A Spinal Cord Sarcoma With EWSR1::WT1 Fusion Exhibited Ewing Sarcoma-Like Pattern Rather Than Desmoplastic Small Round Cell Tumor.

We received a case of a 22 years old female, with a spinal cord sarcoma. The tumor exhibited a morphology similar to that of a small blue round cell tumor, lack of desmoplastic stroma. Immunohistochemical analysis demonstrated diffusely membranal expression of CD99 and cytoplasmic expression of Desmin, but was negative of NKX2.2. Fluorescence in situ hybridization indicated the EWSR1 gene rearrangement. Next generation sequencing (NGS) identified a gene fusion of EWSR1(E9)::WT1(E8). The fusion gene was confirmed by RNA NGS as well. There was a challenge on diagnosis whether should it be diagno

Cardiac 5-HT(4) serotoninergic receptors, 52kD SSA/Ro and autoimmune-associated congenital heart block.

It was recently reported that sera from patients with systemic lupus erythematosus contain antibodies reactive with the second extracellular loop of the serotoninergic 5-HT(4) receptor expressed in the human heart. This antibody response was associated with antibodies to 52kD SSA/Ro, a reactivity prevalent in mothers of children with congenital heart block (CHB). The current study was undertaken to determine whether the 5-HT(4) receptor is a target of the immune response in these mothers. Initial experiments demonstrated mRNA expression of the 5-HT(4) receptor in the human foetal atrium. Elect

Alu elements and the human genome.

Alu insertional elements, the most abundant class of SINEs in humans are dimeric sequences approximately 300 bp in length derived from the 7SL RNA gene. These sequences contain a bipartite RNA pol III promoter, a central poly A tract, a 3' poly A tail, numerous CpG islands and are bracketed by short direct repeats. An estimated 500,000 to 1 x 10(6) units are dispersed throughout the human haploid genome primarily in AT rich neighborhoods located within larger GC dense chromosomal regions via a mechanism known as retroposition. Retroposition activity of Alu elements is determined by both intern

Importance of the immune response to the Ro/La particle in the development of congenital heart block and neonatal lupus erythematosus.

Previous studies have suggested a role for anti-Ro antibodies in the pathogenesis of neonatal lupus erythematosus (NLE). We reexamined the role of the immune response to the Ro/La particle in the development of NLE using newer and more definitive assay techniques. All 15 infants with congenital heart block and NLE had both anti-Ro and anti-La antibodies. Of 8 patients with cutaneous NLE alone, 2 had anti-Ro antibodies alone while the remaining 6 had both anti-Ro and anti-La antibodies. One woman gave birth to 2 children; one had both Ro and La antibodies and developed congenital heart block, t

Protection from interferon-induced apoptosis by Epstein-Barr virus small RNAs is not mediated by inhibition of PKR.

The Epstein-Barr virus (EBV) EBER transcripts are small, highly structured RNAs able to bind to and inhibit activation of the double-stranded RNA-dependent protein kinase PKR in cell-free systems, and within latently infected B-cell lines they inhibit alpha interferon-induced apoptosis that is believed to be mediated through PKR. Here, we address the consequences of EBER expression for PKR activation in vivo in response to alpha interferon. In agreement with published findings, either EBV infection or the EBERs alone protected Burkitt lymphoma cells from alpha-interferon-induced apoptosis. How

ZPR1 is essential for survival and is required for localization of the survival motor neurons (SMN) protein to Cajal bodies.

Mutation of the survival motor neurons 1 (SMN1) gene causes motor neuron apoptosis and represents the major cause of spinal muscular atrophy in humans. Biochemical studies have established that the SMN protein plays an important role in spliceosomal small nuclear ribonucleoprotein (snRNP) biogenesis and that the SMN complex can interact with the zinc finger protein ZPR1. Here we report that targeted ablation of the Zpr1 gene in mice disrupts the subcellular localization of both SMN and spliceosomal snRNPs. Specifically, SMN localization to Cajal bodies and gems was not observed in cells derive

Genome stability disrupted by the circRBM39(4,5,6)-RPA2 axis represses breast tumorigenesis.

Genome instability (GI) is a hallmark of cancer. GI is associated with accumulative DNA damage and tumor-specific defects in DNA repair. Multiple antitumor drugs have been developed to promote persistent GI and to drive DNA damage beyond a threshold that tumor cells can survive. However, regulatory roles of circular RNAs (circRNAs) in GI and DNA damage remain elusive. Through circRNA profiling and a small interfering RNA (siRNA)-mediated screen, we have identified a circRNA termed circRBM39(4,5,6), which is significantly decreased in breast cancer (BC) and disrupts DNA damage response. CircRBM

The Cytoplasmic MicroRNA-Production Complex Regulates MicroRNA Sorting to Extracellular Vesicles During Synaptogenesis.

Extracellular vesicles (EVs) have emerged as novel players of cell communication, in part via the transfer of microRNA (miRNA) cargo to recipient cells. Although several RNA-binding proteins (RBPs) were shown to shuttle miRNAs to EVs, how they are loaded with miRNA is unknown. This study demonstrates that the core accessory protein of the cytoplasmic miRNA production machinery TRBP interacts with several EV-RBPs and differentially regulates miRNA secretion in small EVs. TRBP-regulated EV-miRNAs were highly enriched in a Syncrip-recognition sequence motif. Moreover, we show that Syncrip associa

mTORC1 Controls Phase Separation and the Biophysical Properties of the Cytoplasm by Tuning Crowding.

Macromolecular crowding has a profound impact on reaction rates and the physical properties of the cell interior, but the mechanisms that regulate crowding are poorly understood. We developed genetically encoded multimeric nanoparticles (GEMs) to dissect these mechanisms. GEMs are homomultimeric scaffolds fused to a fluorescent protein that self-assemble into bright, stable particles of defined size and shape. By combining tracking of GEMs with genetic and pharmacological approaches, we discovered that the mTORC1 pathway can modulate the effective diffusion coefficient of particles ≥20 nm in d

The zinc finger RNA binding protein, ZFR, contributes to axon guidance in Caenorhabditis elegans.

ZFR is an ancient and highly conserved chromosome-associated protein from nematodes to mammals, embryologically expressed in most species, with the exception of the nematode Caenorhabditis elegans. The ZFR encodes zinc and RNA binding protein, and in rat, the nuclear-cytoplasmic shuttling ZFR has been found with transport and translation-associated RNA granule-like structures in the somatodendritic compartments of hippocampal neurons. The majority of axons cross the midline before projecting to their contralateral synaptic target and this crossing decision is under tight control. Molecular fac

Luteinizing hormone activates the Hippo pathway to promote progesterone synthesis in bovine luteal cells.

BACKGROUND: Progesterone production by the corpus luteum is essential for embryo implantation and early pregnancy maintenance and is acutely stimulated by luteinizing hormone (LH). While LH signaling through protein kinase A (PKA) is well established, downstream regulatory networks that constrain or shape luteal steroidogenesis remain incompletely defined. Here, we identify a previously unrecognized role for the Hippo signaling pathway in regulating luteal progesterone production. METHODS: Using primary bovine luteal cells isolated from corpora lutea, we examined the relationship between LH/P

Distribution of divalent small interfering RNA into neurons of sensory ganglia produces selective, durable knockdown of Nav1.7 and strong analgesia.

Humans missing the SCN9A gene encoding the Nav1.7 sodium ion channel are wholly insensitive to pain, but Nav1.7 inhibitors to date have not produced correspondingly strong analgesia in clinical trials. In this study, we address whether potential challenges of selectivity, tissue distribution, and target engagement might be addressed by divalent small interfering RNA (di-siRNA), a modality for transcript knockdown throughout the CNS following dosing into cerebrospinal fluid. Small interfering RNA sequences catalyzing SCN9A transcript cleavage were identified, and in live-cell assays, a di-siRNA

Multi-platform analysis of the tumor immune microenvironment associated with breast cancer subtypes.

Breast cancer is genetically and histologically heterogenous, and is influenced by a variety of factors, including the tumor microenvironment (TME). The PAM50 subtypes; Luminal A, Luminal B, Normal-like, Basal-like and Her2-enriched, are associated with different tumor phenotypes and overall survival. The quantity and quality of immune cell infiltration in breast tumors play a key role in cancer development and progression and are associated with survival and treatment response. We used multiplex immunohistochemistry, single-cell RNA sequencing, and two deconvolution algorithms to explore the

Anti-Ro60 Seropositivity Determines Anti-Ro52 Epitope Mapping in Patients With Systemic Sclerosis.

Epitope mapping of anti-Ro52 antibodies (Abs) has been extensively studied in patients with Sjögren's syndrome (SjS) and systemic lupus erythematosus (SLE). Comprehensive epitope mapping in systemic sclerosis (SSc), where anti-Ro52 antibodies are also frequently detected, has not been performed. The aim of the present study was to fully characterize Ro52 epitopes in anti-Ro52-positive SSc using Ro52 fragments spanning the full antigen. Further analysis was made according to anti-Ro60 status. Epitope mapping was performed in 43 anti-Ro52-positive SSc patients. Seventy eight anti-Ro52-positive p

Enhanced Cytoplasmic Delivery of RAGE siRNA Using Bioreducible Polyethylenimine-based Nanocarriers for Myocardial Gene Therapy.

This study aims to develop bioreducible polyethylenimine (rPEI)/siRNA polyplexes with high stability, high transfection efficiency, and low cytotoxicity for efficient cytoplasmic siRNA delivery. rPEI successfully incorporated siRNA into stable and compact nanocomplexes, and the disulfide linkages in rPEI/siRNA were cleaved under reductive environments, resulting in efficient intracellular translocation and siRNA release. In this study, receptor for advanced glycation end-products (RAGE) was selected as a therapeutic target gene because it is associated with inflammatory responses in ischemia/r

B cell apotopes of the 60-kDa Ro/SSA and La/SSB autoantigens.

Apoptosis has been proposed to influence the initiation and diversification of autoimmunity to the Ro (SSA)/La (SSB) ribonucleoprotein (RNP) particle and serve as a target for autoantibody-mediated tissue injury. We have developed a new approach to B cell epitope mapping which identifies "apotopes," defined as epitopes expressed on the surface of apoptotic cells. Preliminary studies support a role for apotopes as diagnostic markers in systemic lupus erythematosus (SLE) and primary Sjögren's syndrome. For example, apotopes within the NH(2)-terminal and central regions of La react with the major

Autoimmune thyroid disease in anti-Ro/SS-A-positive children with annular erythema: report of two cases.

Anti-Ro/SS-A-associated recurrent annular erythema is a rare disorder, and represents a cutaneous manifestation of primary Sjögren's syndrome (SS). We report two childhood cases complicated with autoimmune thyroid disease, one with Graves' disease and the other with autoimmune thyroiditis. Both children were positive for anti-Ro/SS-A and anti-La/SS-B antibodies. One patient was lacking clinical SS with objective evidence of salivary gland involvement, while the other was diagnosed with primary SS. Our observation suggests that autoimmune thyroid disease in the subset of anti-Ro/SS-A-positive c

[Sjögren's syndrome].

Sjögren's syndrome(SS) is a chronic inflammatory disease characterized by keratoconjunctivitis sicca and xerostomia. According to the epidemiological survey done by the Japan Ministry of Health and Welfare(MHW) in 1994, about 17,000 SS patients visit hospitals and the prevalence rate is approximately 0.06% for females. SS is diagnosed mainly by the criteria of MHW in 1977, and the revised ones will be published in the near future. Several candidates for the corresponding antigens for SS autoimmunity have discovered, such as alpha-fodrin and 203-211 amino acid residues of Ro/SSA-52 kDa. The sic
