificial organic matter (AOM), i.e., biochar, wherein the phenol-quinone ratio and associated redox properties varied, was used as a redox-tunable model NOM to investigate the impact of the phenol-quinone redox couples on mercury methylation in Hg-contaminated paddy soils. Our findings confir

N-(1,3-Dimethyl butyl)-N'-phenyl-phenylenediamine-quinone (6ppd-quinone) is of emerging concern due to its widespread presence and toxicity to aquatic species. The chemical has been detected in human biofluids though little is known about its effects on human tissues. The objective of this study was to increase understanding of 6ppd-quinone's potential human health effects by deriving transcriptomic points of departure (tPOD) values in two human cell lines using the TPD-seq workflow. An EC20 for cytotoxicity was calculated for Caco-2 (104 μg/L) but not for HepG2 cells. Even in the absence of c

Retinal ganglion cells are highly metabolically active requiring strictly regulated metabolism and functional mitochondria to keep ATP levels in physiological range. Imbalances in metabolism and mitochondrial mechanisms can be sufficient to induce a depletion of ATP, thus altering retinal ganglion cell viability and increasing cell susceptibility to death under stress. Altered metabolism and mitochondrial abnormalities have been demonstrated early in many optic neuropathies, including glaucoma, autosomal dominant optic atrophy, and Leber hereditary optic neuropathy.
